1Hydrogen Sulfide Alleviates Aluminum Toxicity in Germinating Wheat Seedlings显示文摘Protective role of hydrogen sulfide(H2S) on seed germination and seedling growth was studied in wheat(Triticum) seeds subjected to aluminum(Al3+) stress.We show that germination and seedling growth of wheat is inhibited by high concentrations of AlCl3.At 30 mmol/L AlCl3 germination is reduced by about 50% and seedling growth is more dramatically inhibited by this treatment.Pre-incubation of wheat seeds in the H2S donor NaHS alleviates AlCl3-induced stress in a dose-dependant manner at an optimal concentration of 0.3 mmol/L.We verified that the role of NaHS in alleviating Al3+ stress could be attributed to H2S/HS-by showing that the level of endogenous H2S increased following NaHS treatment.Furthermore,other sodium salts containing sulfur were ineffective in alleviating Al3+ stress.NaHS pretreatment significantly increased the activities of amylases and esterases and sustained much lower levels of MDA and H2O2 in germinating seeds under Al3+ stress.Moreover,NaHS pretreatment increased the activities of guaiacol peroxidase,ascorbate peroxidase,superoxide dismutase and catalase and decreased that of lipoxygenase.NaHS pretreatment also decreased the uptake of Al3+ in AlCl3-treated seed.Taken together these results suggest that H2S could increase antioxidant capability in wheat seeds leading to the alleviation of Al3+ stress.Hua Zhang Zhu-Qin Tan Lan-Ying Hu Song-Hua Wang Jian-Ping Luo Russell L. 2Combination of Total Astragalus Extract and Total Panax Notoginseng Saponins Strengthened the Protective Effects on Brain Damage through Improving Energy Metabolism and Inhibiting Apoptosis after Cerebral Ischemia-Reperfusion in Mice显示文摘Objective:To explore the effects and molecular mechanisms of the combination between total Astragalus extract(TAE) and total Panax notoginseng saponins(TPNS) against cerebral ischemiareperfusion injury.Methods:C57BL/6 mice were randomly divided into sham-operated group,model group,TAE(110 mg/kg) group,TPNS(115 mg/kg) group,TAE-TPNS combination group and Edaravone(4 mg/kg) group,treated for 4 days,then,cerebral ischemia-reperfusion injury was established by bilateral common carotid artery(CCA) ligation for 20 min followed by reperfusion for 1 and 24 h.Results:TPNS could increase adenosine triphosphate(ATP) level,TAE and TAE-TPNS combination increased ATP,adenosine diphosphate(ADP) contents and Na+-K+-ATPase activity,and the effects of TAE-TPNS combination were stronger than those of TAE or TPNS alone after reperfusion for 1 h.After reperfusion for 24 h,TAE,TPNS and TAE-TPNS combination significantly increased neurocyte survival rate and decreased the apoptosis rate as well as down-regulated the expression of phosphorylated c-June N-terminal kinase1/2(p-JNK1/2),cytochrome C(Cyt C),cysteine aspartic acid-specific protease(Caspase)-9 and Caspase-3.Furthermore,the effects in TAE-TPNS combination were better than those in TAE or TPNS alone.Conclusion:The combination of TAE 110 mg/kg and TPNS 115 mg/kg could strengthen protective effects on cerebral ischemia injury,the mechanism underlying might be related to improving jointly the early energy metabolism,and relieving the delayed apoptosis via inhibiting the mitochondrial apoptosis pathway of JNK signal transduction.HUANG Xiao-ping TAN Hua CHEN Bei-yang DENG Chang-qing 2017Chinese Journal of Integrative Medicine2017,23,6:46

5Regulation of multidrug resistance by microRNAs in anti-cancer therapy显示文摘Multidrug resistance(MDR) remains a major clinical obstacle to successful cancer treatment.Although diverse mechanisms of MDR have been well elucidated, such as dysregulation of drugs transporters, defects of apoptosis and autophagy machinery, alterations of drug metabolism and drug targets, disrupti on of redox homeostasis, the exact mechanisms of MDR in a specific cancer patient and the cross-talk among these different mechanisms and how they are regulated are poorly understood.Micro RNAs(mi RNAs) are a new class of small noncoding RNAs that could control the global activity of the cell by post-transcriptionally regulating a large variety of target genes and proteins expression.Accumulating evidence shows that mi RNAs play a key regulatory role in MDR through modulating various drug resistant mechanisms mentioned above, thereby holding much promise for developing novel and more effective individualized therapies for cancer treatment. 