1Gait Recognition by Cross Wavelet Transform and Graph Model显示文摘In this paper, a multi-view gait based human recognition system using the fusion of two kinds of features is proposed.We use cross wavelet transform to extract dynamic feature and bipartite graph model to extract static feature which are coefficients of quadrature mirror filter(QMF)-graph wavelet filter bank. Feature fusion is done after normalization. For normalization of features, min-max rule is used and mean-variance method is used to find weights for normalized features. Euclidean distance between each feature vector and center of the cluster which is obtained by k-means clustering is used as similarity measure in Bayesian framework. Experiments performed on widely used CASIA B gait database show that, the fusion of these two feature sets preserve discriminant information. 3Absolute copy number differences of Y chromosomal genes between crossbred (Bos taurus×Bos indicus) and Indicine bulls显示文摘Background: The Y chromosome in mammal is paternally inherited and harbors genes related to male fertility and spermatogenesis. The unique intra-chromosomal recombination pattern of Y chromosome and morphological difference of this chromosome between Bos taurus and Bos indicus make it an ideal model for studying structural variation, especially in crossbred(Bos taurus × Bos indicus) bulls. Copy Number Variation(CNV) is a type of genomic structural variation that gives information complementary to SNP data. The purpose of this study was to find out copy number differences of four Y chromosomal spermatogenesis-related candidate genes in genomic DNA of crossbred and purebred Indicine bulls. Result: Four Y chromosomal candidate genes of spermatogenesis namely, sex determining gene on Y chromosome(SRY), DEAD box polypeptide 3-Y chromosome(DDX3Y), Ubiquitin specific peptidase 9, Y-linked(USP9Y), testis-specific protein on Y chromosome(TSPY) were evaluated. Absolute copy numbers of Y chromosomal genes were determined by standard curve-based quantitative real time PCR. Copy numbers of SRY and TSPY genes per unit amount of genomic DNA are higher in crossbred than Indicine bulls. However, no difference was observed in DDX3Y and USP9Y gene copy numbers between two groups. Conclusion: The present study demonstrates that the structural organization of Y chromosomes differs between crossbred and Indicine bulls which are reproductively healthy as observed from analysis of semen attributes. The absolute copy numbers of SRY and TSPY genes in unit mass of genomic DNA of crossbred bulls are significantly higher than Indicine bulls. No alteration in absolute copies of DDX3Y and USP9Y gene was found between the genome of crossbred and Indicine bulls. 4Carvacrol Promotes Cell Cycle Arrest and Apoptosis through PI3K/AKT Signaling Pathway in MCF-7 Breast Cancer Cells显示文摘Objective:To examine the role of carvacrol in modulating PI3 K/AKT signaling involved in human breast cancer pathogenesis using in vitro experimental model MCF-7 cells.Methods:MTT and lactate dehydrogenase assays were performed with cells treated with different doses of carvacrol(0–250μmol/L)at different time points(24 and 48 h).The nuclear morphology was assessed in MCF-7 cells with propidium iodide(PI)and acridine orange/ethidium bromide(AO/EB)staining and analyzed by fluorescence microscopy.Events like cell cycle arrest and apoptosis were observed by flow cytometric analysis and expressions of p-Rb,cyclin D1,cyclin-dependent kinase 4(CDK4),CDK6,Bax,Bcl-2,PI3 K/p-AKT were analyzed by immunoblot.Results:Carvacrol significantly reduced cell viability with the half maximal inhibitory concentration value of 200μmol/L at 24 and 48 h(P<0.05).importantly,there was a significant increase in the accumulation of the G0/G1 phase upon treatment with carvacrol in MCF-7 cells(P<0.05 or P<0.01).A remarkable decrease in protein expressions of p-Rb,cyclin D1,CDK4 and CDK6 denoted cell cycle arrest(P<0.05 or P<0.01).In addition,carvacrol treatment significantly inhibited PI3 K/p-AKT protein expressions leading to induction of apoptosis mediated by decreased Bcl2 and increased Bax protein expressions.Further,Annexin V/PI staining by FACS analysis,dual staining by AO/EB and PI staining studies suggested induction of apoptosis by carvacrol through PI3 K/Akt signaling pathway in MCF-7 cells.Conclusion:Carvacrol significantly inhibited the breast cancer MCF-7 cell proliferation and induced apoptosis via suppressing PI3/AKT signaling pathway.Ashok Mari Gopikrishnan Mani Sirpu Natesh Nagabhishek Gopaiakrishnan Balaraman Nirmala Subramanian Fathima Bushra Mirza Jagan Sundaram Devaki Thiruvengadam 2021Chinese Journal of Integrative Medicine2021,27,9:3
