2Quantitative study of tectonic geomorphology along Haiyuan fault based on airborne LiDAR显示文摘High-precision and high-resolution topography are the fundamental data for active fault research.Light detection and ranging(LiDAR)presents a new approach to build detailed digital elevation models effectively.We take the Haiyuan fault in Gansu Province as an example of how LiDAR data may be used to improve the study of active faults and the risk assessment of related hazards.In the eastern segment of the Haiyuan fault,the Shaomayin site has been comprehensively investigated in previous research because of its exemplary tectonic topographic features.Based on unprecedented LiDAR data,the horizontal and vertical coseismic offsets at the Shaomayin site are described.The measured horizontal value is about 8.6 m,and the vertical value is about 0.8 m.Using prior dating ages sampled from the same location,we estimate the horizontal slip rate as4.0±1.0 mm/a with high confidence and define that the lower bound of the vertical slip rate is 0.4±0.1 mm/a since the Holocene.LiDAR data can repeat the measurements of field work on quantifying offsets of tectonic landform features quite well.The offset landforms are visualized on an office computer workstation easily,and specialized software may be used to obtain displacement quantitatively.By combining precious chronological results,the fundamentallink between fault activity and large earthquakes is better recognized,as well as the potential risk for future earthquake hazards.Tao Chen Pei Zhen Zhang Jing Liu Chuan You Li Zhi Kun Ren Kenneth W. 3Primary 4-3-oxosteroid 5β-reductase deficiency:Two cases in China显示文摘Aldo-keto reductase 1D1(AKR1D1) deficiency,a rare but life-threatening form of bile acid deficiency,has not been previously described in China.Here,we describe the first two primary 4-3-oxosteroid 5β-reductase deficiency patients in China's Mainland diagnosed by fast atom bombardment-mass spectroscopy of urinary bile acids and confirmed by genetic analysis.A high proportion of atypical 3-oxo-4-bile acids in the urine indicated a deficiency in 4-3-oxosteroid 5β-reductase.All of the coding exons and adjacent intronic sequence of the AKR1D1 gene were sequenced using peripheral lymphocyte genomic DNA of two patients and one of the patient's parents.One patient exhibited compound heterozygous mutations:c.396C>A and c.722A>T,while the other was heterozygous for the mutation c.797G>A.Based on these mutations,a diagnosis of primary 4-3-oxosteroid 5β-reductase deficiency could be confirmed.With ursodeoxycholic acid treatment and fat-soluble vitamin supplements,liver function tests normalized rapidly,and the degree of hepatomegaly was markedly reduced in both patients.Jing Zhao Ling-Juan Fang Kenneth DR Setchell Rui Chen Li-Ting Li Jian-She Wang 2012World Journal of Gastroenterology2012,18,47:9
4Olmutinib(HM61713) reversed multidrug resistance by inhibiting the activity of ATPbinding cassette subfamily G member 2 in vitro and in vivo显示文摘Overexpressing of ATP-binding cassette(ABC) transporters is the essential cause of multidrug resistance(MDR), which is a significant hurdle to the success of chemotherapy in many cancers.Therefore, inhibiting the activity of ABC transporters may be a logical approach to circumvent MDR.Olmutinib is an epidermal growth factor receptor(EGFR) tyrosine kinase inhibitor(TKI), which has been approved in South Korea for advanced EGFR T790 M-positive non-small cell lung cancer(NSCLC). Here,we found that olmutinib significantly increased the sensitivity of chemotherapy drug in ABCG2-overexpressing cells. Furthermore, olmutinib could also increase the retention of doxorubicin(DOX) and rhodamine 123(Rho 123) in ABC transporter subfamily G member 2(ABCG2)-overexpressing cells. In addition, olmutinib was found to stimulate ATPase activity and inhibit photolabeling of ABCG2 with [^(125) I]-iodoarylazidoprazosin(IAAP). However, olmutinib neither altered ABCG2 expression at protein and m RNA levels nor blocked EGFR, Her-2 downstream signaling of AKT and ERK. Importantly,olmutinib enhanced the efficacy of topotecan on the inhibition of S1-MI-80 cell xenograft growth. All the results suggest that olmutinib reverses ABCG2-mediated MDR by binding to ATP bind site of ABCG2 and increasing intracellular chemotherapeutic drug accumulation. 