5Transcriptomic landscape regulated by the 14 types of bone morphogenetic proteins(BMPs)in lineage commitment and differentiation of mesenchymal stem cells(MSCs)显示文摘Mesenchymal stem cells(MSCs)are ubiquitously-existing multipotent progenitors that can self-renew and differentiate into multiple lineages including osteocytes,chondrocytes,adipocytes,tenocytes and myocytes.MSCs represent one of the most commonly-used adult progenitors and serve as excellent progenitor cell models for investigating lineagespecific differentiation regulated by various cellular signaling pathways,such as bone morphogenetic proteins(BMPs).As members of TGFb superfamily,BMPs play diverse and important roles in development and adult tissues.At least 14 BMPs have been identified in mammals.Different BMPs exert distinct but overlapping biological functions.Through a comprehensive analysis of 14 BMPs in MSCs,we demonstrated that BMP9 is one of the most potent BMPs in inducing osteogenic differentiation of MSCs.Nonetheless,a global mechanistic view of BMP signaling in regulating the proliferation and differentiation of MSCs remains to be fully elucidated.Here,we conducted a comprehensive transcriptomic profiling in the MSCs stimulated by 14 types of BMPs.Hierarchical clustering analysis classifies 14 BMPs into three subclusters:an osteo/chondrogenic/adipogenic cluster,a tenogenic cluster,and BMP3 cluster.We also demonstrate that six BMPs(e.g.,BMP2,BMP3,BMP4,BMP7,BMP8,and BMP9)can induce ISmads effectively,while BMP2,BMP3,BMP4,BMP7,and BMP11 up-regulate Smad-independent MAP kinase pathway.Furthermore,we show that many BMPs can upregulate the expression of the signal mediators of Wnt,Notch and PI3K/AKT/mTOR pathways.While the reported transcriptomic changes need to be further validated,our expression profiling represents the first-of-its-kind to interrogate a comprehensive transcriptomic landscape regulated by the 14 types of BMPs in MSCs.Linghuan Zhang Qing Luo Yi Shu Zongyue Zeng Bo Huang Yixiao Feng Bo Zhang Xi Wang Yan Lei Zhenyu Ye Ling Zhao Daigui Cao Lijuan Yang Xian Chen Bin Liu William Wagstaff Russell R*Reid Hue H*Luu Rex C*Haydon Michael J*Lee Jennifer Moriatis Wolf Zhou Fu Tong-Chuan He Quan Kang 2019Genes & Diseases2019,6,3:11

7Connective tissue growth factor is overexpressed in human hepatocellular carcinoma and promotes cell invasion and growth显示文摘AIM:To determine the expression characteristics of connective tissue growth factor(CTGF/CCN2) in human hepatocellular carcinoma(HCC) in histology and to elucidate the roles of CCN2 on hepatoma cell cycle progression and metastasis in vitro.METHODS:Liver samples from 36 patients(who underwent hepatic resection for the first HCC between 2006 and 2011) and 6 normal individuals were examined for transforming growth factor β1(TGF-β1) or CCN2 mRNA by in situ hybridization.Computer image analysis was performed to measure integrated optimal density of CCN2 mRNA-positive cells in carcinoma foci and the surrounding stroma.Fibroblast-specific protein-1(FSP-1) and E-cadherin were examined to evaluate the process of epithelial to mesenchymal transition,α-smooth muscle actin and FSP-1 were detected to identify hepatic stellate cells,and CD34 was measured to evaluate the extent of vascularization in liver tissues by immunohistochemical staining.CCN2 was assessed for its stimulation of HepG2 cell migration and invasion using commercial kits while flow cytometry was used to determine CCN2 effects on HepG2 cell-cycle.RESULTS:In situ hybridization analysis showed that TGF-β1 mRNA was mainly detected in connective tissues and vasculature around carcinoma foci.In comparison to normal controls,CCN2 mRNA was enhanced 1.9-fold in carcinoma foci(12.36 ± 6.08 vs 6.42 ± 2.35) or 9.4-fold in the surrounding stroma(60.27 ± 28.71 vs 6.42 ± 2.35),with concomitant expression of CCN2 and TGF-β1 mRNA in those areas.Epithelial-mesenchymal transition phenotype related with CCN2 was detected in 12/36(33.3%) of HCC liver samples at the edges between carcinoma foci and vasculature.Incubation of HepG2 cells with CCN2(100 ng/mL) resulted in more of the cells transitioning into S phase(23.85 ± 2.35 vs 10.94 ± 0.23),and induced a significant migratory(4.0-fold) and invasive(5.7-fold) effect.TGF-β1-induced cell invasion was abrogated by a neutralizing CCN2 antibody showing that CCN2 is a downstream mediator of TGF-β1-induced hepatoma cell invasion.CONCLUSION:These data support a role for CCN2 in the growth and metastasis of HCC and highlight CCN2 as a potential novel therapeutic target.Ming Xiu Ya-Hui Liu David R Brigstock Fang-Hui He Rui-Juan Zhang Run-Ping Gao 2012World Journal of Gastroenterology2012,18,47:7
