Evolution of the novel coronavirus from the ongoing Wuhan outbreak and modeling of its spike protein for risk of human transmission显示文摘Dear Editor,The occurrence of concentrated pneumonia cases in Wuhan city,Hubei province of China was first reported on December 30,2019 by the Wuhan Municipal Health Commission(WHO,2020).The pneumonia cases were found to be linked to a large seafood and animal market in Wuhan, and measures for sanitation and disinfection were taken swiftly by the local government agency. The Centers for Disease Control and Prevention(CDC)and Chinese health authorities later determined and announced that a novel coronavirus(CoV),denoted as 2019-nCoV,had caused the pneumonia outbreak in Wuhan city(CDC,2020).Scientists from multiple groups had obtained the virus samples from hospitalized patients(Normile,2020).The isolated viruses were morphologically identical when observed under electron microscopy.Xintian Xu Ping Chen Jingfang Wang Jiannan Feng Hui Zhou Xuan Li Wu Zhong Pei Hao 2020Science China(Life Sciences)2020,63,3:737

Fuzzy adaptive robust control for space robot considering the effect of the gravity显示文摘Space robot is assembled and tested in gravity environment, and completes on-orbit service(OOS) in microgravity environment. The kinematic and dynamic characteristic of the robot will change with the variations of gravity in different working condition. Fully considering the change of kinematic and dynamic models caused by the change of gravity environment, a fuzzy adaptive robust control(FARC) strategy which is adaptive to these model variations is put forward for trajectory tracking control of space robot. A fuzzy algorithm is employed to approximate the nonlinear uncertainties in the model, adaptive laws of the parameters are constructed, and the approximation error is compensated by using a robust control algorithm. The stability of the control system is guaranteed based on the Lyapunov theory and the trajectory tracking control simulation is performed. The simulation results are compared with the proportional plus derivative(PD) controller, and the effectiveness to achieve better trajectory tracking performance under different gravity environment without changing the control parameters and the advantage of the proposed controller are verified.Qin Li Liu Fucai Liang Lihuan Gao Jingfang 2014Chinese Journal of Aeronautics2014,27,6:14
Mobility Analysis of Altmann Overconstrained Linkages by Modified Grübler-Kutzbach Criterion显示文摘Developing a general mobility method/formula is a hot topic lasting for more than 150 years in kinematics. It is necessary to apply any mobility method to puzzling overconstrained mechanisms for verification of its generality. Altmann linkages are such recognized puzzling mechanisms that their mobility analysis is of important significance. A necessary condition for judging a general mobility method is that the method can be fit for Altmann linkages. Firstly, this study classes Altmann linkages into 17 types in terms of the numbers and types of kinematic pairs, and then Altmann overconstrained linkages are further classified into 4 types. Secondly, the mobility of Altmann overconstrained linkages is systematically analyzed by the Modified Grübler-Kutzbach criterion based on screw theory, where passive freedoms are defined as limb passive freedoms and mechanism passive freedoms. In addition, the full-cycle mobility is judged, which overcomes the shortcoming of instantaneous property of screw theory. It is shown that Modified Grübler-Kutzbach criterion not only obtains the correct numerical mobility, but also gives the mobility character by resolving reciprocal screws for the constraint system. This study lays the foundation of verification for the generality of Modified Grübler-Kutzbach criterion. Besides, Altmann overconstrained linkages almost comprise all kinds of modern parallel mechanisms and some classical mechanisms, which provides an important reference for mechanism mobility calculation.LIU Jingfang LI Yanwen HUANG Zhen 2011Chinese Journal of Mechanical Engineering2011,24,4:7
