1Wharton's jelly mesenchymal stem cells differentiate into retinal progenitor cells显示文摘Human Wharton's jelly mesenchymal stem cells were isolated from fetal umbilical cord. Cells were cultured in serum-free neural stem cell-conditioned medium or neural stem cell-conditioned medium supplemented with Dkk-1, a Wnt/β catenin pathway antagonist, and LeftyA, a Nodal signaling pathway antagonist to induce differentiation into retinal progenitor cells. Inverted microscopy showed that after induction, the spindle-shaped or fibroblast-like Wharton's jelly mesenchymal stem cells changed into bulbous cells with numerous processes. Immunofluorescent cytochemical staining and reverse-transcription PCR showed positive expression of retinal progenitor cell markers, Pax6 and Rx, as well as weakly down-regulated nestin expression. 2Preparation and characterization of 3-aminopropyl-triethoxysilane grafted montmorillonite and acid-activated montmorillonite显示文摘3-aminopropyltriethoxysilane grafted montmorillonites were synthesized with montmorillonite, acidactivated montmorillonites and 3-aminopropyltriethoxysilane in ethanol-water mixture. The resulting products were investigated using Fourier transform infrared (FTIR) spectroscopy, X-ray diffraction (XRD), thermogravimetric analysis (TG). XRD patterns demonstrated that silane was intercalated into the montmorillonite gallery, indicated by increasing basal spacings. These intercalated silanes probably adopt bilayer arrangement models within the montmorillonite and acid-activated montmorillonites interlayer spaces. TG and DTG curves indicate that acidification results in a decrease of the thermal stability of the resultant montmorillonites. 4Numerical study of three-dimensional wave-induced longshore current's effects on sediment spreading of the Huanghe River mouth显示文摘A three-dimensional wave radiation stress is introduced into the hydrodynamic sediment coupled model COHERENS-SED,which has been developed through introducing wave-enhanced bottom shear stress,wave dependent surface drag coefficient,wave-induced surface mixing,SWAN,damping function of sediment on turbulence,sediment model and depth-dependent wave radiation stress to COHERENS.The COHERENS-SED is adopted to study the effects induced by wave-induced three-dimensional longshore current on suspended sediment spreading of the Huanghe River (Yellow River) mouth.Several different cases divided by setting different wave parameters of inputting boundary waves are carried out.The modeling results agree with measurement data.In terms of simulation results,it is easy to know that three-dimensional wave radiation stress plays an obvious role when inputting boundary wave height is stronger than 3 m.Moreover,wave direction also affects the sediment spreading rules of the mouth strongly too.LIANG Bingchen ZHAO Hongping LI Huajun WU Guoxiang 2012Acta Oceanologica Sinica2012,31,2:4
5Effect of Honghua (Flos Carthami) on nitric oxide production in RAW 264.7 cells and α-glucosidase activity显示文摘OBJECTIVE: To study the effects of extracts from Honghua(Flos Carthami) on lipopolysaccharide induced nitric oxide(NO) production in RAW 264.7cells and the influence of the extracts on yeastα-glucosidase activity. The total flavonoid content of the extracts was also determined.METHODS: Cytotoxicity of the extracts to RAW264.7 cells was evaluated by the ATPlite? method.Inhibitory effects of the extracts on NO production were evaluated by Griess assay. Curcumin was used as a positive control. Screening of extracts for potential α-glucosidase inhibitors was done by a fluorometric assay.The assay was based on the hydrolysis of 4-methylumbelliferyl-a-D-glucopyranoside to form the fluorescent product, 4-methylumbelliferone. Acarbose was used as a positive control. The total flavonoid content was tested using kaempferol as the standard.RESULTS: There were significant inhibitory effects on NO production when the extracts were 25-100μg/mL(P<0.05) and curcumin was 2-4 μg/mL(P<0.001). The extracts showed an inhibitory effect onα-glucosidase activity at the concentrations of15.6-125 μg/mL with a half maximal(50%) inhibitory concentration(IC50) of(32.8±5.7) μg/mL, compared with the IC 50 of acarbose at(1.8±0.4) μg/mL.There was a significant difference between the two IC50values(P<0.001). The total content of flavonoids per gram of dried herb was1.14 mg.CONCLUSION:Honghua(Flos Carthami)showed in hibitory effects on NO production in activated RAW264.7 macrophage cells and an inhibitory effect on yeast α-glucosidase. 