2Expression of Livin in tissues of lung cancer and its correlation with the expression of caspase-3显示文摘Objective: To study the expressions of two isoforms of Livin in tissues of lung cancer and their relations to histological types and chemotherapy, and to study their correlations to the expression of caspase-3 as well. Methods: Ex-pressions of Livin isoforms α, β and caspase-3 were detected by reverse transcription polymerase chain reaction (RT-PCR) assay in lung cancer tissues as well as in controls. Results: Livin isoforms α and β were expressed in 12 of 27, and 19 of 27 lung cancer tissues respectively, much more than those in lung para-cancerous [both were (0/6)] or benign disease lung tissues (0/12, 1/12; P < 0.01 and P < 0.01). Moreover, they were detected in 7/14, 9 /14 lung adenocarcinomas and 4/12, 9/12 squamocellular and large cell carcinomas, respectively, and both showed expressions in one small cell carcinoma. The levels of these two isoforms in lung cancer were significantly higher than those in controls by Gel imaging system (P < 0.05 and P < 0.05), the former was higher in adenocarcinoma than that in squamocellular carcinoma (P < 0.05), while the latter was the same in both (P > 0.05). Meanwhile, the levels of caspase-3 in lung cancer were significantly lower than those in controls, and it was suggested to be negatively associated with either each of two isoforms or their sum (P < 0.05, P < 0.01 and P < 0.01). Two isoforms of Livin expression seemed to increase after chemotherapy but not related to clinical stages (P > 0.05). Conclusion: Two isoforms of Livin are differently expressed in different histological types of lung cancer and may contribute to corresponding cancerous development; the levels of Livin are negatively associated with those of caspase-3, this may be due to the fact that Livin could resist against apoptosis; high expression of Livin seems to be related to chemotherapy but not clinical stages.Hongru Li Yusheng Chen Gang Chen Baosong Xie Lifang Lin 2008The Chinese-German Journal of Clinical Oncology2008,7,7:3

4TRIM47 is a novel endothelial activation factor that aggravates lipopolysaccharide-induced acute lung injury in mice via K63-linked ubiquitination of TRAF2显示文摘Endothelial activation plays an essential role in the pathogenesis of sepsis-induced acute lung injury,however,the detailed regulatory mechanisms remain largely unknown.Here,we reported that TRIM47,an E3 ubiquitin ligase of the tripartite motifcontaining protein family,was highly expressed in vascular endothelial cells.TRIM47-deficient mice were effectively resistant to lipopolysaccharide(LPS)-induced acute lung injury and death by attenuating pulmonary inflammation.TRIM47 was upregulated during TNFα-induced endothelial activation in vitro.Knockdown of TRIM47 in endothelial cells inhibited the transcription of multiple pro-inflammatory cytokines,reduced monocyte adhesion and the expression of adhesion molecules,and suppressed the secretion of IL-1βand IL-6 in endothelial cells.By contrast,overexpression of TRIM47 promoted inflammatory response and monocyte adhesion upon TNFαstimulation.In addition,TRIM47 was able to activate the NF-κB and MAPK signaling pathways during endothelial activation.Furthermore,our experiments revealed that TRIM47 resulted in endothelial activation by promoting the K63-linked ubiquitination of TRAF2,a key component of the TNFαsignaling pathway.Taken together,our studies demonstrated that TRIM47 as a novel activator of endothelial cells,promoted LPS-induced pulmonary inflammation and acute lung injury through potentiating the K63-linked ubiquitination of TRAF2,which in turn activates NF-κB and MAPK signaling pathways to trigger an inflammatory response in endothelial cells.Yisong Qian Ziwei Wang Hongru Lin Tianhua Lei Zhou Zhou Weilu Huang Xuehan Wu Li Zuo Jie Wu Yu Liu Ling-Fang Wang Xiao-Hui Guan Ke-Yu Deng Mingui Fu Hong-Bo Xin 2022Signal Transduction and Targeted Therapy2022,7,6:2
5A potent PGK1 antagonist reveals PGK1 regulates the production of IL-1βand IL-6显示文摘Glycolytic metabolism enzymes have been implicated in the immunometabolism field through changes in metabolic status. PGK1 is a catalytic enzyme in the glycolytic pathway. Here, we set up a high-throughput screen platform to identify PGK1 inhibitors. DC-PGKI is an ATPcompetitive inhibitor of PGK1 with an affinity of Kd= 99.08 nmol/L. DC-PGKI stabilizes PGK1in vitro and in vivo, and suppresses both glycolytic activity and the kinase function of PGK1. In addition,DC-PGKI unveils that PGK1 regulates production of IL-1β and IL-6 in LPS-stimulated macrophages.Mechanistically, inhibition of PGK1 with DC-PGKI results in NRF2(nuclear factor-erythroid factor 2-related factor 2, NFE2L2) accumulation, then NRF2 translocates to the nucleus and binds to the proximity region of Il-1β and Il-6 genes, and inhibits LPS-induced expression of these genes. DC-PGKI ameliorates colitis in the dextran sulfate sodium(DSS)-induced colitis mouse model. 13Atomistic modeling and rational design of optothermal tweezers for targeted applications显示文摘Optical manipulation of micro/nanoscale objects is of importance in life sciences,colloidal science,and nanotechnology.Optothermal tweezers exhibit superior manipulation capability at low optical intensity.However,our implicit understanding of the working mechanism has limited the further applications and innovations of optothermal tweezers.Herein,we present an atomistic view of opto-thermo-electro-mechanic coupling in optothermal tweezers,which enables us to rationally design the tweezers for optimum performance in targeted applications.Specifically,we have revealed that the non-uniform temperature distribution induces water polarization and charge separation,which creates the thermoelectric field dominating the optothermal trapping.We further design experiments to systematically verify our atomistic simulations.Guided by our new model,we develop new types of optothermal tweezers of high performance using low-concentrated electrolytes.Moreover,we demonstrate the use of new tweezers in opto-thermophoretic separation of colloidal particles of the same size based on the difference in their surface charge,which has been challenging for conventional optical tweezers.With the atomistic understanding that enables the performance optimization and function expansion,optothermal tweezers will further their impacts.Hongru Ding Pavana Siddhartha Kollipara Linhan Lin Yuebing Zheng 2021Nano Research2021,14,1:0
