1Vaccine against Helicobacter pylori : Inevitable approach显示文摘Over three decades have passed since the discovery of Helicobacter pylori(H. pylori), and yet many questions about its treatment remain unanswered. For example, there is no certainty regarding continued use of current antibiotic therapy against H. pylori. The bad news is that even combined regimens are also unable to eradicate bacterial colonization. The worst problem with H. pylori chemotherapy is that even if we identify the most successful regimen, it cannot eliminate the risk of re-infection. This problem is further complicated by the fact that clinicians have no information as to whether probiotics are useful or not. Moreover, to date, we have no large scale produced vaccine effective against H. pylori. Due to the relatively rapid and abundant dissemination of guidelines globally reported concerning management of gastric cancer prevention and therapeutic regimens, clinicians may choose a vaccine as better effective weapon against H. pylori. Therefore, a radical shift in adopted strategies is needed to guide ultimate decisions regarding H. pylori management. In light of failures in vaccine projects, we should identify better vaccine design targeting conserved/essential genes. The unique character and persistence of H. pylori pose obstacles to making an effective vaccine. Preferably, in developing countries, the best reasonable and logical approach is to recommend prophylactic H. pylori vaccine among children as an obligatory national program to limit primary colonization. Trying to produce a therapeutic vaccine would be postponed until later. In reality, we should not forget to prescribe narrow spectrum antibiotics. 2Strategies used by helicobacter pylori to establish persistent infection显示文摘Helicobacter pylori(H. pylori) is a Gram-negative and motile bacterium that colonizes the hostile microniche of the human stomach, then persists for the host's entire life, if not effectively treated. Clinically, H. pylori plays a causative role in the development of a wide spectrum of diseases including chronic active gastritis, peptic ulceration, gastric adenocarcinoma, and gastric mucosaassociated lymphoid tissue lymphoma. Due to the global distribution of H. pylori, it is no exaggeration to conclude that smart strategies are contributing to adaptation of the bacterium to its permanent host. Thirty-four years after the discovery of this bacterium, there are still many unanswered questions. For example, which strategies help the bacterium to survive in this inhospitable microniche? This question is slightly easier to answer if we presume the same clinical concept for both persistent infection and disease. Understanding the mechanisms governing H. pylori persistence will improve identification of the increased risk of diseases such as gastric cancer in patients infected with this bacterium. A well-defined and longterm equilibrium between the human host and H. pylori allows bacterial persistence in the gastric microniche; although this coexistence leads to a high risk of severe diseases such as gastric cancer. To escape the bactericidal activity of stomach acid, H. pylori secretes large amounts of surface-associated and cytosolic urease. The potential to avoid acidic conditions and immune evasion are discussed in order to explain the persistence of H. pylori colonization in the gastric mucosa, and data on bacterial genetic diversity are included. Information on the mechanisms related to H. pylori persistence can also provide the direction for future research concerning effective therapy and management of gastroduodenal disorders. 3Application of Trichoderma harzianum (T22) and Trichoderma atroviride (P1) as plant growth promoters, and their compatibility with copper oxychloride显示文摘Trichoderma strains are used in agriculture because they provide to the plants the following benefits: i) are rhizosphere competence and establish stable rhizosphere microbial communities; ii) control plant disease caused by pathogenic and competitive microflora, by using a variety of mechanisms; iii) improve vegetative growth, root development and yield; iv) make nutrients more available to the plant. In this work we have investigated the ability of T. harzianum T22 and T. atroviride P1 to improve plant growth of locally important horticultural crops: lettuce, tomatoes and peppers and to prevent disease in the greenhouse and field. The effect of the Trichoderma treatment was evaluated by determining the weight of fresh and dry roots and above ground plant biomass, measuring plants height, counting the number of emerged leaves (lettuce, tomatoes and peppers) and quantifying production (tomatoes and peppers). No disease symptoms were found during production, although Fusarium sp. strains pathogenic to tomato were detected in the soil. Compounds containing copper oxychloride are frequently used for fungal disease control in agriculture. In order to investigate the compatibility of T. harzianum T22 and T. atroviride P1 with copper oxychloride applications, the effect on mycelia growth was monitored in both liquid and solid medium. 