4DELLA proteins interact with FLC to repress flowering transition显示文摘Flowering is a highly orchestrated and extremely ct critical process in a plant's life cycle. Previous study has ademonstrated that SUPPRESSOR OF OVEREXPRESSION OF pCONSTANS 1(SOC1) and FLOWERING LOCUS T(FT) integrate m-I the gibberellic acid(GA) signaling pathway and vernalization higpathway in regulating flowering time, but detailed molecular Hmechanisms remain largely unclear. In GA signaling pathway,DELLA proteins are a group of master transcriptional regulators, while in vernalization pathway FLOWERING LOCUS C(FLC) is a core transcriptional repressor that down-regulates the expression of SOC1 and FT. Here, we report that DELLA proteins interact with FLC in vitro and in vivo, and the LHRI domains of DELLAs and the C-terminus of MADS domain of FLC are required for these interactions.Phenotypic and gene expression analysis showed that mutation of FLC reduces while over-expression of FLC enhances the GA response in the flowering process. Further,DELLA-FLC interactions promote the repression ability of FLC on its target genes. 6Perception of people for the risk of Tianwan nuclear power plant显示文摘A questionnaire survey of residents’risk perceptions related to Taiwan nuclear power plant in China was carried out to explore the determining factors that affect individual risk perception.This study proposed to pursue a more comprehensive understanding of factors that affected individual risk perception to nuclear power plants.Covariance structure analysis was conducted using risk perceptions of nuclear power as dependent variable and including interest and knowledge levels of nuclear power,acceptability,benefit perception,trust in nuclear power operation,and trust in government as independent variables.The use of the hypothesis of Elaboration likelihood model(ELM)was also proposed.The results showed that persons with higher levels of interest and knowledge of nuclear power had their own perceptions of risk closely associated with acceptability and potential benefits of nuclear power.In contrast,persons with no interest in and knowledge of nuclear power would have risk perceptions related to their trust in nuclear operation and the government,which partially supported the ELM hypothesis.All these results indicated that the government in China plays an important role in rational risk perceptions,and well-designed communication of risks will help the public to be involved in risk management and improve people’s rational acceptance of risk.Lei HUANG Jun BI Bing ZHANG Fengying LI Changsheng QU 2010Frontiers of Environmental Science & Engineering2010,4,1:6
7The effects of modulation period,modulation ratio,and deposition temperature on microstructure and mechanical properties of ZrB_2/W multilayers显示文摘Monolithic ZrB2,W coatings and ZrB2/W multilayers with different modulation periods and modulation ratios were synthesized by ion beam assisted deposition at room temperature and 400°C.X-ray diffraction (XRD),scanning electron microscopy (SEM),surface profiler,and nanoindention were employed to investigate the influences of the deposition temperature and the modulation period on the growth,textures,interface structure,and mechanical properties of the multilayers.The results indicated that the multilayer with modulation period of 13 nm synthesized at room temperature possessed a higher hardness of 23.8 GPa.Deposition temperature gave a significant contribution to mechanical property enhancement.The 400°C-deposition temperature led to a maximum hardness and elastic modulus value of 32.1 and 399.1 GPa for ZrB2/W multilayer with a modulation period of 6.7 nm.Its critical load increased to 42.8 mN and residual stress decreased to -0.7 GPa.A higher deposition temperature can cause an increase in interfacial atomic mixture and mobility of surface species,which induceds an increase in areal atomic density and dislocation pinning.These results as well as small nanoscale grain sizes should be related to hardness increase.LIU MengYin,TAN Ming,LIU GuangQing,WANG HaiYuan,XUE FengYing,DENG XiangYun & LI DeJun College of Physics and Electronic Information Science,Tianjin Normal University,Tianjin 300387,China 2010Science China(Technological Sciences)2010,53,9:4

