9NMDA Receptor Antagonist MK801 Protects Against 1-Bromopropane-Induced Cognitive Dysfunction显示文摘Occupational exposure to 1-bromopropane(1-BP) induces learning and memory deficits. However, no therapeutic strategies are currently available. Accumulating evidence has suggested that N-methyl-D-aspartate receptors(NMDARs) and neuroinflammation are involved in the cognitive impairments in neurodegenerative diseases. In this study we aimed to investigate whether the noncompetitive NMDAR antagonist MK801 protects against 1-BPinduced cognitive dysfunction. Male Wistar rats were administered with MK801(0.1 mg/kg) prior to 1-BP intoxication(800 mg/kg). Their cognitive performance was evaluated by the Morris water maze test. The brains of rats were dissected for biochemical, neuropathological,and immunological analyses. We found that the spatial learning and memory were significantly impaired in the1-BP group, and this was associated with neurodegeneration in both the hippocampus(especially CA1 and CA3)and cortex. Besides, the protein levels of phosphorylated NMDARs were increased after 1-BP exposure. MK801 ameliorated the 1-BP-induced cognitive impairments and degeneration of neurons in the hippocampus and cortex.Mechanistically, MK801 abrogated the 1-BP-induced disruption of excitatory and inhibitory amino-acid balance and NMDAR abnormalities. Subsequently, MK801 inhibited the microglial activation and release of pro-inflammatory cytokines in 1-BP-treated rats. 10Curcuminoids Target Decreasing Serum Adipocyte-fatty Acid Binding Protein Levels in Their Glucose-lowering Effect in Patients with Type 2 Diabetes显示文摘Whether supplementation of curcuminoids decreases serum adipocyte-fatty acid binding protein(A-FABP) level and whether this decrease benefits glucose control is unclear.One-hundred participants(n=50 administered curcuminoids,n=50 administered placebo) from our previous report on the effect of curcuminoids on type 2 diabetes in a 3-month intervention were assessed for levels of serum A-FABP,oxidative stress,and inflammatory biomarkers.Curcuminoids supplementation led to significant decreases in serum A-FABP,C-reactive protein(CRP),tumor necrosis factor-α,and interleukin-6 levels.Curcuminoids supplementation also significantly increased serum superoxide dismutase(SOD) activity.The change in serum A-FABP levels showed positive correlations with changes in levels of glucose,free fatty acids(FFAs),and CRP in subjects supplemented with curcuminoids.Further stepwise regression analysis showed that A-FABP was an independent predictor for levels of FFAs,SOD,and CRP.These results suggest that curcuminoids may exert anti-diabetic effects,at least in part,by reductions in serum A-FABP level.A-FABP reduction is associated with improved metabolic parameters in human type 2 diabetes.NA Li Xin YAN Bo Lin JIANG Shuo CUI Hong Li LI Ying SUN Chang Hao 2014Biomedical and Environmental Sciences2014,27,11:6
11Optimal power allocation for secure directional modulation networks with a full-duplex UAV user显示文摘This paper makes an investigation of a secure unmanned aerial vehicle(UAV)-aided communication network based on directional modulation(DM). In this network, ground base station(GBS) acts as a control center to transmit confidential message and artificial noise(AN). The UAV user, moving along a linear flight trajectory, is intended to receive the useful information from GBS. At the same time, it also sends AN signals to further interference eavesdropper’s channel. Aiming at maximizing secrecy rate during the UAV flight process, a joint optimization problem is formulated with respect to power allocation(PA)factors, beamforming vector and AN projection matrices. For simplicity, maximum rate transmission, nullspace projection and the leakage-based method are applied to form the transmit beamforming vector, AN projection matrix at GBS, and AN projection vector at UAV user, respectively. Following this, the optimization problem reduces to a bivariate optimization programme with two PA factors. An alternating iterative algorithm(AIA) is proposed to optimize the two PA factors. 