1A rapid advice guideline for the diagnosis and treatment of 2019 novel coronavirus(2019-nCoV) infected pneumonia(standard version)显示文摘In December 2019, a new type viral pneumonia cases occurred in Wuhan, Hubei Province;and then named '2019 novel coronavirus(2019-nCoV)' by the World Health Organization(WHO) on 12 January 2020. For it is a never been experienced respiratory disease before and with infection ability widely and quickly, it attracted the world’s attention but without treatment and control manual. For the request from frontline clinicians and public health professionals of 2019-nCoV infected pneumonia management, an evidence-based guideline urgently needs to be developed. Therefore, we drafted this guideline according to the rapid advice guidelines methodology and general rules of WHO guideline development;we also added the first-hand management data of Zhongnan Hospital of Wuhan University. This guideline includes the guideline methodology, epidemiological characteristics, disease screening and population prevention, diagnosis, treatment and control(including traditional Chinese Medicine), nosocomial infection prevention and control, and disease nursing of the 2019-nCoV. Moreover, we also provide a whole process of a successful treatment case of the severe 2019-nCoV infected pneumonia and experience and lessons of hospital rescue for 2019-nCoV infections. 3Effectiveness of Different Waist Circumference Cut-off Values in Predicting Metabolic Syndrome Prevalence and Risk Factors in Adults in China显示文摘Objective To study the effectiveness of waist circumference cut-off values in predicting the prevalence of metabolic syndrome(MetS) and risk factors in adults in China. Methods A cross-sectional survey was condcuted in 14 provinces(autonomous region, municipality) in China. A total of 47 325 adults aged ≥20 years were selected by multistage stratified sampling, and questionnaire survey and physical and clinical examination were conducted among them. MetS was defined according to the International Diabetes Federation(IDF) criteria and modified IDF criteria.Results The age-standardized prevalence of MetS was 24.2%(22.1% in men and 25.8% in women) and 19.5%(22.1% in men and 18.0% in women) according to the IDF criteria and modified IDF criteria respectively. The age-standardized prevalence of pre-MetS was 8.1%(8.6% in men and 7.8% in women) according to the modified IDF criteria. The prevalence of MetS was higher in urban residents than rural residents and in northern China residents than in southern China residents. The prevalence of central obesity was about 30% in both men and women according to the ethnicity-specific cut-off values of waist circumference for central obesity(90 cm for men and 85 cm for women). Multivariate regression analysis revealed no significant difference in risk factors between the two MetS definitions. 5Total mesopancreas excision for pancreatic head cancer: analysis of 120 cases显示文摘Objective: To evaluate the feasibility and safety of total mesopancreas excision(TMpE) in the treatment of pancreatic head cancer. Methods: The clinical and pathological data of 120 patients with pancreatic head cancer who had undergone TMpE in our center from May 2010 to January 2014 were retrospectively analyzed. Results: The mean operative time was(275.0±50.2) min and the average intra-operative blood loss was(390.0±160.5) mL. Post-operative complications were reported in 45 patients, while no peri-operative death was noted. The specimen margins were measured in three dimensions, and 86 patients(71.6%) achieved R0 resection. 8Genetic characterization and linkage disequilibrium mapping of resistance to gray leaf spot in maize(Zea mays L.)