6Mineral Coated Fertilizer Effect on Nitrogen-Use Efficiency and Yield of Wheat显示文摘A field experiment with winter wheat (Triticum aestivum L.) was conducted on a silt loam calcaric endorusti-ustic Cambosols derived from the Yellow River alluvial deposits in Henan, China, from 2001 to 2002 to evaluate N recovery and agronomic performance of different mineral coated fertilizers (MiCFs) compared to normal urea used in wheat cropping systems under field conditions. Five treatments, including CK (check, no N fertilizer), urea and three different MiCFs at an equivalent N application rate were established in a randomized complete block design. N release from MiCFs in soil was more synchronous with the N requirement of wheat throughout the growth stages than that from urea, with grain yield of the MiCF treatments significantly higher (P < 0.05) than that of the treatment urea. Correspondingly, the N recovery rate was greater for all MiCFs compared to urea, increasing from 32.8% up to 50.1%. 7Simvastatin inhibits apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax显示文摘BACKGROUND: Many studies have showed that apoptosis of endothelial cells plays a curial role in the progress of sepsis. But the role of simvastatin in apoptosis of endothelial cells induced by sepsis is not clear. The present study aimed to investigate the role of simvastatin in apoptosis of endothelial cells induced by sepsis and its mechanism.METHODS: Human umbilical vein endothelial cells(HUVECs) were randomly divided into three groups: control group, sepsis serum intervention group(sepsis group) and simvastatin+sepsis serum intervention group(simvastatin group). After 24-hour incubation with corresponding culture medium, the relative growth rate of HUVECS in different groups was detected by MTT assay; the apoptosis of HUVECs was detected by Hoechst33258 assay and fl ow cytometry; and the expression of the Bcl-2 and Bax genes of HUVECs was detected by PCR.RESULTS: Compared with the sepsis group, HUVECs in the simvastatin group had a higher relative growth rate. Apoptotic HUVECs decreased significantly in the simvastatin group in comparison with the sepsis group. Expression of the Bcl-2 gene in HUVECs decreased obviously, but the expression of the Bax gene increased obviously after 24-hour incubation with sepsis serum; however, the expression of the Bcl-2 and Bax genes was just the opposite in the simvastatin group.CONCLUSIONS: Our study suggests that simvastatin can inhibit apoptosis of endothelial cells induced by sepsis through upregulating the expression of Bcl-2 and downregulating Bax. 8Antigenic variation of the human influenza A(H3N2) virus during the 2014–2015 winter season显示文摘The human influenza A(H3N2) virus dominated the 2014–2015 winter season in many countries and caused massive morbidity and mortality because of its antigenic variation. So far, very little is known about the antigenic patterns of the recent H3N2 virus. By systematically mapping the antigenic relationships of H3N2 strains isolat ed since 2010, we discov ered that two groups with obvious antigenic divergence, named SW13(A/Switzerland/9715293/2013-like strains) and HK14(A/Hong Kong/5738/2014-like strains), co-circulated during the 2014–2015 winter season. HK14 group co-circulated with SW13 in Europe and the United States during this season, while there were few strains of HK14 in China's Mainland, where SW13 has dominated since 2012. Furthermore, we found that substitutions near the receptor-binding site on hemagglutinin played an important role in the antigenic variation of both the groups. 