10Berberine mitigates hepatic insulin resistance by enhancing mitochondrial architecture via the SIRT1/Opa1 signalling pathway显示文摘The aberrant changes of fussion/fission-related proteins can trigger mitochondrial dynamics imbalance,which cause mitochondrial dysfunctions and result insulin resistance(IR).However,the relationship between the inner mitochondrial membrane fusion protein optic atrophy 1(Opa1)and hepatic IR as well as the specific molecular mechanisms of signal transduction has not been fully elucidated.In this study,we explore whether abnormalities in the Opa1 cause hepatic IR and whether berberine(BBR)can prevent hepatic IR through the SIRT1/Opa1 signalling pathway.High-fat diet(HFD)-fed mice and db/db mice are used as animal models to study hepatic IR in vivo.IR,morphological changes,and mitochondrial injury of the liver are examined to explore the effects of BBR.SIRT1/Opa1 protein expression is determined to confirm whether the signalling pathway is damaged in the model animals and is involved in BBR treatment-mediated mitigation of hepatic IR.A palmitate(PA)-induced hepatocyte IR model is established in HepG2 cells in vitro.Opa1 silencing and SIRT1 overexpression are induced to verify whether Opa1 deficiency causes hepatocyte IR and whether SIRT1 improves this dysfunction.BBR treatment and SIRT1 silencing are employed to confirm that BBR can prevent hepatic IR by activating the SIRT1/Opa1 signalling pathway.Western blot analysis and JC-1 fluorescent staining results show that Opa1 deficiency causes an imbalance in mitochondrial fusion/fission and impairs insulin signalling in HepG2 cells.SIRT1 and BBR overexpression ameliorates PA-induced IR,increases Opa1,and improves mitochondrial function.SIRT1 silencing partly reverses the effects of BBR on HepG2 cells.SIRT1 and Opa1 expressions are downregulated in the animal models.BBR attenuates hepatic IR and enhances SIRT1/Opa1 signalling in db/db mice.In summary,Opa1 silencing-mediated mitochondrial fusion/fission imbalance could lead to hepatocyte IR.BBR may improve hepatic IR by regulating the SIRT1/Opa1 signalling pathway,and thus,it may be used to treat type-2 diabetes.Jia Xu Yining Zhang Zhiyi Yu Yueqi Guan Yuqian Lv Meishuang Zhang Ming Zhang Li Chen Xiaoyan Lv Fengying Guan 2022Acta Biochimica et Biophysica Sinica2022,54,10:3

12Formamidinium Lead Bromide (FAPbBr_3) Perovskite Microcrystals for Sensitive and Fast Photodetectors显示文摘Because of the good thermal stability and superior carrier transport characteristics of formamidinium lead trihalide perovskite HC(NH_2)_2 PbX_3(FAPbX_3), it has been considered to be a better optoelectronic material than conventional CH_3NH_3-PbX_3(MAPbX_3). Herein, we fabricated a FAPbBr_3 microcrystal-based photodetector that exhibited a good responsivity of 4000 A W-1 and external quantum efficiency up to 106% under one-photon excitation, corresponding to the detectivity greater than 1014 Jones. The responsivity is two orders of magnitude higher than that of previously reported formamidinium perovskite photodetectors. Furthermore, the FAPbBr_3 photodetector's responsivity to two-photon absorption with an 800-nm excitation source can reach 0.07 A W^(-1), which is four orders of magnitude higher than that of its MAPbBr_3 counterparts. 16Numerical Study on Seasonal Transportation of the Suspended Sediments in the Modern Yellow River Mouth Effected by the Artificial Water and Sediment Regulation显示文摘Since 2002, an artificial water and sediment regulation(AWSR) has been carried out, which largely reduced water and sediment discharged from the Yellow River into the Bohai Sea. Although the sediment transport in the Yellow River Mouth(YRM) has been observed and modeled intensively since AWSR, but preferentially for the non-storm conditions. In this study, a three-dimensional current-wave-sediment coupled model, DHI-MIKE numerical model, was used to examine the seasonal suspended-sediment transport in the YRM after the AWSR. Results show that the seasonal distribution of suspended-sediments in the YRM is dominated by wind and wave rather than river input. The major transport pathway of suspended-sediments is from the western Laizhou Bay to the Bohai Strait during the winter monsoon, especially in storm events. In addition, about 66% of the river sediments deposit within 30 km of the YRM, which is smaller than previous estimations. This review describes the advances of SFC in targeted and untargeted lipid profiling. The advancement of the SFC instruments and the stationary phases are summarized. Typical applications of SFC to the targeted and untargeted lipid profiling are discussed in detail. Moreover, the perspectives of SFC in the lipid profiling are also proposed. 