4From Golden Rice to aSTARice:Bioengineering Astaxanthin Biosynthesis in Rice Endosperm显示文摘Carotenoids are important phytonutrients with antioxidant properties,and are widely used in foods and feedstuffs as Supplements.Astaxanthin,a red-colored ketocarotenoid,has strong antioxidant activity and thus can benefit human health.However,astaxanthin is not produced in most higher plants.Here we report the bioengineering of astaxanthin biosynthesis in rice endosperm by introducing four synthetic genes,sZmPSY1,sPaCrtl,sCrBKT,and sHpBHY,which encode the enzymes phytoene synthase,phytoene desaturase,β-carotene ketolase,and β-carotene hydroxylase,respectively.Transgneic overexpression of two (sZmPSY1 and sPaCrtl),three (sZmPSY1,sPaCrtl and sCrBKT),and all these four genes driven by rice endosperm-specific promoters established the Carotenoid/ketocarotenoid/astaxanthin biosynthetic pathways in the endosperm and thus resulted in various types of germplasm,from the yellow-grained β-caro- tene-enriched Golden Rice to orange-red-grained Canthaxanthin Rice and Astaxanthin Rice,respectively. 8Motion trajectory prediction based on a CNN-LSTM sequential model显示文摘Accurate monitoring the surrounding environment is an important research direction in the field of unmanned systems such as bio-robotics,and has attracted much research attention in recent years.The trajectories of surrounding vehicles should be predicted accurately in space and time to realize active defense and running safety of an unmanned system.However,there is uncertainty and uncontrollability in the process of trajectory prediction of surrounding obstacles.In this study,we propose a trajectory prediction method based on a sequential model,that fuses two neural networks of a convolutional neural network(CNN)and a long short-term memory network(LSTM).First,a box plot is used to detect and eliminate abnormal values of vehicle trajectories,and valid trajectory data are obtained.Second,the trajectories of surrounding vehicles are predicted by merging the characteristics of CNN space expansion and LSTM time expansion;the hyper-parameters of the model are optimized according to a grid search algorithm,which satisfies the double-precision prediction requirement in space and time.Finally,data from next generation simulation(NGSIM)and Creteil roundabout in France are taken as test cases;the correctness and rationality of the method are verified by prediction error indicators.Experimental results demonstrate that the proposed CNNLSTM method is more accurate and features a shorter time cost,which meets the prediction requirements and provides an effective method for the safe operation of unmanned systems.Guo XIE Anqi SHANGGUAN Rong FEI Wenjiang JI Weigang MA Xinhong HEI 2020Science China(Information Sciences)2020,63,11:24

14Reinfection rate and endoscopic changes after successful eradication of Helicobacter pylori显示文摘AIM:To determine the long-term outcomes regarding reinfection with Helicobacter pylori(H.pylori)and endoscopic changes after successful H.pylori eradication. METHODS:From June 1994 to January 2007,186 patients(M:F=98:88;mean age 50.0±11.4 years), in whom H.pylori had been successfully eradicated, were enrolled.The mean duration of follow up was 41.2±24.0 mo. 15Effect of Astragalus membranaceus injection on the activity of the intestinal mucosal mast cells after hemorrhagic shock-reperfusion in rats显示文摘Background The mechanism of mucosal damage induced by ischemia-reperfusion (IR) after hemorrhagic shock is complex; mast cells (MC) degranulation is associated with the mucosal damage. Astragalus membranaceus can protect intestinal mucosa against intestinal oxidative damage after hemorrhagic shock, and some antioxidant agents could prevent MC against degranulation. This study aimed to observe the effects of astragalus membranaceus injection on the activity of intestinal mucosal mast cells (IMMC) after hemorrhage shock-reperfusion in rats Methods Thirty-two Wistar rats were randomly divided into the normal group, model group, low dosage group, (treated with Astragalus membranacaus injection, 10 g crude medication/kg) and high dosage group (treated with Astragalus membranacaus injection, 20 g crude medication/kg). The rat model of hemorrhagic shock-reperfusion was induced by hemorrhage for 60 minutes followed by 90 minutes of reperfusion. The animals were administrated with 3 ml of the test drug solution before reperfusion. At the end of study, intestinal pathology, ultrastructure of IMMC, and expression of tryptase were assayed. The levels of malondisldehyde (MDA), TNF-α, histamine, and superoxide dismutase (SOD) activity in intestine were detected, and the number of IMMC was counted. Results The Chiu’s score of the rats in the model group was higher than in other groups (P<0.01). The Chiu’s score in the high dosage group was higher than that in the low dosage group (P<0.05). Hemorrhage-reperfusion induced IMMC degranulation: Astragalus membranaceus injection attenuated this