4Resistance to clarithromycin and gastroenterologist's persistence roles in nomination for Helicobacter pylori as high priority pathogen by World Health Organization显示文摘Due to the increasing prevalence of clarithromycin resistance, future of management of Helicobacter pylori(H. pylori) infections need to be recognized. To now, clarithromycin was the best effective, well-tolerated and safe antibiotic used in treatment of the bacterium, but, increasing trend of resistance reduced efficacy of recommended regimens. Indeed, gastroenterologists are mostly unable to start appropriate therapyaccording to the sensitivity profile-due to the certain difficulties in routine H. pylori culture procedure and being time consuming method. This announcement by World Health Organization(WHO) was an onset to reconsider current challenging dilemma about H. pylori clarithromycin resistant isolates. Therefore, investigating of various factors affecting this nomination by WHO is highly welcomed. In fact, WHO enumerated more than 16 pathogens which seriously threats human life and public health, thus better management or effective guidelines are necessary. Here for the first time, we nominated this phenomenon as ‘‘gastroenterologist's persistence'' which should be equally investigated as antibiotic resistance. The ability of gastroenterologists to win the game against H. pylori infections is highly influenced by their collaboration with diagnostic laboratories to apply susceptibility patterns before any prescription. 5Development of a procedure for estimating the parameters of mechanistic VOC emission source models from chamber testing data显示文摘In order to evaluate the impacts of volatile organic compounds(VOCs)emissions from building materials on the indoor air quality beyond the standard chamber test conditions and test period,mechanistic emission source models have been developed in the past.However,very limited data are available for the required model parameters including the initial concentration(C_(m0)),in-material diffusion coefficient(D_(m)),partition coefficient(Kma),and convective mass transfer coefficient(k_(m)).In this study,a procedure was developed for estimating the model parameters by using VOC emission data from standard small chamber tests.In the procedure,initial values of the model parameters were refined by multivariate regression analysis of the measured emission data.To verify the procedure and estimate its uncertainty,simulated chamber test data were generated by adding 10% experimental uncertainties on the theoretical curve from the analytical solution to a mechanistic emission model.Then the procedure was applied to the generated data to estimate the model parameters.Results indicated that estimates converged to the original parameter values used for the data generation and the error of estimated parameters D_(m1)C_(m0) and K_(ma) were within±10%,±23%,and±25%of the true values,respectively.The procedure was further demonstrated by applying it to estimate the model parameters from real chamber test data.Wide application of the procedure would result in a database of mechanistic source model parameters for assessing the impact of VOC emissions on indoor pollution load,which are essential input data for evaluating the effectiveness of various indoor air quality(IAQ)design and control strategies as well as the energy required for meeting given IAQ requirements.Zhenlei Liu Andreas Nicolai Marc Abadie Menghao Qin John Grunewald Jianshun Zhang 2021Building Simulation2021,14,2:4
6Synergists action of piperonyl butoxide and S,S,S-tributyl phosphorotrithioate on toxicity of carbamate insecticides against Blattella germanica显示文摘Objective: To determine the synergists action of piperonyl butoxide(PBO) and S,S,Stributyl phosphorotrithioate(DEF) on toxicity of carbamate insecticides against Blattella germanica in Tehran city.Methods: In the current study, German cockroach strains were collected from several hospitals and dormitories in Tehran.At the beginning, different concentrations of bendiocarb and carbaryl(insecticides belong to carbamate group) were determined by surface contact on a susceptible strain.Then, the level of susceptibility and type of resistance mechanisms in the collected strains from contaminated sites to the aforementioned insecticides were studied by using PBO and DEF synergists with different insecticide ratios to synergist(1:0, 1:1, 1:2, 1:3).Results: The DEF synergist along with bendiocarb and carbaryl completely eliminated the resistance in all strains but PBO did not completely eliminate the resistance in the strains of Mofid, Alvand, Valiasr hospitals and Shariati dormitory.Generally, the impact of DEF was observed in the removing resistance more than PBO.Conclusions: In most of these strains, resistance to bendiocarb and carbaryl is completely eliminated by DEF, showing a very high role of estraze enzymes in resistance to bendiocarb and carbaryl.But in most strains PBO does not remove the resistance because other mechanisms, such as reduced cuticle penetration and insensitivity to the acetylcholine esterase enzyme, may be involved.Alireza Sanei Dehkordi Yaser Salim Abadi Hasan Nasirian Teymour Hazratian Mohammad Amin Gorouhi Saideh Yousefi Azim Paksa 2017Asian Pacific Journal of Tropical Medicine2017,10,10:3
7Role of dup A in virulence of Helicobacter pylori显示文摘Helicobacter pylori(H. pylori) is a gastric human pathogen associated with acute and chronic gastritis, 70% of all gastric ulcers, 85% of all duodenal ulcers, and both forms of stomach cancer, mucosal-associated lymphoid tissue(MALT) lymphoma and adenocarcinoma. Recently, attention has focused on possible relationship between presence of certain virulence factor and H. pylori-associated diseases. Some contradictory data between this bacterium and related disorders has been observed since not all the colonized individuals develop to severe disease. The reported diseases plausibility related to H. pylori specific virulence factors became an interesting story about this organism. Although a number of putative virulence factors have been identified including cytotoxin-associated gene a(cag A) and vac A, there are conflicting data about their actual participation as specific risk factor for H. pylori-related diseases. Duodenal ulcer promoting gene a(dup A) is a virulence factor of H. pylori that is highly associated with duodenal ulcer development and reduced risk of gastric cancer. The prevalence of dup A in H. pylori strains isolated from western countries is relatively higher than in H. pylori strains from Asian countries. Current confusing epidemiological reports will continue unless future sophisticated and molecular studies provide data on functional and complete dup A cluster in H. pylori infected individuals. 