1Wheat powdery mildew resistance gene Pm64 derived from wild emmer (Triticum turgidum var.dicoccoides) is tightly linked in repulsion with stripe rust resistance gene Yr5显示文摘Stripe rust and powdery mildew are both devastating diseases for durum and common wheat.Pyramiding of genes conferring resistance to one or more diseases in a single cultivar is an important breeding approach to provide broader spectra of resistances in wheat improvement. A new powdery mildew resistance gene originating from wild emmer(Triticum turgidum var.dicoccoides) backcrossed into common wheat(T. aestivum) line WE35 was identified. It conferred an intermediate level of resistance to Blumeria graminis f. sp. tritici isolate E09 at the seedling stage and a high level of resistance at the adult plant stage. Genetic analysis showed that the powdery mildew resistance in WE35 was controlled by a dominant gene designated Pm64. Bulked segregant analysis(BSA) and molecular mapping indicated that Pm64 was located in chromosome bin 2 BL4-0.50–0.89. Polymorphic markers were developed from the corresponding genomic regions of Chinese Spring wheat and wild emmer accession Zavitan to delimit Pm64 to a 0.55 cM genetic interval between markers WGGBH1364 and WGGBH612, corresponding to a 15 Mb genomic region on Chinese Spring and Zavitan 2 BL, respectively. The genetic linkage map of Pm64 is critical for fine mapping and cloning. Pm64 was completely linked in repulsion with stripe rust resistance gene Yr5. 4Highly Sensitive Refractive Index Sensor Based on Polymer Long-Period Waveguide Grating With Liquid Cladding显示文摘We propose a novel structure and unique sensing mechanism bio-chemical sensor which is fabricated by a polymer long-period waveguide grating with the detection liquid directly as the waveguide cladding.Quantitative detection is realized from analyzing the output absorption spectrum and resonant wavelength shift related to the liquid detection concentration.The proposed polymer long-period waveguide grating based liquid refractive-index sensor is developed experimentally,the high sensitivity of 1.01×10^4nm/RIU is achieved,and the temperature stability coefficient is 1.47nm/℃.Theoretically and experimentally,this work has been demonstrated to have potential application in chemical and biological detections and may provide an important technical support for solving today's increasingly serious civil problems such as food safety and drug safety, which will also have the important scientific significance and application prospects.Lingfang WANG Keyu REN Bao SUN Kaixin CHEN 2019Photonic Sensors2019,9,1:1
5Anthropogenic pressures increase extinction risk of an isolated Asian elephant(Elephas maximus)population in southwestern China,as revealed by a combination of molecular-and landscape-scale approaches显示文摘Identification of the effect of anthropogenic threats on ecosystem is crucial.We used molecular tools and remote sensing to evaluate the population status of an isolated Asian elephant population in southwestern China in response to changes in habitat suitability between 1989 and 2019.A total of 22 unique genotypes were identified from 117 dung samples collected between March and June 2018 using microsatellite DNA analysis,including 13 males and 9 females.Based on the size of fecal boli,1 animal was a juvenile,9 were subadults,and 12 were adults,indicating that recruitment was limited.The effective population size was small(15.3)but there was no signature of a recent population bottleneck.We observed a low genetic diversity(He=0.46±0.05)and a high level of inbreeding(Fis of 0.43±0.11),suggesting low population viability and high risk of extinction.In total,these elephants lost nearly two thirds(62%)of their habitat in 3 decades.The expansion of agriculture and rubber plantations followed by an increase in human settlements after 1989 increased the isolation of this population.We recommend that resettlement of 800 inhabitants of 2 villages and the abandonment of associated farmland and rubber plantations would make an additional 20 km2 of suitable habitat available.This could allow a population increase of 14 elephants,possibly by translocating individuals from elsewhere in China.Ourfindings can be applied to the management and conservation of other fragmented populations in China or in other range countries of Asian elephants.Ying CHEN Yakuan SUN Luciano ATZENI Luke GIBSON Mei HUA Keyu LI Kun SHI David DUDGEON 2022Integrative Zoology2022,17,6:1