Applicability and Generality of the Modified Grübler-Kutzbach Criterion显示文摘A generally applicable criterion for all mechanism mobility has been an active domain in mechanism theory lasting more than 150 years. It is stated that the Modified Grübler-Kutzbach criterion for mobility has been successfully used to solve the mobility of many more kinds of mechanisms, but never before has anyone proven the applicability and generality of the Modified Grübler-Kutzbach criterion in theory. In order to fill the gap, the applicability and generality of the Modified Grübler-Kutzbach Criterion of mechanism mobility is systematically demonstrated. Firstly, the mobility research background and the Modified Grübler-Kutzbach criterion are introduced. Secondly, some new definitions, such as half local freedom, non-common constraint space of a mechanism and common motion space of a mechanism, etc, are given to demonstrate the correctness and broad applicability of the Modified Grübler-Kutzbach criterion. Thirdly, the general applicability of the Modified Grübler-Kutzbach criterion is demonstrated based on screw theory. The mobilities of the classical DELASSUS mechanisms and a modern planar parallel mechanism, are determined through the Modified Grübler-Kutzbach criterion, which are as examples to show the practical application of the Modified Grübler-Kutzbach criterion.LI Yanwen WANG Lumin LIU Jingfang HUANG Zhen 2013Chinese Journal of Mechanical Engineering2013,26,2:6

Influence of CeO2 loading on structure and catalytic activity for NH3-SCR over TiO2-supported CeO2显示文摘A series of supported CeO2/TiO2 catalysts were prepared to explore the influence of CeO2 loading on these catalysts for the selective catalytic reduction of NO3 by NH3(NH3-SCR).The catalysts were investigated in detail by means of XRD,Raman,H2-TPR,NH3-TPD,XPS,in situ DRIFTS,and NH3-SCR reaction. The activity of the catalyst is closely related to the content of CeO2.When the loading of CeO2 is near the dispersion capacity(1.16 mmol Ce4+/100 m^2 TiO2),the catalytic activity is better. This may be because that the dispersed CeO2 is the active species and the catalyst has appropriate redox property, along with the larger amounts of surface Ce content and surface adsorbed oxygen species. Finally,a possible reaction mechanism via the Langmuir-Hinshelwood(L-H)mechanism is tentatively proposed to further understand the NH3-SCR reaction.Hongliang Zhang Long Ding Hongming Long Jiaxin Li Wei Tan Jiawei Ji Jingfang Sun Changjin Tang Lin Dong 2020Journal of Rare Earths2020,38,8:5
Eigen microstates and their evolutions in complex systems显示文摘Emergence refers to the existence or formation of collective behaviors in complex systems. Here,we develop a theoretical framework based on the eigen microstate theory to analyze the emerging phenomena and dynamic evolution of complex system. In this framework, the statistical ensemble composed of M microstates of a complex system with N agents is defined by the normalized N×M matrix A,whose columns represent microstates and order of row is consist with the time. The ensemble matrix A can be decomposed as■,where r=min(N,M),eigenvalueσIbehaves as the probability amplitude of the eigen microstate U_I so that■and U_I evolves following V_I.In a disorder complex system, there is no dominant eigenvalue and eigen microstate. When a probability amplitudeσIbecomes finite in the thermodynamic limit, there is a condensation of the eigen microstate UIin analogy to the Bose–Einstein condensation of Bose gases. This indicates the emergence of U_I and a phase transition in complex system. Our framework has been applied successfully to equilibrium threedimensional Ising model, climate system and stock markets. We anticipate that our eigen microstate method can be used to study non-equilibrium complex systems with unknown orderparameters, such as phase transitions of collective motion and tipping points in climate systems and ecosystems.Yu Sun Gaoke Hu Yongwen Zhang Bo Lu Zhenghui Lu Jingfang Fan Xiaoteng Li Qimin Deng Xiaosong Chen 2021Communications in Theoretical Physics2021,73,6:5
