| 6 | Gene expression changes in spleens of the wildlife reservoir species,Eurasian wild boar(Sus scrofa),naturally infected with Brucella suis biovar 2显示文摘Brucella suis is responsible for swine brucellosis worldwide.Of the five different B.suis biovars(bv.),bv.2 appears restricted to Europe where it is frequently isolated from wild boar and hares,can infect pigs and can cause human brucellosis.In this study,the differential gene expression profile was characterized in spleens of Eurasian wild boar naturally infected with B.suis bv.2.Of the 20,201 genes analyzed in the microarray,633 and 1,373 were significantly(fold change > 1.8;P < 0.01) upregulated and downregulated,respectively,in infected wild boar.The analysis was focused on genes that were over represented after conditional test for biological process gene ontology.Upregulated genes suggested that B.suis bv.2 infection induced cell maturation,migration and/or proliferation in infected animals.The genes downregulated in infected wild boar impaired the activity of several important cellular metabolic pathways such as metabolism,cytoskeleton organization and biogenesis,immune response and lysosomal function and vesicle-mediated transport.In addition,the response to stress,sperm fertility,muscle development and apoptosis seemed to be also impaired in infected animals.These results suggested that B.suis bv.2 may use strategies similar to other smooth brucellae to facilitate intracellular multiplication and the development of chronic infections.To our knowledge,this is the first report of the analysis of gene expression profile in hosts infected with B.suis bv.2,which is important to understand the molecular mechanisms at the host-pathogen interface in the main reservoir species with possible implications in the zoonotic cycle of the pathogen. | Ruth C.Galindo Pilar M.Muoz María J.de Miguel Clara M.Marin Javier Labairu Miguel Revilla José M.Blasco Christian Gortazar José de la Fuente | 2010 | Journal of Genetics and Genomics2010,37,11: | 6 |

| 10 | Utilizing the ultrasensitive Schistosoma up-converting phosphor lateral flow circulating anodic antigen(UCP-LF CAA)assay for sample pooling-strategies显示文摘Background:Methodological applications of the high sensitivity genus-specific Schistosoma CAA strip test,allowing detection of single worm active infections(ultimate sensitivity),are discussed for efficient utilization in sample pooling strategies.Besides relevant cost reduction,pooling of samples rather than individual testing can provide valuable data for large scale mapping,surveillance,and monitoring.Method:The laboratory-based CAA strip test utilizes luminescent quantitative up-converting phosphor(UCP)reporter particles and a rapid user-friendly lateral flow(LF)assay format.The test includes a sample preparation step that permits virtually unlimited sample concentration with urine,reaching ultimate sensitivity(single worm detection)at 100%specificity.This facilitates testing large urine pools from many individuals with minimal loss of sensitivity and specificity.The test determines the average CAA level of the individuals in the pool thus indicating overall worm burden and prevalence.When requiring test results at the individual level,smaller pools need to be analysed with the pool-size based on expected prevalence or when unknown,on the average CAA level of a larger group;CAA negative pools do not require individual test results and thus reduce the number of tests.Results:Straightforward pooling strategies indicate that at sub-population level the CAA strip test is an efficient assay for general mapping,identification of hotspots,determination of stratified infection levels,and accurate monitoring of mass drug administrations(MDA).At the individual level,the number of tests can be reduced i.e.in low endemic settings as the pool size can be increased as opposed to prevalence decrease.Conclusions:At the sub-population level,average CAA concentrations determined in urine pools can be an appropriate measure indicating worm burden.Pooling strategies allowing this type of large scale testing are feasible with the various CAA strip test formats and do not affect sensitivity and specificity.It allows cost efficient stratified testing and monitoring of worm burden at the sub-population level,ideally for large-scale surveillance generating hard data for performance of MDA programs and strategic planning when moving towards transmission-stop and elimination. | Paul L.A.M.Corstjens Pytsje T.Hoekstra Claudia J.de Dood Govert J.van Dam | 2017 | Infectious Diseases of Poverty2017,6,1: | 2 |

| 13 | Late Palaeogene emplacement and late Neogene-Quaternary exhumation of the Kuril island-arc root (Kunashir island) constrained by multi-method thermochronometry显示文摘The Kuril islands constitute a volcanic island arc-trench system,stretching from eastern Hokkaido(Japan)to Kamchatka(Russia) along the northwestern Pacific subduction system.The current arc consists of several volcanic islands mainly with Neogene basement and capped by several,predominantly andesitic,active subduction stratovolcanoes.Kunashir Island is the southwestern-most island of the arc,just off the Hokkaido coast and represents the study area in this paper.The island is composed of a Lower Complex of mainly late Miocene to Pliocene volcanic rocks,covered by an Upper Complex of younger(basaltic)andesitic lava flows and tuffs on which currently four active volcanic edifices are built.In the Lower Complex sub-volcanic and deeper-seated intrusives of the so-called Prasolov and Dokuchaev magmatic complexes are found.More differentiated,tonalitic-granodioritic rocks were collected from these small intrusive bodies.An early Oligocene zircon LA-ICP-MS U/Pb age of 31 Ma for the Prasolov Complex was obtained,showing that the basement of Kunashir Island is older than previously thought.Thermochronometry(apatite fission-track and U-Th-Sm/He and zircon U-Th/He analyses) further shows that the magmatic basement of the island was rapidly exhumed in the Pleistocene to present levels in a differential pattern,with He-ages ranging from 1.9 to 0.8 Ma.It is shown that the northern section of the island was hereby exhumed more intensely. | J.De Grave F.I.Zhimulev S.Glorie G.V.Kuznetsov N.Evans F.Vanhaecke B.McInnes | 2016 | Geoscience Frontiers2016,7,2: | 2 |