8Galvanically induced potentials to enable minimal tribochemical wear of stainless steel lubricated with sodium chloride and ionic liquid aqueous solution显示文摘The effect of galvanically induced potentials on the friction and wear behavior of a 1 RK91 stainless steel regarding to tribocorrosion was investigated using an oscillating ball-on-disk tribometer equipped with an electrochemical cell. The aim of this investigation is to develop a water-based lubricant. Therefore 1 molar sodium chloride(NaCl) and 1% 1-ethyl-3-methylimidazolium chloride [C_2 mim][Cl] water solutions were used. Tribological performance at two galvanically induced potentials was compared with the non-polarized state: cathodic potential-coupling with pure aluminum- and anodic potential-coupling with pure copper. Frictional and electrochemical response was recorded during the tests. In addition, wear morphology and chemical composition of the steel were analyzed using scanning electron microscopy(SEM) and X-ray photoelectron spectroscopy(XPS), respectively. The galvanically induced cathodic polarization of the stainless steel surface results in electrochemical corrosion protection and the formation of a tribolayer. Cations from the electrolyte(sodium Na^+ and 1-ethyl- 3-methylimidazolium [C_2 mim]^+) interact and adhere on the surface. These chemical interactions lead to considerably reduced wear using 1 NaC l(86%) and 1% 1-ethyl-3-methylimidazolium chloride [C_2 mim][Cl](74%) compared to the nonpolarized system. In addition, mechanical and corrosive part of wear was identified using this electrochemical technique. 9A draft sequence of the rice (Oryza sativa ssp. indica) genome显示文摘The sequence of the rice genome holds fundamental information for its biology, including physiology, genetics, development, and evolution, as well as information on many beneficial phenotypes of economic significance. Using a 'whole genome shotgun' approach, we have pro-duced a draft rice genome sequence of Oryza sativa ssp. in-dica, the major crop rice subspecies in China and many other regions of Asia. The draft genome sequence is constructed from over 4.3 million successful sequencing traces with an accumulative total length of 2214.9 Mb. The initial assembly of the non-redundant sequences reached 409.76 Mb in length, based on 3.30 million successful sequencing traces with a total length of 1797.4 Mb from an indica variant cultivar 93-11, giving an estimated coverage of 95.29% of the rice genome with an average base accuracy of higher than 99%. 12Prognostic value of perioperative leukocyte count in resectable gastric cancer显示文摘AIM: To investigate the prognostic significance of perioperative leukopenia in patients with resected gastric cancer.METHODS: A total of 614 eligible gastric cancer patients who underwent curative D2 gastrectomy and adjuvant chemotherapy were enrolled in this study. The relationship between pre- and postoperative hematologic parameters and overall survival was assessed statistically, adjusted for known prognostic factors.RESULTS: The mean white blood cell count(WBC) significantly decreased after surgery, and 107/614(17.4%) patients developed p-leukopenia, which was defined as a preoperative WBC ≥ 4.0 × 109/L and postoperative WBC < 4.0 × 109/L, with an absolute decrease ≥ 0.5 × 109/L. The neutrophil count decreased significantly more than the lymphocyte count. P-leukopenia significantly correlated with poor tumor differentiation and preoperative WBC. A higher preoperative WBC and p-leukopenia were independent negative prognostic factors for survival [hazard ratio(HR) = 1.602, 95% confidence interval(CI): 1.185-2.165; P = 0.002, and HR = 1.478, 95%CI: 1.149-1.902; P = 0.002, respectively] after adjusting for histology, Borrmann type, p TNM stage, vascular or neural invasion, gastrectomy method, resection margins, chemotherapy regimens, and preoperative WBC count. The patients with both higher preoperative WBC and p- leukopenia had a worse prognosis compared to those with lower baseline WBC and no p-leukopenia(27.5 mo vs 57.3 mo, P < 0.001). 