Mesoscale fracture behavior of Longmaxi outcrop shale with different bedding angles:Experimental and numerical investigations显示文摘The mechanical properties and fracturing mechanism of shale containing beddings are critically important in shale gas exploitation and wellbore stability. To investigate the effects of shale bedding on crack behavior and fracturing mechanism, scanning electron microscope(SEM)with a loading system was employed to carry out three-point bending tests on Longmaxi outcrop shale. The crack initiation and propagation of Longmaxi shale were observed and recorded by taking photos during loading. The cracking paths were extracted to calculate the crack length through a MATLAB program. The peak load, fracture toughness and fracture energy all increase with the bedding angle from 0°to 90°.The crack length and energy were also found to increase with the bedding angle in the range of 0°-600 and then drop slightly. The fracturing mechanism of shale includes the main crack affected by the bedding angle and disturbed by randomly distributed particles. The main cracking path was accompanied by several microcrack branches which could form an interconnected crack system. When the main crack encounters larger sedimentary particles,it will deflect around the particles and then restore to the initial direction.A numerical technique using extended finite element method(XFEM)coupled with anisotropic cohesive damage criteria was developed, which is able to capture the dependence of crack propagations on bedding angle and sedimentary particles. This study sheds light on understanding and predicting mesoscale fracture behavior of shale with different bedding angles.Jianping Zuo Jingfang Lu Rojin Ghandriz Jintao Wang Yanhong Li Xiaoyan Zhang Jun Li Hongtao Li 2020Journal of Rock Mechanics and Geotechnical Engineering2020,12,2:4
Mobility of the Myard 5R Linkage Involved in “Gogu Problem”显示文摘Since the traditional Grbler-Kutzbach criterion fails in many overconstrained mechanisms, developing a general mobility formula is a hot topic lasting for more than 150 years in mechanisms. GOGU systematically investigated various mobility methods, and pointed that the methods were not fit for two kinds of paradoxical overconstrained mechanisms. The mobility on the two kinds of mechanisms is regarded as 'Gogu problem' . The Modified Grbler-Kutzbach criterion has solved the mobility of the second kind of mechanisms in 'Gogu problem' , and has developed into a systematic mobility methodology. Myard 5R linkage is one of the single-loop mechanisms involved in 'Gogu problem' , its joint axes are distributed in space with special geometric conditions, which increases the difficulty of mobility analysis. The study is to calculate the global mobility of the Myard 5R linkage using the mobility methodology. Firstly, the mobility methodology based on screw theory is briefly introduced. Secondly, some homogeneous transforms are performed according to the D-H parameters and the invariance of the linkage plane symmetry is revealed, which provides an idea to judge a plane-symmetric loop. The special geometric features of the axes distribution are discussed as well. Finally, the global mobility of the Myard 5R linkage is determined by the Modified Grbler-Kutzbach criterion. The results show that the methodology can be applied to more paradoxical mechanisms.LIU Jingfang HUANG Zhen LI Yanwen 2009Chinese Journal of Mechanical Engineering2009,22,3:3
Mechanisms and Effects on HBV Replication of the Interaction between HBV Core Protein and Cellular Filamin B显示文摘Hepatitis B virus(HBV) infection is one of the major problems that threatens global health. There have been many studies on HBV, but the relationship between HBV and host factors is largely unexplored and more studies are needed to clarify these interactions. Filamin B is an actin-binding protein that acts as a cytoskeleton protein, and it is involved in cell development and several signaling pathways. In this study, we showed that filamin B interacted with HBV core protein, and the interaction promoted HBV replication. The interaction between filamin B and core protein was observed in HEK293T, Huh7 and HepG2 cell lines by co-immunoprecipitation and co-localization immnofluoresence. Overexpression of filamin B increased the levels of HBV total RNAs and pre-genome RNA(pg RNA), and improved the secretion level of hepatitis B surface antigen(HBsAg) and hepatitis B e antigen(HBeAg). In contrast, filamin B knockdown inhibited HBV replication, decreased the level of HBV total RNAs and pgRNA, and reduced the secretion level of HBsAg and HBeAg. In addition, we found that filamin B and core protein may interact with each other via four blocks of argentine residues at the C-terminus of core protein. In conclusion, we identify filamin B as a novel host factor that can interact with core protein to promote HBV replication in hepatocytes. Our study provides new insights into the relationship between HBV and host factors and may provide new strategies for the treatment of HBV infection.Yilin Li Yishuang Sun Fuyun Sun Rong Hua Chenlin Li Lang Chen Deyin Guo Jingfang Mu 2018Virologica Sinica2018,33,2:3
