| 1 | Experimental study on anaphylaxis of Qingkailing injection and its components on Beagle dogs显示文摘OBJECTIVE:To study the anaphylaxis of Qingkailing injection(QI) and its components.METHODS:Experimental anaphylactoid and allergic reactions were used.Changes in the behaviors of Beagles and serum levels of histamine,immunoglobulin(Ig)E,IgG,IgM,eosinophil cationic protein(ECP),and interleukin(IL)-4,as well as blood pressure,after injecting QI and its components on the forelimb veins of Beagles were observed.RESULTS:According to comprehensive determination of abnormal behavior scores and changes in serum levels of histamine,IgE,IgG,IgM,ECP,and IL-4,as well as in blood pressure,radix isatidis and hyodeoxycholic acid caused anaphylactoid reactions,and honeysuckle,radix isatidis,hydrolysate,cholic acid and Gardenia jasminoides caused allergic reactions.The anaphylaxis of QI involved anaphylactoid and allergic reactions.CONCLUSION:QI and its components need to be refined further to improve the safety,efficacy,and quality of its use in clinical settings. | Zhiguo Wang Danqiao Wang Yu Sui Haifeng Cui Youhua Yu | 2012 | Journal of Traditional Chinese Medicine2012,32,4: | 6 |
| 2 | Chinese herbal medicine enhances sexual function and c-Fos/nNOS expression in the nucleus accumbens of orchidectomized rats显示文摘OBJECTIVE: To determine whether the central nervous system is involved in the effect of Chinese herbal medicine on sexual function recovery in orchidectomized rats.METHODS: Orchidectomized rats were administered intragastrically with a decoction of 'kidneynourishing' Chinese herbal medicine once per day for 28 days. Accessory genital organ weight, plasma testosterone, and mating behavior were investigated. The expression of c-Fos and neuronal nitric oxide synthase(n NOS) in neuronal cells in the nucleus accumbens(NAc) was analyzed by immunohistochemistry.RESULTS: There was a decrease in accessory genital organ weight, plasma testosterone, and sexual behavior, as well as a low number of c-Fos-positive cells and a large n NOS-positive cell area in orchidectomized rats. Administration of the herbal medicine increased accessory genital organ weight, testosterone level, mating behavior, and c-Fos-positive cell number, while it decreased the n NOS-positive cell area in orchidectomized rats.CONCLUSION: An increase of plasma testosterone after administration of 'kidney-nourishing' herbal medicine might contribute to the elevated sexual function and activity in orchidectomized rats. | 6 | Protection of 3'-methoxy-puerarin against focal brain ischemia and its association with c-fos expression显示文摘The present study established a rat model of focal brain ischemia by occlusion of the middle cerebral artery covered with FeCl3, and investigated the protective effect of 3'-methoxy-puerarin. Hippocampal and cortical c-fos gene expression was determined using in situ hybridization. Results showed that 3'-methoxy-puerarin reduced neurological deficit scores, cerebral infarcted zone and water content of brain tissues, dramatically increased the activity of catalase and glutathione peroxidase in the ischemia zone of the hippocampus, increased the activity of catalase in the cortex, decreased lipid peroxide and lactic acid contents in the hippocampus and cerebral cortex, and down-regulated c-fos gene expression in brain ischemic rats. | 7 | Homeobox B8 Targets Sterile Alpha Motif Domain-Containing Protein 9 and Drives Glioma Progression显示文摘Gliomas are the most commonly occurring tumors of the central nervous system. Glioblastoma multiforme (GBM) is the most malignant and aggressive brain cancer in adults. Further understanding of the mechanisms underlying the aggressive nature of GBM is urgently needed. Here we identified homeobox B8(HOXB8), a member of the homeobox family, as a crucial contributor to the aggressiveness of GBM. Data mining of publicly accessible RNA sequence datasets and our patient cohorts confirmed a higher expression of HOXB8 in the tumor tissue of GBM patients, and a strong positive correlation between the expression level and pathological grading of tumors and a negative correlation between the expression level and the overall survival rate. We next showed that HOXB8 promotes the proliferation and migration of glioblastoma cells and is crucial for the activation of the PI3K/AKT pathway and expression of epithelial–mesenchymal transition-related genes, possibly through direct binding to the promoter of SAMD9 (Sterile Alpha Motif Domain-Containing Protein 9) and activating its transcription. | 10 | Vasorelaxant effect of osthole on isolated thoracic aortic rings in rats显示文摘OBJECTIVE: To investigate the effect of osthole on isolated thoracic aortic rings, and to determine the potential mechanism of action.METHODS: Thoracic aortas were isolated from Wistar rats, and were suspended in tissue organ chambers for vascular tension measurement. The effect of cumulative osthole(10-9, 10-8, 10-7, 10-6, and 10-5 mol/L) on endothelium-intact and endothelium-denuded thoracic aortic rings pre-contracted with phenylephrine(PE, 10-6 mol/L) or KCl(6 × 10-2 mol/L) was recorded. Histomorphological changes of thoracic aorta were analyzed by hematoxylin-eosin. The effects of different osthole concentrations on endothelium-intact aortic rings, which were pre-inhibited with the non-selective nitric oxide synthase inhibitor L-Arg(NO2)-OMe·HCl(3 × 10-4 mol/L), endothelium-derived nitric oxide synthase inhibitor Nω-nitro-L-arginine(3 × 10-4 mol/L), guanylate cyclase inhibitor 1 H-[1,2,4] oxadiazolo [4,3-α]quinoxaline-1-one(10-5 mol/L), cyclooxygenase inhibitor indometacin(10-5 mol/L), and the Ca2+-activated potassium channel inhibitor tetraethylammonium nitrate(10-5 mol/L), and then contracted with PE, were examined. Aortic rings incubated with osthole(10-5 mol/L), phentolamine(10-5 mol/L), or verapamil(10-5 mol/L) in Ca2+-free Krebs-Henseleit solution(KHS) were stimulated with PE or