3Partially hydrolyzed guar gum attenuates non-alcoholic fatty liver disease in mice through the gut-liver axis显示文摘BACKGROUND The gut-liver axis has attracted much interest in the context of chronic liver disease pathogenesis.Prebiotics such as dietary fibers were shown to attenuate non-alcoholic fatty liver disease(NAFLD)by modulating gut microbiota.Partially hydrolyzed guar gum(PHGG),a water-soluble dietary fiber,has been reported to alleviate the symptoms of various intestinal diseases and metabolic syndromes.However,its effects on NAFLD remain to be fully elucidated.To determine whether treatment with PHGG attenuates NAFLD development in mice through the gut-liver axis.METHODS Seven-week-old male C57BL/6J mice with increased intestinal permeability were fed a control or atherogenic(Ath)diet(a mouse model of NAFLD)for 8 wk,with or without 5%PHGG.Increased intestinal permeability was induced through chronic intermittent administration of low-dose dextran sulfate sodium.Body weight,liver weight,macroscopic findings in the liver,blood biochemistry[aspartate aminotransferase(AST)and alanine aminotransferase(ALT),total cholesterol,triglyceride,free fatty acids,and glucose levels],liver histology,myeloperoxidase activity in liver tissue,mRNA expression in the liver and intestine,serum endotoxin levels in the portal vein,intestinal permeability,and microbiota and short-chain fatty acid(SCFA)profiles in the cecal samples were investigated.RESULTS Mice with increased intestinal permeability subjected to the Ath diet showed significantly increased serum AST and ALT levels,liver fat accumulation,liver inflammatory(tumor necrosis factor-αand monocyte chemotactic protein-1)and fibrogenic(collagen 1a1 andαsmooth muscle actin)marker levels,and liver myeloperoxidase activity,which were significantly attenuated by PHGG treatment.Furthermore,the Ath diet combined with increased intestinal permeability resulted in elevated portal endotoxin levels and activated toll-like receptor(TLR)4 and TLR9 expression,confirming that intestinal permeability was significantly elevated,as observed by evaluating the lumen-to-blood clearance of fluorescein isothiocyanate-conjugated dextran.PHGG treatment did not affect fatty acid metabolism in the liver.However,it decreased lipopolysaccharide signaling through the gut-liver axis.In addition,it significantly increased the abundance of cecal Bacteroides and Clostridium subcluster XIVa.Treatment with PHGG markedly increased the levels of SCFAs,particularly,butyric acid,acetic acid,propionic acid,and formic acid,in the cecal samples.CONCLUSION PHGG partially prevented NAFLD development in mice through the gut-liver axis by modulating microbiota and downstream SCFA profiles.Shun Takayama Kazuhiro Katada Tomohisa Takagi Takaya Iida Tomohiro Ueda Katsura Mizushima Yasuki Higashimura Mayuko Morita Tetsuya Okayama Kazuhiro Kamada Kazuhiko Uchiyama Osamu Handa Takeshi Ishikawa Zenta Yasukawa Tsutomu Okubo Yoshito Itoh Yuji Naito 2021World Journal of Gastroenterology2021,27,18:4

5Critical function of DNA methyltransferase 1 in tomato development and regulation of the DNA methylome and transcriptome显示文摘DNA methylation confers epigenetic regulation on gene expression and thereby on various biological processes.Tomato has emerged as an excellent system to study the function of DNA methylation in plant development.To date,regulation and function of DNA methylation maintenance remains unclear in tomato plants.Here,we report the critical function of tomato(Solanum lycopersicum)Methyltransferase 1(Sl MET1)in plant development and DNA methylome and transcriptome regulation.Using CRISPR-Cas9 gene editing,we generated slmet1 mutants and observed severe developmental defects with a frame-shift mutation,including small and curly leaves,defective inflorescence,and parthenocarpy.In leaf tissues,mutations in Sl MET1 caused CG hypomethylation and CHH hypermethylationon a whole-genome scale,leading to a disturbed transcriptome including ectopic expression