5儿童NTRK重排间叶源性肿瘤临床病理学观察显示文摘目的:探讨儿童NTRK重排间叶源性肿瘤临床和病理学特征,以提高对该类疾病的认识。方法:收集上海交通大学医学院附属上海儿童医学中心及新加坡KK Women′s and Children′s Hospital从2017年1月至2019年9月5例手术切除标本,采用EnVision法检测免疫组织化学表型,荧光原位杂交(FISH)检测相关基因并对NTRK基因重排进行克隆性分析。结果:该组5例患儿,3例男性,2例女性。年龄从3个月到13岁,部位包括软组织(膝、胸腔、腹壁)和肾,肿瘤大小为4.5~12.5 cm。组织学检查主要呈梭形细胞肿瘤,浸润性生长,可伴有炎性细胞。免疫表型上,肿瘤细胞阳性表达Pan-TRK,分子检测均存在NTRK基因重排,包括DCTN1-NTRK1的发现。除2例目前接受靶向药物治疗,其余均为无疾病进展病例,随访时间9~29个月。结论:NTRK重排间叶源性肿瘤,其肿瘤发生部位广泛,组织学图像多变。免疫组织化学Pan-TRK可以帮助诊断,NTRK基因检测确认其存在重排是诊断的金标准,对不能完整切除或有复发、转移病例建议靶向药物治疗。殷敏智 马靖 何巧 沈萍 陈洁枫 金晓婷 张忠德 Chik Hong Kuick Chen Huiyi Eileen Hui Qi Ng Sze Jet Aw Kenneth Tou En Chang 2020中华病理学杂志2020,49,7:6

7Particle shape characterisation and its application to discrete element modelling显示文摘Increasing importance has been placed on particle shape implementation within discrete element modelling(DEM) in order to more accurately reflect the non-spherical behaviour of the bulk material being handled. As computational resources grow, complex particle shapes are increasingly being modelled as the associated simulation times become more realistic to provide timely solutions. The objective of this research is to assess particle shape descriptors through a digital image segmentation technique, and to further implement particle shape parameters into generation of corresponding irregular shaped DEM particles. Separated and lumped particle images were analysed and reconstructed through the development of two distinct methodologies. Subsequently, various particle shape descriptors were obtained using combinations of image segmentation algorithms, including mathematical morphology processing,thresholding, edge detection, region growing, region splitting and region merging. DEM particles were subsequently created using particle shape results obtained above. 8The Genomes of the Allohexaploid Echinochloa crus-galli and Its Progenitors Provide Insights into Polyploidization-Driven Adaptation显示文摘The hexaploid species Echinochloa crus-galli is one of the most detrimental weeds in crop fields,especially in rice paddies.Its evolutionary history is similar to that of bread wheat,arising through polyploidization after hybridization between a tetraploid and a diploid species.In this study,we generated and analyzed high-quality genome sequences of diploid(E.haploclada),tetraploid(E.oryzicola),and hexaploid(E.crus-galli)Echinochloa species.Gene family analysis showed a significant loss of disease-resistance genes such as those encoding NB-ARC domain-containing proteins during Echinochloa polyploidization,contrary to their significant expansionduring wheat polyploidization,suggesting that natural selection might favor reduced investment in resistance in this weed to maximize its growth and reproduction.In contrast to the asymmetric patterns of genome evolution observed in wheat and other crops,no significant differences in selection pressure were detected between the subgenomes in E.oryzicola and E.crus-galli.In addition,distinctive differences in subgenome transcriptome dynamics during hexaploidization were observed between E.crus-galli and bread wheat.Collectively,our study documents genomic mechanisms underlying the adaptation of a major agricultural weed during polyploidization.The genomic and transcriptomic resources of three Echinochloa species and new insights into the polyploidization-driven adaptive evolution would be useful for future breeding cereal crops.Chu-Yu Ye Dongya Wu Lingfeng Mao Lei Jia Jie Qiu Sangting Lao Meihong Chen Bowen Jiang Wei Tang Qiong Peng Lang Pan Lifeng Wang Xiaoxiao Feng Longbiao Guo Chulong Zhang Elizabeth A.Kellogg Kenneth M.Olsen Lianyang Bai Longjiang Fan 2020Molecular Plant2020,13,9:4
