2Correlation of E-selectin gene polymorphisms with risk of ischemic stroke A meta-analysis显示文摘OBJECTIVE: To investigate the association of E-selectin S128R polymorphisms with ischemic stroke. DATA SOURCES: A computer-based online search was conducted in PubMed, Elsevier, Ovid Database, the China National Knowledge Infrastructure, and Wanfang Database between January 1998 and December 2010. STUDY SELECTION: Case-controlled studies addressing the association of the E-selectin polymorphism and ischemic stroke were included in this review. The genotype distribution complied with the Hardy-Weinberg genetic equilibrium. The included reports were evaluated by two authors for strict quality screening. Meta-analysis software, REVMAN 5.1, was used to investigate heterogeneity, pooled odds ratio (OR) and 95% confidence interval (CI) in individual studies. MAIN OUTCOME MEASURES: Genotype and allele distributions at the E-selectin S128R site. RESULTS: Six case-controlled studies were included after screening and application of inclusion and exclusion criteria. There was no heterogeneity in the genotype and allele frequencies, and no publication bias was found. Meta-analysis of the pooled data showed that the OR value of the (AC+CC)/AA genotype was 1.93 (95% CI: 1.55-2.41, Z = 5.80, P < 0.000 01), and the OR for the C/A allele was 1.80 (95% CI: 1.47-2.22, Z = 5.59, P < 0.000 01) in the ischemic stroke group, compared with control group. Results of pooled data in Chinese subjects showed that the OR value of (AC+CC)/AA was 2.36 (95% CI: 1.68-3.31, Z = 4.99, P < 0.000 01), and the OR value of the C/A allele was 2.25 (95% CI: 1.63-3.12, Z = 4.89, P < 0.000 01). 3Ecological restoration and recovery in the wind-blown sand hazard areas of northern China: relationship between soil water and carrying capacity for vegetation in the Tengger Desert显示文摘The main prevention and control area for wind-blown sand hazards in northern China is about 320000 km2 in size and includes sandlands to the east of the Helan Mountain and sandy deserts and desert-steppe transitional regions to the west of the Helan Mountain.Vegetation recovery and restoration is an important and effective approach for constraining wind-blown sand hazards in these areas.After more than 50 years of long-term ecological studies in the Shapotou region of the Tengger Desert,we found that revegetation changed the hydrological processes of the original sand dune system through the utilization and space-time redistribution of soil water.The spatiotemporal dynamics of soil water was significantly related to the dynamics of the replanted vegetation for a given regional precipitation condition.The long-term changes in hydrological processes in desert areas also drive replanted vegetation succession.The soil water carrying capacity of vegetation and the model for sand fixation by revegetation in aeolian desert areas where precipitation levels are less than 200 mm are also discussed.LI XinRong ZHANG ZhiShan TAN HuiJuan GAO YanHong LIU LiChao WANG XinPing 2014Science China(Life Sciences)2014,57,5:39
4Comparative study of the net exchange of CO_(2) in 3 types of vegetation ecosystems on the Qinghai-Tibetan Plateau显示文摘Using the eddy covariance method,from 1 July2003 to 30 June 2004, we conducted the observation andanalysis of ecosystem CO2 flux in 3 types of alpine meadowvegetation (Kobresia humilis, Potentilla fruticosa shrub andKobresia tibetica swamp meadows) on the Qinghai-TibetanPlateau. The results show that the Kobresia humilis meadow,the shrub meadow and the swamp meadow’s highest CO2uptake rates are 16.78, 10.42 and 16.57 μmol·m?2·s?1respectively, while their highest CO2 release rates are 8.22,7.73 and 18.67 μmol·m?2·s?1 respectively. The Kobresiahumilis meadow and shrub meadow’s annual atmosphericuptakes are 282 g CO2/m2 and 53 g CO2/m2, respectively,while swamp meadow’s annual atmospheric release is 478 gCO2/m2. This proves that the Kobresia humilis meadow andthe shrub meadow on the Qinghai-Tibetan Plateau haverelatively low potential for CO2 uptake and release comparedto C4 grasslands, a number of lowland grasslands, andforests. Moreover, swamp meadow has relatively high releasepotential. This, in turn, reveals clear differences in carbonsource/sink between different types of vegetation in theQinghai-Tibetan Plateau alpine meadow ecosystem. 