10The herbal compound geniposide rescues formaldehyde-induced apoptosis in N2a neuroblastoma cells显示文摘The herbal medicine Tong Luo Jiu Nao(TLJN)contains geniposide(GP)and ginsenoside Rg1 at a molar ratio of 10:1.Rg1 is the major component of another herbal medicine,panax notoginseng saponin(PNS).TLJN has been shown to strengthen brain function in humans,and in animals it improves learning and memory.We have previously shown that TLJN reduces amyloidogenic processing in Alzheimer’s disease(AD)mouse models.Together this suggests TLJN may be a potential treatment for patients with dementia.Because chronic damage of the central nervous system by formaldehyde(FA)has been presented as a risk factor for age-associated cognitive dysfunction,in the present study we investigated the protective effect of both TLJN and GP in neuron-like cells exposed to FA.FA-exposed murine N2a neuroblastoma cells were incubated with TLJN,its main ingredient GP,as well as PNS,to measure cell viability and morphology,the rate of apoptosis and expression of genes encoding Akt,FOXO3,Bcl2 and p53.The CCK-8 assay,cytoskeletal staining and flow cytometry were used to test cell viability,morphology and apoptosis,respectively.Fluorescent quantitative real-time PCR(qRT-PCR)was used to monitor changes in gene expression,and HPLC to determine the rate of FA clearance.Treatment of N2a cells with 0.09 mmol L?1 FA for 24 h significantly reduced cell viability,changed cell morphology and promoted apoptosis.Both TLJN and GP conferred neuroprotection to FA-treated N2a cells,whereas PNS,which had to be used at lower concentrations because of its toxicity,did not.Our data demonstrate that TLJN can rescue neuronal damage caused by FA and that its main ingredient,GP,has a major role in this efficacy.This presents purified GP as a drug or lead compound for the treatment of AD.CHEN JinYan SUN MengRu WANG XingHua LU Jing WEI Yan TAN Yan LIU Ying GTZ Jürgen HE RongQiao HUA Qian 2014Science China(Life Sciences)2014,57,4:15
11Influence of α/β interface phase on the tensile properties of laser cladding deposited Ti–6Al–4V titanium alloy显示文摘Laser cladding deposited Ti–6Al–4V titanium alloy universally shows more complex microstructures,each of which has significant effect on mechanical properties. Of particular α/β interface phase has been observed in this paper under certain conditions. It demonstrates that the influence of the α/β interface phase on the tensile properties is closely associated with dislocations and twin substructure through comparison experiments. The results show that the α/β interface phase hinders dislocation motion and decreases effective slip length. In addition, the twin substructure has been activated in the α/β interface phase during tensile process and has acted somehow like grain boundaries. Therefore, the strength and the work-hardening rate of the laser cladding deposited Ti–6Al–4V titanium alloy have been significantly improved due to the dynamic Hall–Petch effect. Besides, the α/β interface phase leads to more uniform dislocations distribution, which implies that relative lower local concentrated stress will be produced along the α/β interface phase or colony boundary after the same amount of plastic deformation. Moreover,the twinning-induced plasticity effects in the α/β interface phase further increase the plastic deformation capacity. 12Modeling framework of voltage-source converters based on equivalence with synchronous generator显示文摘Along with the increasing penetration of distributed generation with voltage-source converters(VSCs),there are extensive concerns over the potential virtual rotor angle stability, which is characterized by oscillations of power and frequency during the dynamic process of synchronization in the grid. Several control strategies have been developed for VSCs to emulate rotating inertia as well as damping of oscillations. This paper classifies these strategies and provides a small-signal modeling framework including all kinds of VSCs in different applications for virtual rotor angle stability. A unified perspective based on the famous Phillips–Heffron model is established for various VSCs. Thus, the concepts of equivalent inertia and the synchronizing and damping coefficients in different VSCs are highlighted, based on the similarities with the synchronous generator(SG) system in both physical mechanisms and mathematical models. It revealed the potentiality of various VSCs to achieve equivalence with the SG. This study helps promote the unity of VSCs and traditional SGs in both theories and methods for analyzing the dynamic behavior and enhancing the stability. 