1SDF-1/CXCR4 axis modulates bone marrow mesenchymal stem cell apoptosis, migration and cytokine secretion显示文摘Bone marrow mesenchymal stem cells(MSCs)are considered as a promising cell source to treat the acute myocardial infarction.However,over 90%of the stem cells usually die in the first three days of transplantation.Survival potential,migration ability and paracrine capacity have been considered as the most important three factors for cell transplantation in the ischemic cardiac treatment.We hypothesized that stromal-derived factor-1(SDF-1)/CXCR4 axis plays a critical role in the regulation of these processes.In this study,apoptosis was induced by exposure of MSCs to H2O2 for 2 h.After re-oxygenation,the SDF-1 pretreated MSCs demonstrated a significant increase in survival and proliferation.SDF-1 pretreatment also enhanced the migration and increased the secretion of pro-survival and angiogenic cytokines including basic fibroblast growth factor and vascular endothelial growth factor.Western blot and RT-PCR demonstrated that SDF-1 pretreatment significantly activated the pro-survival Akt and Erk signaling pathways and up-regulated Bcl-2/Bax ratio.These protective effects were partially inhibited by AMD3100,an antagonist of CXCR4.We conclude that the SDF-1/CXCR4 axis is critical for MSC survival,migration and cytokine secretion.Xiaolei Liu Biyan Duan Zhaokang Cheng Xiaohua Jia Lina Mao Hao Fu Yongzhe Che Lailiang Ou Lin Liu Deling Kong 2011Protein & Cell2011,2,10:53
2Deferoxamine promotes recovery of traumatic spinal cord injury by inhibiting ferroptosis显示文摘Ferroptosis is an iron-dependent novel cell death pathway. Deferoxamine, a ferroptosis inhibitor, has been reported to promote spinal cord injury repair. It has yet to be clarified whether ferroptosis inhibition represents the mechanism of action of Deferoxamine on spinal cord injury recovery. A rat model of Deferoxamine at thoracic 10 segment was established using a modified Allen's method. Ninety 8-week-old female Wistar rats were used. Rats in the Deferoxamine group were intraperitoneally injected with 100 mg/kg Deferoxamine 30 minutes before injury. Simultaneously, the Sham and Deferoxamine groups served as controls. Drug administration was conducted for 7 consecutive days. The results were as follows:(1) Electron microscopy revealed shrunken mitochondria in the spinal cord injury group.(2) The Basso, Beattie and Bresnahan locomotor rating score showed that recovery of the hindlimb was remarkably better in the Deferoxamine group than in the spinal cord injury group.(3) The iron concentration was lower in the Deferoxamine group than in the spinal cord injury group after injury.(4) Western blot assay revealed that, compared with the spinal cord injury group, GPX4, xCT, and glutathione expression was markedly increased in the Deferoxamine group.(5) Real-time polymerase chain reaction revealed that, compared with the Deferoxamine group, mRNA levels of ferroptosis-related genes Acyl-CoA synthetase family member 2(ACSF2) and iron-responsive element-binding protein 2(IREB2) were up-regulated in the Deferoxamine group.(6) Deferoxamine increased survival of neurons and inhibited gliosis. These findings confirm that Deferoxamine can repair spinal cord injury by inhibiting ferroptosis. 4Bi-level Planning for Integrated Energy Systems Incorporating Demand Response and Energy Storage Under Uncertain Environments Using Novel Metamodel显示文摘The optimal planning and design of an integrated energy system(IES)is of great significance to facilitate distributed renewable energy(DRE)technology and improve the overall energy efficiency of the energy system.With the increased penetration of distributed generation(DG),the power supply and load sides of an IES present more increased levels of uncertainties.Demand response(DR)and the energy storage system(ESS)serve as important means to shift energy supply and use across time to counter the indeterminate variations.However,the current IES planning methods are unable to effectively deal with the uncertainties of DREs and loads,and to optimize the operations of DG-DR-ESS due to the enormous possible combinations.In this paper,a new method for the optimal planning and design of an integrated energy system