6Unveiling mitophagy-mediated molecular heterogeneity and development of a risk signature model for colorectal cancer by integrated scRNA-seq and bulk RNA-seq analysis显示文摘Background:Accumulating researchers have recognized mitophagy as a key player in tumors,but few studies have investigated its role in the tumor microenvironment(TME).Advances in the technology of single-cell RNA sequencing(scRNA-seq)have allowed unveiling the concealed features of the TME at cellular resolution.This study aimed to elucidate the role of mitophagy within the TME of colorectal cancer(CRC)and to establish a mitophagy-mediated risk model.Methods:We assessed mitophagy-related pathway activities at both single-cell and tissue levels.Subsequently,an unsupervised clustering algorithm was employed to identify mitophagy-mediated subtypes.Furthermore,we developed a mitophagy-mediated risk signature(MMRS)using least absolute shrinkage and selection operator(LASSO)Cox analysis and constructed a MMRS model incorporating the risk score and clinical variables.Subsequently,we used quantitative reverse transcription polymerase chain reaction analysis to verify the expression of the screened genes.Results:We retrieved and annotated a total of 14,719 cells from eight samples in the scRNA-seq GSE132465 data set.The activities of mitophagy-related pathways were uniformly upregulated in cancer cells.Integrating with bulk RNA-seq data,we identified two mitophagy-mediated clusters(C1 and C2)with distinct characteristics and prognoses.C2 was identified as a mitophagy-high cluster.Then,we developed a five-gene MMRS via LASSO Cox analysis in The Cancer Genome Atlas(TCGA)cohort.We utilized the GSE39582 cohort to validate the efficacy of our model.The expression of CX3CL1 and INHBB was upregulated in CRC tissues.Conclusions:The present study identified two mitophagy-mediated CRC subtypes with distinct features.Our MMRS may provide potential therapeutic strategies for CRC.The findings of our work offer novel insights into the involvement of mitophagy in CRC.Han Gao Qi Zou Linyun Ma Keyu Cai Yi Sun Li Lu Donglin Ren Bang Hu 2023Gastroenterology Report2023,11,1:0
7Advances in the Application of Perovskite Materials显示文摘Nowadays, the soar of photovoltaic performance of perovskite solar cells has set off a fever in the study of metal halide perovskite materials. The excellent optoelectronic properties and defect tolerance feature allow metal halide perovskite to be employed in a wide variety of applications. This article provides a holistic review over the current progress and future prospects of metal halide perovskite materials in representative promising applications, including traditional optoelectronic devices(solar cells, light-emitting diodes, photodetectors, lasers), and cutting-edge technologies in terms of neuromorphic devices(artificial synapses and memristors) and pressure-induced emission. 8Stochastic simulation of ground motions based on NGA-West2 strong motion records显示文摘Stochastic modeling of ground motion is a simple tool to predict ground shaking level for future earthquake and less time consuming than physics-based deterministic modeling.In this paper,a record-based stochastic method that considers the time-and frequency-evolution of ground motion is used to estimate ground motion for scenario earthquakes in tectonic active region.The stochastic method employs a time-domain modulation function to describe the temporal nonstationarity and a filter impulse response function that describe the evolution of frequency content.For characterizing the modulation function and the filter impulse function,six parameters(Ia,D5-95,tmid,ωmid,ω',ξf)are defined,and 2,571 pairs of ground motion recording in the NGA-west2 database are selected to identify the six parameters.Probabilistic density function is assigned to each of the parameter by fitting the frequency distribution histogram.The parameters are then transformed into standard normal space where regression analysis is performed by considering each parameter as function of moment magnitude,rupture distance,vS30(The time-averaged shear wave velocity of the top 30 m of soil).The prediction equations are used to generate ground motions for several scenario earthquakes and compared to NGA-West2 GMPEs.Peng Tian Xiaodan Sun Xin Li Keyu Wan 2019Earthquake Science2019,32,3:0
9Evaluation of N_2-Fixing Capacities of Herbaceous andShrub Legumes Under Field Condition显示文摘Natural 15N abundance method was used in this study to investigate the N2-fixing capacities of several herbaceous and shrub legumes by a field experiment. For herbaceous legumes, the results were in consistent with those obtained by a pot experiment. Crotalaria mucronata Desv. had a higher N2-fixing capacity than Vigna sinensis (L.) Savi. For shrub legumes, N2-fixing capacity under field condition was slightly different from that in pot experiment. These results demonstrated that the natural 15N abundance method was applicable to evaluating N2-fixing capacities of herbaceous and shrub legumes, and that pot experiment was suitable for evaluating the N2-fixing capacities of not only herbaceous legumes but also shrub legumes.Leguminous N2-fixing plants differed in response to phosphorus fertilization. Phosphorus fertilizer application greatly increased the percentage of nitrogen derived from air (% N dfa) and total amount of N2 fixed by Lespedeza formosa (Vog.) Koehne (Jiangxi). Phosphorus fertilizer showed no significant effect on the N2-fixing percentage of Cajanus cajan (L.) 10Interactions between hydrocarbon-bearing fluids and calcite in fused silica capillary capsules and geological implications for deeply-buried hydrocarbon reservoirs显示文摘During