显示文摘Gray leaf spot(GLS),caused by Cercospora zeae-maydis,is an important foliar disease of maize(Zea mays L.)worldwide,resistance to which is controlled by multiple quantitative trait loci(QTL).To gain insights into the genetic architecture underlying the resistance to this disease,an association mapping population consisting of 161 inbred lines was evaluated for resistance to GLS in a plant pathology nursery at Shenyang in 2010 and 2011.Subsequently,a genome-wide association study,using 41,101 single-nucleotide polymorphisms(SNPs),identified 51 SNPs significantly(P<0.001)associated with GLS resistance,which could be converted into 31 QTL.In addition,three candidate genes related to plant defense were identified,including nucleotidebinding-site/leucine-rich repeat,receptor-like kinase genes similar to those involved in basal defense.Two genic SNPs,PZE-103142893 and PZE-109119001,associated with GLS resistance in chromosome bins 3.07 and 9.07,can be used for marker-assisted selection(MAS)of GLS resistance.These results provide an important resource for developing molecular markers closely linked with the target trait,enhancing breeding efficiency.Liyu Shi Xiangling Lv Jianfeng Weng Hanyong Zhu Changlin Liu Zhuanfang Hao Yu Zhou Degui Zhang Mingshun Li Xiaoke Ci Xinhai Li Shihuang Zhang 2014The Crop Journal2014,2,Z1:9
9Duct-to-duct biliary reconstruction after radical resection of Bismuth Ⅲ a hilar cholangiocarcinoma显示文摘At present, radical resection remains the only effective treatment for patients with hilar cholangiocarcinoma. The surgical approach for R0 resection of hilar cholangiocarcinoma is complex and diverse, but for the biliary reconstruction after resection, almost all surgeons use Roux-en-Y hepaticojejunostomy. A viable alternative to Roux-en-Y reconstruction after radical resection of hilar cholangiocarcinoma has not yet been proposed. We report a case of performing duct-to-duct biliary reconstruction after radical resection of Bismuth Ⅲa hilar cholangiocarcinoma. End-to-end anastomosis between the left hepatic duct and the distal common bile duct was used for the biliary reconstruction, and a singlelayer continuous suture was performed along the bile duct using 5-0 prolene. The patient was discharged favorably without biliary fistula 2 wk later. Evidence for tumor recurrence was not found after an 18 mo follow- up. Here, an Os CIPK7 mutant via TILLING procedure with a point mutation in the kinase domain showed increased chilling tolerance, which could be potentially used in the molecular breeding. 11High glucose causes apoptosis of rabbit corneal epithelial cells involving activation of PERK-eIF2α-CHOP-caspase-12 signaling pathway显示文摘AIM: To investigate the effect of high concentration of glucose(HCG) on double stranded RNA-activated protein kinase-like ER kinase(PERK)-eukaryotic initiation factor-2α(eIF2α)-transcription factor C/EBP homologous protein(CHOP)-cysteine aspartate specific proteinase(caspase-12) signaling pathway activation and apoptosis in rabbit corneal epithelial cells(RCECs). METHODS: RCECs were treated by different concentrations of glucose for 0-48 h. The expressions of PERK, p-PERK, eIF2α, p-eIF2α, 78 k Da glucose-regulated protein 78(GRP78), CHOP, B-cell lymphoma 2(Bcl-2), B-cell lymphoma-2-associated X protein(Bax) and caspase-12 were determined by Western blot. Apoptosis was detected by TUNEL assay. Meanwhile, the function of PERK-eI F2α-CHOP-caspase-12 signaling pathway activation in high glucose-induced apoptosis was evaluated using PERK inhibitor, GSK2606414. RESULTS: HCG significantly promoted the expression of p-PERK, p-eIF2α, GRP78, CHOP, Bax and cleaved caspase-12 in RCECs(P<0.05), while remarkably decreased the expression of Bcl-2 and caspase-12(P<0.05), and the alterations caused by glucose were in concentration-and time-dependent manners. Meanwhile, PERK and eIF2α expressions were not affected in all groups(P>0.05). TUNEL assay showed that the apoptosis rate of RCECs in the HCG group increased significantly in contrast with that in the normal concentration of glucose or osmotic pressure control group(P<0.05), and the apoptosis rate increased with the increase of glucose concentration within limits(P<0.05). GSK2606414 down-regulated the expression of p-PERK and p-eI F2α in the HCG group(P<0.05), while still did not affect the expression of PERK and eIF2α among groups(P>0.05). Correspondingly, GSK2606414 also significantly reduced the apoptosis rate induced by high glucose(P<0.05). 