1Codon 249 mutation in exon 7 of p53 gene in plasma DNA:maybe a new early diagnostic marker of hepatocellular carcinoma in Qidong risk area,China显示文摘AIM: One of the characteristics of hepatocellular carcinoma (HCC) in Qidong area is the selective mutation resulting in a serine substitution at codon 249 of the p53 gene (1,20),and it has been identified as a 'hotspot' mutation in heptocellular carcinomas occurring in populations exposed to aflatoxin and with high prevalence of hepatitis B virus carriers (2, 3, 9, 10, 16, 24). We evaluated in this paper whether this 'hotspot' mutation could be detected in cellfree DNA circulating in plasma of patients with hepatocellular carcinoma and cirrhosis in Qidong, China, and tried to illustrate the significance of the detection of this molecular biomarker.METHODS: We collected blood samples from 25hepatocellular carcinoma patients, 20 cirrhotic patients and 30 healthy controls in Qidong area. DNA was extracted and purified from 200 μl of plasma from each sample. The 249ser p53 mutation was detected by restriction digestion analysis and direct sequencing of exon-7 PCR products.RESULTS: We found in exon 7 of p53 gene G→T transversion at the third base of codon 249 resulting 249Arg→249ser mutation in 10/25 (40%) hepatocellular carcinoma cases,4/20 (20%) cirrhotics, and 2/30 (7 %) healthy controls.The adjusted odds ratio for having the mutation was 22.1(95 % CI, 3.2~91.7) for HCC cases compared to controls.CONCLUSION: These data show that the 249ser p53mutation in plasma is strongly associated with hepatocellular carcinoma in Qidong patients. 3Stress Effect of Cadmium Absorption between Aloe and Solanum nigrum L显示文摘The stress effect of cadmium absorption between aloe and Solanum nigrum L was studied through a simulation experiment with different cadmium contents in soil.The results showed that the growth characteristics,biomass and Cd content in the plants of aloe and S.nigrum L were closely related to Cd content in the soil,and S.nigrum L and aloe had a strong tolerance to Cd in the experiment.pH of rhizosphere soil with S.nigrum L and aloe planted was generally higher than that of non-rhizosphere soil,which is related to the planting pattern.Compared with aloe,S.nigrum L had a stronger capacity to accumulate Cd,and intercropping S.nigrum L and aloe could inhibit aloe's absorption of Cd,which is favorable to the safe planting of aloe.In addition,S.nigrum L has the potential to restore polluted soil.Chen Yingzhi Fu Shanming Wang Jianhuang Pei Jie Li Wenhao He Caizhen 2015Meteorological and Environmental Research2015,6,3:4
4An Adaptive PM Model Based on Difference Eigenvalue for Image Restoration显示文摘Classic Perona-Malik(PM) model is usually used to smooth noise for the degraded image. However, to our knowledge, a well-known defect of the PM model is prone to cause ‘staircase' effect and blur image fine feature because of the lack of correct diffusion strength in diffusion process. To tackle the problem, we will improve the PM model by developing a new structure descriptor to adjust anisotropic diffusion strength for smoothing noise and preserving image feature adaptively. The new structure descriptor is based on image local geometry-Hessian matrix, called difference eigenvalue, which can adaptively track edge feature and high degree of homogeneity in an image, even when the observed image is blurred. 5The in vitro reconstitution of nucleosome and its binding patterns with HMG1/2 and HMG14/17 proteins显示文摘Using atomic force microscopy (AFM), the dynamic process of the in vitro nucleosome reconstitution followed by slow dilution from high salt to low salt was visualized. Data showed that the histone octamers were dissociated from DNA at 1M NaCl. When the salt concentration was slowly reduced to 650 mMand 300 mM, the core histones bound to the naked DNA gradually. Once the salt concentration was reduced to 50 mM the classic 'beads-on-a-string' structure was clearly visualized. Furthermore, using the technique of the in vitro reconstitution ofnucleosome,the mono- and di- nucleosomes were assembled in vitro with both HS2core (-10681 to -10970 bp) and NCR2 (-372to -194 bp) DNA sequences in the 5'flanking sequence of human b-globin gene. Data revealed that HMG 1/2 and HMG 14/17 proteins binding to both DNA sequences are changeable following the assembly and disassembly of nucleosomes. 9A virtual reality based surgical skills training simulator for catheter ablation with real-time and robust interaction显示文摘Background Atrial fibrillation(AF)is the most common sustained cardiac arrhythmia that can cause severe heart problems.Catheter ablation is one of the most ideal procedures for the treatment of AF.Physicians qualified to perform this procedure need to be highly skilled in manipulating the relevant surgical devices.This study proposes an interactive surgical simulator with high fidelity to facilitate efficient training and low-cost medical education.Methods We used a shared centerline model to simulate the interaction between multiple surgical devices.An improved adaptive deviation-feedback approach is proposed to accelerate the convergence of each iteration.The periodical beating of the human heart was also simulated in real time using the position-based dynamics(PBD)framework to achieve higher fidelity.We then present a novel method for handling the interaction between the devices and the beating heart mesh model.Experiments were conducted in a homemade simulator prototype to evaluate the robustness,performance,and flexibility of the proposed method.Preliminary evaluation of the simulator was performed by medical students,residents,and surgeons.Results The interaction between surgical devices,static vascular meshes,and beating heart mesh was stably simulated in a frame rate suitable for interaction.Conclusion Our simulator is capable of simulating the procedure of catheter ablation with high fidelity and provides immersive visual experiences and haptic feedback.Haoyu WANG Jianhuang WU 2021Virtual Reality & Intelligent Hardware2021,3,4:1
10Relationships between the Extratropical ENSO Precursor and Leading Modes of Atmospheric Variability in the Southern Hemisphere显示文摘Previous studies suggest that the atmospheric precursor of El Ni ?no–Southern Oscillation(ENSO) in the extratropical Southern Hemisphere(SH) might trigger a quadrapole sea surface temperature anomaly(SSTA) in the South Pacific and subsequently influence the following ENSO. Such a quadrapole SSTA is referred to as the South Pacific quadrapole(SPQ).The present study investigated the relationships between the atmospheric precursor signal of ENSO and leading modes of atmospheric variability in the extratropical SH [including the SH annular mode(SAM), the first Pacific–South America(PSA1) mode, and the second Pacific–South America(PSA2) mode]. The results showed that the atmospheric precursor signal in the extratropical SH basically exhibits a barotropic wavenumber-3 structure over the South Pacific and is significantly correlated with the SAM and the PSA2 mode during austral summer. Nevertheless, only the PSA2 mode was found to be a precursor for the following ENSO. It leads the SPQ-like SSTA by around one month, while the SAM and the PSA1 mode do not show any obvious linkage with either ENSO or the SPQ. 12A Novel Method for Conversion of Valuable BDS Intermediates HPBS/Cx-HPBS from DBT/Cx-DBT显示文摘A simple recombinant PCR method was used to delete the dszB gene responsible for the slowest step of the Dsz pathway and allow the accumulation of hydroxyphenyl benzene sulfinate (HPBS) in the recombinant E. coli. Using GC/MS, HPBS accumulation was confirmed. The recombinant E. coli could also desulfurize Cx-DBT to corresponding Cx-HPBS.
