3Effect of welding parameters on the heat-affected zone of AISI409 ferritic stainless steel显示文摘One of the main problems during the welding of ferritic stainless steels is severe grain growth within the heat-affected zone(HAZ).In the present study,the microstructural characteristics of tungsten inert gas(TIG) welded AISI409 ferritic stainless steel were investigated by electron backscattered diffraction(EBSD),and the effects of welding parameters on the grain size,local misorientation,and low-angle grain boundaries were studied.A 3-D finite element model(FEM) was developed to predict the effects of welding parameters on the holding time of the HAZ above the critical temperature of grain growth.It is found that the base metal is not fully recrystallized.During the welding,complete recrystallization is followed by severe grain growth.A decrease in the number of low-angle grain boundaries is observed within the HAZ.FEM results show that the final state of residual strains is caused by competition between welding plastic strains and their release by recrystallization.Still,the decisive factor for grain growth is heat input.Eslam Ranjbarnodeh Stefanie Hanke Sabine Weiss Alfons Fischer 2012International Journal of Minerals,Metallurgy and Materials2012,19,10:6
4Progressive loss of striatal dopamine terminals in MPTP-induced acute parkinsonism in cynomolgus monkeys using vesicular monoamine transporter type 2 PET imaging([^(18)F]AV-133)显示文摘The 1-methyl-4-phenyl-1,2,3,4-tetrahydropyridine(MPTP)-induced parkinsonism model, particularly in non-human primates, remains the gold-standard for studying the pathogenesis and assessing novel therapies for Parkinson's disease. However, whether the loss of dopaminergic neurons in this model is progressive remains controversial, mostly due to the lack of objective in vivo assessment of changes in the integrity of these neurons. In the present study, parkinsonism was induced in cynomolgus monkeys by intravenous administration of MPTP(0.2 mg/kg) for 15 days; stable parkinsonism developed over 90 days, when the symptoms were stable. Noninvasive positron emission tomographic neuroimaging of vesicular monoamine transporter 2 with 9-[18F]fluoropropyl-(+)-dihydrotetrabenazine([18F]AV-133) was used before, and 15 and 90 days after the beginning of acute MPTP treatment. The imaging showed evident progressive loss of striatal uptake of [18F]AV-133. The dopaminergic denervation severity had a significant linear correlation with the clinical rating scores and the bradykinesia subscores. 7Genome-wide DNA hypermethylation and homocysteine increase a risk for myopia显示文摘AIM: To test for the association between genome-wide methylation and myopia in human and mice. METHODS: Long interspersed nucleotide element 1(LINE-1) methylation levels were used to surrogate genome-wide methylation level. We first tested for the association between high myopia(<-6 D) and LINE-1 methylation in leukocytes in 220 cases and 220 control subjects. Secondly, we validated the results of LINE-1 methylation in eyes from the form deprivation myopia(FDM) mice. Furthermore,we calculated the correlation of LINE-1 methylation levels between leukocyte DNA and ocular DNA in the mice. We also tested whether dopamine can alter LINE-1 methylation levels. RESULTS: The LINE-1 methylation level was significantly higher in the myopic human subjects than controls. The upper and middle tertiles of the methylation levels increased an approximately 2-fold(P≤0.002) risk for myopia than the lower tertile. Similarly, FDM mice had high LINE-1 methylation levels in the leukocyte, retina and sclera, and furthermore the methylation levels detected from these three tissues were significantly correlated. Immunohistochemical staining revealed higher levels of homocysteine and methionine in the rodent myopic eyes than normal eyes. Dopamine treatment to the cells reduced both LINE-1 methylation and DNA methyltransferase levels. CONCLUSION: LINE-1 hypermethylation may be associated with high myopia in human and mice. 8G-Quadruplex(G4) Motifs in the Maize(Zea mays L.) Genome Are Enriched at Specific Locations in Thousands of Genes Coupled to Energy Status,Hypoxia,Low Sugar,and Nutrient Deprivation显示文摘The G-quadruplex(G4) elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix.This G4 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA.The reversibility and structural diversity of G4 s make them highly versatile genetic structures,as demonstrated by their roles in various functions including telomere metabolism,genome maintenance,immunoglobulin gene diversification,transcription,and translation.Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci.To investigate their potential roles in a large-genome model plant species,we computationally identified 149,988 non-telomeric G4 motifs in maize(Zea mays L.,B73 AGPv2),29%of which were in non-repetitive genomic regions.G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand,one in the 5'UTR and the other at the 5'end of the first intron.Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro.The G4 motifs were prevalent in key regulatory genes associated with hypoxia(group VII ERFs),oxidative stress(DJ-1/GATase1),and energy status(AMPK/SnRK) pathways.They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis,sugar degradation,inositol metabolism,and base excision repair.Collectively,the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses.Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize,a model genetic system and major world crop species for feed,food,and fuel.Carson M.Andorf Mykhailo Kopylov Drena Dobbs Karen E.Koch M.Elizabeth Stroupe Carolyn J.Lawrence Hank W.Bass 2014Journal of Genetics and Genomics2014,41,12:2

