1Locomotor analysis identifies early compensatory changes during disease progression and subgroup classification in a mouse model of amyotrophic lateral sclerosis显示文摘Amyotrophic lateral sclerosis is a motoneuron degenerative disease that is challenging to diagnose and presents with considerable variability in survival.Early identification and enhanced understanding of symptomatic patterns could aid in diagnosis and provide an avenue for monitoring disease progression.Use of the m SOD1 G93 A mouse model provides control of the confounding environmental factors and genetic heterogeneity seen in amyotrophic lateral sclerosis patients,while investigating underlying disease-induced changes.In the present study,we performed a longitudinal behavioral assessment paradigm and identified an early hindlimb symptom,resembling the common gait abnormality foot drop,along with an accompanying forelimb compensatory mechanism in the m SOD1 G93 A mouse.Following these initial changes,m SOD1 mice displayed a temporary hindlimb compensatory mechanism resembling an exaggerated steppage gait.As the disease progressed,these compensatory mechanisms were not sufficient to sustain fundamental locomotor parameters and more severe deficits appeared.We next applied these initial findings to investigate the inherent variability in B6 SJL m SOD1 G93 A survival.We identified four behavioral variables that,when combined in a cluster analysis,identified two subpopulations with different disease progression rates:a fast progression group and a slow progression group.This behavioral assessment paradigm,with its analytical approaches,provides a method for monitoring disease progression and detecting m SOD1 subgroups with different disease severities.This affords researchers an opportunity to search for genetic modifiers or other factors that likely enhance or slow disease progression.Such factors are possible therapeutic targets with the potential to slow disease progression and provide insight into the underlying pathology and disease mechanisms.Melissa M.Haulcomb Rena M.Meadows Whitney M.Miller Kathryn P.Mc Millan Me Kenzie J.Hilsmeyer Xuefu Wang Wesley T.Beaulieu Stephanie L.Dickinson Todd J.Brown Virginia M.Sanders Kathryn J.Jones 2017Neural Regeneration Research2017,12,10:3
2Caregiver burden is increased in Parkinson’s disease with mild cognitive impairment (PD-MCI)显示文摘Background:There is limited evidence on caregiver outcomes associated with mild cognitive impairment in patients with Parkinson’s disease(PD-MCI)and the coping strategies used by these caregivers.Methods:To investigate this relationship,we examined levels of burden,depression,anxiety,coping strategies and positive aspects of caregiving in the informal caregivers of 96 PD patients.The PD patients were classified using MDS-Task Force Level II criteria as showing either normal cognition(PD-N;n=51),PD-MCI(n=30)or with dementia(PDD;n=15).Results:Mean Zarit Burden Interview(ZBI)score increased significantly between carers of PD-N(M=13.39,SD=12.22)compared to those of PD-MCI patients(M=22.00,SD=10.8),and between carers of PD-MCI and PDD patients(M=29.33,SD=9.59).Moreover,the proportion of carers showing clinically significant levels of burden(ZBI score≥21)also increased as the patients’cognitive status declined(18% for PD-N;60% for PD-MCI;and 80%for PDD)and was mirrored by an increasing amount of time spent providing care by the caregivers.Caregiver ZBI score was independent of patient neuropsychiatric symptoms,motor function,disease duration and time that caregivers spent caregiving.Caregiver use of different coping strategies increased with worsening cognition.However,we found only equivocal evidence that the use of problem-focused,emotion-focused and dysfunctional coping mediated the association between patient cognitive status and caregiver burden,because the inverse models that used caregiver burden as the mediator were also significant.Conclusions:The study highlights the impact of Parkinson’s disease on those providing care when the patient’s cognition is poor,including those with MCI.Caregiver well-being has important implications for caregiver support,nursing home placement and disease course.Ann J.Jones Roeline G.Kuijer Leslie Livingston Daniel Myall Kyla Horne Michael MacAskill Toni Pitcher Paul T.Barrett Tim J.Anderson John C.Dalrymple-Alford 2017Translational Neurodegeneration2017,6,1:2

