2Plecanatide and dolcanatide, novel guanylate cyclase-C agonists, ameliorate gastrointestinal inflammation in experimental models of murine colitis显示文摘AIM: To evaluate the effect of orally administeredplecanatide or dolcanatide, analogs of uroguanylin, on amelioration of colitis in murine models.METHODS: The cyclic guanosine monophosphate(cG MP) stimulatory potency of plecanatide and dolcanatide was measured using a human colon carcinoma T84 cellbased assay. For animal studies all test agents were formulated in phosphate buffered saline. Sulfasalazine or 5-amino salicylic acid(5-ASA) served as positive controls. Effect of oral treatment with test agents on amelioration of acute colitis induced either by dextran sulfate sodium(DSS) in drinking water or by rectal instillation of trinitrobenzene sulfonic(TNBS) acid, was examined in BALB/c and/or BDF1 mice. Additionally, the effect of orally administered plecanatide on the spontaneous colitis in T-cell receptor alpha knockout(TCRα-/-) mice was also examined. Amelioration of colitis was assessed by monitoring severity of colitis, disease activity index and by histopathology. Frozen colon tissues were used to measure myeloperoxidase activity.RESULTS: Plecanatide and dolcanatide are structurally related analogs of uroguanylin, which is an endogenous ligand of guanylate cyclase-C(GC-C). As expected from the agonists of GC-C, both plecanatide and dolcanatide exhibited potent cG MP-stimulatory activity in T84 cells. Once-daily treatment by oral gavage with either of these analogs(0.05-0.5 mg/kg) ameliorated colitis in both DSS and TNBS-induced models of acute colitis, as assessed by body weight, reduction in colitis severity(P < 0.05) and disease activity index(P < 0.05). Amelioration of colitis by either of the drug candidates was comparable to that achieved by orally administered sulfasalazine or 5-ASA. Plecanatide also effectively ameliorated colitis in TCRα-/- mice, a model of spontaneous colitis. As dolcanatide exhibited higher resistance to proteolysis in simulated gastric and intestinal juices, it was selected for further studies. 3原发性肾平滑肌肉瘤13例临床病理分析显示文摘目的分析13例肾脏平滑肌肉瘤患者的生存情况,评估预后相关因素。方法回顾性分析1996年至2008年间我院诊治的13例肾脏平滑肌肉瘤患者的临床症状、病理学特征、治疗方式及随访资料,运用logistic回归评估各项观察指标与预后的相关性,分析各指标对预后的影响。结果患者平均年龄为45.7岁,61.5%(8/13)为女性。平均肿瘤直径为〔11.1±10.2(3.0~34.0)〕cm,53.9%(7/13)累及肾包膜,23.1%(3/13)淋巴结阳性,完成随访11例,中位生存时间为39.2月(9~81月),1年、3年、5年生存率分别为81.8%,54.5%、16.2%。8例患者出现远处转移,截止本次随访9例患者死亡。相关性分析显示:患者的生存与年龄、病理分级、是否完整切除、是否淋巴结转移、是否侵犯包膜或肾盂、是否化疗等无相关性。结论原发性肾脏平滑肌肉瘤为高致死率的肾脏恶性肿瘤,需尽早发现、手术切除,否则生存预后差。熊子兵 石明 Kunwar Ashok 陈铌 张朋 曾浩 李响 2012四川大学学报(医学版)2012,43,1:5
4Plecanatide-mediated activation of guanylate cyclase-C suppresses inflammation-induced colorectal carcinogenesis in Apc+/Min-FCCC mice显示文摘AIM To evaluate the effect of orally administered plecanatide on colorectal dysplasia in Apc^(+/Min-FCCC) mice with dextran sodium sulfate(DSS)-induced inflammation. METHODS Inflammation driven colorectal carcinogenesis was induced in Apc^(+/Min-FCCC) mice by administering DSS in their drinking water. Mice were fed a diet supplemented with plecanatide(0-20 ppm) and its effect on the multiplicity of histopathologically confirmed polypoid,flat and indeterminate dysplasia was evaluated. Plecanatide-mediated activation of guanylate cyclase-C(GC-C) signaling was assessed in colon tissues by measuring cyclic guanosine monophosphate(cG MP) by ELISA, protein kinase G-II and vasodilator stimulated phosphoprotein by immunoblotting. Ki-67, c-myc and cyclin D1 were used as markers of proliferation. Cellular levels and localization of b-catenin in colon tissues were assessed by immunoblotting and immunohistochemistry, respectively. Uroguanylin(UG) and GC-C transcript levels were measured by quantitative reverse