11Pediatric obesity prevention:From na?ve examination of energy imbalance towards strategies that influence the competition for nutrient resources among tissues显示文摘Current pediatric obesity interventions have collectively yielded relatively unsuccessful results. In this Field of Vision, we present plausible physiologic underpinnings fostering ineffectiveness of conventional strategies grounded in requisite induction of negative energy imbalance. Moreover, such recommendations exacerbate the underlying metabolic dysfunction by further limiting metabolic fuel availability, lowering energy expenditure, and increasing hunger(recapitulating the starvation response amid apparent nutritional adequacy) which precede and promote obesity during growth and development. The qualitative aspects of musculoskeletal system(i.e., endocrine response, muscle functional capacity) are likely to improve metabolic function and increase nutrient delivery and utilization. An intricate and complex system including multiple feedback mechanisms operates to homeostatically regulate energy balance and support optimal body composition trajectories and metabolic health, during growth and development. Thus, ignoring the interdependencies of regulatory growth processes initiates a nuanced understanding of energy regulation and thus misguided attempts at preventive strategies. Importantly, these gains are not dependent upon weight-loss, rather we suggest can be achieved through resistance training. Collectively, optimizing musculoskeletal health via resistance training elicits augmentation of competitive capacity across systems. 15Inhibition of autophagy rescues HT22 hippocampal neurons from erastin-induced ferroptosis显示文摘Ferroptosis is a regulated form of cell death which is considered an oxidative iron-dependent process.The lipid hydroperoxidase glutathione peroxidase 4 prevents the iron(Fe2+)-dependent formation of toxic lipid reactive oxygen species.While emerging evidence indicates that inhibition of glutathione peroxidase 4 as a hallmark of ferroptosis in many cancer cell lines,the involvement of this biochemical pathway in neuronal death remains largely unclear.Here,we investigate,first whether the ferroptosis key players are involved in the neuronal cell death induced by erastin.The second objective was to examine whether there is a cross talk between ferroptosis and autophagy.The third main was to address neuron response to erastin,with a special focus on ferritin and nuclear receptor coactivator 4-mediated ferritinophagy.To test this in neurons,erastin(0.5-8μM)was applied to hippocampal HT22 neurons for 16 hours.In addition,cells were cultured with the autophagy inhibitor,3-methyladenin(10 mM)and/or ferroptosis inhibitors,ferrostatin 1(10-20μM)or deferoxamine(10-200μM)before exposure to erastin.In this study,we demonstrated by immunofluorescence and western blot analysis,that erastin downregulates dramatically the expression of glutathione peroxidase 4,the sodium-independent cystine-glutamate antiporter and nuclear receptor coactivator 4.The protein levels of ferritin and mitochondrial ferritin in HT22 hippocampal neurons did not remarkably change following erastin treatment.In addition,we demonstrated that not only the ferroptosis inhibitor,ferrostatin1/deferoxamine abrogated the ferroptotic cell death induced by erastin in hippocampal HT22 neurons,but also the potent autophagy inhibitor,3-methyladenin.We conclude that(1)erastin-induced ferroptosis in hippocampal HT22 neurons,despite reduced nuclear receptor coactivator 4 levels,(2)that either nuclear receptor coactivator 4-mediated ferritinophagy does not occur or is of secondary importance in this model,(3)that ferroptosis seems to share some features of the autophagic cell death process.Nora Hanke Abdelhaq Rami 2023Neural Regeneration Research2023,18,7:2

19Compliant Mechanism Synthesis by Using Elastic Similitude显示文摘Compliant mechanisms have several advantages, especially smaller number of elements and therefore less movable joints. The flexural members furthermore allow an integration of special functions like balancing or locking. Especially fiber reinforced materials exhibit a wide range of function integration considering their compliance in passive as well active applications. To take advantage of compliant elements in applications a robust synthesis tool is needed. The synthesis based on topology optimization method or the pseudo rigid body approach leads to complex structures. Considering the use of fiber reinforced material a synthesis approach which leads to less complex structures is more suitable. For building up simple structures, with only one cantilever beam as compliant element(B) a graphical approach using the elastic similitude is the most efficient method. A step-by-step synthesis procedure is presented to synthesize compliant mechanisms with rotatory joints(R) and prismatic joints(P) to develop RRB/PRB- and RPB-linkages. Using the elastic similitude to implement these results into a graphical synthesis algorithm is the innovation part of this paper. It can be shown that this approach leads to a comfortable handling of beam elements during the synthesis, where the two free parameters can be directly coupled to scale and fix the orientation of the beam element. This advantage inherently shortens the development process. In giving an example the focus lies of the experimental approach, which also shows that the simple BERNOULLI beam model is valid and so the synthesis by using the elastic similitude.