9Cell osteogenic bioactivity mediated precisely by varying scaled micro-pits on ordered micro/nano hierarchical structures of titanium显示文摘Hierarchical surface structures with micro–nano scale play a crucial role in regulation of cell proliferation and osteogenic differentiation.It has been proven that cells are extremely sensitive to the nanoscaled structure and show multifarious phenotypes.Though a vital function of microstructure on osseointegration has been confirmed,the cell performances response to different microscaled structure is needed to be further dissected and in depth understood.In this work,the ordered micro–nano hierarchical structures with varying micro-scaled pits were precisely fabricated on titanium successfully by the combination of electrochemical,chemical etching and anodization as well.In vitro systematical assessments indicated that the micro–nano multilevel structures on titanium exhibited excellent cells adhesion and spreading ability,as well as steerable proliferation and osteogenic differentiation behaviors.It is shown that smaller micro-pits and lower roughness of the hierarchical structures enabled faster cell propagation.Despite cell growth was delayed on micro–nano titanium with relatively larger cell-match-size micro-pits and roughness,osteogenic-specific genes were significantly elevated.Furthermore,the alkaline phosphatase activity,collagen secretion and extracellular matrix mineralization of MC3T3-E1 on multiscaled titanium were suppressed by a large margin after adding IWP-2(an inhibitor of Wnt/b-catenin signal pathway),indicating this pathway played a crucial part in cell osteogenic differentiation modulated by micro–nano structures.Yanmei Zhang Xiankuan Wang Yaxian Li Jianhe Liang Pinliang Jiang Qiaoling Huang Yun Yang Hongping Duan Xiang Dong Gang Rui Changjian Lin 2022Regenerative Biomaterials2022,9,1:1

14Constraint of Isomorphous Substitution in Magnetite on Its Catalytic Activity in Heterogeneous Fenton Reaction显示文摘In nature, iron cations in magnetite are usually isomorphically substituted by divalent (Co, Ni, Zn, Cu, Mn, etc.), trivalent (Al, V, Cr, etc.) and tetravalent (Ti) cations while maintaining the spinel structure. 16Woody Plants as Resources in China, Their Conservation and Use显示文摘This paper deals with species of the woody plants in China and their conservation problems. The floristic elements of woody plants was computed and the results indicate that the elements of Tropical Asia accounts for the largest number of genus while the proportion of the elements of Temperate Asia is relatively small. For each of eight vegetation region, the number of woody plants was established and a list of the endangered and rare species was compiled. 17Rapid Acquisition of High-Quality SARS-CoV-2 Genome via Amplicon-Oxford Nanopore Sequencing显示文摘Genome sequencing has shown strong capabilities in the initial stages of the COVID-19 pandemic such as pathogen identification and virus preliminary tracing.While the rapid acquisition of SARS-Co V-2 genome from clinical specimens is limited by their low nucleic acid load and the complexity of the nucleic acid background.To address this issue,we modified and evaluated an approach by utilizing SARS-Co V-2-specific amplicon amplification and Oxford Nanopore Prometh ION platform.This workflow started with the throat swab of the COVID-19 patient,combined reverse transcript PCR,and multi-amplification in one-step to shorten the experiment time,then can quickly and steadily obtain high-quality SARS-Co V-2 genome within 24 h.A comprehensive evaluation of the method was conducted in 42 samples:the sequencing quality of the method was correlated well with the viral load of the samples;high-quality SARS-Co V-2 genome could be obtained stably in the samples with Ct value up to 39.14;data yielding for different Ct values were assessed and the recommended sequencing time was 8 h for samples with Ct value of less than 