1miR-7/TGF-β2 axis sustains acidic tumor microenvironment-induced lung cancer metastasis显示文摘Acidosis,regardless of hypoxia involvement,is recognized as a chronic and harsh tumor microenvironment(TME)that educates malignant cells to thrive and metastasize.Although overwhelming evidence supports an acidic environment as a driver or ubiquitous hallmark of cancer progression,the unrevealed core mechanisms underlying the direct effect of acidification on tumorigenesis have hindered the discovery of novel therapeutic targets and clinical therapy.Here,chemical-induced and transgenic mouse models for colon,liver and lung cancer were established,respectively.miR-7 and TGF-β2 expressions were examined in clinical tissues(n=184).RNA-seq,miRNA-seq,proteomics,biosynthesis analyses and functional studies were performed to validate the mechanisms involved in the acidic TME-induced lung cancer metastasis.Our data show that lung cancer is sensitive to the increased acidification of TME,and acidic TME-induced lung cancer metastasis via inhibition of miR-7-5 p.TGF-β2 is a direct target of miR-7-5 p.The reduced expression of miR-7-5 p subsequently increases the expression of TGF-β2 which enhances the metastatic potential of the lung cancer.Indeed,overexpression of miR-7-5 p reduces the acidic p H-enhanced lung cancer metastasis.Furthermore,the human lung tumor samples also show a reduced miR-7-5 p expression but an elevated level of activated TGF-β2;the expressions of both miR-7-5 p and TGF-β2 are correlated with patients’survival.We are the first to identify the role of the miR-7/TGF-β2 axis in acidic p H-enhanced lung cancer metastasis.Our study not only delineates how acidification directly affects tumorigenesis,but also suggests miR-7 is a novel reliable biomarker for acidic TME and a novel therapeutic target for non-small cell lung cancer(NSCLC)treatment.Our study opens an avenue to explore the p H-sensitive subcellular components as novel therapeutic targets for cancer treatment.Tao Su Suchao Huang Yanmin Zhang Yajuan Guo Shuwei Zhang Jiaji Guan Mingjing Meng Linxin Liu Caiyan Wang Dihua Yu Hiu-Yee Kwan Zhiying Huang Qiuju Huang Elaine Lai-Han Leung Ming Hu Ying Wang Zhongqiu Liu Linlin Lu 2022Acta Pharmaceutica Sinica B2022,12,2:2

4Experimental study of two saturated natural soils and their saturated remoulded soils under three consolidated undrained stress paths显示文摘In this paper,an experimental investigation is conducted to study the mechanical behavior of saturated natural loess,saturated natural filling in ground fissure and their corresponding saturated remoulded soils under three consolidated undrained triaxial stress tests,namely,conventional triaxial compression test(CTC),triaxial compression test(TC)and reduced triaxial compression test(RTC).The test results show that stress-strain relation,i.e.strain-softening or strain-hardening,is remarkably influenced by the structure,void ratio,stress path and confining pressure.Natural structure,high void ratio,TC stress path,RTC stress path and low confining pressures are favorable factors leading to strain-softening.Excess pore pressure during shearing is significantly affected by stress path.The tested soils are different from loose sand on character of strain-softening and are different from common clay on excess pore water pressure behavior.The critical states in p′-q space in CTC,TC and RTC tests almost lie on one line,which indicates that the critical state is independent of the above stress paths.As for remoulded loess or remoulded filling,the critical state line(CSL)and isotropic consolidation line(ICL)in e-log p′space are almost straight,while for natural loess or natural filling,in e-log p′space there is a turning point on the CSL,which is similar to the ICL.Mingjing JIANG Haijun HU Jianbing PENG Serge LEROUEIL 2011Frontiers of Structural and Civil Engineering2011,5,2:1

6Two chemical mutagens modulate the seed germination,growth,and phenotypic characteristics of Chinese fir(Cunninghamia lanceolata)显示文摘Chinese fir(Cunninghamia lanceolata(Lamb.)Hook),a fast-growing and economically important timber tree species in China,is widely used in construction,furniture,and paper manufacture but has a long breeding cycle.Chemical mutagens,such as ethyl methane sulfonate(EMS)and sodium azide(SA),are widely used in crops such as rice,wheat,cotton,soybean and sugarcane but their utility for tree breeding is unknown.In this study we examined the effects of EMS and S A on Chinese fir seed germination and growth.Chinese fir seeds were treated with the two chemical mutagens;were planted in Jiangle County,Fujian Province,China;and their heights were measured from 2011 to2017.The concentrations and durations of treatment with the two chemical mutagens were significantly associated with the Chinese fir seedling and mortality rates,as well as with the heights of trees from the seedling stage to 3 years old.We also generated 127 mutants with abnormal branches and reproductive growth.We report here the effects of two chemical mutagens on Chinese fir breeding;our data will contribute to knowledge of the utility of EMS and SA in forestry.Sen Cao Ruiyang Hu Xialei Wu Yuhan Sun Bo Wu Hongjing Duan Huazhong Lin Mingjing Wu Luming Fang Xiaolong Yu Wei Wu Yun Li 2021Journal of Forestry Research2021,32,5:0
