1The advance of adjuvant treatment for triple-negative breast cancer显示文摘Triple-negative breast cancer(TNBC)is a subtype of breast cancer characterized by its highly aggressive behavior,early recurrence,and poor outcomes,when compared with other subtypes.Due to the absence of the estrogen receptor,progesterone receptor,and human epidermal growth factor receptor 2 expression,TNBC lacks meaningful biomarkers and an effective therapeutic strategy.Chemotherapy remains the main adjuvant treatment for patients with TNBC.Anthracycline/taxane-based regimens are the standard of care in adjuvant settings.The addition of capecitabine or platinum may offer extra benefits to patients with TNBC,but at the cost of increased toxicity or adverse events.Dose-dense chemotherapy may enhance treatment efficacy in patients who are able to tolerate the treatment regimen,especially in high-risk patients.As a heterogenous disease,TNBC can be classified into several molecular subtypes according to genomic or transcriptional features,which may indicate potential targets for more precise and individualized treatment strategies.With our increased understanding of signal pathways associated with TNBC,as well as the discovery of novel biomarkers indicative of TNBC prognosis,several new therapeutic options are under investigation,and some have already reported good results.In this review,we summarized the current conventional therapeutic strategies and emerging clinical trials regarding adjuvant treatment for TNBC.Furthermore,we evaluated the prognostic value of several potential targets and the progress of targeted therapy in TNBC,both in neoadjuvant and adjuvant settings.Jingyu Ge Wenjia Zuo Yiyu Chen Zhiming Shao Keda Yu 2022Cancer Biology & Medicine2022,19,2:2

3Feasibility of sentinel lymph node biopsy omission after integration of ^(18)F-FDG dedicated lymph node PET in early breast cancer: a prospective phase II trial显示文摘Objective:Sentinel lymph node biopsy(SLNB)is currently the standard of care in clinically node negative(cN0)breast cancer.The present study aimed to evaluate the negative predictive value(NPV)of 18F-FDG dedicated lymph node positron emission tomography(LymphPET)in cN0 patients.Methods:This was a prospective phase II trial divided into 2 stages(NCT04072653).In the first stage,cN0 patients underwent axillary LymphPET followed by SLNB.In the second stage,SLNB was omitted in patients with a negative preoperative axillary assessment after integration of LymphPET.Here,we report the results of the first stage.The primary outcome was the NPV of LymphPET to detect macrometastasis of lymph nodes(LN-macro).Results:A total of 189 patients with invasive breast cancer underwent LymphPET followed by surgery with definitive pathological reports.Forty patients had LN-macro,and 16 patients had only lymph node micrometastasis.Of the 131 patients with a negative LymphPET result,16 patients had LN-macro,and the NPV was 87.8%.After combined axillary imaging evaluation with ultrasound and LymphPET,100 patients were found to be both LymphPET and ultrasound negative,9 patients had LN-macro,and the NPV was 91%.Conclusions:LymphPET can be used to screen patients to potentially avoid SLNB,with an NPV>90%.The second stage of the SOAPET trial is ongoing to confirm the safety of omission of SLNB according to preoperational axillary evaluation integrating LymphPET.Junjie Li Jingyi Cheng Guangyu Liu Yifeng Hou Genghong Di Benglong Yang Yizhou Jiang Liang Huang Feilin Qu Sheng Chen Yan Wang Keda Yu Zhimin Shao 2022Cancer Biology & Medicine2022,19,7:1
4Magnesium-based materials in orthopaedics:material properties and animal models显示文摘As a new generation of medical metal materials,degradable magnesium-based materials have excellent mechanical properties and osteogenic promoting ability,making them promising materials for the treatment of refractory bone diseases.Animal models can be used to understand and evaluate the performance of materials in complex physiological environments,providing relevant data for preclinical evaluation of implants and laying the foundation for subsequent clinical studies.To date,many researchers have studied the biocompatibility,degradability and osteogenesis of magnesium-based materials,but there is a lack of review regarding the effects of magnesium-based materials in vivo.In view of the growing interest in these materials,this review briefly describes the properties of magnesium-based materials and focuses on the safety and efficacy of magnesium-based