| 4 | Resveratrol inhibits collagen Ⅰ synthesis by suppressing IGF-1R activation in intestinal fibroblasts显示文摘AIM:To investigate whether resveratrol(3,4,5-trihydroxy-trans-stilbene)inhibits collagenⅠsynthesis induced by insulin growth factor-1(IGF-1)in intestinal fibroblasts,and to explore the possible molecular mechanisms.METHODS:Male Sprague-Dawley rats were randomly divided into two groups:a control group and a2,4,6-trinitrobenzenesulfonic acid(TNBS)-induced colitis group.After 21 d of TNBS administration,the degree of inflammation and fibrosis in colon was measured by HE staining and Masson’s trichrome staining.Western blotting was used to examine collagenⅠ,IGF-1 and silent information regulator 1(SIRT1)protein expression in colitis tissues.Western blotting and quantitative realtime polymerase chain reaction were used to characterize collagenⅠprotein and col1a2 mRNA expression in mouse intestinal fibroblasts and CCD-18Co cells treated with IGF-1.A MEK inhibitor(U0126)was used to determine whether IGF-1-induced collagenⅠexpression was mediated by extracellular signal-regulated kinase 1/2(ERK1/2)-dependent mechanism.Effects of resveratrol on collagenⅠprotein level,insulin growth factor-1receptor(IGF-1R)and ERK1/2 phosphorylation levels were also examined after IGF-1 treatment in fibroblasts.To evaluate whether SIRT1 was necessary for the anti-fibrosis effect of resveratrol,cells were transfected with SIRT1-specific small interfering RNAs,wildtype SIRT1,and deacetylase-inactive mutant SIRT1.RESULTS:CollagenⅠand IGF-1 expression was increased,and SIRT1 expression was decreased(0.67±0.04 vs 1.05±0.07,P<0.001)in TNBS-induced colitis compared with the control group.In vitro,IGF-1 could induce collagenⅠexpression,mainly through the ERK1/2 signal pathway.Resveratrol reduced basal and IGF-1-induced collagenⅠ?gene and protein expression in intestinal fibroblasts.Overexpression of wild-type SIRT1,not deacetylase-inactive mutant SIRT1,decreased expression of collagenⅠinduced by IGF-1.Moreover,silencing SIRT1 restored collagen?Ⅰ?expression in fibroblasts challenged with resveratrol.However,disruption of SIRT1 did not influence the anti-fibrotic effects of resveratrol and IGF-1-induced collagenⅠexpression.Further analysis revealed that resveratrol significantly decreased phosphorylation of IGF-1R and its downstream signaling molecules by inhibiting IGF-1 binding to its receptor.CONCLUSION:Our data suggest that resveratrol effectively inhibits collagenⅠsynthesis in IGF-1-stimulated fibroblasts,partly by inhibiting IGF-1R activation,and SIRT1 is also responsible for the process. | Ping Li Mei-Lan Liang Ying Zhu Yao-Yao Gong Yun Wang Ding Heng Lin Lin | 2014 | World Journal of Gastroenterology2014,20,16: | 6 |

| 9 | Combined detection and subclass characteristics analysis of CTCs and CTECs by SE-iFISH in ovarian cancer显示文摘Objective:Hematogenous metastasis is essential for the progression of ovarian cancer(OC),and circulating tumor cells(CTCs)are part of the metastatic cascade.However,the detection rate of CTC is low due to the use of less sensitive detection methods.Therefore,this study aimed to detect CTCs and circulating tumorigenic endothelial cells(CTECs)in patients with OC using subtraction enrichment and immunostaining and fluorescence in situ hybridization(SE-iFISH).Methods:We enrolled a total of 56 subjects,including 20 OC patients and 36 ovarian benign tumor patients.CTCs and CTECs were captured by subtraction enrichment(SE)and counted and classified according to immunofluorescence staining of tumor markers(TMs)carbohydrate antigen 125(CA125)and human epididymis protein 4(HE4)combined with fluorescence in situ hybridization(iFISH)of chromosome 8(Chr8)aneuploidy.The diagnostic value and subtype characteristics of CTCs and CTECs were investigated.Results:The detection rate of CTCs by SE-iFISH was high.Compared with CA125 and HE4,Chr8 aneuploidy was the major identification feature of CTC.CTC counts in OC were statistically higher than those in benign groups.CTC and CTEC with≥pentaploidy were detected in both groups,illustrating the poor diagnostic value of CTC or CTEC.Distributions of triploid and tetraploid CTC subtypes were significantly