| 1 | Mitochondrial protection by low doses of insulin-like growth factor-Ⅰin experimental cirrhosis显示文摘AIM: To characterize the mitochondrial dysfunction in experimental cirrhosis and to study whether insulin-like growth factor-Ⅰ(IGF-Ⅰ) therapy (4 wk) is able to induce beneficial effects on damaged mitochondria leading to cellular protection. METHODS: Wistar rats were divided into three groups: Control group, untreated cirrhotic rats and cirrhotic rats treated with IGF-Ⅰtreatment (2 μg/100 g bw/d). Mitochondrial function was analyzed by flow cytometry in isolated hepatic mitochondria, caspase 3 activation was assessed by Western blot and apoptosis by TUNEL in the three experimental groups. RESULTS: Untreated cirrhotic rats showed a mitochon- drial dysfunction characterized by a significant reduction of mitochondrial membrane potential (in status 4 and 3); an increase of intramitochondrial reactive oxigen species (ROS) generation and a significant reduction of ATPase activity. IGF-Ⅰtherapy normalized mitochondrial func-tion by increasing the membrane potential and ATPase activity and reducing the intramitochondrial free radical production. Activity of the electron transport complexes Ⅰand Ⅲ was increased in both cirrhotic groups. In addition, untreated cirrhotic rats showed an increase of caspase 3 activation and apoptosis. | 3 | The effect of total carbon on microscopic soil properties and implications for crop production显示文摘Soil structure is a dynamic property affected by physical, chemical, and microbiological processes. Addition of organic matter to soils and the use of different management practices have been reported to impact soil structure and crop production. Moderation in soil temperature and increases in microbial activity and soil water retention are often suggested as reasons for the rise in crop yield when organic matter is added to the soil. Less is known about the direct effect of changes in soil structure on crop production. A field experiment was conducted to study the effect of summer cover crop and in-season management system on soil structure. The experiment was a nested design with summer cover crop as the main plot and management system as the subplot. Summer cover crop treatments included cowpea (Vigna unguiculata L. Walp.) incorporated into the soil in the fall (CI), cowpea used as mulch in the fall (CM), sudangrass (Sorghum vulgare) incorporated into the soil in the fall (S), and dry fallow or bare ground (B). Management systems were organic (ORG) and conventional (CNV) systems. Lettuce (Lactuca sativa L.) and cantaloupes (Cucumis melo L.) were cultivated in rotation in the plots for three consecutive years using the same cover crops and management systems for each plot. Disturbed and undisturbed soil cores were collected at the end of the third year and used for laboratory experiments to measure physical, chemical, and hydraulic properties. Image analysis was used to quantify soil structure properties using a scanning electron microscope on thin sections prepared from the undisturbed soil cores. We found that total soil carbon was correlated with porosity, saturation percentage, and pore roughness. Pore roughness was correlated with crop production in general and with marketable production in particular. Previous results from our laboratory showed that IGF-1 deficiency might play a pathogenetic role in hypogonadism of cirrhosis. The administration of IGF-1 for a short period of time reverted the testicular atrophy associated with advanced experimental cirrhosis.The aim of this study was to establish the historical progression of the described alterations in the testes,explore testicular morphology, histopathology, cellular proliferation, integrity of testicular barrier and hypophysogonadal axis in rats with no ascitic cirrhosis.METHODS: Male Wistar rats with histologically-proven cirrhosis induced with carbon tetrachloride (CC14) for 11 wk,were allocated into two groups (n = 12, each) to receive recombinant IGF-1 (2 μg/100 g·d, sc) for two weeks or vehicle. Healthy rats receiving vehicle were used as control group (n = 12).RESULTS: Compared to controls, rats with compensated cirrhosis showed a normal testicular size and weight and very few histopathological testicular abnormalities.However, these animals showed a significant diminution of cellular proliferation and a reduction of testicular transferrin expression. In addition, pituitary-gonadal axis was altered, with significant higher levels of FSH (P<0.001 vs controls) and increased levels of LH in untreated cirrhotic animals.