9Diversity and distribution patterns of non-volant smallmammals along different elevation gradients on Mt. Kenya,Kenya显示文摘The distribution of small mammals in mountainous environments across different elevations can provide important information on the effects of climate change on the dispersal of species. However, few studies conducted on Afromontane ecosystems have compared the altitudinal patterns of small mammal diversity. We investigated the species diversity and abundance of non-volant small mammals(hereafter ‘small mammals')on Mt. Kenya, the second tallest mountain in Africa,using a standard sampling scheme. Nine sampling transects were established at intervals of 200 m on the eastern(Chogoria) and western(Sirimon) slopes.A total of 1 905 individuals representing 25 species of small mammals were trapped after 12 240 trap-nights.Abundance was highest at mid-elevations on both slopes.However, species richness and their distribution patterns differed between the two slopes. More species were recorded on Chogoria(24) than on Sirimon(17). On Chogoria, species richness was higher at mid-high elevations, with a peak at mid-elevation(2 800 m a.s.l.),whereas species richness showed little variation on the Sirimon slope. These results indicate that patterns of species diversity can differ between slopes on the same mountain. In addition, we extensively reviewed literature on Mt. Kenya's mammals and compiled a comprehensive checklist of 76 mammalian species. 101.7-μm dissipative soliton Tm-doped fiber laser显示文摘We report on the dissipative soliton generation in a 1.7-μm net-normal dispersion Tm-doped fiber laser by nonlinear polarization rotation technique. An intra-cavity bandpass filter was employed to suppress the longwavelength emission, while the cavity dispersion was compensated by a segment of ultrahigh numerical aperture(UHNA4) fiber. The dissipative soliton with a central wavelength of 1746 nm was obtained, covering a spectral range from 1737 nm to 1754 nm. The de-chirped duration and energy of the dissipative soliton were 370 fs and 0.2 nJ, respectively. In addition, the dynamics of multiple dissipative solitons were also investigated. Through optimization of the cavity dispersion, the 50 nm broadband dissipative soliton with de-chirped pulse duration of 230 fs could be achieved. 11The effect of adjunct caudal block on postoperative analgesia in robot-assisted laparoscopic radical prostatectomy: A prospective randomized controlled, single blinded pilot study in a tertiary centre显示文摘Objective:Caudal block provides satisfactory postoperative pain relief in lower abdominal operations.This pilot study explores its safety and effect on postoperative pain control in patients who underwent robot-assisted laparoscopic radical prostatectomy(RARP).Methods:From 2013 to 2014,40 consecutive patients were randomized into two groups d one received caudal block using ropivacaine immediately after operation,the other received standard analgesia.Primary outcome measure was pain score based on 11-point Likert scale(0e10)recorded at recovery room,and at 6,12,24,48,and 72 h after operation.All analgesic requirements,opioid-related adverse events and time to passage of flatus were examined.Results:Mean age of the two groups was similar(60.4 vs.62.3 years,p Z 0.33),as was American Society of Anaesthesiologists(ASA)class,body mass index(BMI)and operation times.No significant difference in median pain scores was reported in recovery room(2 vs.3,p Z 0.34),and at 6 h(2 vs.2,p Z 0.94),12 h(0 vs.0,p Z 0.62),24 h(1 vs.0,p Z 0.58),48 h(1 vs.0,p Z 0.36)and 72 h(0 vs.0,p Z 0.78)postoperatively between control and caudal block groups,respectively.There was a higher mean opioid usage in the caudal block group which was not statistically significant.Although this was statistically insignificant while no significant difference in mean paracetamol usage was observed postoperatively.Median time to passage of flatus was similar(2.0 vs.2.0 days,p Z 0.97).There was one case of superficial wound infection and no opioid-related adverse events observed.Hospital stay was similar in both groups(2.5 vs.2.5 days,p Z 0.96).Kenneth Chen Allen Sim Alex Ford Kan 2018Asian Journal of Urology2018,5,2:3