transcription polymerase chain reaction(RT-PCR). A mouse cytokine array panel was used to detect cytokines in the supernatant of colon explant cultures. RESULTS Oral treatment of Apc^(+/Min-FCCC) mice with plecanatide produced a statistically significant reduction in the formation of inflammation-driven polypoid, flat and indeterminate dysplasias. This anti-carcinogenic activity of plecanatide was accompanied by activation of cG MP/GC-C signaling mediated inhibition of Wnt/b-catenin signaling and reduced proliferation. Plecanatide also decreased secretion of pro-inflammatory cytokines(IL-6, IL-1 TNF), chemokines(MIP-1, IP-10) and growth factors(GCSF and GMCSF) from colon explants derived from mice with acute DSS-induced inflammation. The effect of plecanatidemediated inhibition of inflammation/dysplasia on endogenous expression of UG and GC-C transcripts was measured in intestinal tissues. Although GC-C expression was not altered appreciably, a statistically significant increase in the level of UG transcripts was detected in the proximal small intestine and colon, potentially due to a reduction in intestinal inflammation and/or neoplasia. 5Oral treatment with plecanatide or dolcanatide attenuates visceral hypersensitivity via activation of guanylate cyclase-C in rat models显示文摘AIM To investigate the effects of plecanatide and dolcanatide on maintenance of paracellular permeability, integrity of tight junctions and on suppression of visceral hypersensitivity. METHODS Transport of fluorescein isothiocyanate(FITC)-dextran was measured to assess permeability across cell monolayers and rat colon tissues. Effects of plecanatide and dolcanatide on the integrity of tight junctions in Caco-2 and T84 monolayers and on the expression and localization of occludin and zonula occludens-1(ZO-1) were examined by immunofluorescence microscopy. Anti-nociceptive activity of these agonists was evaluated in trinitrobenzene sulfonic acid(TNBS)-induced inflammatory as well as in non-inflammatory partial restraint stress(PRS) rat models. Statistical significance between the treatment groups in the permeability studies were evaluated using unpaired t-tests.RESULTS Treatment of T84 and Caco-2 monolayers with lipopolysaccharide(LPS) rapidly increased permeability, which was effectively suppressed when monolayers were also treated with plecanatide or dolcanatide. Similarly, when T84 and Caco-2 monolayers were treated with LPS, cell surface localization of tight junction proteins occludin and ZO-1 was severely disrupted. When cell monolayers were treated with LPS in the presence of plecanatide or dolcanatide, occludin and ZO-1 were localized at the cell surface of adjoining cells, similar to that observed for vehicle treated cells. Treatment of cell monolayers with plecanatide or dolcanatide without LPS did not alter permeability, integrity of tight junctions and cell surface localization of either of the tight junction proteins. In rat visceral hypersensitivity models, both agonists suppressed the TNBS-induced increase in abdominal contractions in response to colorectal distension without affecting the colonic wall elasticity, and both agonists also reduced colonic hypersensitivity in the PRS model. CONCLUSION Our results suggest that activation of GC-C signaling might be involved in maintenance of barrier function, possibly through regulating normal localization of tight junction proteins. Consistent with these findings, plecanatide and dolcanatide showed potent antinociceptive activity in rat visceral hypersensitivity models. 