12In vivo flow cytometry reveals a circadian rhythm of circulating tumor cells显示文摘Circulating tumor cells(CTCs)is an established biomarker of cancer metastasis.The circulation dynamics of CTCs are important for understanding the mechanisms underlying tumor cell dissemination.Although studies have revealed that the circadian rhythm may disrupt the growth of tumors,it is generally unclear whether the circadian rhythm controls the release of CTCs.In clinical examinations,the current in vitro methods for detecting CTCs in blood samples are based on a fundamental assumption that CTC counts in the peripheral blood do not change significantly over time,which is being challenged by recent studies.Since it is not practical to draw blood from patients repeatedly,a feasible strategy to investigate the circadian rhythm of CTCs is to monitor them by in vivo detection methods.Fluoresce nee in vivo flow cytometry(IVFC)is a powerful optical technique that is able to detect fluoresce nt circulating cells directly in living animals in a noninvasive manner over a long period of time.In this study,we applied fluorescence IVFC to monitor CTCs noninvasively in an orthotopic mouse model of human prostate cancer.We observed that CTCs exhibited stochastic bursts over cancer progression.The probability of the bursting activity was higher at early stages than at late stages.We longitudinally monitored CTCs over a 24-h period,and our results revealed striking daily oscillations in CTC counts that peaked at the onset of the night(active phase for rodents),suggesting that the release of CTCs might be regulated by the circadian rhythm.Xi Zhu Yuanzhen Suo Yuting Fu Fuli Zhang Nan Ding Kai Pang Chengying Xie Xiaofu Weng Meilu Tian Hao He Xunbin Wei 2021Light(Science & Applications)2021,10,6:6
13Zea mays(L.) P1 locus for cob glume color identified as a post-domestication selection target with an effect on temperate maize genomes显示文摘Artificial selection during domestication and post-domestication improvement results in loss of genetic diversity near target loci. However, the genetic locus associated with cob glume color and the nature of the genomic pattern surrounding it was elusive and the selection effect in that region was not clear. An association mapping panel consisting of 283 diverse modern temperate maize elite lines was genotyped by a chip containing over 55,000 evenly distributed SNPs. Ten-fold resequencing at the target region on 40 of the panel lines and 47 tropical lines was also undertaken. A genome-wide association study(GWAS) for cob glume color confirmed the P1 locus, which is located on the short arm of chromosome 1, with a-log10 P value for surrounding SNPs higher than the Bonferroni threshold(α/n, α < 0.001) when a mixed linear model(MLM) was implemented. A total of 26 markers were identified in a 0.78 Mb region surrounding the P1 locus, including 0.73 Mb and 0.05 Mb upstream and downstream of the P1 gene, respectively. A clear linkage disequilibrium(LD) block was found and LD decayed very rapidly with increasing physical distance surrounding the P1 locus. The estimates of π and Tajima's D were significantly(P < 0.001) lower at both ends compared to the locus. Upon comparison of temperate and tropical lines at much finer resolution by resequencing(180-fold finer than chip SNPs), a more structured LD block pattern was found among the 40 resequenced temperate lines. All evidence indicates that the P1 locus in temperate maize has not undergone neutral evolution but has been subjected to artificial selection during post-domestication selection or improvement. An Illumina GoldenGate assay with 1 536 SNP markers was used to genotype maize inbred lines and identified the functional genetic variations underlying drought tolerance by association analysis. Across 80 lines, 1 006 polymorphic SNPs (65.5% of the total) in the assay with good call quality were used to estimate the pattern of genetic diversity, population structure, and familial relatedness. The analysis showed the best number of fixed subgroups was six, which was consistent with their original sources and results using only simple sequence repeat markers. Pairwise linkage disequilibrium (LD) and association mapping with phenotypic traits investigated under water-stressed and well-watered regimes showed rapid LD decline within 100-500 kb along the physical distance of each chromosome, and that 29 SNPs were associated with at least two phenotypic traits in one or more environments, which were related to drought-tolerant or drought-responsive genes. 