degranulation. Expression of tryptase and the number of IMMC in the model group increased compared with the other groups (P<0.01) and was significantly reduced by the treatments of Astragalus membranaceus injection at both doses. There was no significant difference between the two treatment groups (P>0.05). MDA content and concentration of TNF-α in the model group were higher than that in the other three groups (P<0.05), and the concentration of TNF-α in the low dosage group was higher than that in the high dosage group (P<0.05). SOD activity and the concentration of histamine in the model group were lower than the other three groups (P<0.05). There was a negative correlation betweenthe Chiu’s score and the concentration of histamine and a positive correlation between the Chiu’s score and the concentration of TNF-α and between the SOD activity and the concentration of histamine in the four groups (P<0.05). 17Kai Xin San ameliorates scopolamine-induced cognitive dysfunction显示文摘Kai Xin San(KXS, containing ginseng, hoelen, polygala, and acorus), a traditional Chinese herbal compound, has been found to regulate cognitive dysfunction; however, its mechanism of action is still unclear. In this study, 72 specific-pathogen-free male Kunming mice aged 8 weeks were randomly divided into a vehicle control group, scopolamine group, low-dose KXS group, moderate-dose KXS group, high-dose KXS group, and positive control group. Except for the vehicle control group and scopolamine groups(which received physiological saline), the doses of KXS(0.7, 1.4 and 2.8 g/kg per day) and donepezil(3 mg/kg per day) were gastrointestinally administered once daily for 2 weeks. On day 8 after intragastric treatment, the behavioral tests were carried out. Scopolamine group and intervention groups received scopolamine 3 mg/kg per day through intraperitoneal injection. The effects of KXS on spatial learning and memory, pathological changes of brain tissue, expression of apoptosis factors, oxidative stress injury factors, synapse-associated protein, and cholinergic neurotransmitter were measured. The results confirmed the following.(1) KXS shortened the escape latency and increased residence time in the target quadrant and the number of platform crossings in the Morris water maze.(2) KXS increased the percentage of alternations between the labyrinth arms in the mice of KXS groups in the Y-maze.(3) Nissl and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling staining revealed that KXS promoted the production of Nissl bodies and inhibited the formation of apoptotic bodies.(4) Western blot assay showed that KXS up-regulated the expression of anti-apoptotic protein Bcl-2 and inhibited the expression of pro-apoptotic protein Bax. KXS up-regulated the expression of postsynaptic density 95, synaptophysin, and brain-derived neurotrophic factor in the cerebral cortex and hippocampus.(5) KXS increased the level and activity of choline acetyltransferase, acetylcholine, superoxide dismutase, and glutathione peroxidase, and reduced the level and activity of acetyl cholinesterase, reactive oxygen species, and malondialdehyde through acting on the cholinergic system and reducing oxidative stress damage. 20Bifurcation and Chaos Analysis of Nonlinear Rotor System with Axial-grooved Gas-lubricated Journal Bearing Support显示文摘Axial-grooved gas-lubricated journal bearings have been widely applied to precision instrument due to their high accuracy, low friction, low noise and high stability. The rotor system with axial-grooved gas-lubricated journal bearing support is a typical nonlinear dynamic system. The nonlinear analysis measures have to be adopted to analyze the behaviors of the axial-grooved gas-lubricated journal bearing-rotor nonlinear system as the linear analysis measures fail. The bifurcation and chaos of nonlinear rotor system with three axial-grooved gas-lubricated journal bearing support are investigated by nonlinear dynamics theory. A time-dependent mathematical model is established to describe the pressure distribution in the axial-grooved compressible gas-lubricated journal bearing. The time-dependent compressible gas-lubricated Reynolds equation is solved by the differential transformation method. The gyroscopic effect of the rotor supported by gas-lubricated journal bearing with three axial grooves is taken into consideration in the model of the system, and the dynamic equation of motion is calculated by the modified Wilson-θ-based method. To analyze the unbalanced responses of the rotor system supported by finite length gas-lubricated journal bearings, such as bifurcation and chaos, the bifurcation diagram, the orbit diagram, the Poincaré map, the time series and the frequency spectrum are employed. The numerical results reveal that the nonlinear gas film forces have a significant influence on the stability of rotor system and there are the rich nonlinear phenomena, such as the periodic, period-doubling, quasi-periodic, period-4 and chaotic motion, and so on.