has been introduced and verified.The new method consists of three integrated elements.First,the method of the probability scenario has been used to model the uncertainties of the DREs and loads so as to better characterize the impact of uncertainty on the planning and design of the IES.Secondly,the optimal operation of the IES under different probability scenarios is ensured using the second-order cone optimization for quick solutions due to the simplicity of this sub-problem,serving as the bottom-level optimization.Thirdly,the optimal planning and design of IES through optimal sizing of the power generating components and ESS are performed using a special meta-model based global optimization method due to the complex,black-box,and computation intensive nature of this top-level optimization in a nested,bi-level global optimization problem.The combined approach takes full account of the interrelated operations of DG-DR-ESS under different design configurations to support a better optimal planning and design of the IES.The simulation has been carried out on an IES system modified from the IEEE 33-node distribution system.The simulation results show that the proposed method and model are effective.Hao Xiao Wei Pei Zuomin Dong Li Kong 2018CSEE Journal of Power and Energy Systems2018,4,2:27

6Catalytic oxidation of gas-phase elemental mercury by nano-Fe_2 O_3显示文摘Heterogeneous oxidation of gas-phase Hg 0 by nano-Fe 2 O 3 was investigated on a fixed bed reactor, and the effects of oxygen concentration, bed temperature, water vapour concentration and particle size have been discussed. The results showed that Hg 0 could be oxidized by active oxygen atom on the surface of nano-Fe 2 O 3 as well as lattice oxygen in nano-Fe 2 O 3 . Among the factors that affect Hg 0 oxidation by nano-Fe 2 O 3 , bed temperature plays an important role. More than 40% of total mercury was oxidized at 300°C, however, the test temperature at 400°C could cause sintering of nano-catalyst, which led to a lower efficiency of Hg 0 oxidation. The increase of oxygen concentration could promote mercury oxidation and led to higher Hg 0 oxidation efficiency. No obvious mercury oxidation was detected in the pure N 2 atmosphere, which indicates that oxygen is required in the gas stream for mercury oxidation. The presence of water vapour showed different effects on mercury oxidation depending on its concentration. 7Structure evolution of oxygen removal from porous carbon for optimizing supercapacitor performance显示文摘The presence of oxygen functional groups is detrimental to the capacitive performance of porous carbon electrode in organic electrolyte. In this regards, hydrogen thermal reduction has been demonstrated effective approach in removing the unstable surface oxygen while maintaining the high porosity of carbon matrix. However, the exact evolution mechanism of various oxygen species during this process, as well as the correlation with electrochemical properties, is still under development. Herein, biomass-based porous carbon is adopted as the model material to trace its structure evolution of oxygen removal under hydrogen thermal reduction process with the temperature range of 400–800 °C. The optimum microstructure with low oxygen content of 0.90% and proper pore size distribution was achieved at 700°C. XPS, TPRMS and Boehm titration results indicate that the oxygen elimination undergoes three distinctive stages(intermolecular dehydration, hydrogenation and decomposition reactions). The optimum microstructure with low oxygen content of 0.90% and proper pore size distribution was achieved at 700 °C. Benefiting from the stable electrochemical interface and the optimized porous structure, the as-obtained HAC-700 exhibit significantly suppressed self-discharge and leak current, with improved cycling stability, which is attributable to the stabilization of electrochemical interface between carbon surface and electrolyte. 