17Long-term outcomes after radical gastrectomy in gastric cancer patients with overt bleeding显示文摘AIM: To investigate the difference in long-term outcomes between gastric cancer patients with and without a primary symptom of overt bleeding(OB).METHODS: Consecutive patients between January 1, 2007 and March 1, 2012 were identified retrospectively by reviewing a gastric cancer database at Xinhua Hospital Affiliated to Shanghai Jiao Tong University School of Medicine. A follow-up examination was performed on patients who underwent a radical gastrectomy. OB due to gastric cancer included hematemesis, melena or hematochezia, and gastric cancer was confirmed as the source of bleeding by endoscopy. Patients without OB were defined as cases with occult bleeding and those with other initial presentations, including epigastric pain, weakness, weight loss and obstruction. The 3-year overall survival(OS) rate, age, gender, AJCC T stage, AJCC N stage, overall AJCC stage, tumor size, histological type, macroscopic(Borrmann) type, lymphovascular invasion and R status were compared between patients with and without OB. Moreover, we carried out a subgroup analysis based on tumor location(upper, middle and lower).RESULTS: We identified 939 patients. Of these, 695(74.0%) were hospitalized for potential radical gastrectomy and another 244 received palliative resection, rerouting of the gastrointestinal tract, chemotherapy, radiotherapy or no treatment due to the presence of unresectable tumors. Notably, there was no significant difference in the percentage of OB patients between resectable cases and unresectable cases(20.3 % vs 22.1 %, P = 0.541). Fo l l o w- u p examination was performed on 653 patients(94%) who underwent radical gastrectomy. We found no significant difference in 3-year OS rate(68.2% vs 61.2%, P = 0.143) or clinicopathological characteristics(P > 0.05) between these patients with and without OB. Subgroup analysis based on tumor location showed that the 3-year OS rate of upper gastric cancer was significantly higher in patients with OB(84.6%) than in those without OB(48.1%, P < 0.01) and that AJCC stagesⅠ-Ⅱ(56.4% vs 35.1%, P = 0.017) and T1-T2 category tumors(30.8% vs 13%, P = 0.010) were more frequent in patients with OB than in those without OB. 18Recent advances of pharmaceutical crystallization theories显示文摘As a technology of separation and purification,crystallization plays a vital role in diverse industries such as inorganic salt,pharmaceutical,and food industries,which has a huge impact on purity,crystal polymorph,crystal morphology,and particle size distribution of final products.In the past few decades,with the rapid advancement of experimental approaches and molecular simulation methods,considerable advances in the interpretation of crystallization mechanisms have been obtained,promoting the investigation and understanding of crystallization theories greatly.In this review,the advances of pharmaceutical crystallization theories in recent years from the perspectives of nucleation and crystal growth are summarized and discussed.Two thermodynamic models that are helpful in the study of the crystallization mechanisms will be introduced.In this section,the perturbed-chain statistical associating fluid theory(PC-SAFT)and a chemical-potential-gradient model will be introduced,which have been successfully applied in pharmaceutical solubility prediction,the research of dissolution mechanism as well as dissolution kinetics analysis.These two models are expected to be applied to the study of pharmaceutical crystallization process and mechanism.Furthermore,molecular simulation based on the interaction between particles can provide structural information,thermodynamics,and dynamics properties of complex systems at the molecular level,like intermolecular interaction and surface adsorption energies.Application and some shortcomings of molecular simulation,especially molecular dynamics simulation,in the field of pharmaceutical crystallization will be expounded.Jingyun Weng Yiping Huang Dule Hao Yuanhui Ji 2020Chinese Journal of Chemical Engineering2020,28,4:5