Spectrofluorimetric analysis of captopril based on its obstruction effect of the nanomaterial surface energy transfer between acridine orange and gold nanoparticles显示文摘A simple and sensitive method for detection of captopril was established based on its obstructive effect on nanomaterial surface energy transfer(NSET). It was found that the acridine orange(AO) could be adsorbed onto the surface of citrated-gold nanoparticles(Au NPs) through electrostatic interaction. Incidentally, the fluorescence of AO was quenched owing to the dipole-dipole interaction of NSET between AO fluorophore and the Au NPs. However, captopril could obstruct the occurrence of NSET between AO and Au NPs effectively with the formation of Au-S covalent bonds between it and the Au NPs. Consequently, AO molecules were moved away from the surface of Au NPs leading to a decline of the energy transfer efficiency. Moreover, the fluorescence of AO could be gradually restored with the addition of captopril. Under the optimal conditions, the recovered fluorescence intensity correlated linearly with the concentration of captopril in the range of 400 nmol/L–2.0 μmol/L with a detection limit of 71 nmol/L. Besides, the proposed method was successfully applied for the detection of captopril in troches with the recovery of 93%–102% and the RSD lower than 2.24%. The results were in good agreement with those obtained from the HPLC method.Jingfang Guo Yamei Yang Xiaoli Hu Yuanfang Li 2015Science China Chemistry2015,58,5:2
Docking and molecular dynamics studies on CYP2D6显示文摘Drug-metabolizing enzymes, also known as cytochrome P450s,are a superfamily of hemoglobin responsible for metabolizing more than 90% clinical drugs.Cytochrome P450 2D6(CYP2D6) is a significant member of cytochrome P450s for the reason of metabolizing about 20% clinical drugs. In this paper, molecular docking and molecular dynamic simulations are used to investi-gate the active site of CYP2D6,roles of essential amino acids within the active site and time-dependent protein energy changes. The results suggest that amino acids Glu216,Asp301,Ser304 and Ala305 in the active site are likely to form hydrogen bonding interactions with substrates;the benzene ring of Phe120 and aromatic ring in the substrates form Π-Π interactions. In addition, molecular dynamics simulations prove that the catalytic conformation of CYP2D6 without ligands can be obtained by their own atomic fluctuations. The impact of ligands on protein system energy and large conformational shift is not very large.Cytochrome P450s is known for their genetic polymorphisms, which will result in severe adverse drug reactions.Ideally,we hope to use mo-lecular modeling to investigate the differences between the substrates of wild-type and mutants while they are bonded with drugs, and predict the drug metabolizing ability of mutants.Reduce the possibility for people taking drugs that they can not metabolize, therefore reduce the rate of adverse drug reactions, and eventually establish a platform of personalized drugs to largely benefit human health.WANG JingFang ZHANG ChengCheng WEI DongQing LI YiXue 2010Chinese Science Bulletin2010,55,18:2
Rice FLOURY ENDOSPERM 18 encodes a pentatricopeptide repeat protein required for 5′ processing of mitochondrial nad5 messenger RNA and endosperm development显示文摘Pentatricopeptide repeat(PPR) proteins, composing one of the largest protein families in plants, are involved in RNA binding and regulation of organelle RNA metabolism at the posttranscriptional level. Although several PPR proteins have been implicated in endosperm development in rice(Oryza sativa), the molecular functions of many PPRs remain obscure. Here, we identified a rice endosperm mutant named floury endosperm 18(flo18) with pleiotropic defects in both reproductive and vegetative development.Map-based cloning and complementation tests showed that FLO18 encodes a mitochondriontargeted P-type PPR protein with 15 PPR motifs.Mitochondrial function was disrupted in the flo18 mutant, as evidenced by decreased assembly of Complex I in the mitochondrial electron transport chain and altered mitochondrial morphology. Loss of FLO18 function resulted in defective 5′-end processing of mitochondrial nad5 transcripts encoding subunit 5 of nicotinamide adenine dinucleotide hydrogenase. These results suggested that FLO18 is involved in 5′-end processing of nad5 messenger RNA and plays an important role in mitochondrial function and endosperm development.Mingzhou Yu Mingming Wu Yulong Ren Yihua Wang Jingfang Li Cailin Lei Yinglun Sun Xiuhao Bao Hongming Wu Hang Yang Tian Pan Yongfei Wang Ruonan Jing Mengyuan Yan Houda Zhang Lei Zhao Zhichao Zhao Xin Zhang Xiuping Guo Zhijun Cheng Bing Yang Ling Jiang Jianmin Wan 2021Journal of Integrative Plant Biology2021,63,5:2
