1Control mechanism for the upper airway collapse in patients with obstructive sleep apnea syndrome: a finite element study显示文摘Obstructive sleep apnea syndrome (OSAS) is characterized by recurrent collapses of the upper airway, which lead to repetitive transient hypoxia, arousals and finally sleep fragmentation. Both anatomical and neuromuscular factors may play key roles in the pathophysiology of OSAS. The purpose of this paper was to study the control mechanism of OSAS from the mechanical point of view. A three-dimensional finite element model was developed, which not only reconstructed the realistic anatomical structure of the human upper airway, but also included surrounding structures such as the skull, neck, hyoid, cartilage and soft tissues. The respiration process during the normal and apnea states was simulated with the fluid-structure interaction method (FSI) and the computational fluid dynamics method (CFD). The airflow and deformation of the upper airway obtained from the FSI and the CFD method were compared and the results obtained under large negative pressure during an apnea episode were analyzed. The simulation results show that the FSI method is more feasible and effective than the CFD method. The concave configuration of the upper airway may accelerate the collapse of the upper airway in a positive feedback mechanism, which supplies meaningful information for clinical treatment and further research of OSAS.HUANG RenHan LI XiPing RONG QiGuo 2013Science China(Life Sciences)2013,56,4:12
2Erucic acid from Isatis indigotica Fort. suppresses influenza A virus replication and inflammation in vitro and in vivo through modulation of NF-kB and p38 MAPK pathway显示文摘Isatis indigotica Fort.(Ban-Lan-Gen)is an herbal medicine prescribed for influenza treatment.However,its active components and mode of action remain mostly unknown.In the present study,erucic acid was isolated from Isatis indigotica Fort.,and subsequently its underlying mechanism against influenza A virus(IAV)infection was investigated in vitro and in vivo.Our results demonstrated that erucic acid exhibited broad-spectrum antiviral activity against IAV resulting from reduction of viral polymerase transcription activity.Erucic acid was found to exert inhibitory effects on IAV or viral(v)RNA-induced pro-inflam-matory mediators as well as interferons(IFNs).The molecular mechanism by which erucic acid with antiviral and anti-inflammatory properties was attributed to inactivation of NF-kB and p38 MAPK signaling.Furthermore,the NF-kB and p38 MAPK inhibitory effect of erucic acid led to diminishing the transcriptional activity of interferon-stimulated gene factor 3(ISGF-3),and thereby reducing IAV-triggered pro-inflammatory response amplification in IFN-β-sensitized cells.Additionally,IAV-or vRNA-triggered apoptosis of alveolar epithelial A549 cells was prevented by erucic acid.In vivo,erucic acid administration consistently displayed decreased lung viral load and viral antigens expression.Meanwhile,erucic acid markedly reduced CD8+cytotoxic T lymphocyte(CTL)recruitment,pro-apoptotic signaling,hyperactivity of multiple signaling pathways,and exacerbated immune inflammation in the lung,which resulted in decreased lung injury and mortality in mice with a mouse-adapted A/FM/1/47-MA(H1N1)strain infection.Our findings provided a mechanistic basis for the action of erucic acid against IAV-mediated inflammation and injury,suggesting that erucic acid may have a therapeutic potential in the treatment of influenza.Xiaoli Liang Yuan Huang Xiping Pan Yanbing Hao Xiaowei Chen Haiming Jiang Jing Li Beixian Zhou Zifeng Yang 2020Journal of Pharmaceutical Analysis2020,10,2:10
33D printing of multilayered scaffolds for rotator cuff tendon regeneration显示文摘Repairing massive rotator cuff tendon defects remains a challenge due to the high retear rate after surgical intervention.3D printing has emerged as a promising technique that enables the fabrication of engineered tissues with heterogeneous structures and mechanical properties,as well as controllable microenvironments for tendon regeneration.In this study,we developed a new strategy for rotator cuff tendon repair by combining a 3D printed scaffold of polylactic-co-glycolic acid(PLGA)with cell-laden collagen-fibrin hydrogels.We designed and fabricated two types of scaffolds:one featuring a separate layer-by-layer structure and another with a tri-layered structure as a whole.Uniaxial tensile tests showed that both types of scaffolds had improved mechanical properties compared to single-layered PLGA scaffolds.The printed scaffold with collagen-fibrin hydrogels effectively supported the growth,proliferation,and tenogenic differentiation of human adipose-derived mesenchymal stem cells.Subcutaneous implantation of the multilayered scaffolds demonstrated their excellent in vivo biocompatibility.This study demonstrates the feasibility of 3D printing multilayered scaffolds for application in rotator cuff tendon regeneration.Xiping Jiang Shaohua Wu Mitchell Kuss Yunfan Kong Wen Shi Philipp N.Streubel Tieshi Li Bin Duan 2020Bioactive Materials2020,5,3:7