different,and combined detection of triploid and tetraploid CTCs showed the best diagnostic value.In contrast,the distribution of CTECs in the OC and benign groups had no statistically significant difference.Small CTCs accounted for over 1/3 of the total CTC count.We also found that small CTCs and CTECs primarily comprised triploid cells,while large CTCs and CTECs mainly comprised pentaploidy and beyond.Conclusions:The application of SE-iFISH offered a more comprehensive understanding of heterogeneous CTCs and CTECs in OC.Analysis of subclass characteristics of the CTCs and CTECs according to Chr8 aneuploidy and cell size may broaden their potential clinical utility and deepen mechanistic studies in OC. | Hongyan Cheng Shang Wang Wenqing Luan Xue Ye Sha Dou Zhijian Tang Honglan Zhu Peter Ping Lin Yi Li Heng Cui Xiaohong Chang | 2021 | Chinese Journal of Cancer Research2021,33,2: | 3 |
| 10 | Role of Diabetes Mellitus on Treatment Effects in Drug-susceptible Initial Pulmonary Tuberculosis Patients in China显示文摘We assessed the role of diabetes mellitus(DM) on treatment effects in drug‐susceptible initial pulmonary tuberculosis(PTB) patients. A prospective study was conducted in eight provinces of China from October 2008 to December 2010. We enrolled 1,313 confirmed drug‐susceptible initial PTB patients, and all subjects received the treatment regimen(2 H3 R3 E3 Z3/4 H3 R3) as recommended by the national guidelines. Of the 1,313 PTB patients, 157(11.9%) had DM; these patients had more sputum smear‐positive rates at the end of the second month [adjusted odds ratios(aO R) 2.829, 95% confidence intervals(CI) 1.783‐4.490], and higher treatment failure(aO R 2.120, 95% CI 1.565‐3.477) and death rates(aO R 1.536, 95% CI 1.011‐2.628). | 11 | Genomic organization and sequence dynamics of the AvrPiz-t locus in Magnaporthe oryzae显示文摘Plants utilize multiple layers of defense mechanisms to fight against the invasion of diverse pathogens.The R gene mediates resistance,in most cases,dependent on the co-existence of its cognate pathogen-derived avirulence (Avr) gene.The rice blast R gene Piz-t corresponds in gene-for-gene fashion to the Magnaporthe oryzae Avr gene AvrPiz-t.In this study,we determined and compared the genomic sequences surrounding the AvrPiz-t gene in both avirulent and virulent isolates,designating as AvrPiz-t-ZB15 and avrPiz-t-70-15 regions,respectively.The sequence of the AvrPiz-t-ZB15 region is 120966 bp whereas avrPiz-t-70-15 is 146292 bp in length.The extreme sequence similarity and good synteny in gene order and content along with the absence of two predicted genes in the avrPiz-t-70-15 region were observed in the predicted protein-coding regions in the AvrPiz-t locus.Nevertheless,frequent presence/absence and highly dynamic organization of transposable elements (TEs) were identified,representing the major variation of the AvrPiz-t locus between different isolates.Moreover,TEs constitute 27.3% and 43.2% of the genomic contents of the AvrPiz-t-ZB15 and avrPiz-t-70-15 regions,respectively,indicating that TEs contribute largely to the organization and evolution of AvrPiz-t locus.The findings of this study suggest that M.oryzae could benefit in an evolutionary sense from the presence of active TEs in genes conferring avirulence and provide an ability to rapidly change and thus to overcome host R genes. | Ping LI Bin BAI Hong-yan ZHANG Heng ZHOU Bo ZHOU | 2012 | Journal of Zhejiang University-Science B(Biomedicine & Biotechnology)2012,13,6: | 3 |

Syngenetic brine sedimentation, hot submarine spring eruption—accumulation, syngenetic sedimentation—vadose brine replacement had been supposed as the genetic models before. Under these models, syngenetic mineralization should be dominant in ore formation. However, a series of geological and geochemical properties investigated by authors of this paper are contrary to these models. | 13 | Effect of Electroacupuncture on Anxiety and Craving in Heroin Addicts During Detoxification显示文摘Objective:To observe the effect of electroacupuncture at Jiaji (EX-B 2) points for anxiety and craving in heroin addicts during detoxification. Methods:A total of 60 cases were randomized into an