15Lignocellulose as an insoluble fiber source in poultry nutrition:a review显示文摘Extensive research in recent years into the use of various fiber sources in poultry nutrition has led to the perception that dietary fiber is more than a simple diet diluent.Several studies showed that the feeding of insoluble fiber sources such as oat hulls,sunflower hulls or wood shavings may affect digestive physiology and function improving chickens health and growth performance.In this context,the effect of lignocellulose as an insoluble dietary fiber source is increasingly being investigated.Lignocellulose is a component of plant cell walls and consists mainly of the insoluble carbohydrate polymers cellulose and hemicelluloses as well as the phenolic polymer lignin.Lignocellulose is chemically and physicochemically different from other insoluble fiber sources and thus possibly has different effects on poultry compared to traditional fiber sources.Several studies investigated the effect of dietary lignocellulose on growth performance,nutrient digestibility,gastrointestinal tract development and intestinal microbiota in broilers and laying hens.Studies differed in terms of feed formulation and lignocellulose inclusion level as well as products of different suppliers were used.The results obtained are inconsistent;beneficial,indifferent or detrimental effects of feeding lignocellulose were observed,so that a final assessment of lignocellulose as a“novel”insoluble fiber source is difficult.This review article summarizes the results of studies in connection with the feeding of lignocellulose to poultry,compares them with those that have used other insoluble fiber sources and illuminates the possible mechanisms of action.Ilen Röhe Jürgen Zentek 2021Journal of Animal Science and Biotechnology2021,12,4:2
16Deoxynivalenol accumulation and detoxification in cereals and its potential role in wheat-Fusarium graminearum interactions显示文摘Deoxynivalenol(DON)is a prominent mycotoxin showing significant accumulation in cereal plants during infection by the phytopathogen Fusarium graminearum.It is a virulence factor that is important in the spread of F.graminearum within cereal heads,and it causes serious yield losses and significant contamination of cereal grains.In recent decades,genetic and genomic studies have facilitated the characterization of the molecular pathways of DON biosynthesis in F.graminearum and the environmental factors that influence DON accumulation.In addition,diverse scab resistance traits related to the repression of DON accumulation in plants have been identified,and experimental studies of wheat–pathogen interactions have contributed to understanding detoxification mechanisms in host plants.The present review illustrates and summarizes the molecular networks of DON mycotoxin production in F.graminearum and the methods of DON detoxification in plants based on the current literature,which provides molecular targets for crop improvement programs.This review also comprehensively discusses recent advances and challenges related to genetic engineering-mediated cultivar improvements to strengthen scab resistance.Furthermore,ongoing advancements in genetic engineering will enable the application of these molecular targets to develop more scab-resistant wheat cultivars with DON detoxification traits.Kun Luo Jiao Guo Dejia He Guangwei Li Thérèse Ouellet 2023aBIOTECH2023,4,2:2

18Demand for hospital emergency departments:a conceptual understanding显示文摘BACKGROUND: Emergency departments (EDs) are critical to the management of acute illnessand injury, and the provision of health system access. However, EDs have become increasinglycongested due to increased demand, increased complexity of care and blocked access to ongoingcare (access block). Congestion has clinical and organisational implications. This paper aims todescribe the factors that appear to infl uence demand for ED services, and their interrelationships asthe basis for further research into the role of private hospital EDs.DATA SOURCES: Multiple databases (PubMed, ProQuest, Academic Search Elite and ScienceDirect) and relevant journals were searched using terms related to EDs and emergency health needs.Literature pertaining to emergency department utilisation worldwide was identified, and articlesselected for further examination on the basis of their relevance and signifi cance to ED demand.RESULTS: Factors influencing ED demand can be categorized into those describing thehealth needs of the patients, those predisposing a patient to seeking help, and those relating topolicy factors such as provision of services and insurance status.