8The Prospective Two-Dimensional Graphene Nanosheets: Preparation, Functionalization, and Applications显示文摘Graphene, as an intermediate phase between fullerene and carbon nanotube, has aroused much interests among the scientific community due to its outstanding electronic, mechanical, and thermal properties.With excellent electrical conductivity of 6000 S/cm, which is independent on chirality, graphene is a promising material for high-performance nanoelectronics, transparent conductor, as well as polymer composites. On account of its Young's Modulus of 1 TPa and ultimate strength of 130 GPa, isolated graphene sheet is considered to be among the strongest materials ever measured. Comparable with the single-walled carbon nanotube bundle,graphene has a thermal conductivity of 5000 W/(m·K), which suggests a potential application of graphene in polymer matrix for improving thermal properties of the graphene/polymer composite. Furthermore, graphene exhibits a very high surface area, up to a value of 2630 m^2/g. All of these outstanding properties suggest a wide application for this nanometer-thick, two-dimensional carbon material. This review article presents an overview of the significant advancement in graphene research: preparation, functionalization as well as the properties of graphene will be discussed. 9The geothermal formation mechanism in the Gonghe Basin: Discussion and analysis from the geological background显示文摘The Gonghe Basin,a Cenozoic down-warped basin,is located in the northeastern part of the Qinghai-Xizang (Tibetan) Plateau,and spread over important nodes of the transfer of multiple blocks in the central orogenic belt in the NWW direction.It is also called “Qin Kun Fork” and “Gonghe Gap”.The basin has a high heat flow value and obvious thermal anomaly.The geothermal resources are mainly hot dry rock and underground hot water.In recent years,the mechanism of geothermal formation within the basin has been controversial.On the basis of understanding the knowledge of predecessors,this paper proposes the geothermal formation mechanism of the “heat source–heat transfer–heat reservoir and caprock–thermal system”of the Gonghe Basin from the perspective of a geological background through data integrationintegrated research-expert,discussion-graph,compilation-field verification and other processes: (1) Heat source: geophysical exploration and radioisotope calculations show that the heat source of heat in the basin has both the contribution of mantle and the participation of the earth's crust,but mainly the contribution of the deep mantle.(2) Heat transfer: The petrological properties of the basin and the exposed structure position of the surface hot springs show that one transfer mode is the material of the mantle source upwells and invades from the bottom,directly injecting heat;the other is that the deep fault conducts the deep heat of the basin to the middle and lower parts of the earth's crust,then the secondary fracture transfers the heat to the shallow part.(3) Heat reservoir and caprock: First,the convective strip-shaped heat reservoir exposed by the hot springs on the peripheral fault zone of the basin;second,the underlying hot dry rock layered heat reservoir and the upper new generation heat reservoir and caprock in the basin revealed by drilling data.(4) Thermal system: Based on the characteristics of the “heat source-heat transfer-heat reservoir and caprock”,it is preliminarily believed that the Gonghe Basin belongs to the non-magmatic heat source hydrothermal geothermal system (type II21) and the dry heat geothermal system (type II22).Its favorable structural position and special geological evolutionary history have given birth to a unique environment for the formation of the geothermal system.There may be a cumulative effect of heat accumulation in the eastern part of the basin,which is expected to become a favorable exploration area for hot dry rocks.Yan-fang Feng Xiao-xu Zhang Bo Zhang Jiang-tao Liu Yang-gang Wang De-long Jia Li-rong Hao Zhao-yu Kong 2018China Geology2018,1,3:15