9Fecal microbiota transplantation against irritable bowel syndrome?Rigorous randomized clinical trials are required显示文摘Halkj?r et al searched systematically nine articles including 48 patients, and concluded that fecal microbiota transplantation(FMT) can be an ideal treatment option for irritable bowel syndrome(IBS) subjects. Regardless of the few successes in current traditional therapies(change in diet, herbal medicine and antibiotics) in IBS, a sharp increase in interests in the FMT option has been reported in the current century. However, there is a long list of unclear issues concerning the application of FMT for the treatment of IBS. 10Chinese Helicobacter pylori vaccine: Solution for an old challenge?显示文摘Helicobacter pylori(H. pylori) is an important cause for gastric cancer in high risk individuals. H. pylori colonizes more than 50% of the world's population and associated peptic ulcer disease and gastric malignancy have important public health implications. It has been classified as a class Ⅰ carcinogen in 1994 by the World Health Organization. Clinicians are often prompted to eliminate the infection the moment it is detected. This also, unfortunately, led to reckless use of antibiotics and reports of increasing resistance are now worldwide. Each year, many of people die from gastric cancer; thus application of effective vaccine can reduce this relatively high mortality worldwide. H. pylori can be eliminated by antibiotics but efficacy is sharply decreasing. Moreover, current therapy is also expensive and with side effects. Vaccine may be the best solution to the above problem but there are many challenges in producing such an effective therapeutic vaccine. Recently, the Chinese group published in Lancet, a single-center, randomized, phase Ⅲ study of an oral recombinant vaccine(Urease B subunit fused with heat-labile enterotoxin B derived from Escherichia coli) prescribed in the Chinese children(6-15 years) without a history of H. pylori infection. 12Identification and characterization of Trichoderma species aggressive to Pleurotus in Italy显示文摘In the late 1980’s the deve lop ment of a severe epidemic of green mold caused by Trichoderma spp. was not ed in the commercial production of Agaricus bisporus (champignon) in the U nited Kingdom, North America, Spain and Holland, which caused extensive economic losses. The parasitic fungi isolated from the edible mushroom belonged to four biotypes, Th1, Th2, Th3 and Th4 of T. harzianum. However, among these biotypes, only Th2 (since c lassified as T. aggressivum f. europaeum) and Th4 (T. aggressivum f. aggressivum) were identified as the fungi causing problems in Agaricus production. In general, mushroom compost hosts both aggressive and innocuous is olates of Trichoderma, which are not morphologically distinguishable. Abo ut four years ago, a problem with green mold became apparent in the production o f Pleurotus ostreatus in Northern Italy, which eventually developed to a c risis situation in the South two years later and threatened to seriously comprom ise the Pleurotus market. This study was initiated to: isolate and identif y the aggressive fungi, then morphologically, physiologically and genetically characterize the isolates, dete rmine the source and phases of infection, and study methods of control. Samples were obtained from different phases of compost preparation at the locality of a major producer and supplier of compost to the mushroom industry in Southern Ital y, and microbial counts were conducted. Although the presence of Trichoderma was detected in the initial stages of composting, this value was reduced to zero from the phase of pasteurization to seeding with Pleurotus. Trichoderma infestations were noted in the packaged Pleurotus bales at various time s during the incubation phase (7-15 days after seeding) and after shipping to th e mushroom greenhouses, where the pathogen infestations greatly reduced the qual ity and quantity of the mushroom yield, as well as the number of potential harvest cycles. Preliminary r esults from the morphological and genetic characterization of Trichoderma isolates parasitic to Pleurotus indicated that they are different from bot h T. aggressivum forms parasitic to Agaricus, and the majority of the isolates probably belong to the species T. harzianum. In vitr o confrontation plates were performed with 26 isolates of aggressive Trich oderma obtained from compost, three Trichoderma isolates used in biolog ical control and 12 varieties of Pleurotus. No inhibitory effect was obse rved between any of the Trichoderma isolates with Pleurotus, althou gh some growth inhibition was caused by the biocontrol isolates of Trichoderm a on some of the aggressive isolates. The temperature optimum for Pleurotus growth was at 28 ℃, whereas Trichoderma grew well at a wider range (20- 28 ℃), and exceeded the growth rate of Pleurotus by three times at 25 ℃. T he pH optimum for the growth of Pleurotus was alkaline (pH 8-9) whereas Trichoderma preferred acidic-neutral pH (5-7) . Various commercial fungicides used in agriculture (procloraz, thiabendazole, dichloran, benomyl, p r opiconazole, thiofanatomethyl) were tested against the aggressive and biocontro l isolates of Trichoderma, as well as the different varieties of Pleuro tus to determine dose response curves and combinations that would inhibit spo re germination, mycelial growth and subsequent sporulation.