6Cell–cell contact with proinflammatory macrophages enhances the immunotherapeutic effect of mesenchymal stem cells in two abortion models显示文摘Mesenchymal stem cells(MSCs),which are pluripotent cells with immunomodulatory properties,have been considered good candidates for the therapy of several immune disorders,such as inflammatory bowel diseases,concanavalin A-induced liver injury,and graft-versus-host disease.The embryo is a natural allograft to the maternal immune system.A successful pregnancy depends on the timely extinction of the inflammatory response induced by embryo implantation,followed by the switch to a tolerant immune microenvironment in both the uterus and the system.Excessive infiltration of immune cells and serious inflammatory responses are triggers for embryo rejection,which results in miscarriage.Here,we demonstrated that adoptive transfer of MSCs could prevent fetal loss in a lipopolysaccharide(LPS)-induced abortion model and immune response-mediated spontaneous abortion model.The immunosuppressive MSCs alleviated excessive inflammation by inhibiting CD4+T cell proliferation and promoting the decidual macrophage switch to M2 in a tumor necrosis factor-stimulated gene-6(TSG-6)-dependent manner.Cell-tocell contact with proinflammatory macrophages increased the TSG-6 production by the MSCs,thereby enhancing the suppressive regulation of T cells and macrophages.Moreover,proinflammatory macrophages in contact with the MSCs upregulated the expression of CD200 on the stem cells and facilitated the reprogramming of macrophages towards an anti-inflammatory skew through the interaction of CD200 with CD200R on proinflammatory macrophages.Therefore,the results demonstrate that a TSG-6-mediated paracrine effect,reinforced by cell-to-cell contact between MSCs and proinflammatory macrophages,is involved in the mechanism of MSC-mediated abortion relief through the induction of immune tolerance.Our study also indicates the potential application of MSCs in clinical recurrent miscarriages.Yanhong Li Di Zhang Ling Xu Lin Dong Ji Zheng Yikong Lin Jiefang Huang Yanyun Zhang Yu Tao Xingxing Zang Dajin Li Meirong Du 2019Cellular & Molecular Immunology2019,16,12:23
7BMAL1 knockout macaque monkeys display reduced sleep and psychiatric disorders显示文摘Circadian disruption is a risk factor for metabolic, psychiatric and age-related disorders, and non-human primate models could help to develop therapeutic treatments. Here, we report the generation of BMAL1 knockout cynomolgus monkeys for circadian-related disorders by CRISPR/Cas9 editing of monkey embryos. These monkeys showed higher nocturnal locomotion and reduced sleep, which was further exacerbated by a constant light regimen. Physiological circadian disruption was reflected by the markedly dampened and arrhythmic blood hormonal levels. Furthermore, BMAL1-deficient monkeys exhibited anxiety and depression, consistent with their stably elevated blood cortisol, and defective sensory processing in auditory oddball tests found in schizophrenia patients. Ablation of BMAL1 up-regulated transcriptional programs toward inflammatory and stress responses, with transcription networks associated with human sleep deprivation, major depressive disorders, and aging. 8Neutrophil extracellular traps mediate the crosstalk between glioma progression and the tumor microenvironment via the HMGB1/RAGE/IL-8 axis显示文摘Objective:Neutrophil extracellular traps(NETs)produced by tumor-infiltrating neutrophils(TINs)are associated with poor prognosis in patients with several types of cancer.However,the mechanisms underlying the involvement of NETs in glioma progression remain largely unknown.This study aimed to elucidate the roles of NETs in biological processes that drive the crosstalk between glioma progression and the tumor microenvironment.Methods:Neutrophil infiltration and NETs formation were investigated in glioma tissue through immunohistochemistry,and their relationships with clinicopathological features and outcomes were statistically evaluated.The effects of NETs on glioma cell progression were studied in a co-culture system.In vivo and in vitro experiments validated the reactive oxygen species activity and cytokine production of TINs,as well as the ERK signaling pathway activation and the metastasis of gliomas.Results:Neutrophil infiltration and NETs formation were induced in high-grade glioma compared with low-grade glioma.NETs induced by TINs were determined to be an oncogenic marker of high-grade gliomas and to be involved in cell proliferation and invasion.NETs overproduction promoted glioma cell proliferation,migration,and invasion.Furthermore,HMGB1 was found to bind to RAGE and activate the NF-κB signaling pathway in vitro.In addition,NETs stimulated the NF-κB signaling pathway,thus promoting IL-8 secretion in glioblastoma.Subsequently,IL-8 recruited neutrophils which in turn mediated NETs formation via the PI3 K/AKT/ROS axis in TINs.Conclusions:Our results suggest that NETs produced by TINs mediate the crosstalk between glioma progression and the tumor microenvironment by regulating the HMGB1/RAGE/IL-8 axis.Targeting NETs formation or IL-8 secretion may be an effective approach to inhibit glioma progression.Caijun Zha Xiangqi Meng Lulu Li Shan Mi Da Qian Ziwei Li Pengfei Wu Shaoshan Hu Shihong Zhao Jinquan Cai Yanhong Liu 2020Cancer Biology & Medicine2020,17,1:19