11Antisense MMP-9 RNA inhibits malignant glioma cell growth in vitro and in vivo显示文摘The matrix-degrading metalloproteinases (MMPs), particularly MMP-9, play important roles in the pathogenesis and development of malignant gliomas. In the present study, the oncogenic role of MMP-9 in malignant glioma cells was investigated via antisense RNA blockade in vitro and in vivo. TJ905 malignant glioma cells were transfected with pcDNA3.0 vector expressing antisense MMP-9 RNA (pcDNA-AS-MMP9), which significantly decreased MMP-9 expression, and cell proliferation was assessed. For in vivo studies, U251 cells, a human malignant glioma cell line, were implanted subcutaneously into 4-to 6-week-old BALB/c nude mice. The mice bearing well-established U251 gliomas were treated with intratumoral pcDNA-AS-MMP9-Lipofectamine complex (AS-MMP-9-treated group), subcutaneous injection of endostatin (endostatin-treated group), or both (combined therapy group). Mice treated with pcDNA (empty vector)-Lipofectamine served as the control group. Four or eight weeks later, the volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity were assayed. We demonstrate that pcDNA-AS-MMP9 significantly decreased MMP-9 expression and inhibited glioma cell proliferation. Volume and weight of tumor, MMP-9 expression, microvessel density and proliferative activity in the antisense-MMP-9-treated and therapeutic alliance groups were significantly lower than those in the control group. The results suggest that MMP-9 not only promotes malignant glioma cell invasiveness, but also affects tumor cell proliferation. Blocking the expression of MMP-9 with antisense RNA substantially suppresses the malignant phenotype of glioma cells, and thus can be used as an effective therapeutic strategy for malignant gliomas.Cuiyun Sun Qian Wang Hongxu Zhou Shizhu Yu Alain R. 12Cardiac autonomic nerve fiber regeneration in chronic heart failure Do Akt gene-transduced mesenchymal stem cells promote repair?显示文摘BACKGROUND: Transplantation of Akt-over-expressing mesenchymal stem cells (Akt-MSCs) has been shown to repair infarcted myocardium and improve cardiac function. However, little is known about the therapeutic effects of Akt-MSCs on cardiac autonomic neuropathy in chronic heart failure (CHF). OBJECTIVE: The present study used adriamycin-induced CHF rat models to observe the effect of Akt-MSCs on cardiac autonomic nervous regeneration and the factors mediating this effect. DESIGN, TIME AND SETTING: A randomized, controlled animal experiment was performed at the Central Laboratory of Basic Medical College, China Medical University, between September 2008 and April 2009. MATERIALS: Rabbit anti-choline acetyltransferase (ChAT), growth associated protein-43 (GAP-43), synaptophysin (SYN) polyclonal antibodies and the secondary antibody (goat anti-rabbit IgG) were purchased from Boster, China. Cat-A-Kit assay system was provided by Amersham, USA. METHODS: (1) Adult rat MSCs were isolated and cultured for the preparation of Akt-MSCs. (2) Forty male Wistar rats were intramyocardially administered adriamycin at 2 mg/kg over 3 days for a total of five times and once a week for additional five times thereafter to establish CHF models. At 2 weeks after final adriamycin treatment, 34 successful CHF rat models were randomized to three groups: Akt-MSCs (n = 11), simple MSCs (s-MSCs, n =11), and control (n = 12). Each group was intravenously administered Akt-MSCs (2×106 cells in 100 μL PBS), s-MSCs (2×106 cells in 100 μL PBS) or equal volume of phosphate buffered saline, once a day for a total of three times. MAIN OUTCOME MEASURES: At 4 weeks after final adriamycin treatment, myocardial norepi- nephrine (NE) content was detected using a Cat-A-Kit assay system. Myocardial ChAT, SYN and GAP-43 were performed by immunohistochemistry and Western blot analysis. Prior to, 2 and 4 weeks after adriamycin treatment, echocardiographic examination was performed and left ventricular ejection fraction (LVEF) was determined. RESULTS: Myocardial NE content, as well as SYN-positive and GAP-43-positive nerve fiber density and expression, and LVEF, was the greatest in the Akt-MSCs group, followed by the s-MSCs group, and lastly the control group (P < 0.05 or P < 0.01). ChAT expression was similar between Akt-MSCs and s-MSCs groups, but it was higher compared with the control group (P < 0.05). NE contents were negatively correlated to LVEF (r = -0.64, P = 0.015). 