of many RIN target genes such as ACC2 in leaf tissues,which are normally expressed in fruits.Neither the CG hypomethylation nor CHH hypermethylation in the slmet1 mutants is related to tissue culture.Meanwhile,tissue culture induces non-CG hypomethylation,which occurs more frequently at gene regions than at TE regions.Our results depict Sl MET1-and tissue culture-dependent tomato DNA methylomes,and that Sl MET1 is required for maintaining a normal transcriptome and normal development of tomato.Yu Yang Kai Tang Tatsiana U Datsenka Wenshan Liu Suhui Lv Zhaobo Lang Xingang Wang Jinghui Gao Wei Wang Wenfeng Nie Zhaoqing Chu Heng Zhang Avtar K HANDa Jian-Kang Zhu Huiming Zhang 2019Journal of Integrative Plant Biology2019,61,12:3
6Mitochondrial DNA from hepatocytes as a ligand for TLR9: Drivers of nonalcoholic steatohepatitis?显示文摘Nonalcoholic fatty liver disease(NAFLD) is the most common liver disease worldwide, affecting approximately one third of the Western world. It consists of a wide spectrum of liver disorders, ranging from fatty liver to nonalcoholic steatohepatitis(NASH), which consists of steatosis, ballooning injury and inflammation. Despite an alarming growth in the statistics surrounding NAFLD, there are as yet no effective therapies for its treatment. Innate immune signaling has been thought to play a significant role in initiating and augmenting hepatic inflammation, contributing to the transition from nonalcoholic fatty liver to NASH. An immune response is triggered by countless signals called damage-associated molecular patterns(DAMPs) elicited by lipid-laden and damaged hepatocytes, which are recognized by pattern recognition receptors(PRRs) on hepatic immune cells to initiate inflammatory signaling. In this editorial, in addition to summarizing innate immune signaling in NAFLD and discussing potential therapies that target innate immune pathways, we have described a recent study that demonstrated that mitochondrial DNA serves as a DAMP activating a hepatic PRR, TLR9, in mice and in the plasma of NASH patients. 11Primary rhegmatogenous retinal detachment:evaluation of a minimally restricted face-down positioning after pars plana vitrectomy and gas tamponade显示文摘AIM:To evaluate the safety and efficacy of a minimally restricted face-down postoperative positioning following pars plana vitrectomy(PPV)with gas tamponade for primary rhegmatogenous retinal detachment(RRD).METHODS:Patients with primary RRD treated with PPV and gas tamponade and followed up for at least 6 mo were selected for the study.All phakic eyes underwent simultaneous cataract surgery.The patients were required to be in a postoperative position that prevented downward flow of retinal tears.Patients with macular detachment were positioned face-down for only a couple of hours.The patients were assessed for preoperative and postoperative best-corrected visual acuity(BCVA),anatomical retinal reattachment rate,and postoperative complications.RESULTS:In total,40 eyes of 39 patients with primary RRD were included in the study.A single tear was present in 30 eyes(75.0%),multiple retinal tears were present in nine eyes(22.5%),and oral dialysis was present in one eye(2.5%).The anatomical success rate was 90.0%(36 cases)after the primary surgery,and the final anatomical success rate was 100%.The BCVA improved significantly(P<0.001)from 0.75 logarithm angle of resolution(log MAR)preoperatively to 0.12 log MAR at the final visit.Postoperative complications included intraocular pressure elevation(≥25 mm Hg)in 11 patients(27.5%),fibrin formation in two patients(5.0%),pupillary capture of the intraocular lens in two patients(5.0%),and posterior synechia in one patient(2.5%).CONCLUSION:A minimally restricted face-down and flexible postoperative positioning after PPV and gas tamponade for primary RRD is effective and safe.Kiichiro Kusaba Kotaro Tsuboi Tsuneaki Handa Yukihiko Shiraki Takuya Kataoka Motohiro Kmaei 2021International Journal of Ophthalmology(English edition)2021,14,6:2