10Recombinant Newcastle disease virus expressing African swine fever virus protein 72 is safe and immunogenic in mice显示文摘African swine fever(ASF) is a lethal hemorrhagic disease that affects wild and domestic swine. The etiological agent of ASF is African swine fever virus(ASFV). Since the first case was described in Kenya in 1921, the disease has spread to many other countries. No commercial vaccines are available to prevent ASF. In this study, we generated a recombinant Newcastle disease virus(r NDV) expressing ASFV protein 72(p72) by reverse genetics and evaluated its humoral and cellular immunogenicity in a mouse model. The recombinant virus, r NDV/p72, replicated well in embryonated chicken eggs and was safe to use in chicks and mice. The p72 gene in r NDV/p72 was stably maintained through ten passages. Mice immunized with r NDV/p72 developed high titers of ASFV p72 specific Ig G antibody, and had higher levels of Ig G1 than IgG2 a. Immunization also elicited T-cell proliferation and secretion of IFN-γ and IL-4. 11L-3-n-butylphthalide protects against vascular dementia via activation of the Akt kinase pathway显示文摘As a neuroprotective drug for the treatment of ischemic stroke, 3-n-butylphthalide, a celery seed extract, has been approved by the State Food and Drug Administration of China as a clinical therapeutic drug for ischemic stroke patients. L-3-n-butylphthalide possesses significant efficacy in the treatment of acute ischemic stroke. The activated Akt kinase pathway can prevent the death of nerve cells and exhibit neuroprotective effects in the brain after stroke. This study provides the hypothesis that l-3-n-butylphthalide has a certain therapeutic effect on vascular dementia, and its mechanism depends on the activation of the Akt kinase pathway. A vascular dementia mouse model was established by cerebral repetitive ischemia/reperfusion, and intragastrically administered l-3-n-butylphthalide daily for 28 consecutive days after ischemia/reperfusion, or 7 consecutive days before ischemia/reperfusion. The Morris water maze test showed significant impairment of spatial learning and memory at 4 weeks after operation, but intragastric administration of l-3-n-butylphthalide, especially pretreatment with l-3-n-butylphthalide, significantly reversed these changes. Thionine staining and western blot analylsis showed that preventive and therapeutic application of l-3-n-butylphthalide can reduce loss of pyramidal neurons in the hippocampal CA1 region and alleviate nerve damage in mice with vascular demen-tia. In addition, phosphorylated Akt expression in hippocampal tissue increased significantly after l-3-n-butylphthalide treatment. 14Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 15Cloning of a gene-edited macaque monkey by somatic cell nuclear transfer显示文摘Cloning of macaque monkeys by somatic cell nucleus transfer(SCNT) allows the generation of monkeys with uniform genetic backgrounds that are useful for the development of non-human primate models of human diseases. Here, we report the feasibility of this approach by SCNT of fibroblasts from a macaque monkey(Macaca fascicularis), in which a core circadian transcription factor BMAL1 was knocked out by clustered regularly interspaced short palindromic repeat/Cas9 gene editing(see accompanying paper).Out of 325 SCNT embryos transferred into 65 surrogate monkeys, we cloned five macaque monkeys with BMAL1 mutations in both alleles without mosaicism, with nuclear genes identical to that of the fibroblast donor monkey. Further peripheral blood m RNA analysis confirmed the complete absence of the wild-type BMAL1 transcript. 