13Dynamic CCT Diagram of Automobile Beam Steel With High Strength Produced by FTSR Technology显示文摘The dynamic continuous cooling transformation(CCT)diagram and phase transformation rules of 510 MPa automobile beam steel,which is produced by a continuous casting of thin slab of FTSR technology in Tangshan Iron and Steel Co.Ltd.,are researched by thermal simulation experiment.The microstructure characteristics of the beam steel under different test conditions are studied by means of optical microscope and scanning electron microscope.The test results show that the critical temperatures of phase transformation Ar3 and Ar1 will all decrease with the increase of the cooling rate.When the cooling rate is lower than 20 ℃·s-1,the ferrite and pearlite phase transformations are the main parts;when the cooling rate is higher than 20 ℃·s-1,the bainite phase appears.Moreover,the microstructures of 510 MPa automobile beam steel produced by FTSR technology are also studied,and the results are basically in accordance with the CCT diagram gained from the test.WANG Xin KANG Yong-lin YU Hao CHEN Li-bin KONG Qing-fu 2008Journal of Iron and Steel Research(International)2008,15,2:11
14The diurnal and seasonal characteristics of urban heat island variation in Beijing city and surrounding areas and impact factors based on remote sensing satellite data显示文摘Based on the land surface temperature (LST), the land cover classification map,vegetation coverage, and surface evapotranspiration derived from EOS-MODIS satellite data, and by the use of GIS spatial analytic technique and multivariate statistical analysis method, the urban heat island (UHI) spatial distribution of the diurnal and seasonal variabilities and its driving forces are studied in Beijing city and surrounding areas in 2001. The relationships among UHI distribution and landcover categories, topographic factor, vegetation greenness, and surface evapotranspiration are analyzed. The results indicate that: (i) The significant UHI occur in Beijing city areas in the four seasons due to high heat capacity and multi-reflection of compression building, as well as with special topographic features of its three sides surrounded by mountains,especially in the summer. The UHI spatial distribution is corresponding with the urban geometry structure profile. The LST difference is approximately 4-6℃ between Beijing city and suburb areas, comparatively is 8- 10℃ between Beijing city area and outer suburb area in northwestern regions. (ii) The UHI distribution and intensity in daytime are different from nighttime in Beijing city area, the nighttime UHI is obvious. However, in the daytime, the significant UHI mainly appears in the summer, the autumn takes second place, and the UHI in the winter and the spring seem not obvious. The surface evapotranspiration in suburb areas is larger than that in urban areas in the summer, and high latent heat exchange is evident, which leads to LST difference between city area and suburb area. 16Effect of Shen-Fu Injection(参附注射液)on Hemodynamics in Early Volume Resuscitation Treated Septic Shock Patients显示文摘Objective: To investigate the hemodynamic effect of Shen-Fu Injection(参附注射液, SFI) in early volume resuscitation treated septic shock patients by monitoring pulse indicator continuous cardiac output(PICCO). Methods: All septic shock patients admitted in the Intensive Care Unit of the Affiliated Hospital of Shandong University of Traditional Chinese Medicine from January 1 st, 2014 to December 31 th, 2015, were reviewed, and totally 65 were enrolled in this study. They were assigned to SFI group(33 cases) and control group(32 cases). All 65 patients underwent conventional treatment mainly including volume resuscitation, antibiotics and vasoactive drugs therapy. The patients of the SFI group received additional 100 mL of SFI intravenously every 12 h. In all 65 patients, the PICCO arterial catheter and vein catheter were implanted within 1 h after the diagnosis of septic shock. In the course of early volume resuscitation, hemodynamic data of patients were recorded by PICCO monitor at 0, 12, and 24 h after the catheter implantation. Results: The hemodynamic indices of the two groups showed no significant differences at the beginning of 0 h(P>0.05). At 12 and 24 h, the hemodynamic indices of SFI group were significantly improved in comparison with the control group(P<0.05), including cardiac index(CI), global end diastolic volume index(GEDI), mean arterial pressure(MAP) and heart rate(HR). In addition, there was no significant change of extra-vascular lung water index between the two groups(P>0.05). 