14Comparison of cell salvage with one and two suction devices during cesarean section in patients with placenta previa and/or accrete: a randomized controlled trial显示文摘Background:Cell salvage has recently been recommended for obstetric use in cases with a high risk of massive hemorrhage during cesarean section(CS).However,limited data are available to support the use of one suction device to collect lost blood.This study aimed to investigate the volume of red blood cells(RBCs)salvaged and the components of amniotic fluid(AF)in blood salvaged by one suction device or two devices during CS in patients with placenta previa and/or accrete.Methods:Thirty patients with placenta previa and/or accrete undergoing elective CS in the Women's Hospital of Zhejiang University School of Medicine were recruited for the present study from November 1,2017 to December 1,2018.The patients were randomly assigned to one of the two groups according to an Excel-generated random number sheet:Group 1(w=15),in which only one suction device was used to aspirate all blood and AF,and Group 2(w=15),in which a second suction device was mainly used to aspirate AF before the delivery of the placenta.Three samples of blood per patient(pre-wash,post-wash,and post-filtration)were collected to measure AF components.The salvaged RBC volumes were recorded.Continuous data of pre-wash,post-wash,and postfiltration samples were analyzed by using one-way analysis of variance with Tukey5s test for multiple comparisons,or Kruskal-Wallis test with Dunn test for multiple comparisons.Comparisons of continuous data between Group 1 and Group 2 were conducted using Student's t test or Mann-Whitney U test.Results:The salvaged RBC volume was significantly higher in Group 1 than that in Group 2(401.6±77.2 mL vs.330.1土53.3 mL,?=4.175,P<0.001).In both groups,squamous cells,lamellar bodies,and fat were significantly reduced by washing(all P<0.001)and squamous cells were further reduced by filtering(P<0.001).Squamous cells were found in six post-filtration samples(three from each group).Lamellar bodies and fat were completely removed by filtering.Insulin-like growth factor binding protein 1,alphafetoprotein,albumin,lactate dehydrogenase,and potassium were significantly reduced post-wash(all P<0.05),with no further significant reduction after filtration in either group(all P>0.05).The mean percentage of fetal RBCs post-filtration was(1.8±0.8)%with a range of 1.0%to 3.5%and(1.9±0.9)%with a range of 0.7%to 4.0%in Groups 1 and 2,respectively,showing no significant difference between the two groups(U=188.5,P=0.651).Conclusion:Cell salvage performed by one suction device could result in higher volume of salvaged RBCs and can be used safely for CS in patients with placenta previa and/or accrete when massive hemorrhage occurs.Trial registration number:ChiCTR-INR-17012926,http://gffzz62a03ee25e6f4a77ho6wk5wvqqv6566xu.ffgz.tsg.suse.edu.cn/Chinese Clinical Trial Registry.Hong Chen Hua Tan Pei-Xin Luo Yi-Fang Shen Chang-Cheng Lyu Xiao-Wei Qian Xin-Zhong Chen 2020Chinese Medical Journal2020,,6:12
15Garcinol inhibits esophageal cancer metastasis by suppressing the p300 and TGF-β1 signaling pathways显示文摘Metastasis causes the main lethality in esophageal cancer patient.Garcinol,a natural compound extracted from Gambogic genera,is a histone acetyltransferase(HAT)inhibitor that has shown anticancer activities such as cell cycle arrest and apoptosis induction.In this study,we investigated the effects of garcinol on the metastasis of esophageal cancer in vitro and in vivo.We found that garcinol(5-15μM)dose-dependently inhibited the migration and invasion of human esophageal cancer cell lines KYSE150 and KYSE450 in wound healing,transwell