17Comparison of cancer incidence and mortality in three GDP per capita levels in China, 2013显示文摘Objective:In this research,the patterns of cancer incidence and mortality in areas with different gross domestic product per capita(GDPPC)levels in China were explored,using data from population-based cancer registries in 2013,collected by the National Central Cancer Registry(NCCR).Methods:Data from 255 cancer registries were qualified and included in this analysis.Based on the GDPPC data of 2014,cities/counties were divided into 3 levels:high-,middle-and low-GDPPC areas,with 40,000 and 80,000 RMB per year as cut points.We calculated cancer incidences and mortalities in these three levels,stratified by gender and age group.The national population of the Fifth Census in 2000 and Segi’s population were applied for age-standardized rates.Results:The crude incidence and mortality rates as well as age-standardized incidence rate(ASIR)showed positive associations with GDPPC level.The age-standardized mortality rate(ASMR)nevertheless showed a negative association with GDPPC level.The ASMR in high-,middle-and low-GDPPC areas was 103.12/100,000,112.49/100,000 and 117.43/100,000,respectively.Lung cancer was by far the most common cancer in all three GDPPC levels.It was also the leading cause of cancer death,regardless of gender and GDPPC level.Negative associations with GDPPC level were found for the ASIRs of lung,stomach,esophageal and liver cancer,whereas colorectal and breast cancer showed positive associations.Except for breast cancer,the ASMRs of the other five cancers were always higher in middle-and low-GDPPC areas than in high-GDPPC areas.Conclusions:The economic development is one of the main factors of the heavy cancer burden on Chinese population.It would be reasonable to implement cancer control strategies referring to the local GDPPC level.Zhixun Yang Rongshou Zheng Siwei Zhang Hongmei Zeng Changfa Xia He Li Li Wang Yanhong Wang Wanqing Chen 2017Chinese Journal of Cancer Research2017,29,5:13

19Feedback control of nonlinear differential algebraic systems using Hamiltonian function method显示文摘The stabilization and H∞ control of nonlinear differential algebraic systems (NDAS) are investigated using the Hamiltonian function method. Firstly, we put forward a novel dissipative Hamiltonian realization (DHR) structure and give the condition to com- plete the Hamiltonian realization. Then, based on the DHR, we present a criterion for the stability analysis of NDAS and construct a stabilization controller for NDAS in absence of disturbances. Finally, for NDAS in presence of disturbances, the L2 gain is analyzed via generalized Hamilton-Jacobi inequality and an H∞ control strategy is constructed. 20Soil water repellency and influencing factors of Nitraria tangutorun nebkhas at different succession stages显示文摘Soil water repellency(WR) is an important physical characteristic of soil surface. It is capable of largely influencing the hydrological and geomorphological processes of soil, as well as affecting the ecological processes of plants, such as growth and seed germination, and has thus been a hot topic in recent research around the world. In this paper, the capillary rise method was used to study the soil WR characteristics of Nitraria tangutorun nebkhas. Soil water repellencies at different succession stages of Nitraria tangutorun were investigated, and the relationships between soil WR and soil organic matter, total N, and total P, soil texture, pH, and concentrations of CO3 2-, HCO3-, Cl-, SO4 2-, Na+, K+, Ca2+and Mg2+were discussed. Soil WR may be demonstrated at the following nebkhas dune evolvement stages: extremely degraded>degraded>stabilized>well developed>newly developed>quick sand. Apart from some soil at the bottom, the WR of other soils(crest and slope of dune) was found to be largest at the topsoil, and decreased as the soil depth increased. The results showed that multiple factors affected soil WR characteristics, e.g. WR increased significantly as the contents of soil organic matter and total N increased, but did not change as the total P content increased. Soil texture was a key factor affecting soil WR; soil WR increased significantly as clay content increased, and decreased significantly as sand content increased. Low pH was shown to be more suitable for the occurrence of soil WR. Four cations(Ca2+, Mg2+, K+and Na+) and two anions(Cl-and SO4 2-) enhanced soil WR, while CO3 2-decreased it. HCO3-did not show any observable effect. Finally, we established a best-fit general linear model(GLM) between soil-air-water contact angle(CA) and influencing factors(CA=5.606 sand+6.496(clay and silt)–2.353 pH+470.089 CO3 2-+11.346 Na+–407.707 Cl-–14.245 SO4 2-+0.734 total N–519.521).