materials in vivo.Various animal models including rats,rabbits,dogs and pigs are covered to better understand and evaluate the progress and future of magnesium-based materials.This literature analysis reveals that the magnesium-based materials have good biocompatibility and osteogenic activity,thus causing no adverse reaction around the implants in vivo,and that they exhibit a beneficial effect in the process of bone repair.In addition,the degradation rate in vivo can also be improved by means of alloying and coating.These encouraging results show a promising future for the use of magnesium-based materials in musculoskeletal disorders.Xirui Jing Qiuyue Ding Qinxue Wu Weijie Su Keda Yu Yanlin Su Bing Ye Qing Gao Tingfang Sun Xiaodong Guo 2021Biomaterials Translational2021,2,3:1
5In situ bone regeneration with sequential delivery of aptamer and BMP2 from an ECM-based scaffold fabricated by cryogenic free-form extrusion显示文摘In situ tissue engineering is a powerful strategy for the treatment of bone defects.It could overcome the limitations of traditional bone tissue engineering,which typically involves extensive cell expansion steps,low cell survival rates upon transplantation,and a risk of immuno-rejection.Here,a porous scaffold polycaprolactone(PCL)/decellularized small intestine submucosa(SIS)was fabricated via cryogenic free-form extrusion,followed by surface modification with aptamer and PlGF-2_(123-144)*-fused BMP2(pBMP2).The two bioactive molecules were delivered sequentially.The aptamer Apt19s,which exhibited binding affinity to bone marrow-derived mesenchymal stem cells(BMSCs),was quickly released,facilitating the mobilization and recruitment of host BMSCs.BMP2 fused with a PlGF-2_(123-144)peptide,which showed“super-affinity”to the ECM matrix,was released in a slow and sustained manner,inducing BMSC osteogenic differentiation.In vitro results showed that the sequential release of PCL/SIS-pBMP2-Apt19s promoted cell migration,proliferation,alkaline phosphatase activity,and mRNA expression of osteogenesis-related genes.The in vivo results demonstrated that the sequential release system of PCL/SIS-pBMP2-Apt19s evidently increased bone formation in rat calvarial critical-sized defects compared to the sequential release system of PCL/SIS-BMP2-Apt19s.Thus,the novel delivery system shows potential as an ideal alternative for achieving cell-free scaffold-based bone regeneration in situ.Tingfang Sun Chunqing Meng Qiuyue Ding Keda Yu Xianglin Zhang Wancheng Zhang Wenqing Tian Qi Zhang Xiaodong Guo Bin Wu Zekang Xiong 2021Bioactive Materials2021,6,11:1

7A machine learning model to predict efficacy of neoadjuvant therapy in breast cancer based on dynamic changes in systemic immunity显示文摘Objective:Neoadjuvant therapy(NAT)has been widely implemented as an essential treatment to improve therapeutic efficacy in patients with locally-advanced cancer to reduce tumor burden and prolong survival,particularly for human epidermal growth receptor 2-positive and triple-negative breast cancer.The role of peripheral immune components in predicting therapeutic responses has received limited attention.Herein we determined the relationship between dynamic changes in peripheral immune indices and therapeutic responses during NAT administration.Methods:Peripheral immune index data were collected from 134 patients before and after NAT.Logistic regression and machine learning algorithms were applied to the feature selection and model construction processes,respectively.Results:Peripheral immune status with a greater number of CD3^(+)T cells before and after NAT,and a greater number of CD8^(+)T cells,fewer CD4^(+)T cells,and fewer NK cells after NAT was significantly related to a pathological complete response(P<0.05).The post-NAT NK cell-to-pre-NAT NK cell ratio was negatively correlated with the response to NAT(HR=0.13,P=0.008).Based on the results of logistic regression,14 reliable features(P<0.05)were selected to construct the machine learning model.The random forest model exhibited the best power to predict efficacy of NAT among 10 machine learning model approaches(AUC=0.733).Conclusions:Statistically significant relationships between several specific immune indices and the efficacy of NAT were revealed.A random forest model based on dynamic changes in peripheral immune indices showed robust performance in predicting NAT efficacy.Yusong Wang Mozhi Wang Keda Yu Shouping Xu Pengfei Qiu Zhidong Lyu Mingke Cui Qiang Zhang Yingying Xu 2023Cancer Biology & Medicine2023,20,3:0