19Urban flood inundation and damage assessment based on numerical simulations of design rainstorms with different characteristics显示文摘In the context of global climate change and urbanization,urban flooding is an important type of natural disaster that affects urban development,especially in China,which is experiencing rapid urbanization.In the past 10 years,urban flooding events have caused huge disaster losses in Chinese cities.This has resulted in significant negative effects on the urban infrastructure,socioeconomic systems,and urban residents,thus causing widespread concern.Studies have confirmed the change in extreme rainstorms is due to the changing environments in many cities globally.Conducting studies on the impact of these rainstorms with different characteristics for urban flooding is valuable for coping with unfavorable situations.In addition,numerical simulations provide an economical and viable means to perform these studies.This paper presents a numerical model of Xiamen Island in China.Simulations were conducted for 12 design rainstorm events with different return periods,rain patterns,and durations.The results indicate that,in the case of an equal rainfall amount,the rainfall intensity is the key factor that affects the inundated area,depth,and damages.However,the rainfall intensity is not the only determining factor;the rainfall pattern also affects the inundations.In regard to the rainfall pattern,a higher rainfall peak coefficient usually leads to severe urban inundation and damage.As a result,the lag time would be shorter,which may further exacerbate the impact of urban flood disasters.The results of this study provide insights into managing flood risks,developing urban flood prevention strategies,and designing flood prevention measures.MEI Chao LIU JiaHong WANG Hao LI ZeJin YANG ZhiYong SHAO WeiWei DING XiangYi WENG BaiSha YU YingDong YAN DianYi 2020Science China(Technological Sciences)2020,63,11:5
20Maxingxiongting mixture attenuates hypoxia pulmonary arterial hypertension to improve right ventricular hypertrophy by inhibiting the rho-kinase signaling pathway显示文摘OBJECTIVE:To explore the mechanism of Maxingxiongting mixture(MXXTM)on pulmonary hypertension in a rat model established by intraperitoneal injection of monocrotaline solution,smoking and forced swimming.METHODS:A total of 30 male Sprague-Dawley rats were randomly divided into five groups:control group,model group,high-dose of MXXTM group(HM),low-dose of MXXTM group(LM),and fasudil group.The mean pulmonary artery pressure(m PAP)was measured by using a miniature catheter.Lung tissue and right ventricular tissue sections were stained with hematoxylin-eosin.The right ventricle(RV)and left ventricle+septum(LV+S)were weighted.RV/(LV+S)was calculated to reflect the degree of right ventricular hypertrophy.Rho/Rho-kinase signaling pathway key proteins(Rho A,ROCKⅠand ROCKⅡ)in rat right ventricular tissue were measured by Western blot analysis.The levels of serum hypoxia-inducible factor-1α(HIF-1α),vascular endothelial growth factor(VEGF)and the levels of plasma renin activity(PRA),angiotensinⅡ(ANG-Ⅱ),aldosterone(ALD)in rat anticoagulated plasma were all measured by enzyme-linked immunosorbent assay.RESULTS:Compared with the control group,the m PAP and RV/(LV+S)in the model group were significantly increased.Administration of fasudil resulted in a significant decrease of m PAP and RV/(LV+S).In the HM group and LM group,m PAP and RV/(LV+S)were significantly lower than the model group.Compared with the control group,the contents of HIF-1α,VEGF,PRA,ANG-Ⅱand ALD in the model group were significantly increased.The administration of fasudil and high-dose MXXTM significantly reduced the contents of HIF-1α,VEGF,PRA,ANG-II and ALD.Compared with the control group,the expression of Rho A,ROCKⅠand ROCKⅡin the right ventricle of the model group were significantly increased.The administration of fasudil and high-dose MXXTM significantly reduced the expression of Rho A and RockⅡproteins.Our results indicated that high-dose of MXXTM had similar effects on reducing pulmonary artery pressure and improving right ventricular remodeling to fasudil.However,MXXTM was unable to restore parameters above to control levels.CONCLUSIONS:MXXTM attenuates hypoxia pulmonary arterial hypertension to improve right ventricular hypertrophy by inhibiting the Rho-kinase signaling pathway.An Xing Li Songtao Weng Xiangwen Wang Xian Wu Hao Zhang Xinyue Gao Jian Yang Renxu Peng Bo 2020Journal of Traditional Chinese Medicine2020,40,6:4