migration,and Matrigel invasion assays.Furthermore,garcinol treatment dose-dependently decreased the protein levels of p300/CBP(transcriptional cofactors and HATs)and p-Smad2/3 expression in the nucleus,thus impeding tumor cell proliferation and metastasis.Knockdown of p300 could inhibit cell metastasis,but CBP knockdown did not affect the cell mobility.It has been reported that TGF-β1 stimulated the phosphorylation of Smad2/3,which directly interact with p300/CBP in the nucleus,and upregulating HAT activity of p300.We showed that garcinol treatment dose-dependently suppressed TGF-β1-activated Smad and non-Smad pathway,inhibiting esophageal cancer cell metastasis.In a tail vein injection pulmonary metastasis mouse model,intraperitoneal administration of garcinol(20 mg/kg)or 5-FU(20 mg/kg)significantly decreased the number of lung tumor nodules and the expression levels of Ki-67,p300,and p-Smad2/3 in lung tissues.In conclusion,our study demonstrates that garcinol inhibits esophageal cancer metastasis in vitro and in vivo,which might be related to the suppression of p300 and TGF-β1 signaling pathways,suggesting the therapeutic potential of Garcinol for metastatic tumors.Jing Wang Man Wu Dan Zheng Hong Zhang Yue Lv Li Zhang Hong-sheng Tan Hua Zhou Yuan-zhi Lao Hong-xi Xu 2020Acta Pharmacologica Sinica2020,41,1:12
16The enhanced X-ray Timing and Polarimetry mission—eXTP显示文摘In this paper we present the enhanced X-ray Timing and Polarimetry mission—eXTP. eXTP is a space science mission designed to study fundamental physics under extreme conditions of density, gravity and magnetism. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. 19The prognostic impact of traditional Chinese medicine monomers on tumor-associated macrophages in non-small cell lung cancer显示文摘Non-small cell lung cancer(NSCLC) accounts for 80%-85% of all lung malignancies and good diagnosis and prognosis of NSCLC are critical to the increase of its survival rate. Tumor-associated macrophages(TAM) abundantly present in numerous cancer types, and the role of TAMs in tumor biology and their prognostic value in cancer become major topics of interest. After various stimulations in the tumor microenvironment, TAMs develop into a M1(tumor-inhibitory) phenotype or M2(tumor-promoting) phenotype. Recent studies show that traditional Chinese medicine(TCM) monomers have markedly inhibitory actions for NSCLC through M1/M2 modulation. Due to the TCM monomers mainly covered five categories, i.e. terpenoids, flavonoids, polysaccharides, natural polyphenols, and alkaloids. Thus, we will discuss the regulation of TCM monomers on TAM involve in these five parts in this review. 20Neddylation of PTEN regulates its nuclear import and promotes tumor development显示文摘PTEN tumor suppressor opposes the PI3K/Akt signaling pathway in the cytoplasm and maintains chromosomal integrity in the nucleus.Nucleus–cytoplasm shuttling of PTEN is regulated by ubiquitylation,SUMOylation and phosphorylation,and nuclear PTEN has been proposed to exhibit tumor-suppressive functions.Here we show that PTEN is conjugated by Nedd8 under high glucose conditions,which induces PTEN nuclear import without effects on PTEN stability.PTEN neddylation is promoted by the XIAP ligase and removed by the NEDP1 deneddylase.We identify Lys197 and Lys402 as major neddylation sites on PTEN.Neddylated PTEN accumulates predominantly in the nucleus and promotes rather than suppresses cell proliferation and metabolism.The nuclear neddylated PTEN dephosphorylates the fatty acid synthase(FASN)protein,inhibits the TRIM21-mediated ubiquitylation and degradation of FASN,and then promotes de novo fatty acid synthesis.In human breast cancer tissues,neddylated PTEN correlates with tumor progression and poor prognosis.Therefore,we demonstrate a previously unidentified pool of nuclear PTEN in the Nedd8-conjugated form and an unexpected tumor-promoting role of neddylated PTEN.Ping Xie Zhiqiang Peng Yujiao Chen Hongchang Li Mengge Du Yawen Tan Xin Zhang Zhe Lu Chun-Ping Cui Cui Hua Liu Fuchu He Lingqiang Zhang 2021Cell Research2021,31,3:11
