1Mitophagy links oxidative stress conditions and neurodegenerative diseases显示文摘Mitophagy is activated by a number of stimuli, including hypoxia, energy stress, and increased oxidative phosphorylation activity. Mitophagy is associated with oxidative stress conditions and central neurodegenerative diseases. Proper regulation of mitophagy is crucial for maintaining homeostasis; conversely, inadequate removal of mitochondria through mitophagy leads to the generation of oxidative species, including reactive oxygen species and reactive nitrogen species, resulting in various neurodegenerative diseases, such as Alzheimer's disease, Parkinson's disease, Huntington's disease, and amyotrophic lateral sclerosis. These diseases are most prevalent in older adults whose bodies fail to maintain proper mitophagic functions to combat oxidative species. As mitophagy is essential for normal body function, by targeting mitophagic pathways we can improve these disease conditions. The search for effective remedies to treat these disease conditions is an ongoing process, which is why more studies are needed. Additionally, more relevant studies could help establish therapeutic conditions, which are currently in high demand. In this review, we discuss how mitophagy plays a significant role in homeostasis and how its dysregulation causes neurodegeneration. 2Grapevine VlbZIP30 improves drought resistance by directly activating VvNAC17 and promoting lignin biosynthesis through the regulation of three peroxidase genes显示文摘Drought stress severely affects grapevine quality and yield,and recent reports have revealed that lignin plays an important role in protection from drought stress.Since little is known about lignin-mediated drought resistance in grapevine,we investigated its significance.Herein,we show that VlbZIP30 mediates drought resistance by activating the expression of lignin biosynthetic genes and increasing lignin deposition.Transgenic grapevine plants overexpressing VlbZIP30 exhibited lignin deposition(mainly G and S monomers)in the stem secondary xylem under control conditions,which resulted from the upregulated expression of VvPRX4 and VvPRX72.Overexpression of VlbZIP30 improves drought tolerance,characterized by a reduction in the water loss rate,maintenance of an effective photosynthesis rate,and increased lignin content(mainly G monomer)in leaves under drought conditions.Electrophoretic mobility shift assay,luciferase reporter assays,and chromatin immunoprecipitation-qPCR assays indicated that VlbZIP30 directly binds to the G-box cis-element in the promoters of lignin biosynthetic(VvPRX N1)and drought-responsive(VvNAC17)genes to regulate their expression.In summary,we report a novel VlbZIP30-mediated mechanism linking lignification and drought tolerance in grapevine.The results of this study may be of value for the development of molecular breeding strategies to produce drought-resistant fruit crops.Mingxing Tu Xianhang Wang Wuchen Yin Ya Wang Yajuan Li Guofeng Zhang Zhi Li Junyang Song Xiping Wang 2020Horticulture Research2020,7,1:5
3On the reversibility of the α_(2)/ω_(o) phase transformation in a high Nb containing TiAl alloy during high temperature deformation显示文摘The transformations between the phasesα_(2)(Ti_(3)Al)andω_(o)were investigated in a lamellar multiphase titanium aluminide alloy based onγ(TiAl).The paper complements an earlier investigation performed on the same material in which the importance of deformation-induced twin structures for theα_(2)→ω_(o) transformation was demonstrated.The present study shows that the reverse transformationω_(o)→α_(2) can also occur during high-temperature deformation.The transformation is probably triggered by constraint stresses,which exist between the different constituents due to the crystalline mismatch.The combined operation of mechanical twinning of theα_(2) phase and the reversible transformation fully converts theα_(2) lamellae into a mixture ofα_(2) andω_(o).This conversion greatly reduces the mechanical anisotropy existing in formerα_(2) lamellae.Regarding the technical use of the alloy,the stability of the converted structure with respect to further annealing was also examined.The reported processes occur at the nano-meter and sub nano-meter scale,thus,advanced characterization techniques were applied,such as high-resolution transmission electron microscopy(HRTEM)and atom probe tomography(APT).Lin Song Fritz Appel Andreas Stark Uwe Lorenz Junyang He Zhanbing He Junpin Lin Tiebang Zhang Florian Pyczak 2021Journal of Materials Science & Technology2021,,34:1