7A tale of motor neurons and CD4+ T cells: moving forward by looking back显示文摘Amyotrophic lateral sclerosis(ALS) is a fatal progressive disorder characterized by the selective degeneration of motor neurons(MN). The impact of peripheral immune status on disease progression and MN survival is becoming increasingly recognized in the ALS research field. In this review, we briefly discuss findings from mouse models of peripheral nerve injury and immunodeficiency to understand how the immune system regulates MN survival. We extend these observations to similar studies in the widely used superoxide dismutase 1(SOD1) mouse model of ALS. Last, we present future hypotheses to identify potential causative factors that lead to immune dysregulation in ALS. 10Manipulating active layer morphology of molecular donor/polymer acceptor based organic solar cells through ternary blends显示文摘The development of molecular donor/polymer acceptor blend(MD/PA)-type organic solar cells(OSCs) lags far behind other type OSCs. It is due to the large-size phase separation morphology of MD/PAblend, which results from the high crystallinity of molecular donors. In this article, to suppress the crystallinity of molecular donors, we use ternary blends to develop OSCs based on one polymer acceptor(P-BNBP-f BT) and two molecular donors(DR3 TBDTT and BTR) with similar chemical structures.The ternary OSC exhibits a power conversion efficiency(PCE) of 4.85%, which is higher than those of the binary OSCs(PCE=3.60% or 3.86%). To our best knowledge, it is the first report of ternary MD/PA-type OSCs and this PCE is among the highest for MD/PA-type OSCs reported so far. Compared with the binary blends, the ternary blend exhibits decreased crystalline size and improved face-on orientation of the donors. As a result, the ternary blend exhibits improved and balanced charge mobilities, suppressed charge recombination and increased donor/acceptor interfacial areas, which leads to the higher shortcircuit current density. 11Reservoir quality of fluvial sandstone reservoirs in salt-walled mini- basins: an example from the Seagull field, Central Graben, North Sea, UK显示文摘The Triassic fluvial sandstones of the Skagerrak Formation were deposited in a series of salt-walled mini-basins and act as important hydrocarbon reservoirs for several high-pressure, high-temperature(HPHT) fields in the Central Graben, North Sea. The HPHT reservoirs exhibit excellent reservoir quality considering their depth of burial and hence have been of high interest for hydrocarbon exploration. This research uses a multidisciplinary approach to assess the Skagerrak Formation fluvial reservoir quality from the Seagull field incorporating core analysis, petrography, electron microscopy, XRD analysis, fluid inclusion appraisal and burial history modelling. Halokinesis and salt withdrawal at the margin of the saltwalled mini-basin induced early disaggregation bands and fractures at shallow burial and led to increased influx of meteoric water and clay mineral infiltration from overlying sedimentation. The density of disaggregation bands correlates with the occurrence and magnitude of pore-filling authigenic clay minerals, concentrated along the margin of the saltwalled mini-basin. 14Using Twitter Bios to Measure Changes in Self-Identity:Are Americans Defining Themselves More Politically Over Time?显示文摘Are Americans weaving their political views more tightly into the fabric of their self-identity over time?If so,then we might expect partisan disagreements to continue becoming more emotional,tribal,and intractable.Much recent scholarship has speculated that this politicization of Americans’identity is occurring,but there has been little compelling attempt to quantify the phenomenon,largely because the concept of identity is notoriously difficult to measure.We introduce here a methodology,Longitudinal Online Profile Sampling(LOPS),which affords quantifiable insights into the way individuals amend their identity over time.Using this method,we analyze millions of“bios”on the microblogging site Twitter over a 4-year span,and conclude that the average American user is increasingly integrating politics into their social identity.Americans on the site are adding political words to their bios at a higher rate than any other category of words we measured,and are now more likely to describe themselves by their political affiliation than their religious affiliation.The data suggest that this is due to both cohort and individual-level effects.Nick Rogers Jason J.Jones 2021Journal of Social Computing2021,2,1:0