7Investigation of Optimal Megnetic Properties in NdFeB Magnets by Artificial Neural Network显示文摘In order to study the effect of alloy component on magnetic properties of NdFeB magnets, the experiment schemes are carried out by the uniform design theory, and the relationship between the component and the magnetic properties is established by artificial neural network(ANN) predicting model.The element contents of alloys are optimized by the ANN model.Meanwhile, the influences of mono-factor or multi-factor interaction on alloy magnetic properties are respectively discussed according to the curves ploted by ANN model.Simulation result shows that the predicted and measured results are in good agreement.The relative error is every low, the error is not more than 1.68% for remanence Br, 1.56% for maximal energy product (BH)m, and 7.73% for coercivity Hcj.Hcj can be obviously improved and Br can be reduced by increasing Nd or Zr content.Co and B have advantageous effects on increasing Br and disadvantageous effects on increasing Hcj.Influence of alloying elements on Hcj and Br are inverse, and the interaction among the alloying elements play an important role in the magnetic properties of NdFeB magnets.The ANN prediction model presents a new approach to investigate the nonlinear relationship between the component and the magnetic properties of NdFeB alloys.Lian Lixian Liu Ying Hu Wang Hou Tinghong Gao Shengji Tu Mingjing 2004Journal of Rare Earths2004,22,z1:0
8Inhibition of cystathionine β-synthase promotes apoptosis and reduces cell proliferation in chronic myeloid leukemia显示文摘Increased endogenous hydrogen sulfide(H_(2)S)level by cystathionine β-synthase(CBS)has been shown to closely relate tumorigenesis.H_(2)S promotes angiogenesis,stimulates bioenergy metabolism and inhibits selective phosphatases.However,the role of CBS and H_(2)S in chronic myeloid leukemia(CML)remains elusive.In this study,we found that CBS and H_(2)S levels were increased in the bone marrow mononuclear cells of pediatric CML patients,as well as in the CML-derived K562 cells and CBS expression levels were correlated with different disease phases.Inhibition of CBS reduced the proliferation of the CML primary bone marrow mononuclear cells and induced growth inhibition,apoptosis,cell cycle arrest,and migration suppression in K562 cells and tumor xenografts.The knockdown of CBS expression by shRNA and inhibiting CBS activity by AOAA decreased the endogenous H_(2)S levels,promoted mitochondrial-related apoptosis and inhibited the NF-xB-mediated gene expression.Our study suggests that inhibition of CBS induces cell apoptosis,as well as limits cell proliferation and migration,a potential target for the treatment of chronic myeloid leukemia.Dan Wang Huan Yang Yun Zhang Rong Hu Dongjie Hu Qunxian Wang Yannan Liu Mingjing Liu Zijun Meng Weihui Zhou Weihong Song 2021Signal Transduction and Targeted Therapy2021,6,3:0
9Safety and activity of WX-0593(Iruplinalkib)in patients with ALK-or ROS1-rearranged advanced non-small cell lung cancer:a phase 1 dose-escalation and dose-expansion trial显示文摘WX-0593(Iruplinalkib)is a novel,highly selective oral ALK and ROS1 tyrosine kinase inhibitor(TKI).In this study,the safety,antitumor activity,and pharmacokinetics of WX-0593 were evaluated in advanced non-small cell lung cancer(NSCLC)patients with ALK or ROS1 rearrangement.In the dose-escalation phase and dose-expansion phase,patients were treated with WX-0593 until disease progression,unacceptable toxicity,or subject withdrawal.In the dose-escalation phase,the primary endpoints were maximum tolerated dose(MTD),dose-limiting toxicity(DLT),and safety assessed by investigators.In the dose-expansion phase,the primary endpoint was objective response rate(ORR)assessed by investigators.Between September 25,2017 and October 15,2018,a total of 153 patients received WX-0593 treatment.Two dose-limiting toxicities(DLTs)including one grade 3 QT interval prolonged and one grade 2 chronic heart failure were reported at the dose of 300 mg in one patient.MTD was not reached.Overall,140 of the 152(92%)patients experienced treatment-related adverse events(TRAEs)and 35 of the 152(23%)patients had TRAEs≥grade 3.The overall ORR was 59.3%(32 of 54)for the dose-escalation phase and 56.6%(56 of 99)for the dose-expansion phase.For patients who were ALK-rearranged and ALK TKI naive,the ORR were 81.0%(17 of 21)in the dose-escalation phase and 76.3%(29 of 38)in the dose-expansion phase,and for patients who previously received crizotinib as the only ALK TKI,the ORR were 38.1%(8 of 21)and 45.7%(21 of 46)for the two phases,respectively.For patients who were ROS1-rearranged,the ORR were 30.0%(3 of 10)in the dose-escalation phase and 44.4%(4 of 9)in the dose-expansion phase.WX-0593 showed favorable safety and promising antitumor activity in advanced NSCLC patients with ALK or ROS1 rearrangement.Yuankai Shi Jian Fang Xuezhi Hao Shucai Zhang Yunpeng Liu Lin Wang Jianhua Chen Yi Hu Xiaosheng Hang Juan Li Chunling Liu Yiping Zhang Zhehai Wang Yanping Hu Kangsheng Gu Jian’an Huang Liangming Zhang Jinlu Shan Weiwei Ouyang Yanqiu Zhao Wu Zhuang Yan Yu Jun Zhao Helong Zhang Pei Lu Weidong Li Meimei Si Mingjing Ge Huaize Geng 2022Signal Transduction and Targeted Therapy2022,7,2:0