4Anatomical distributional defects in mutant genes associated with dominant intermediate Charcot-Marie-Tooth disease type C in an adenovirus-mediated mouse model显示文摘Dominant intermediate Charcot-Marie-Tooth disease type C(DI-CMTC) is a dominantly inherited neuropathy that has been classified primarily based on motor conduction velocity tests but is now known to involve axonal and demyelination features.DI-CMTC is linked to tyrosyl-t RNA synthetase(YARS)-associated neuropathies,which are caused by E196 K and G41 R missense mutations and a single de novo deletion(153-156 del VKQV).It is well-established that these YARS mutations induce neuronal dysfunction,morphological symptoms involving axonal degeneration,and impaired motor performance.The present study is the first to describe a novel mouse model of YARS-mutation-induced neuropathy involving a neuron-specific promoter with a deleted mitochondrial targeting sequence that inhibits the expression of YARS protein in the mitochondria.An adenovirus vector system and in vivo techniques were utilized to express YARS fusion proteins with a Flag-tag in the spinal cord,peripheral axons,and dorsal root ganglia.Following transfection of YARS-expressing viruses,the distributions of wild-type(WT) YARS and E196 K mutant proteins were compared in all expressed regions; G41 R was not expressed.The proportion of Flag/green fluorescent protein(GFP) double-positive signaling in the E196 K mutant-type mice did not significantly differ from that of WT mice in dorsal root ganglion neurons.All adenovirus genes,and even the empty vector without the YARS gene,exhibited GFP-positive signaling in the ventral horn of the spinal cord because GFP in an adenovirus vector is driven by a cytomegalovirus promoter.The present study demonstrated that anatomical differences in tissue can lead to dissimilar expressions of YARS genes.Thus,use of this novel animal model will provide data regarding distributional defects between mutant and WT genes in neurons,the DICMTC phenotype,and potential treatment approaches for this disease.Seo Jin Lee Sandesh Panthi Hyun Woo Jo Jaeyoung Cho Min-Sik Kim Na Young Jeong In Ok Song Junyang Jung Youngbuhm Huh 2017Neural Regeneration Research2017,12,3:1
5Long-term case-fatality rate of nontuberculous mycobacterial disease in people living with HIV显示文摘Background:Few data are available regarding the long-term case-fatality rate(CFR)among people living with HIV(PLWH)with nontuberculous mycobacteria(NTM)disease.The aim of this study is to analyze the long-term CFR in patients with NTM disease and to identify risk factors for their death.Methods:A retrospective cohort study of 379 cases of microbiologically confirmed NTM disease in PLWH was conducted from January 1,2012,to December 31,2020,in Shanghai,China.We used Kaplan-Meier survival analysis and the log-rank test to compare the long-term CFR in patients with disseminated NTM(DNTM)and localized NTM disease.Univariate Cox proportional hazards regression analysis and a stepwise Cox proportional hazards regression model were used to estimate the predictors of long-term CFR.Results:The cohort was followed up for a median of 26 months.The total CFR was 15.7%by one year and increased to 22.6%at 5 years after the diagnosis of NTM disease.The 5-year CFR of PLWH with DNTM was significantly higher than that of PLWH with localized NTM(26.7%vs 19.6%for DNTM and localized NTM disease,respectively).Older age[hazard ratio(HR)=1.04,95%confidence interval(CI):1.02-1.06,P<0.001],comorbidity(HR=2.05,95%CI:1.21-3.49,P<0.01),DNTM(HR=2.08,95%CI:1.17-3.68,P<0.05),and HIV viral load(HR=1.32,95%CI:1.12-1.55,P<0.001)were all independent risk factors for long-term CFR.In the subgroup analysis,time to culture positivity was negatively correlated with CFR in patients with DNTM(HR=0.90,95%CI:0.82-0.98,P<0.05).Conclusions:NTM was associated with a high long-term CFR in PLWH.Further approaches to prevent NTM disease in PLWH are urgently needed.Jingjing Hu Ling Gu Yueming Shao Renfang Zhang Tangkai Qi Jianjun Sun Zhenyan Wang Wei Song Yang Tang Jiangrong Wang Shuibao Xu Junyang Yang Yinzhong Shen Li Liu Jun Chen Hongzhou Lu 2022Infectious Diseases of Poverty2022,11,1:0

