| 1 | Cloning of 9-cis-epoxycarotenoid dioxygenase (NCED) gene encoding a key enzyme during abscisic acid (ABA) biosynthesis and ABA-regulated ethylene production in detached young persimmon calyx显示文摘Unlike the typical climacteric fruits, persimmons (Diospyros kaki Thunb.) produce higher levels of ethylene when they are detached from trees at a younger stage. In order to obtain detailed information on the role of abscisic acid (ABA) in ripening, we cloned the DKNCED1, DKACS2, and DKACO1 genes from the calyx. Water loss was first noted in the calyx lobe, and DKNCED1 was highly expressed 1 d after the fruits were detached, coinciding with an increase in the ABA content. Then, the DKACS2 and DKACO1 genes were expressed after some delay. In the calyx, the ABA peak was observed 2 d after the fruits were harvested, and this peak preceded the ethylene peak observed on day 3. The fruit firmness rapidly decreased on day 4, and the fruits softened completely 6 d after they were harvested. The increases in the expressions of ABA, ethylene, and the genes in the calyxes occurred earlier than the corresponding increases in the pulp, although the 3 increases occurred on different days. Exogenous ABA treatment increased ABA concentration, induced expression of both ACS and ACO, and promoted ethylene synthesis and young-fruit softening; by contrast, treatment with NDGA inhibited the gene expressions and ethylene synthesis and delayed young-fruit softening. These results indicate that ethylene biosynthesis in the detached young persimmon fruits is initially triggered by ABA, which is induced by water loss in the calyx, through the induction of DKACS2 and DKACO1 expressions. | 3 | A remote sensing monitoring method for alpine grasslands desertification in the eastern Qinghai–Tibetan Plateau显示文摘Alpine grassland is the typical vegetation in the eastern Qinghai–Tibetan Plateau,which has important ecological service functions,and also supports the development of alpine stock farming.In recent years,under both the natural and human disturbance,alpine grasslands in this area have appeared to different degrees of desertification.A diagnosis of the desertification degree serves as the basis for grassland ecological restoration.This study constructs a comprehensive index based on remote sensing called alpine grassland desertification index(AGDI)to monitor the areas and degree of desertification.The most relevant indicators of desertification,namely,vegetation fraction,aboveground biomass,soil moisture,and land surface temperature,were selected to establish AGDI.The geographical detector is used to reselect and assess these indicators.The results show that the overall verification accuracy of AGDI is 82.05%.In particular,the accuracy of identifying severe desertification is the highest.Our study confirms that the desertification of alpine grasslands in the eastern Qinghai–Tibetan Plateau is characterized by fragmentation.Thus,Landsat-8 OLI data with a spatial resolution of 30 m is more suitable than MODIS data for alpine grasslands desertification monitoring.The research results can provide a methodological reference for monitoring desertification of alpine grasslands and other grassland regions in the world. | KUANG Qian YUAN Quan-zhi HAN Ji-chong LENG Rong WANG Yu-shuang ZHU Ke-hong LIN Shuo REN Ping | 2020 | Journal of Mountain Science2020,17,6: | 4 |

| 6 | COVID-19-associated monocytic encephalitis(CAME):histological and proteomic evidence from autopsy显示文摘Severe neurological symptoms are associated with Coronavirus disease 2019(COVID-19).However,the morphologic features,pathological nature and their potential mechanisms in patient brains have not been revealed despite evidence of neurotropic infection.In this study,neuropathological damages and infiltrating inflammatory cells were quantitatively evaluated by immunohistochemical staining,ultrastructural examination under electron microscopy,and an image threshold method,in postmortem brains from nine critically ill COVID-19 patients and nine age-matched cadavers of healthy individuals.Differentially expressed proteins were identified by quantitative proteomic assays.Histopathological findings included neurophagocytosis,microglia nodules,satellite phenomena,extensive edema,focal hemorrhage,and infarction,as well as infiltrating mononuclear cells.Immunostaining of COVID-19 brains revealed extensive activation of both microglia and astrocytes,severe damage of the blood-brain barrier(BBB)and various degrees of perivascular infiltration by predominantly CD14+/CD16+/CD141+/CCR7+/CD11c+monocytes and occasionally CD4+/CD8+T lymphocytes.Quantitative proteomic assays combined with bioinformatics analysis identified upregulated proteins predominantly involved in immune responses,autophagy and cellular metabolism in COVID-19 patient brains compared with control brains.Proteins involved in brain development,neuroprotection,and extracellular matrix proteins of the basement membrane were downregulated,potentially caused by the activation of transforming growth factorβreceptor and vascular endothelial growth factor signaling pathways.Thus,our results define histopathological and molecular profiles of COVID-19-associated monocytic encephalitis(CAME)and suggest potential therapeutic targets. | Pei-Pei Zhang Zhi-Cheng He Xiao-Hong Yao Rui Tang Jie Ma Tao Luo Chuhong Zhu Tian-Ran Li Xindong Liu Dingyu Zhang Shuyang Zhang Yi-Fang Ping Ling Leng Xiu-Wu Bian | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 3 |

It was found that catalytic ozonization in the presence of Mn-Zr-O (a modified manganese dioxide supported on silica gel) had significantly enhanced the removal rate (72%) of total organic carbon (TOC) compared with that of ozonization alone (19%).