12Cardiac Na^(+)-Ca^(2+) exchanger 1(ncx1h)is critical for the ventricular cardiomyocyte formation via regulating the expression levels of gata4 and hand2 in zebrafish显示文摘Ca^(2+) signaling is critical for heart development;however,the precise roles and regulatory pathways of Ca^(2+) transport proteins in cardiogenesis remain largely unknown.Sodium-calcium exchanger 1(Ncx1)is responsible for Ca^(2+) efflux in cardiomyocytes.It is involved in cardiogenesis,while the mechanism is unclear.Here,using the forward genetic screening in zebrafish,we identified a novel mutation at a highly-conserved leucine residue in ncx1 gene(mutant^(LDD353)/ncx1h^(L154P))that led to smaller hearts with reduced heart rate and weak contraction.Mechanistically,the number of ventricular but not atrial cardiomyocytes was reduced in ncx1h^(L154P) zebrafish.These defects were mimicked by knockdown or knockout of ncx1h.Moreover,ncx1h^(L154P) had cytosolic and mitochondrial Ca^(2+) overloading and Ca^(2+) transient suppression in cardiomyocytes.Furthermore,ncx1h^(L154P) and ncx1h morphants downregulated cardiac transcription factors hand2 and gata4 in the cardiac regions,while overexpression of hand2 and gata4 partially rescued cardiac defects including the number of ventricular myocytes.These findings demonstrate an essential role of the novel 154th leucine residue in the maintenance of Ncx1 function in zebrafish,and reveal previous unrecognized critical roles of the 154th leucine residue and Ncx1 in the formation of ventricular cardiomyocytes by at least partially regulating the expression levels of gata4 and hand2.Liming Chu Huimin Yin Lei Gao Li Gao Yu Xia Chiyuan Zhang Yi Chen Tingxi Liu Jijun Huang Kenneth R.Boheler Yong Zhou Huang-Tian Yang 2021Science China(Life Sciences)2021,64,2:3

15Outcomes of combination MRI-targeted and transperineal template biopsy in restaging low-risk prostate cancer for active surveillance显示文摘Objective:Active surveillance(AS)offers a strategy to reduce overtreatment and now is a widely accepted treatment option for low-risk prostate cancer.An ideal tool for risk-stratification would detect aggressive cancers and exclude such men from taking up AS in the first place.We evaluate if a combination of transperineal template biopsy with magnetic resonance imaging(MRI)-targeted biopsy identifies significant prostate cancer amongst men initially diagnosed with low-risk prostate cancer.Methods:This prospective,single-blinded study included men with low-risk prostate cancer(D’Amico’s Criteria)diagnosed on conventional transrectal ultrasound-guided biopsy.Patients first underwent multiparametric MRI of the prostate6 weeks after initial biopsy.Each suspicious lesion is mapped and assigned a Prostate Imaging Reporting and Data System(PIRADS)score.Template biopsy is first performed with the surgeon blinded to MRI findings followed by MRI-targeted biopsy using a robotic transperineal biopsy platform.Results:The age of the 19 men included is 65.4±4.9 years(mean±SD).Prostate specific antigen(PSA)at diagnosis and at the time of transperineal biopsy were comparable(7.3±1.7 ng/mL and 7.0±1.8 ng/mL,p Z 0.67),so were prostate volumes(34.2±8.9 mL and 32.1±13.4 mL,p Z 0.28).MRI-targeted biopsy had a higher percentage of cancer detection per core compared to template biopsy(11.7%vs.6.5%,p Z 0.02),this was more than 3 times superior for Gleason 7 disease(5.9%vs.1.6%,p<0.01).Four of 18(22.2%)patients with MRI lesions had significant disease with MRI-targeted biopsy alone.Three of 19 patients(15.8%)had