electro-acupuncture group and a control group, 30 in each group. Cases in the electroacupuncture group were treated with electroacupuncture at Jiaji (EX-B 2) points, whereas cases in the control group were treated with Methadone and Doxepin. Then scores were graded using Zung self-rating anxiety score (SAS) and self-made visual analogue scale (VAS). The assessment was conducted once a day to record the adverse reactions. Results:After treatment, the SAS and VAS scores in the electro-acupuncture group were significantly lower than that in the control group (P<0.01). | 14 | Effects of Multiple Cleaning and Disinfection Interventions on Infectious Diseases in Children: A Group Randomized Trial in China显示文摘Objective To assess the effectiveness of multiple cleaning and disinfection interventions in the homes and kindergartens, in reducing gastrointestinal and respiratory illnesses of children. Methods From October 2010 to September 2011, we performed a prospective, controlled study in China. 408 children under 5 years old were recruited and group randomized into intervention and control groups. Families and kindergartens in the intervention group were provided with antibacterial products for hand hygiene and surface cleaning or disinfection for one year. Each child's illness symptoms and sick leave were recorded every day. Results A total of 393 children completed the study, with similar baseline demographics in each of the 2 groups. Except for abdominal pain, the odds of symptoms(fever, cough and expectoration, runny nose and nasal congestion, diarrhea), illness(acute respiratory illness and gastrointestinal illness), and sick leave per person each month were significantly reduced by interventions. The rates of fever, diarrhea, acute respiratory illness, gastrointestinal illness and sick leave per person per year were significantly decreased as well. | 16 | Numerical Discretization-Based Kernel Type Estimation Methods for Ordinary Differential Equation Models显示文摘We consider the problem of parameter estimation in both linear and nonlinear ordinary differential equation(ODE) models. Nonlinear ODE models are widely used in applications. But their analytic solutions are usually not available. Thus regular methods usually depend on repetitive use of numerical solutions which bring huge computational cost. We proposed a new two-stage approach which includes a smoothing method(kernel smoothing or local polynomial fitting) in the first stage, and a numerical discretization method(Eulers discretization method, the trapezoidal discretization method,or the Runge–Kutta discretization method) in the second stage. Through numerical simulations, we find the proposed method gains a proper balance between estimation accuracy and computational cost.Asymptotic properties are also presented, which show the consistency and asymptotic normality of estimators under some mild conditions. The proposed method is compared to existing methods in term of accuracy and computational cost. The simulation results show that the estimators with local linear smoothing in the first stage and trapezoidal discretization in the second stage have the lowest average relative errors. | 19 | Scientific objectives of the Hot Universe Baryon Surveyor(HUBS) mission显示文摘The Hot Universe Baryon Surveyor(HUBS) is a proposed space-based X-ray telescope for detecting X-ray emissions from the hot gas content in our universe. With its unprecedented spatially-resolved high-resolution spectroscopy and large field of view,the HUBS mission will be uniquely qualified to measure the physical and chemical properties of the hot gas in the interstellar medium, the circumgalactic medium, the intergalactic medium, and the intracluster medium. These measurements will be valuable for two key scientific goals of HUBS, namely to unravel the AGN and stellar feedback physics that governs the formation and evolution of galaxies, and to probe the baryon budget and multi-phase states from galactic to cosmological scales. In addition to these two goals, the HUBS mission will also help us solve some problems in the fields of galaxy clusters, AGNs, difuse X-ray backgrounds, supernova remnants, and compact objects.