13Promoter methylation of Wnt/β-Catenin signal inhibitor TMEM88 is associated with unfavorable prognosis of nonsmall cell lung cancer显示文摘Objective:Recent research has indicated that altered promoter methylation of oncogenes and tumor suppressor genes is an important mechanism in lung cancer development and progression.In this study,we investigated the association between promoter methylation of TMEM88,a possible inhibitor of the Wnt/β-Catenin signaling,and the survival of patients with nonsmall cell lung cancer(NSCLC).Methods:Twelve pairs of tumor and adjacent non-tumor samples were used for microarray analyses of DNA methylation and gene expression.For validation,more than two hundred additional samples were analyzed for methylation using bisulfite pyrosequencing and for gene expression using q RT-PCR.Then the cell function were tested by wound healing,transwell,CCK8 and cell cycle assay.Results:Our analysis of patient specimens showed that TMEM88 methylation was higher in NSCLC tumors(82.2%±10.3,P<0.01)compared with the adjacent normal tissues(65.9%±7.2).The survival analysis revealed that patients with high TMEM88methylation had a shorter overall survival(46 months)compared with patients with low TMEM88 methylation(>56 months;P=0.021).In addition,we found that demethylation treatment could inhibit tumor cell proliferation,migration,and invasion,which was supportive of an association between methylation and survival.Conclusions:Based on these consistent observations,we concluded that TMEM88 may play an important role in NSCLC progression and that promoter methylation of TMEM88 may serve as a biomarker for NSCLC prognosis and treatment.Rongna Ma Nannan Feng Xiao Yu Hongyan Lin Xiaohong Zhang Oumin Shi Huan Zhang Shuo Zhang Lei Li Min Zheng Ming Gao Herbert Yu Biyun Qian 2017Cancer Biology & Medicine2017,14,4:6

15Fatigue Crack Growth Rate of Ti-6Al-4V Considering the Effects of Fracture Toughness and Crack Closure显示文摘Fatigue fracture is one of the main failure modes of Ti-6A1-4V alloy,fracture toughness and crack closure have strong effects on the fatigue crack growth(FCG)rate of Ti-6A1-4V alloy.The FCG rate of Ti-6A1-4V is investigated by using experimental and analytical methods.The effects of stress ratio,crack closure and fracture toughness on the FCG rate are studied and discussed.A modified prediction model of the FCG rate is proposed,and the relationship between the fracture toughness and the stress intensity factor(SIF)range is redefined by introducing a correcting coefficient.Notched plate fatigue tests(including the fracture toughness test and the FCG rate test)are conducted to investigate the influence of affecting factors on the FCG rate.Comparisons between the predicted results of the proposed model,the Paris model,the Walker model,the Sadananda model,and the experimental data show that the proposed model gives the best agreement with the test data particularly in the near-threshold region and the Paris region,and the corresponding calculated fatigue life is also accurate in the same regions.By considering the effects of fracture toughness and crack closure,the novel FCG rate prediction model not only improves the estimating accuracy,but also extends the adaptability of the FCG rate prediction model in engineering.ZHANG Junhong YANG Shuo LIN Jiewei 2015Chinese Journal of Mechanical Engineering2015,28,2:5
16Tetramethylpyrazine prevents liver fibrotic injury in mice by targeting hepatocyte-derived and mitochondrial DNA-enriched extracellular vesicles显示文摘Liver fibrosis is the common consequence of almost all liver diseases and has become an urgent clinical problem without efficient therapies.Recent evidence has shown that hepatocytes-derived extracellular vesicles(EVs)play important roles in liver pathophysiology,but little is known about the role of damaged hepatocytes-derived EVs in hepatic stellate cell(HSC)activation and following fibrosis.Tetramethylpyrazine(TMP)from Ligusticum wallichii Franchat exhibits a broad spectrum of biological activities including liver protection.In this study,we investigated whether TMP exerted liver-protective action through regulating EV-dependent intercellular communication between hepatocytes and HSCs.Chronic liver injury was induced in mice by CCl_(4)(1.6 mg/kg,i.g.)twice a week for 8 weeks.In the last 4 weeks of CCl_(4) administration,mice were given TMP(40,80,160 mg·kg^(-1)·d^(-1),i.g.).Acute liver injury was induced in mice by injection of a single dose of CCl_(4)(0.8 mg/kg,i.p.).After injection,mice were treated with TMP(80 mg/kg)every 24 h.We showed that TMP treatment dramatically ameliorated CCl_(4)-induced oxidative stress and hepatic inflammation as well as acute or chronic liver fibrosis.In cultured mouse primary hepatocytes(MPHs),treatment with CCl_(4) or acetaminophen resulted in mitochondrial dysfunction,release of mitochondrial DNA(mtDNA)from injured hepatocytes to adjacent hepatocytes and HSCs through EVs,mediating hepatocyte damage and fibrogenic responses in activated HSCs;pretreatment of MPHs with TMP(25μM)prevented all these pathological effects.Transplanted serum EVs from TMP-treated mice prevented both initiation and progression of liver fibrosis caused by CCl_(4).Taken together,this study unravels the complex mechanisms underlying the protective effects of TMP against mtDNA-containing EV-mediated hepatocyte injury and HSC activation during liver injury,and provides critical evidence inspiring the development of TMP-based innovative therapeutic agents for the treatment of liver fibrosis.Ya-jing Li Run-ping Liu Ming-ning Ding Qi Zheng Jian-zhi Wu Xiao-yong Xue Yi-qing Gu Bo-ning Ma Ya-jie Cai Shuo Li Sheng Lin Lu-yong Zhang Xiaojiaoyang Li 2022Acta Pharmacologica Sinica2022,43,8:5