6Self-Assembled Al Nanostructure/ZnO Quantum Dot Heterostructures for High Responsivity and Fast UV Photodetector显示文摘Light confinement induced by spontaneous near-surface resonance is inherently determined by the location and geometry of metallic nanostructures(NSs),offering a facile and effective approach to break through the limitation of the light-mater interaction within the photoactive layers.Here,we demonstrate high-performance Al NS/ZnO quantum dots(Al/ZnO) heterostructure UV photodetectors with controllable morphologies of the self-assembled Al NSs.The Al/ZnO heterostructures exhibit a superior light utilization than the ZnO/Al heterostructures,and a strong morphological dependence of the Al NSs on the optical properties of the heterostructures.The inter-diffusion of Al atoms into ZnO matrixes is of a great benefit for the carrier transportation.Consequently,the optimal photocurrent of the Al/ZnO heterostructure photodetectors is significantly increased by 275 times to ~1.065 mA compared to that of the pristine ZnO device,and an outstanding photoresponsivity of 11.98 A W-1 is correspondingly achieved under 6.9 MW cm-2 UV light illumination at 10 V bias.In addition,a relatively fast response is similarly witnessed with the Al/ZnO devices,paving a path to fabricate the high-performance UV photodetectors for applications.Sisi Liu Ming‑Yu Li Jianbing Zhang Dong Su Zhen Huang Sundar Kunwar Jihoon Lee 2020Nano-Micro Letters2020,12,9:3
7A data-driven framework to predict the morphology of interfacial Cu6Sn5 IMC in SAC/Cu system during laser soldering显示文摘A data-driven approach combining together the experimental laser soldering,finite element analysis and machine learning,has been utilized to predict the morphology of interracial intermetallic compound(IMC) in Sn-xAg-yCu/Cu(SAC/Cu) system.Six types of SAC solders with varying weight proportion of Ag and Cu,have been processed with fiber laser at different magnitudes of power(30-50 W) and scan speed(10-240 mm/min),and the resultant IMC morphologies characterized through scanning electron microscope are categorized as prismatic and scalloped ones.For the different alloy composition and laser parameters,finite element method(FEM) is employed to compute the transient distribution of temperature at the interface of solder and substrates.The FEM-generated datasets are supplied to a neural network that predicts the IMC morphology through the quantified values of temperature dependent Jackson parameter(αJ).The numerical value of αJ predicted from neural network is validated with experimental IMC morphologies.The critical scan speed for the morphology transition between prismatic and scalloped IMC is estimated for each solder composition at a given power.Sn-0.7 Cu having the largest critical scan speed at 30 W and Sn-3.5 Ag alloy having the largest critical scan speed at input power values of 40 W and 50 W,thus possessing the greatest likelihood of forming prismatic interfacial IMC during laser soldering,can be inferred as most suitable SAC solders in applications exposed to shear loads.Anil Kunwar Lili An Jiahui Liu Shengyan Shang Peter Raback Haitao Ma Xueguan Song 2020Journal of Materials Science & Technology2020,47,15:3
8Integration of machine learning with phase field method to model the electromigration induced Cu_(6)Sn_(5) IMC growth at anode side Cu/Sn interface显示文摘Currently,in the era of big data and 5G communication technology,electromigration has become a serious reliability issue for the miniaturized solder joints used in microelectronic devices.Since the effective charge number(Z*)is considered as the driving force for electromigration,the lack of accurate experimental values for Z* poses severe challenges for the simulation-aided design of electronic materials.In this work,a data-driven framework is developed to predict the Z* values of Cu and Sn species at the anode based LIQUID,Cu_(6)Sn_(5) intermetallic compound(IMC)and FCC