3Ecological restoration and recovery in the wind-blown sand hazard areas of northern China: relationship between soil water and carrying capacity for vegetation in the Tengger Desert显示文摘The main prevention and control area for wind-blown sand hazards in northern China is about 320000 km2 in size and includes sandlands to the east of the Helan Mountain and sandy deserts and desert-steppe transitional regions to the west of the Helan Mountain.Vegetation recovery and restoration is an important and effective approach for constraining wind-blown sand hazards in these areas.After more than 50 years of long-term ecological studies in the Shapotou region of the Tengger Desert,we found that revegetation changed the hydrological processes of the original sand dune system through the utilization and space-time redistribution of soil water.The spatiotemporal dynamics of soil water was significantly related to the dynamics of the replanted vegetation for a given regional precipitation condition.The long-term changes in hydrological processes in desert areas also drive replanted vegetation succession.The soil water carrying capacity of vegetation and the model for sand fixation by revegetation in aeolian desert areas where precipitation levels are less than 200 mm are also discussed.LI XinRong ZHANG ZhiShan TAN HuiJuan GAO YanHong LIU LiChao WANG XinPing 2014Science China(Life Sciences)2014,57,5:39

6BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 7Effect of dietary soy oil, glucose, and glutamine on growth performance, amino acid profile, blood profile, immunity, and antioxidant capacity in weaned piglets显示文摘Weaning stress results in gastrointestinal dysfunction and depressed performance in pigs. This study aimed to investigate the effect of soy oil, glucose, and glutamine on the growth and health of weaned piglets. Compared with those in the glutamine group, piglets in the glucose and soy oil groups had greater average daily gain, average daily feed intake, and gain: feed ratio from day 0 to 14, and gain: feed ratio for the overall period. There were no differences with regard to serum amino acids among the three groups on day 14, except glycine and threonine. The serum concentration of histidine, serine, threonine, proline, and cysteine was the highest in the glutamine group, while the content of glycine and lysine in the soy oil group on day 28 was the highest among all groups. Piglets fed with glutamine had greater serum glucose and creatinine on day 14, high-density lipoprotein on day 28, and serum IgG and IgM on day 28. Piglets in the glutamine group demonstrated lower serum total superoxide dismutase on day 14 and 28; however, they demonstrated higher total superoxide dismutase and total antioxidant capacity in the duodenum and ileum on day 14. 8Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression.Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu 2020Cancer Biology & Medicine2020,17,1:19

10SHP2 inhibition triggers anti-tumor immunity and synergizes with PD-1 blockade显示文摘Tyrosine phosphatase SHP2 is a promising drug target in cancer immunotherapy due to its bidirectional role in both tumor growth promotion and T-cell inactivation. Its allosteric inhibitor SHP099 is known to inhibit cancer cell growth both in vitro and in vivo. However, whether SHP099-mediated SHP2 inhibition retards tumor growth in vivo via anti-tumor immunity remains elusive. To address this, a CT-26 colon cancer xenograft model was established in mice since this cell line is insensitive to SHP099.Consequently, SHP099 minimally affected CT-26 tumor growth in immuno-deficient nude mice, but significantly decreased the tumor burden in CT-26 tumor-bearing mice with intact immune system.SHP099 augmented anti-tumor immunity, as shown by the elevated proportion of CD8tIFN-γtT cells and the upregulation of cytotoxic T-cell related genes including Granzyme B andPerforin, which decreased the tumor load. In addition, tumor growth in mice with SHP2-deficient T-cells was markedly slowed down because of enhanced anti-tumor responses. Finally, the combination of SHP099 and antiPD-1 antibody showed a higher therapeutic efficacy than either monotherapy in controlling tumor growthin two colon cancer xenograft models, indicating that these agents complement each other. 11Recombinant Newcastle disease virus expressing African swine fever virus protein 72 is safe and immunogenic in mice显示文摘African swine fever(ASF) is a lethal hemorrhagic disease that affects wild and domestic swine. The etiological agent of ASF is African swine fever virus(ASFV). Since the first case was described in Kenya in 1921, the disease has spread to many other countries. No commercial vaccines are available to prevent ASF. In this study, we generated a recombinant Newcastle disease virus(r NDV) expressing ASFV protein 72(p72) by reverse genetics and evaluated its humoral and cellular immunogenicity in a mouse model. The recombinant virus, r NDV/p72, replicated well in embryonated chicken eggs and was safe to use in chicks and mice. The p72 gene in r NDV/p72 was stably maintained through ten passages. Mice immunized with r NDV/p72 developed high titers of ASFV p72 specific Ig G antibody, and had higher levels of Ig G1 than IgG2 a. Immunization also elicited T-cell proliferation and secretion of IFN-γ and IL-4. 14Assessment of heavy metal enrichment and its human impact in lacustrine sediments from four lakes in the mid-low reaches of the Yangtze River, China显示文摘Sediments from four lakes in the mid-low reaches of the Yangtze River, Taibai Lake, Longgan Lake, Chaohu Lake and Xijiu Lake, were chosen to evaluate their enrichment state and history. The state of heavy metal enrichment was at a low level in the sediment of Taibai Lake and Longgan Lake. The enrichment state of Co, Cr and Ni was also low in the sediment of Chaohu Lake and Xijiu Lake, while Cu, Pb and Zn enrichment reached a higher level. Mass accumulation fluxes were calculated to quantitatively evaluate the anthropogenic contribution to heavy metals in the sediment. The anthropogenic accumulation fluxes were lower in the sediment of Taibai Lake and Longgan Lake compared with the other two lakes, where heavy metals, especially Cu, Pb and Zn, were mainly from anthropogenic sources. Heavy metal accumulation did not vary greatly in the sediment of Taibai Lake and Longgan Lake, while that in Chaohu Lake and Xijiu Lake increased since the 1950s and substantially increased since the 1980s, although a decrease occurred since 2000 AD in Xijiu Lake. Heavy metal enrichment was strongly related to human activities in the catchment. The development of urbanization and industrialization was much more rapid in the catchments of Chaohu Lake and Xijiu Lake than of the other two lakes, and thus large amounts of anthropogenically sourced heavy metals were discharged into the lakes, which resulted in a higher contamination risk. 15Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 16Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer显示文摘Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. 