17A novel DOA estimation algorithm using directional antennas in cylindrical conformal arrays显示文摘In this paper, a novel direction of arrival(DOA) estimation algorithm using directional antennas in cylindrical conformal arrays(CCAs) is proposed. To eliminate the shadow effect, we divide the CCAs into several subarrays to obtain the complete output vector. Considering the anisotropic radiation pattern of a CCA, which cannot be separated from the manifold matrix, an improved interpolation method is investigated to transform the directional subarray into omnidirectional virtual nested arrays without non-orthogonal perturbation on the noise vector. Then, the cross-correlation matrix(CCM) of the subarrays is used to generate the consecutive co-arrays without redundant elements and eliminate the noise vector. Finally, the full-rank equivalent covariance matrix is constructed using the output of co-arrays,and the unitary estimation of the signal parameters via rotational invariance techniques(ESPRIT) is performed on the equivalent covariance matrix to estimate the DOAs with low computational complexity. 19Mechanisms underlying the promotion of functional recovery by deferoxamine after spinal cord injury in rats显示文摘Deferoxamine, a clinically safe drug used for treating iron overload, also repairs spinal cord injury although the mechanism for this action remains unknown. Here, we determined whether deferoxamine was therapeutic in a rat model of spinal cord injury and explored potential mechanisms for this effect. Spinal cord injury was induced by impacting the spinal cord at the thoracic T10 vertebra level. One group of injured rats received deferoxamine, a second injured group received saline, and a third group was sham operated. Both 2 days and 2 weeks after spinal cord injury, total iron ion levels and protein expression levels of the proinflammatory cytokines tumor necrosis factor-α and interleukin-1β and the pro-apoptotic protein caspase-3 in the spinal cords of the injured deferoxamine-treated rats were significantly lower than those in the injured saline-treated group. The percentage of the area positive for glial fibrillary acidic protein immunoreactivity and the number of terminal deoxynucleotidyl transferase d UTP nick end labeling-positive cells were also significantly decreased both 2 days and 2 weeks post injury, while the number of Neu N-positive cells and the percentage of the area positive for the oligodendrocyte marker CNPase were increased in the injured deferoxamine-treated rats. At 14–56 days post injury, hind limb motor function in the deferoxamine-treated rats was superior to that in the saline-treated rats. 20Laboratory Characterizations on 2007 Cases of Monoclonal Gammopathies in East China显示文摘Monoclonal gammopathies are characterized by the presence of monoclonal immunoglobulin in patients with or without evidence of multiple myeloma (MM),macroglobulinemia,amyloidosis (AL),or a related plasma cell proliferative disorder. This study aims to evaluate laboratory diagnostic characters of monoclonal gammopathies and investigates the correlation between monoclonal gammopathies and transforming growth factor β1 (TGFβ1). Immunofixation electrophoresis (IFE),serum protein electrophoresis (SPE),nephelometry and urine light chain ELISA were used for laboratory identification of monoclonal immunoglobulins. Plasma TGFβ1 was detected with double-antibodies ELISA. Lightcycler was used for single nucleotide polymorphism (SNP) analysis. Totally 2,007 cases of monoclonal immunoglobulin (M protein) were identified in 10,682 samples. The isotypes of M protein were IgG type 47.1%,IgA 23.0%,IgM 8.7%,IgD 5.3%,free light chain κ 6.1%,λ 9.8%. In reference to IFE,the coherency of diagnosis was serum light chain ratio (κ/λ) 94.4%,quantitation of Igs 83%,light chain quantitation 80.9%,and urine light chain ratio (κ/λ) 58.0%. Plasma TGFβ1 was elevated significantly compared to normal control. The allelic frequency of codon 10 (C > T) was neither associated with the existence of the M protein nor with the M protein isotype. Monoclonal gammopathies can be identified with the combination of IFE,SPE,Igs quantitaion and urine light chain determination.