significant disease with template biopsy alone.In combination,both techniques upclassified five patients(26.3%),all of whom underwent radical prostatectomy.Whole mount histology confirmed tumour location and grade.All six patients with PIRADS 5 lesions had cancer detected(66.6%significant disease).Conclusion:A combination of MRI-targeted and template biopsy may optimally risk-classify“low-risk”patients diagnosed on initial conventional transrectal ultrasonography(TRUS)prostate biopsy.Kenneth Chen Kae Jack Tay Yan Mee Law Hakan Aydin Henry Ho Christopher Cheng John Shyi Peng Yuen 2018Asian Journal of Urology2018,5,3:2

19Isolated alpine habitats reveal disparate ecological drivers of taxonomic and functional beta-diversity of small mammal assemblages显示文摘The interpretation of patterns of biodiversity requires the disentanglement of geographical and environmental variables.Disjunct alpine communities are geographically isolated from one another but experience similar environmental impacts.Isolated homogenous habitats may promote speciation but constrain functional trait variation.In this study,we examined the hypothesis that dispersal limitation promotes taxonomic divergence,whereas habitat similarity in alpine mountains leads to functional convergence.We performed standardized field investigation to sample non-volant small mammals from 18 prominent alpine sites in the Three Parallel Rivers area.We estimated indices quantifying taxonomic and functional alpha-and beta-diversity,as well as beta-diversity components.We then assessed the respective importance of geographical and environmental predictors in explaining taxonomic and functional compositions.No evidence was found to show that species were more functionally similar than expected in local assemblages.However,the taxonomic turnover components were higher than functional ones(0.471±0.230 vs.0.243±0.215),with nestedness components showing the opposite pattern(0.063±0.054 vs.0.269±0.225).This indicated that differences in taxonomic compositions between sites occurred from replacement of functionally similar species.Geographical barriers were the key factor influencing both taxonomic total dissimilarity and turnover components,whereas functional betadiversity was primarily explained by climatic factors such as minimum temperature of the coldest month.Our findings provide empirical evidence that taxonomic and functional diversity patterns can be independently driven by different ecological processes.Our results point to the importance of clarifying different components of beta-diversity to understand the underlying mechanisms of community assembly.These results also shed light on the assembly rules and ecological processes of terrestrial mammal communities in extreme environments.Wen-Yu Song Xue-You Li Zhong-Zheng Chen Quan Li Kenneth Otieno Onditi Shui-Wang He Xue-Long Jiang 2020Zoological Research2020,41,6:2
20Fiber optical parametric oscillator based on highly nonlinear dispersion-shifted fiber显示文摘The development of fiber optical parametric oscillators (FOPO) based on highly nonlinear dispersion- shifted fiber is reviewed in this paper. Firstly, the background and motivation are introduced, and it is pointed out that the FOPO is promising to act as optical source in non-conventional wavelength bands. Subsequently, the context focuses principally on the problem of inherent multiple-longitudinal-mode characteristic of FOPO and the corresponding solutions to it. The primary technique is by locking the phase of multiple longitudinal modes. The first reported actively mode locked FOPO is also presented in this article. However, it is not probable to realize passively mode locked FOPO because of the random phase dithering of the pump required for suppressing stimulated Brillouin scattering. Furthermore, a regeneratively mode locked FOPO is demonstrated, which can generate wide band tunable radiation in non- conventional wavelengths.