7Identification of potential key genes in resveratrol biosynthesis via transcriptional analyses of berry development in grapevine(Vitis spp.)genotypes varying in trans-resveratrol content显示文摘Resveratrol is an important secondary metabolite not only owing to its function as a phytoalexin,but also its potential benefits to human health.In this study,the content of trans-resveratrol was documented in seven accessions of grapevine,in the seed,pulp and skin of berries,and at three developmental stages.The highest amount(2.99μg g^(−1) FW)was found in the skin of berries at the ripe stage from V.amurensis‘Tonghua-3'.Resveratrol was not detected in several samples,including skin of berries at the green hard or véraison stage from V.davidii‘Tangwei'.We carried out transcriptional profiling of developing‘Tonghua-3'and‘Tangwei'berries to identify gene expression patterns that may be linked with the difference in resveratrol content between these accessions.The expression levels of several differentially expressed genes(DEGs)with presumed function in resveratrol biosynthesis,including STILBENE SYNTHASEs(STSs),CINNAMATE 4-HYDROXYLASEs(C4Hs)and 4-COUMARATE-COA LIGASEs(4CLs),were significantly higher in‘Tonghua-3',than in'Tangwei'during the véraison and ripe stages.Gene ontology and Kyoto Encyclopedia of Genes and Genomes analyses suggested that these DEGs were enriched for multiple biological processes at the three stages of fruit development.Additionally,we identified a total of 36 transcription factors,including MYBs,WRKYs,ERFs,bHLHs and bZIPs,that were coexpressed with 17 STSs via a weighted gene co-expression network analysis,suggesting roles as regulators of resveratrol biosynthesis.Overall,these findings provide insight into genotypic differences in resveratrol biosynthesis in grapevine,as well as the molecular genetics of its regulation.Xiuming Zhang Yifei Wu Yichu Zhang Xiangjing Yin Steve van Nocker Junqiang Guo Zhi Li Min Gao Junyang Song Xiping Wang 2022Fruit Research2022,2,1:0
8Gold mineralization in Jiaodong Peninsula and destruction of North China Craton:Insights from Mesozoic granite显示文摘Large-scale tectonic magmatism and extensive gold mineralization have occurred in the eastern North China Craton(NCC)(Jiaodong and Liaodong peninsulas)since Mesozoic,which indicated that the region experienced decratonization process.The genesis type of granites evolved from S-type to I-type and A-type during Late Jurassic to Early Cretaceous,and thus reflects the evolution of geodynamics in the Late Mesozoic,indicating the varied subduction process of the Paleo-Pacific Plate(PPP)and the craton destruction.The evolution of geochemical features shows that the mantle beneath the Jiaodong Peninsula evolved from EM-II in Jurassic to EM-I in Early Cretaceous,demonstrating that the mantle switched from enriched to depleted.The gold of decratonic gold deposits in Jiaodong Peninsula mainly comes from the lithospheric mantle domains metasomatized by fluids derived from the metamorphism and dehydration of the subducted slab in the mantle transition zone.The rapid decomposition of minerals leads to the concentrated release of ore-forming fluids,and this process leads to the explosive gold mineralization during the craton destruction.Extensive magmatic uplift and extensional structures,triggered by the craton destruction in Early Cretaceous formed the extensional tectonic system,providing space for the decratonic gold deposits in Jiaodong Peninsula.WANG Bin ZHOU Jianbo DING Zhengjiang ZHAO Tiqun SONG Mingchun BAO Zhongyi LYU Junyang XU Shaohui YAN Chunming LIU Xiangdong LIU Jialiang 2023Global Geology2023,26,2:0