4Comparison of labeling methods and time course of traumatic brain injury-induced cell death in mice显示文摘BACKGROUND:Various molecular mechanisms of cell death following traumatic brain injury have been previously described.However,the time course of cell death remains unclear.TUNEL and Fluoro-Jade B labeling have been widely used to label apoptotic cells and neuronal degeneration.Propidium iodide (PI) functions as a biomarker of cell death in vivo.OBJECTIVE:To explore the role of PI labeling compared to TUNEL and Fluoro-Jade B staining for detecting neural cell death,and to observe time course of traumatic brain injury-induced cell death in mice.DESIGN,TIME AND SETTING:A randomized,controlled,animal experiment was performed at the Laboratory of Aging and Nervous Diseases,Soochow University from September 2007 to December 2008.MATERIALS:PI (B1221) was purchased from Sigma,USA.TUNEL kit was purchased from Roche Molecular Biochemicals,USA.Fluoro-Jade B was purchased from Chemicon,USA.METHODS:A total of 70 healthy,male,Kunming mice were randomly assigned to sham-surgery (n=5) and model (n=65) groups.Traumatic brain injury was established using the controlled cortical impact method.PI was intraperitoneally injected at 1 hour prior to animal sacrifice.MAIN OUTCOME MEASURES:TUNEL,Fluoro-Jade B,and PI-positive cells were quantified using a double-labeling method to determine the time course of traumatic brain injury-induced cell death.RESULTS:PI labeled cells in an earlier phase of cell death than TUNEL and Fluoro-Jade B labeling.PI-positive cells were observed immediately following injury,and the numbers rapidly increased in injured brain areas at 1 hour,peaked at 24-48 hours,and subsequently decreased at 3-21 days post-injury.TUNEL-labeled cells were significantly increased at 12 hours,while Fluoro-Jade B-labeled cells were increased at 6 hours after injury,with cells still visible at 6-48 hours post-injury.Moreover,a greater number of PI-positive cells were observed compared to TUNEL-and Fluoro-Jade B-labeled cells.CONCLUSION:PI labeling is more sensitive and reliable than TUNEL and Fluoro-Jade B staining for detecting cell death following traumatic brain injury.Moreover,PI labeling can function as a reliable marker to estimate the entire time course of cell death.Chengliang Luo Xiping Chen Hong Ni Qianqian Li Rui Yang Yuxia Sun Guangyou Zhu Luyang Tao 2010Neural Regeneration Research2010,5,9:6

6Grapevine VlbZIP30 improves drought resistance by directly activating VvNAC17 and promoting lignin biosynthesis through the regulation of three peroxidase genes显示文摘Drought stress severely affects grapevine quality and yield,and recent reports have revealed that lignin plays an important role in protection from drought stress.Since little is known about lignin-mediated drought resistance in grapevine,we investigated its significance.Herein,we show that VlbZIP30 mediates drought resistance by activating the expression of lignin biosynthetic genes and increasing lignin deposition.Transgenic grapevine plants overexpressing VlbZIP30 exhibited lignin deposition(mainly G and S monomers)in the stem secondary xylem under control conditions,which resulted from the upregulated expression of VvPRX4 and VvPRX72.Overexpression of VlbZIP30 improves drought tolerance,characterized by a reduction in the water loss rate,maintenance of an effective photosynthesis rate,and increased lignin content(mainly G monomer)in leaves under drought conditions.Electrophoretic mobility shift assay,luciferase reporter assays,and chromatin immunoprecipitation-qPCR assays indicated that VlbZIP30 directly binds to the G-box cis-element in the promoters of lignin biosynthetic(VvPRX N1)and drought-responsive(VvNAC17)genes to regulate their expression.In summary,we report a novel VlbZIP30-mediated mechanism linking lignification and drought tolerance in grapevine.The results of this study may be of value for the development of molecular breeding strategies to produce drought-resistant fruit crops.Mingxing Tu Xianhang Wang Wuchen Yin Ya Wang Yajuan Li Guofeng Zhang Zhi Li Junyang Song Xiping Wang 2020Horticulture Research2020,7,1:5