phases for the binary Cu-Sn system undergoing electromigration at 523.15 K.The growth rate constants(kem)of the anode IMC at several magnitudes of applied low current density(j=1×10^6 to 10×10^6A/m^2)are extracted from simulations based on a 1D multi-phase field model.A neural network employing Z* and j as input features,whereas utilizing these computed kemdata as the expected output is trained.The results of the neural network analysis are optimized with experimental growth rate constants to estimate the effective charge numbers.For a negligible increase in temperature at low j values,effective charge numbers of all phases are found to increase with current density and the increase is much more pronounced for the IMC phase.The predicted values of effective charge numbers Z* are then utilized in a 2D simulation to observe the anode IMC grain growth and electrical resistance changes in the multi-phase system.As the work consists of the aspects of experiments,theory,computation,and machine learning,it can be called the four paradigms approach for the study of electromigration in Pb-free solder.Such a combination of multiple paradigms of materials design can be problem-solving for any future research scenario that is marked by uncertainties regarding the determination of material properties.Anil Kunwar Yuri Amorim Coutinho Johan Hektor Haitao Ma Nele Moelans 2020Journal of Materials Science & Technology2020,59,24:3

10耻骨后顺行前列腺癌根治术与逆行根治术的围手术期情况和长期并发症比较显示文摘目的对比分析耻骨后顺行前列腺癌根治术与逆行根治切除术的围手术期情况和长期并发症。方法回顾性分析2006年4月至2011年5月接受开放性前列腺癌根治性切除术的76例患者的临床资料和随访结果,其中45例行耻骨后顺行前列腺癌根治术,31例行耻骨后逆行前列腺癌根治术,比较两组患者的围手术期情况和长期并发症发生率。结果 76例患者手术均获成功,无围手术期死亡病例,11例出现切缘阳性,顺行组5例,逆行组6例。术中出血(367.2±132.4)mL,其中顺行组出血(324.0±70.3)mL,逆行组出血(429.8±172.3)mL,差异具有统计学意义(P<0.05)。术后3例出现切口感染,顺行组1例(2.2%),逆行组2例(6.5%),两组间差异无统计学意义。随访2~63(32.56±18.25)月,无患者死于前列腺癌,4例生化复发。两组患者长期并发症(控尿情况、尿道狭窄和勃起功能障碍)发生率的差异无统计学意义。结论开放性前列腺癌根治术治疗局限性前列腺癌安全有效,顺行前列腺癌根治术较逆行前列腺癌根治术术中出血少。两种手术方式在围手术期情况和长期并发症上无明显差异。熊子兵 石明 Kunwar Ashok 陈铌 张朋 曾浩 李响 2012四川大学学报(医学版)2012,43,1:2

12Improved Photoresponse of UV Photodetectors by the Incorporation of Plasmonic Nanoparticles on Ga N Through the Resonant Coupling of Localized Surface Plasmon Resonance显示文摘Very small metallic nanostructures,i.e.,plasmonic nanoparticles(NPs),can demonstrate the localized surface plasmon resonance(LSPR)e ect,a characteristic of the strong light absorption,scattering and localized electromagnetic field via the collective oscillation of surface electrons upon on the excitation by the incident photons.The LSPR of plasmonic NPs can significantly improve the photoresponse of the photodetectors.In this work,significantly enhanced photoresponse of UV photodetectors is demonstrated by the incorporation of various plasmonic NPs in the detector architecture.Various size and elemental composition of monometallic Ag and Au NPs,as well as bimetallic alloy Ag Au NPs,are fabricated on Ga N(0001)by the solid-state dewetting approach.The photoresponse of various NPs are tailored based on the geometric and elemental evolution of NPs,resulting in the highly enhanced photoresponsivity of 112 A W-1,detectivity of 2.4×1012 Jones and external quantum e ciency of 3.6×104%with the high Ag percentage of Ag Au alloy NPs at a low bias of 0.1 V.The Ag Au alloy NP detector also demonstrates a fast photoresponse with the relatively short rise and fall time of less than 160 and 630 ms,respectively.The improved photoresponse with the Ag Au alloy NPs is correlated with the simultaneous e ect of strong plasmon absorption and scattering,increased injection of hot electrons into the Ga N conduction band and reduced barrier height at the alloy NPs/Ga N interface.Sundar Kunwar Sanchaya Pandit Jae-Hun Jeong Jihoon Lee 2020Nano-Micro Letters2020,12,7:2