17Generalized mixed finite element method for 3D elasticity problems显示文摘Without applying any stable element techniques in the mixed methods, two simple generalized mixed element(GME) formulations were derived by combining the minimum potential energy principle and Hellinger–Reissner(H–R) variational principle. The main features of the GME formulations are that the common C0-continuous polynomial shape functions for displacement methods are used to express both displacement and stress variables, and the coefficient matrix of these formulations is not only automatically symmetric but also invertible. Hence, the numerical results of the generalized mixed methods based on the GME formulations are stable. Displacement as well as stress results can be obtained directly from the algebraic system for finite element analysis after introducing stress and displacement boundary conditions simultaneously. Numerical examples show that displacement and stress results retain the same accuracy. The results of the noncompatible generalized mixed method proposed herein are more accurate than those of the standard noncompatible displacement method. 18Feedback control of nonlinear differential algebraic systems using Hamiltonian function method显示文摘The stabilization and H∞ control of nonlinear differential algebraic systems (NDAS) are investigated using the Hamiltonian function method. Firstly, we put forward a novel dissipative Hamiltonian realization (DHR) structure and give the condition to com- plete the Hamiltonian realization. Then, based on the DHR, we present a criterion for the stability analysis of NDAS and construct a stabilization controller for NDAS in absence of disturbances. Finally, for NDAS in presence of disturbances, the L2 gain is analyzed via generalized Hamilton-Jacobi inequality and an H∞ control strategy is constructed. 19Soil water repellency and influencing factors of Nitraria tangutorun nebkhas at different succession stages显示文摘Soil water repellency(WR) is an important physical characteristic of soil surface. It is capable of largely influencing the hydrological and geomorphological processes of soil, as well as affecting the ecological processes of plants, such as growth and seed germination, and has thus been a hot topic in recent research around the world. In this paper, the capillary rise method was used to study the soil WR characteristics of Nitraria tangutorun nebkhas. Soil water repellencies at different succession stages of Nitraria tangutorun were investigated, and the relationships between soil WR and soil organic matter, total N, and total P, soil texture, pH, and concentrations of CO3 2-, HCO3-, Cl-, SO4 2-, Na+, K+, Ca2+and Mg2+were discussed. Soil WR may be demonstrated at the following nebkhas dune evolvement stages: extremely degraded>degraded>stabilized>well developed>newly developed>quick sand. Apart from some soil at the bottom, the WR of other soils(crest and slope of dune) was found to be largest at the topsoil, and decreased as the soil depth increased. The results showed that multiple factors affected soil WR characteristics, e.g. WR increased significantly as the contents of soil organic matter and total N increased, but did not change as the total P content increased. Soil texture was a key factor affecting soil WR; soil WR increased significantly as clay content increased, and decreased significantly as sand content increased. Low pH was shown to be more suitable for the occurrence of soil WR. Four cations(Ca2+, Mg2+, K+and Na+) and two anions(Cl-and SO4 2-) enhanced soil WR, while CO3 2-decreased it. HCO3-did not show any observable effect. Finally, we established a best-fit general linear model(GLM) between soil-air-water contact angle(CA) and influencing factors(CA=5.606 sand+6.496(clay and silt)–2.353 pH+470.089 CO3 2-+11.346 Na+–407.707 Cl-–14.245 SO4 2-+0.734 total N–519.521). 20Multi-Stage Hydrocarbon Accumulation and Formation Pressure Evolution in Sinian Dengying FormationCambrian Longwangmiao Formation, Gaoshiti-Moxi Structure, Sichuan Basin显示文摘Sichuan Basin is a typical superimposed basin, which experienced multi-phase tectonic movements, meanwhile Sinian–Cambrian underwent complex hydrocarbon accumulation processes, causing exploration difficulties in the past 60 years. Based on the microscopic evidence of fluid inclusions, combined with basin-modelling, this paper determines stages and time of hydrocarbon accumulation, reconstructs evolution of formation pressure and dynamic processes of hydrocarbon accumulation in Sinian Dengying Formation-Cambrian Longwangmiao Formation of Gaoshiti-Moxi structure. Three stages of inclusions are detected, including a stage of yellow-yellowgreen fluorescent oil inclusions, a stage of blue fluorescent oil-gas inclusions and a stage of non-fluorescent gas inclusions, reflecting the study area has experienced a series of complex hydrocarbon accumulation processes, such as formation of paleo-oil reservoirs, cracking of crude oil, formation of paleo-gas reservoirs and adjustment to present gas reservoirs, which occurred during 219–188, 192–146 and 168–0 Ma respectively. During the period of crude oil cracking, Dengying Formation-Longwangmiao Formation showed weak overpressure to overpressure characteristics, then after adjustment of paleo-gas reservoirs to present gas reservoirs, the pressure in Dengying Formation changed into overpressure but finally reduced to normal pressure system.