7Coarsening behavior of γ' particles in a nickel-base superalloy显示文摘The coarsening behavior of γ' particles in a nickel-base superalloy FGH95 was investigated by means of experimental observations and growth kinetics calculations. The results show that when aging at 1000,1080 and 1140°C for different times,the relation of average particle size to time obeys the cube law ( a /2)3= kt,where k is 15.49 × 103,77.5 × 103 and 230.04 × 103 nm3/min,respectively. The particle size distributions are better fit to the LSW theoretical distributions when aging at 1000°C within 1440 min. The activation energy for γ' particles coarsening is determined to be 288.20±1.79 kJ/mol,which correlates well to the diffusion activation energies of Al,Ti,and Nb in the nickel matrix. This indicates that the coarsening of γ' particles is controlled by the diffusion of Al,Ti,and Nb in the nickel matrix. 9Characterization of γ′ precipitates in a nickel base superalloy quenching from aging temperature at different rates显示文摘The characteristics of γ′ precipitates in a superalloy quenched from 1050°C at different rates were investigated using field emission scanning electron microscope(FESEM).When quenched from 1050°C, the size of primary aging γ′ precipitates has a small increase in the specimens that experienced iced-brine-quenching, oil-quenching, and air-cooling-quenching conditions and a drastic increase in the specimen that experienced a furnace-cooling-quenching condition.The cooling γ′ precipitates have unimodal distributions after quenching at the air-cooling rate and bimodal distributions after quenching at the furnace-cooling rate, but there are not these distributions in the specimens that experienced iced-brine-quenching and oil-quenching conditions.When aging at 760°C, the size of primary aging γ′ precipitates appears unaffected in the specimens that experienced iced-brine-quenching, oil-quenching, and air-cooling-quenching conditions.However, it has a drastic increase in the specimen that experienced a furnace-cooling-quenching condition, and it is interesting that the bigger cooling γ′ precipitates have a coalescence and octodendritic shape.The microhardness study indicates that the hardness has no variation in the specimens that experienced iced-brine-quenching, oil-quenching, and air-cooling-quenching conditions and has a drastic decrease in the specimens that experienced a furnace-cooling-quenching condition and obtains the minimum microhardness value 390.8 HV.LI Hongyu, SONG Xiping, WANG Yanli, and CHEN Guoliang State Key Lab for Advanced Metals and Materials, University of Science and Technology Beijing, Beijing 100083, China 2010Rare Metals2010,29,2:4
10Three-dimensional numerical simulation of obfique wave action on vertical walls显示文摘A three-dimensional numerical model based on the potential theory was developed to study the oblique wave action on vertical walls. A source term inside the domain was used to generate incident waves and outgoing waves were dissipated by sponge layers and trans- mitted by radiation boundaries. The finite difference method was used to solve the governing equations and boundary conditions in the regular transformed domain in σ-coordinate. Satisfactory agreements between the numerical predictions and experimental results of wave force were obtained. 15VlbZIP30 of grapevine functions in dehydration tolerance via the abscisic acid core signaling pathway显示文摘Drought stress limits the growth and development of grapevines,thereby reducing productivity,but the mechanisms by which grapevines respond to drought stress remain largely uncharacterized.Here,we characterized a group A bZIP gene from“Kyoho”grapevine,VlbZIP30,which was shown to be induced by abscisic acid(ABA)and dehydration stress.Overexpression of VlbZIP30 in transgenic Arabidopsis thaliana enhanced dehydration tolerance.Transcriptome analysis revealed that a major proportion of ABA-responsive and/or drought-responsive genes are transcriptionally regulated by VlbZIP30 during ABA or mannitol treatment at the cotyledon greening stage.We identified an A.thaliana G-box motif(CACGTG)and a potential grapevine G-box motif(MCACGTGK)in the promoters of the 39 selected A.thaliana genes upregulated in the transgenic plants and in the 35 grapevine homologs,respectively.Subsequently,using two grapevine-related databases,we found that 74%(23/31)and 84%(21/25)of the detected grapevine genes were significantly upregulated by ABA and drought stress,respectively,suggesting that these genes are involved in ABA or dehydration stress and may be regulated by VlbZIP30 in grapevine.We propose that VlbZIP30 functions as a positive regulator of dehydration-responsive signaling in the ABA core signaling pathway.Mingxing Tu Xianhang Wang Yanxun Zhu Dejun Wang Xuechuan Zhang Ye Cui Yajuan Li Min Gao Zhi Li Yuejin Wang Xiping Wang 2018Horticulture Research2018,5,1:2