19Efficacy of ICIs on patients with oncogene-driven non-small cell lung cancer:a retrospective study显示文摘Aim:The objective of our study was to assess the efficacy of immune checkpoint inhibitors(ICIs)on patients with non-small-cell lung cancer(NSCLC)harboring oncogenic alterations.Methods:We retrospectively enrolled patients with advanced non-squamous NSCLC who were treated with anti-PD-1-based monotherapy or combined immunotherapy.Major characteristics including PD-L1 expression,treatment,and survival were analyzed.Results:In total,309 non-squamous NSCLC patients with a median age of 61 years(range 20-88 years)including 70.9%male were retrospectively enrolled.The molecular alterations involved epidermal growth factor receptor(EGFR)(n=81),V-Ki-ras2 Kirsten rat sarcoma viral oncogene homolog(KRAS)(n=31),anaplastic lymphoma kinase(ALK)(n=1),human epidermal growth factor receptor 2(HER2)(n=12),V-raf murine sarcoma viral oncogene homolog(BRAF)(n=2),rearranged during transfection(n=4),and c-ros oncogene 1(ROS1)(n=3).In the EGFR subset,the ORR was 30.9%(n=81)and PFS was significantly shorter than WT group(median PFS:5.7 months vs.7.1 months;P=0.0061).In subgroup analyses,ICI combined therapy was significantly correlated with a longer PFS compared with ICI monotherapy(median PFS:7.7 months vs.4.7 months;P=0.0112).In KRAS patients,ORR was 51.6%(n=31).No significant difference was found in subgroup analyses.The ORR and PFS were 16.7%(n=12)and 28.6%(n=7),7.8 months and 9.0 months for HER2 and EGFR Exon20 insertion patients,respectively.Three ROS1 patients were enrolled with a PFS of 16.0,34.2,and 45.0 months individually,and one ALK patient with PFS of 4.4 months was identified.No response was found in two BRAF patients.Conclusion:ICI-based combination therapy can bring benefit to patients with EGFR-mutant NSCLC.ICI-based combination therapy could be considered for patients with ROS1 rearrangement,HER2 mutation and EGFR Exon20 insertion NSCLC.Xiaojin Guo He Du Jiayu Li Menghang Yang Anweng Xiong Haiping Zhang Fengying Wu 2022Cancer Drug Resistance2022,5,1:2
20PC4 serves as a negative regulator of skin wound healing in mice显示文摘Background:Human positive cofactor 4(PC4)was initially characterized as a multifunctional transcriptional cofactor,but its role in skin wound healing is still unclear.The purpose of this study was to explore the role of PC4 in skin wound healing through PC4 knock-in mouse model.Methods:A PC4 knock-in mouse model(PC4^(+/+))with a dorsal full-thickness wound was used to investigate the biological functions of PC4 in skin wound healing.Quantitative PCR,Western blot analysis and immunohistochemistry were performed to evaluate the expression of PC4;Sirius red staining and immunofluorescence were performed to explore the change of collagen deposition and angiogenesis.Proliferation and apoptosis were detected using Ki67 staining and TUNEL assay.Primary dermal fibroblasts were isolated from mouse skin to perform cell scratch experiments,cck-8 assay and colony formation assay.Results:The PC4^(+/+)mice were fertile and did not display overt abnormalities but showed an obvious delay in cutaneous healing of dorsal skin.Histological staining showed insufficient re-epithelialization,decreased angiogenesis and collagen deposition,increased apoptosis and decreased cell proliferation in PC4^(+/+)skin.Our data also showed decreased migration rate and proliferation ability in cultured primary fibroblasts from PC4^(+/+)mice in vitro.Conclusions:This study suggests that PC4 might serve as a negative regulator of skin wound healing in mice.Fengying Liao Long Chen Peng Luo Zhongyong Jiang Zelin Chen Ziwen Wang Chi Zhang Yu Wang JintaoHe Qing Wang Yawei Wang Lang Liu Yu Huang Huilan Wang Qingzhi Jiang Min Luo YiboGan Yunsheng Liu Yang Wang Jie Wu Wentao Xie Zhuo Cheng Yali Dai Jialun Li Zujuan Liu Fan Yang Chunmeng Shi 2020Burns & Trauma2020,8,1:1