20;variation analysis indicated that the method can detect the existing and emerging genomic mutations as well;Illumina sequencing verified that ultra-deep sequencing can greatly improve the single read error rate of Nanopore sequencing,making it as low as 0.4/10,000 bp.In summary,high-quality SARS-Co V-2 genome can be acquired by utilizing the amplicon amplification and it is an effective method in accelerating the acquisition of genetic resources and tracking the genome diversity of SARSCo V-2.Yi Yan Ke Wu Jun Chen Haizhou Liu Yi Huang Yong Zhang Jin Xiong Weipeng Quan Xin Wu Yu Liang Kunlun He Zhilong Jia Depeng Wang Di Liu Hongping Wei Jianjun Chen 2021Virologica Sinica2021,36,5:0
18Clinical Application of PES Membrane Dialyzer显示文摘Objective The dialysate membrane can be classified into acetate membrane and synthetic membrane,while the latter can be further divided into polysulfone,polyacrylonitrile,etc.Polyethersulfone membrane is high molecular membrane material with excellent biocompatibility,stable mechanical properties,high temperature and pressure resistance,and good cleaning performance,which has been applied for more than 10 years.This trial is to assess the safety and efficacy of hollow polysulfone membrane fiber dialyzer with pattern number Enttex^(TM)-16LF(E60)for hemodialysis in patients with acute or chronic renal failure due to multiple factors.Methods Totally 76 patients receiving maintenance hemodialysis were included in a two-site,random,open label,two-stage,positive crossover and non inferiority validation study.The clinical efficacy and safety of the test device were observed,which was a hollow fiber dialyzer produced by Guangzhou Enttex Medical Products Co.,Ltd.,with polyethersulfone membrane imported from Germany.It was a low flux dialyzer with the pattern number of EnttexTM-16LF(E60).Inclusion criteria:①patients with acute or chronic renal failure due to various reasons who needed hemodialysis;②18 to 80 years old,gender unlimited;③patients who received stable dialysis for more than 3 months,regular hemodialysis 3 times a week,4 h at each time,blood flow rate of 200~350ml/min;④using bicarbonate dialysate at a flow rate of 500ml/min;⑤patients with arteriovenous fistula or who received artificial blood vessel puncture dialysis;received anticoagulation by heparin or low molecular weight heparin.Exclusion criteria:①patients needing heparin free dialysis for severe anemia,tumor and active bleeding;②patients with severe cardiac(gradeⅢcardiac function),hepatic(severe hepatitis and cirrhosis)and pulmonary diseases(severe respiratory failure).Results Comparison of parameters,including the changes of Kt/V,general creatinine and urea clearance rates and URR(urea reduction rate)shows no statistical significance in differences between the two devices,indicating that the test and control devices can equally and effectively eliminate toxins including creatinine and urea,etc.PPP and MITTP results are consistent.Comparison of ultrafiltration rates between two groups of patients shows no statistical difference.PPP and MITTP results are consistent,indicating that both the test device and control device can effectively eliminate water.In MITTP,electrolytes including K^+,Na^+,Cl^-,Ca^2+,HCO3^-,etc.show no statistical significance in difference between the test and control devices before and after dialysis(P>0.05).After treating by using test and control devices,body weight,serum potassium and phos-phorus all significantly decrease(P<0.001);serum Ca^2+,PO2,PCO2and HCO3^-all significantly increase(P<0.001).These results indicate that both the test and control devices can effectively eliminate serum potassium and phosphorus,and eliminate excess water in body,maintain the balance between liquid and electrolytes in patients.PPP and MITTP results are consistent.Conclusion This clinical trial confirms that hollow polyethersulfone membrane fiber dialyzer can be safely and effectively used in hemodialysis therapy.YUN Chen YANG Peidian LIANG Wenxue CHEN Yu CHEN Gangyi LIU Hongping OUYANG Zihua SONG Huifeng LI Yunyi YU Zongchao GUAN Baozhang DONG Xiangnan LIU Huanhuan Cao Rui ZHANG Zeyu FAN Lijing HU Bo HE Yongcheng SHI Mingjie XU Jun Yang Lifeng TANG Shuifu 2019临床医学工程2019,26,S01:0