16Increasing rainfed wheat yield by optimizing agronomic practices to consume more subsoil water in the Loess Plateau显示文摘Erratic rainfall and misalignment between the rainy season and the growing season of winter wheat greatly limit rainfed winter wheat yield in the Loess Plateau of China. To increase the grain yield of winter wheat in this region, the effects of different agronomic practices, including adjusting planting pattern(NR, narrow row spacing), increasing seeding rate(high seeding rate, HS), decreasing basal nitrogen rate and increasing top-dressed nitrogen rate(DBN), and replacing an old cultivar with a new cultivar(NC) on wheat yield were investigated for two consecutive years. The results showed that the current grain yield of rainfed winter wheat in the Loess Plateau could be increased to 5879–7093 kg ha^(-1) by HS, DBN and NC practices relative to the practice of high-yielding farmers(PF). The increased yield due to HS, DBN and NC was attributed to the higher number of spikes ha^(-1), 1000-grain weight, and kernels spike^(-1). Before the flowering stage, HS increased soil water consumption(SWC) in 1–3 m subsoil due to the higher plant population compared with that of PF, whereas DBN decreased SWC in the 0–2 m soil layer compared with that of PF. After the flowering stage, HS, DBN, and NC increased SWC by 8–16 mm in 2–3 m subsoil compared to PF. The water use efficiency(WUE) was increased under DBN and NC in comparison with PF.However, the WUE did not increase under HS as it had the highest evapotranspiration among the five treatments. 17Wave decomposition phenomenon and spectrum evolution over submerged bars显示文摘Wave decomposition phenomenon and spectrum evolution over submerged bars are investigated by a previously developed numerical model. First, the computed free surface displacements of regular waves at various locations are compared with the available experimental data to confirm the validity of the numerical model, and satisfactory agreements are obtained. In addition, variations of decomposition characteristics with incident wave parameters and the change of energy spectrum for regular waves are also studied. Then the spectrum evolution of irregular waves over submerged bars, as well as the influence of incident peak wave period and the steepness of the front slope of the bar on spectrum evolution, is investigated. Wave decomposition and spectral shape are found to be significantly influenced by the incident wave conditions. 19Detection of QTL and QTN and candidate genes for oil content in soybean using a combination of four-way-RIL and germplasm populations显示文摘Soybean is a source of edible oil for humans and provides a third of the vegetable oil consumed worldwide.Increasing seed oil content in seeds is thus a key objective in soybean breeding.In the present study,a four-way recombinant inbred line(FW-RIL)population comprising 144 lines,planted in 10 environments,and a germplasm panel of 455 accessions,planted in two environments,were used to collect oil-content phenotypes.First,59 quantitative trait loci(QTL)were detected in the FW-RIL population by inclusive complete interval mapping on a linkage map consisting of 2232 single-nucleotide polymorphism(SNP)markers.Also in the FW-RILs,44 quantitative trait nucleotides(QTNs)were detected by association analysis using 109,676 SNP markers and fivemethods of multi-locus genome-wide association study.Second,77 QTN were detected by association analysis in the germplasm panel using 63,306 markers.Comparison of the QTL and QTN suggested four QTN controlling oil content.Pathway analysis was performed on genes in attenuation regions of these four QTN,and two candidate genes involved in the synthesis or metabolism of soybean oil were identified.These findings provide useful information about the genetics of oil content and may contribute to its genetic improvement by marker-assisted selection.Xiyu Li Kaixin Zhang Xu Sun Shanshan Huang Jiajing Wang Chang Yang Mahfishan Siyal Chao Wang Chunlan Guo Xiping Hu Wen-Xia Li Hailong Ning 2020The Crop Journal2020,8,5:2