OsbZIP72 Is Involved in Transcriptional Gene-Regulation Pathway of Abscisic Acid Signal Transduction by Activating Rice High-Affinity Potassium Transporter OsHKT1;1显示文摘We created CRISPR-Cas9 knock-out and overexpressing OsbZIP72 transgenic rice plants to gain a better understanding of the role and molecular mechanism of OsbZIP72 gene in stress tolerance, which has remained largely elusive.OsbZIP72 was expressed and integrated into rice transgenic plant genomes, and the OsbZIP72 transcript in overexpression lines was elicited by salinity, abscisic acid(ABA)and drought stresses.OsbZIP72 overexpressing plants showed higher tolerance to drought and salinity stresses, while knock-out transgenic lines showed higher sensitivity to these stresses. The differentially expressed genes(DEGs)from RNA-sequencing data encompassed several abiotic stress genes, and the functional classification of these DEGs demonstrated the robust transcriptome diversity in OsbZIP72.Yeast one-hybrid,along with luciferase assay, indicated that OsbZIP72 acted as a transcriptional initiator.Remarkably, electrophoresis mobility assay revealed that OsbZIP72 bound directly to the ABAresponsive element in the OsHKT1;1 promoter region and activated its transcription.Overall, our findings revealed that OsbZIP72 can act as a transcriptional modulator with the ability to induce the expression of OsHKT1;1 in response to environmental stress through an ABA-dependent regulatory pathway, indicating that OsbZIP72 can play a crucial role in the ABA-mediated salt and drought tolerance pathway in rice.WANG Baoxiang LIU Yan WANG Yifeng LI Jingfang SUN Zhiguang CHI Ming XING Yungao XU Bo YANG Bo LI Jian LIU Jinbo CHEN Tingmu FANG Zhaowei LU Baiguan XU Dayong Babatunde Kazeem BELLO 2021Rice science2021,28,3:2
Reinforcement learning based energy efficient robot relay for unmanned aerial vehicles against smart jamming显示文摘Unmanned aerial vehicles(UAVs)with limited energy resources, severe path loss, and shadowing to the ground base stations are vulnerable to smart jammers that aim to degrade the UAV communication performance and exhaust the UAV energy. The UAV anti-jamming communication performance, such as the outage probability, degrades if the robot relay is not aware of the jamming policies and the UAV network topology. In this paper,we propose a robot relay scheme for UAVs against smart jamming, which combines reinforcement learning with a function approximation approach named tile coding,to jointly optimize the robot moving distance and relay power with the unknown jamming channel states and locations. The robot mobility and relay policy are chosen based on the received jamming power, the robot received signal quality, location and energy consumption, and the bit error rate of the UAV messages. We also present a deep reinforcement learning version for the robot with sufficient computing resources. It uses three deep neural networks to choose the robot mobility and relay policy with reduced sample complexity,so as to avoid exploring dangerous policies that lead to the high outage probability of the UAV messages. The network architecture of the three networks is designed with fully connected layers instead of convolutional layers to reduce the computational complexity, which is analyzed by theoretical analyses. We provide the performance bound of the proposed schemes in terms of the bit error rate, robot energy consumption and utility based on a game-theoretic study.Simulation results show that the performance of our proposed relay schemes, including the bit error rate, the outage probability, and the robot energy consumption outperforms the existing schemes.Xiaozhen LU Jingfang JIE Zihan LIN Liang XIAO Jin LI Yanyong ZHANG 2022Science China(Information Sciences)2022,65,1:1
