| 3 | Recruitment of endogenous progenitor cells by erythropoietin loaded particles for in situ cartilage regeneration显示文摘Cartilage injury affects millions of people throughout the world,and at this time there is no cure.While transplantation of stem cells has shown some success in the treatment of injured cartilage,such treatment is limited by limited cell sources and safety concerns.To overcome these drawbacks,a microscaffolds system was developed capable of targeting,reducing the inflammatory response and recruiting endogenous progenitor cells to cartilage-defect.Erythropoietin(EPO)-loaded-hyaluronic acid(HA)microscaffolds(HA+EPO)were fabricated and characterized.HA-microscaffolds showed good cell-compatibility and could target chondrocytes via CD44 receptors.HA+EPO was designed to slowly release EPO while recruiting progenitor cells.Finally,the ability of HA+EPO to repair cartilage-defects was assessed using a rabbit model of full-thickness cartilagedefect.Our results showed that the intra-articular administration of EPO,HA,and EPO+HA reduced the number of inflammatory cells inside the synovial-fluid,while EPO+HA had the greatest anti-inflammatory effects.Furthermore,among all groups,EPO+HA achieved the greatest progenitor cell recruitment and subsequent chondrogenesis.The results of this work support that,by targeting and localizing the release of growthfactors,HA+EPO can reduce inflammatory responses and promote progenitor cells responses.This new platform represents an alternative treatment to stem-cell transplantation for the treatment of cartilage injury. | Amirhossein Hakamivala Shuxin li Kayti Robinson YiHui Huang Shuai Yu Baohong Yuan Joseph Borrelli Jr. Liping Tang | 2020 | Bioactive Materials2020,5,1: | 7 |
| 4 | Cell cycle-related kinase reprograms the liver immune microenvironment to promote cancer metastasis显示文摘The liver is an immunologically tolerant organ and a common metastatic site of multiple cancer types.Although a role for cancer cell invasion programs has been well characterized,whether and how liver-intrinsic factors drive metastatic spread is incompletely understood.Here,we show that aberrantly activated hepatocyte-intrinsic cell cycle-related kinase(CCRK)signaling in chronic liver diseases is critical for cancer metastasis by reprogramming an immunosuppressive microenvironment.Using an inducible liverspecific transgenic model,we found that CCRK overexpression dramatically increased both B16F10 melanoma and MC38 colorectal cancer(CRC)metastasis to the liver,which was highly infiltrated by polymorphonuclear-myeloid-derived suppressor cells(PMNMDSCs)and lacking natural killer T(NKT)cells.Depletion of PMN-MDSCs in CCRK transgenic mice restored NKT cell levels and their interferon gamma production and reduced liver metastasis to 2.7% and 0.7%(metastatic tumor weights)in the melanoma and CRC models,respectively.Mechanistically,CCRK activated nuclear factor-kappa B(NF-κB)signaling to increase the PMN-MDSC trafficking chemokine C-X-C motif ligand 1(CXCL1),which was positively correlated with liver-infiltrating PMN-MDSC levels in CCRK transgenic mice.Accordingly,CRC liver metastasis patients exhibited hyperaaivation of hepatic CCRK/NF-κB/CXCL1 signaling,which was associated with accumulation of PMN-MDSCs and paucity of NKT cells compared to healthy liver transplantation donors.In summary,this study demonstrates that immunosuppressive reprogramming by hepatic CCRK signaling undermines antimetastatic immunosurveillance.Our findings offer new mechanistic insights and therapeutic targets for liver metastasis intervention. | Xuezhen Zeng Jingying Zhou Zhewen Xiong Hanyong Sun Weiqin Yang Myth T.S.Mok Jing Wang Jingqing Li Man Liu Wenshu Tang Yu Feng Hector Kwong-Sang W ang Shun-Wa Tsang King-Lau Chow Philip Chun Yeung John Wong Paul Bo-San Lai Anthony Wing-Hung Chan Ka Fai To Stephen Lam Chan Qiang Xia Jing Xue Xiao Chen Jun Yu Sui Peng Joseph Jao-Yiu Sung Ming Kuang Alfred Sze-Lok Cheng | 2021 | Cellular & Molecular Immunology2021,18,4: | 5 |
| 5 | Carbamylated erythropoietin regulates immune responses and promotes long-term kidney allograft survival through activation of PI3K/AKT signaling显示文摘Modulation of alloimmune responses is critical to improving transplant outcome and promoting long-term graft survival.To determine mechanisms by which a nonhematopoietic erythropoietin(EPO)derivative,carbamylated EPO(CEPO),regulates innate and adaptive immune cells and affects renal allograft survival,we utilized a rat model of fully MHC-mismatched kidney transplantation.CEPO administration markedly extended the survival time of kidney allografts compared with the transplant alone control group.This therapeutic effect was inhibited when the recipients were given LY294002,a selective inhibitor of the phosphoinositide 3-kinase(PI3K)/protein kinase B(AKT)signaling pathway or anti-EPO receptor(EPOR)antibody,in addition to CEPO.In vitro,CEPO inhibited the differentiation and function of dendritic cells and modulated their production of proinflammatory and anti-inflammatory cytokines,along with activating the PI3K/AKT signaling pathway and increasing EPOR mRNA and protein expression by these innate immune cells.Moreover,after CD4^(+)T cells were exposed to CEPO the Th1/Th2 ratio decreased and the regulatory T cell(Treg)/Th17 ratio increased.These effects were abolished by LY294002 or anti-EPOR antibody,suggesting that CEPO regulates immune responses and promotes kidney allograft survival by activating the PI3K/AKT signaling pathway in an EPOR-dependent manner.The immunomodulatory and specific signaling pathway effects of CEPO identified in this study suggest a potential therapeutic approach to promoting kidney transplant survival. | Ning Na Daqiang Zhao Jinhua Zhang Jiaqing Wu Bin Miao Heng Li Yingxun Luo Zuofu Tang Wensheng Zhang Joseph A.Bellanti Song Guo Zheng | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 4 |

| 7 | Pathological correlations between traumatic brain injury and chronic neurodegenerative diseases显示文摘Traumatic brain injury is among the most common causes of death and disability in youth and young adults.In addition to the acute risk of morbidity with moderate to severe injuries,traumatic brain injury is associated with a number of chronic neurological and neuropsychiatric sequelae including neurodegenerative diseases such as Alzheimer’s disease and Parkinson’s disease.However,despite the high incidence of traumatic brain injuries and the established clinical correlation with neurodegeneration,the causative factors linking these processes have not yet been fully elucidated.Apart from removal from activity,few,if any prophylactic treatments against post-traumatic brain injury neurodegeneration exist.Therefore,it is imperative to understand the pathophysiological mechanisms of traumatic brain injury and neurodegeneration in order to identify potential factors that initiate neurodegenerative processes.Oxidative stress,neuroinflammation,and glutamatergic excitotoxicity have previously been implicated in both secondary brain injury and neurodegeneration.In particular,reactive oxygen species appear to be key in mediating molecular insult in neuroinflammation and excitotoxicity.As such,it is likely that post injury oxidative stress is a key mechanism which links traumatic brain injury to increased risk of neurodegeneration.Consequently,reactive oxygen species and their subsequent byproducts may serve as novel fluid markers for identification and monitoring of cellular damage.Furthermore,these reactive species may further serve as a suitable therapeutic target to reduce the risk of post-injury neurodegeneration and provide long term quality of life improvements for those suffering from traumatic brain injury. | Marcela Cruz-Haces Jonathan Tang Glen Acosta Joseph Fernandez Riyi Shi | 2017 | Translational Neurodegeneration2017,6,1: | 2 |