the burial processes of deep/ultra-deep hydrocarbon reservoirs,the interactions between hydrocarbon-bearing fluids and reservoirs significantly affect the quality evolution of hydrocarbons and reservoirs;thus,this topic requires further investigation.In this study,the continuous evolution and the coupling mechanisms in various anhydrous and hydrous nC_(16)H_(34)-(water)-(calcite)systems in fused silica capillary capsules(FSCCs)were investigated using laser Raman spectroscopy,fluorescence color analysis,and fluorescence spectroscopy,and the mineral alterations were analyzed using scanning electron microscopy(SEM).The experimental results show that extensive organic-inorganic interactions occur in the systems if water is present,and different inorganic components have different effects on hydrocarbon degradation.Distilled water promotes freeradical thermal cracking and steps oxidation,forming more low-molecular-weight hydrocarbons,CO_(2),and organic acids(e.g.,acetic acids)but suppresses the free-radical cross-linking,generating less high-molecular-weight hydrocarbons.However,in the presence of CaCl_(2) water,the yields of hydrocarbon gases are lower than in the distilled water system because high concentrations of Ca ions inhibit the generation of free radicals.Calcites,which exhibit different surface reactivities in different fluid conditions,affect hydrocarbon degradation in different ways.In the anhydrous nC16H34-calcite system,calcites promote the generation of both hydrocarbon gases and high-molecular-weight hydrocarbons.In contrast,in the hydrous nC16H34-distilled(CaCl_(2))watercalcite system,calcites promote the generation of hydrocarbon gases and suppress the generation of high-molecular-weight hydrocarbons.Calcite also reacts with organic acids via surface reactions to form secondary pores.Therefore,except for the formation temperature and pressure,organic-inorganic interactions are controlled by multiple factors,such as the water saturation,water type,water salinity,and the mineral content,resulting in different evolutions of the hydrocarbon degradation and reservoir properties.Zihao JIN Guanghui YUAN Yingchang CAO Keyu LIU Yanzhong WANG Jianye SUN Xiluo HAO Ling ZHOU Yihan WEI Shuqi WU 2022Science China Earth Sciences2022,65,2:0
11Successive formation of secondary pores via feldspar dissolution in deeply buried feldspar-rich clastic reservoirs in typical petroliferous basins and its petroleum geological significance显示文摘Clastic rock reservoirs in petroliferous basins are generally rich in feldspars. Feldspar dissolution has developed widely in clastic reservoirs, and the resulting secondary pores are crucial in deeply buried reservoirs. Based on a study of the diagenesis of clastic reservoirs in the Bohai Bay Basin, Tarim Basin, and Pearl River Mouth Basin and physical and numerical simulation experiments of fluid-rock interactions, this paper proposed a successive formation model of secondary pores via feldspar dissolution in deeply buried clastic reservoirs, considering the global research progresses in feldspar dissolution in clastic rocks. Feldspar dissolution can occur from shallow open systems to deep-ultra deep closed systems in petroliferous basins, resulting in the successive formation of secondary pores at different diagenetic stages. The successive mechanism includes three aspects. The first aspect is the succession of corrosive fluids that dissolve minerals. Meteoric freshwater dominates at the Earth’s surface and the early diagenetic A stage. Subsequently, organic acids and COformed via kerogen maturation dominate at the early diagenetic B stage to the middle diagenetic stage. COand organic acids formed via hydrocarbon oxidation in hydrocarbon reservoirs dominate at the middle diagenetic B stage to the late diagenetic stage. The second aspect is the successive formation processes of secondary pores via feldspar dissolution. Large-scale feldspar secondary pores identified in deep reservoirs include secondary pores formed at shallow-medium depths that are subsequently preserved into deep layers, as well as secondary pores formed at deep depths. Existing secondary pores in deeply buried reservoirs are the superposition of successively feldspar dissolution caused by different acids at different stages. The third aspect is a successive change in the feldspar alteration pathways and porosity enhancement/preservation effect. Open to semi-open diagenetic systems are developed from the Earth’s surface to the early diagenetic stage, and feldspar dissolution forms enhanced secondary pores. Nearly closed to closed diagenetic systems develop in the middle to late diagenetic stages, and feldspar dissolution forms redistributional secondary pores. The associated cementation causes compression resistance of the rock, which is favorable for the preservation of secondary pores in deep layers. These new insights extend the formation window of secondary pores in petroliferous basins from the traditional acid-oil generation window to a high-temperature gas generation window after hydrocarbon charging.