With a lot of advancement in the field, yet the biological cause of metastasis are poorly understood. The microfluidic system provides advanced technology to reconstruct a variety of in vivo-like environment for studying the interactions between tumor cells(TCs) and endothelial cells(ECs). This review gives a brief account of both two-dimensional models and three-dimensional microfluidic systems for the analysis of TCs-ECs coculture as well as their applications to anti-cancer drug screening. 19Breast cancer stem-like cells can promote metastasis by activating platelets and down-regulating antitumor activity of natural killer cells显示文摘OBJECTIVE:To investigate whether cancer stem cells(CSCs) more efficiently activating platelets and evading immune surveillance than non-CSCs thus promoting metastasis.METHODS:We enriched and identified sphereforming cells(SFCs) and coincubated washed platelets with several platelet activators including collagen,4T1 and SFCs.Platelet-coating tumor cells,platelet activation and TGF-β1 release were analyzed.Then natural kell cells(NK) were incubated with supernatants of different activated platelet samples what we called sample release(SR).The degranulation assay and NKG2 D expression on NK cells were conducted by flow cytometry.Finally tissue factor(TF) expression of SFCs or 4T1 were evaluated by western blot.RESULTS:Breast cancer cell line 4T1 could form spheres in serum-free medium at low adherence.Sphere-forming cells expressed high levels of the CD24-/lowCD44 + stem cell phenotype.Both sphere-forming cells or 4T1 were coated with abundant platelets while sphere-forming cells induced significantly higher expression of platelet activating receptor CD62 p than 4T1 did(P < 0.01).And sphere-forming cells induced platelets to produce more TGF-β1 than 4T1 did(P < 0.01).Furthermore,sample releases induced by sphere-forming cells caused more vigorous inhibition of NK cells antitumor reactivity(P < 0.05) and reduced NKG2 D expression(P < 0.01).The final results showed that sphere-forming cells expressed higher levels of TF than 4T1(P < 0.05).CONCLUSION:Our findings indicate that CSCs could efficiently activate platelets,induce platelets to secrete more TGF-β1,decrease NKG2 D expression and inhibit antitumor activity of NK cell,compared with 4T1.And higher levels of TF expression of CSCs may account for this correlation of CSCs and platelets.Wang Shuo Zhang Ying Cong Weihong Liu Jie Zhang Yuren Fan Huiting XuYonggang Lin Hongsheng 2016Journal of Traditional Chinese Medicine2016,36,4:5
20Fabrication of virus-like particles with strip-pattern surface:A two-step self-assembly approach显示文摘Spherical nanostructures with striped patterns on the surfaces resembling the essential structures of natural virus particles were constructed through a two-step self-assembly approach of polystyrene-boligo(acrylic acid)(PS-b-oligo-AA) and poly(γ-benzyI L-glutamate)-b-poly(ethylene glycol)(PBLG-bPEG) copolymer mixtures in solution.On the basis of difference in hydrophilicity and self-assembly properties of the two copolymers,the two-step self-assembly process is realized.It was found that PS-boligo-AA copolymers formed spherical aggregates by adding a certain amount of water into polymer solutions in the first step.In the second step,two polymer solutions were mixed and water was further added,inducing the self-assembly of PBLG-b-PEG on the surfaces of PS-b-oligo-AA spheres to form striped patterns.In-depth study was conducted for the indispensable defects of striped patterns which are dislocations and +1/2 disclinations.The influencing factors such as the mixing ratio of two copolymers and the added water content in the first step on the morphology and defects of the striped patterns were investigated.This work not only presents an idea to interpret mechanism of the cooperative self-assembly behavior,but also provides an effective approach to construct virus-like particles and other complex structures with controllable morphology.Shuo Zhang Chun-Hua Cai Zhou Guan Jia-Ping Lin Xing-Yu Zhu 2017Chinese Chemical Letters2017,28,4:5