KCl.RESULTS: There was a dose-dependent increase in vasorelaxation of isolated thoracic aortic rings(both with and without endothelium) with increasing osthole concentration. Hematoxylin-eosin staining showed that osthole significantly improved thoracic aorta ring morphology. Compared with the control group, there were also significant differences after incubation with L-Arg(NO2)-OMe ω-nitro-L-arginine, and 1 H-[1,2,4] oxadiazo·lo HCl,N [4,3-α] quinoxaline-1-one(P < 0.05 for all). The relaxation rate of the rings in the osthole group incubated with indometacin and tetraethylammonium nitrate were similar to controls. In Ca2+-free KHS, the PE-induced contraction was similar between the osthole(4.37% ± 0.41%) and control(4.21% ± 1.33%)groups. However, after cumulative CaCl2(0.5, 1, 1.5,2, 2.5, and 3 mmol/L), the Ca2+-induced contraction was significantly inhibited in the osthole and phentolamine groups compared with controls(P < 0.05).After cumulative CaCl2 was added to Ca2+-free KHS(high K+ concentration), the contraction rate was significantly higher than both of the control and the osthole groups(P < 0.05). | 13 | Changes in the epigenome and transcriptome of rice in response to Magnaporthe oryzae infection显示文摘DNA methylation participates in regulating the expression of coding and non-coding regions in plants. To investigate the association between DNA methylation and pathogen infection, we used whole-genome bisulfite sequencing to survey temporal DNA methylation changes in rice after infection with the rice blast fungus Magnaporthe oryzae. In contrast to previous findings in Arabidopsis, global DNA methylation levels in rice increased slightly after rice blast infection. We identified over 38,000 differentially methylated regions(DMRs), and hypermethylated DMRs far outnumbered hypomethylated DMRs. Most DMRs were located in transposable element regions. Using transcriptome analysis, we identified 8830 differentially expressed genes(DEGs) after 1, 3, and 5 days of infection. Over one-third of DEGs, most of which were CHH-type DMRs, were associated with DMRs. Functional analysis of the CHH DMR-DEGs indicated their involvement in many important biological processes, including cell communication and response to external stimulus. The transcription of many NBS-LRR family genes was affected by changes in DNA methylation, suggesting that DNA methylation plays essential roles in the response of rice to M. oryzae infection. | 14 | Multiple Endmember Hyperspectral Sparse Unmixing Based on Improved OMP Algorithm显示文摘In conventional linear spectral mixture analysis model,a class is represented by a single endmember.However,the intra-class spectral variability is usually very large,which makes it difficult to represent a class,and in this case,it leads to incorrect unmixing results. Some proposed algorithms play a positive role in overcoming the endmember variability,but there are shortcomings on computation intensive,unsatisfactory unmixing results and so on. Recently,sparse regression has been applied to unmixing,assuming each mixed pixel can be expressed as a linear combination of only a few spectra in a spectral library. It is essentially the same as multiple endmember spectral unmixing. OMP( orthogonal matching pursuit),a sparse reconstruction algorithm,has advantages of simple structure and high efficiency. However,it does not take into account the constraints of abundance non-negativity and abundance sum-to-one( ANC and ASC),leading to undesirable unmixing results. In order to solve these issues,this paper presents an improved OMP algorithm( fully constraint OMP,FOMP) for multiple endmember hyperspectral sparse unmixing. The proposed algorithm overcomes the shortcomings of OMP,and on the other hand,it solves the problem of endmember variability.The ANC and ASC constraints are firstly added into the OMP algorithm,and then the endmember set is refined by the relative increase in root-mean-square-error( RMSE) to avoid over-fitting,finally pixels are unmixed by their optimal endmember set. The simulated and real hyperspectral data experiments show that FOPM unmixing results are ideally comparable and abundance RMSE reduces much lower than OMP and simple spectral mixture analysis( s SMA),and has a strong anti-noise performance. | 16 | Optical investigation on polyoxymethylene dimethyl ethers spray flame at different oxygen levels in a constant volume vessel显示文摘In this paper,high-speed imaging and spectrometry diagnostics were used to study the spray flame structures and emission spectra of polyoxymethylene dimethyl ethers(PODE)in an optical constant volume vessel.The effects of oxygen volume fraction(15%to 80%)on some typical combustion characteristics such as spatially integrated natural intensity,flame propagation speed,lift-off length and the distribution of flame emission spectra were investigated.The results show that the PODE spray flame mainly exhibits blue chemiluminescence,which is different from the diesel yellow diffusion flame dominated by soot radiation.As the oxygen concentration increases,the flame natural luminosity and propagation speed increases,the position of lift-off region and flame tip move towards the injector nozzle,and the flame width becomes narrower.According to the spectral results,the blue chemiluminescence generated by carbon monoxide oxidation dominates the PODE flame luminescence,suggesting that no soot is formed.The increase in oxygen concentration leads to enlarged intensity and gradient of PODE flame radiation.In summary,the combination of PODE and oxygen-enriched combustion shows no soot formation and promotes the oxidation of carbon monoxide and unburned hydrocarbon.This study can provide more insights into PODE spray combustion,and offer guidelines for achieving efficient and clean combustion. | ZHENG ZunQing LIU Wei LIU HaiFeng FENG Lei CUI YanQing ZHANG Shuo YAO MingFa | 2021 | Science China(Technological Sciences)2021,64,8: | 1 |
