1Advances and challenges in the treatment of esophageal cancer显示文摘Esophageal cancer(EC)is one of the most common cancers with high morbidity and mortality rates.EC includes two histological subtypes,namely esophageal squamous cell carcinoma(ESCC)and esophageal adenocarcinoma(EAC).ESCC primarily occurs in East Asia,whereas EAC occurs in Western countries.The currently available treatment strategies for EC include surgery,chemotherapy,radiation therapy,molecular targeted therapy,and combinations thereof.However,the prognosis remains poor,and the overall five-year survival rate is very low.Therefore,achieving the goal of effective treatment remains challenging.In this review,we discuss the latest developments in chemotherapy and molecular targeted therapy for EC,and comprehensively analyze the application prospects and existing problems of immunotherapy.Collectively,this review aims to provide a better understanding of the currently available drugs through in-depth analysis,promote the development of new therapeutic agents,and eventually improve the treatment outcomes of patients with EC.Shiming He Jian Xu Xiujun Liu Yongsu Zhen 2021Acta Pharmaceutica Sinica B2021,11,11:26

3Blunted nitric oxide regulation in Tibetans under high-altitude hypoxia显示文摘Nitric oxide(NO) is an important molecule for vasomotor tone, and elevated NO signaling was previously hypothesized as a unique and adaptive physiological change in highland Tibetans. However, there has been lack of NO data from Tibetans living at low altitude and lowlander immigrants living at high altitude, which is crucial to test this hypothesis. Here, through cross-altitude(1990–5018 m) and cross-population(Tibetans and Han Chinese) analyses of serum NO metabolites(NOx) of 2086 individuals, we demonstrate that although Tibetans have a higher serum NOx level compared to lowlanders, Han Chinese immigrants living at high altitude show an even higher level than Tibetans. Consequently, our data contradict the previous proposal of increased NO signaling as the unique adaptive strategy in Tibetans.Instead, Tibetans have a relatively lower circulating NOx level at high altitude. This observation is further supported by data from the hypoxic experiments using human umbilical vein endothelial cells and gene knockout mice. No difference is detected between Tibetans and Han Chinese for endothelial nitric oxide synthase(e NOS), the key enzyme for circulating NO synthesis, suggesting that e NOS itself is unlikely to be the cause. We show that other NO synthesis-related genes(e.g. GCH1) carry Tibetan-enriched mutations significantly associated with the level of circulating NOx in Tibetans. 4De novo assembly of a Tibetan genome and identification of novel structural variants associated with high-altitude adaptation显示文摘Structural variants(SVs) may play important roles in human adaptation to extreme environments such as high altitude but have been under-investigated. Here, combining long-read sequencing with multiple scaffolding techniques, we assembled a high-quality Tibetan genome(ZF1), with a contig N50 length of 24.57 mega-base pairs(Mb) and a scaffold N50 length of 58.80 Mb. The ZF1 assembly filled 80 remaining N-gaps(0.25 Mb in total length) in the reference human genome(GRCh38). Markedly, we detected17 900 SVs, among which the ZF1-specific SVs are enriched in GTPase activity that is required for activation of the hypoxic pathway. Further population analysis uncovered a 163-bp intronic deletion in the MKL1 gene showing large divergence between highland Tibetans and lowland Han Chinese. This deletion is significantly associated with lower systolic pulmonary arterial pressure, one of the key adaptive physiological traits in Tibetans. Moreover, with the use of the high-quality de novo assembly, we observed a much higher rate of genome-wide archaic hominid(Altai Neanderthal and Denisovan) shared non-reference sequences in ZF1(1.32%–1.53%) compared to other East Asian genomes(0.70%–0.98%),reflecting a unique genomic composition of Tibetans. One such archaic hominid shared sequence-a662-bp intronic insertion in the SCUBE2 gene-is enriched and associated with better lung function(the FEV1/FVC ratio) in Tibetans. 7Identification of QTL for kernel number-related traits in a rice chromosome segment substitution line and fine mapping of qSP1显示文摘A chromosome segment substitution line (CSSL) is a powerful tool for combining quantitative trait locus (QTL) mapping with the pyramiding of desirable alleles. The rice CSSL Z1364 with increased kernel number was identified in a BC3F8 population derived from a cross of Nipponbare as the recipient with Xihui 18 as the donor parent. Z1364 carried three substitution segments distributed on chromosomes 1, 6, and 8. The mean substitution length was 1.19 Mb. Of 17 QTL identified on the substitution segments, qSP1 for spikelets per panicle, qSSD1 for seed-set density, and qNSB1 for number of secondary branches explained respectively 57.34%, 87.7%, and 49.44% of the corresponding phenotypic variance and were all linked to RM6777. Chi-square analysis showed that the increased kernel number in Z1364 was inherited recessively by a single gene. By fine mapping, qSP1 was delimited to a 50-kb region on the short arm of chromosome 1. Based on DNA sequence, a previously uncharacterized rice homolog of Arabidopsis thaliana AT4G32551 was identified as a candidate gene for qSP1 in which mutation increases the number of spikelets and kernels in Z1364. qSP1 was expressed in all tissues, but particularly in 1-cm panicles. The expression levels of OsMADS22, GN1A, and DST were upregulated and those of LAX2, GNP1, and GHD7 were downregulated in Nipponbare. 8Olfactory and gustatory dysfunctions of COVID-19 patients in China:A multicenter study显示文摘Introduction:With the spread of the epidemic worldwide,an increasing number of doctors abroad have observed the following atypical symptoms of coronavirus disease 2019(COVID-19):olfactory or taste disorders.Therefore,clarifying the incidence and clinical characteristics of olfactory and taste disorders in Chinese COVID-19 patients is of great significance and urgency.Materials and Methods:A retrospective study was conducted,which included 229 severe acute respiratory syndrome coronavirus 2 confirmed patients,through face-to-face interviews and telephone follow-up.Following the completion of questionnaires,the patients participating in the study,were categorized according to the degree of olfactory and taste disorders experienced,and the proportion of each clinical type of patient with olfactory and taste disorders and the time when symptoms appeared were recorded.Results:Among the 229 patients,31(13.54%)had olfactory dysfunction,and 44(19.21%)had gustatory dysfunction.For the patients with olfactory dysfunction,6(19.35%)developed severe disease and became critically ill.Olfactory dysfunction appeared before the other symptoms in 21.43%of cases.The proportion of females with olfactory and gustatory dysfunction was higher than that of males(P<0.001).Conclusions:The incidence of olfactory and gustatory dysfunction was much lower than that reported abroad;the prognosis of patients with olfactory dysfunction is relatively favorable;olfactory and gustatory dysfunction can be used as a sign for early screening;females are more prone to olfactory and gustatory dysfunction.Jianhui Li Yi Sun Enqiang Qin Hu Yuan Mingbo Liu Wenqi Yi Zhu Chen Chengcheng Huang Fengjie Zhou Ruiyao Chen Leibo Zhang Ning Yu Qiong Liu Xuejun Zhou Jingjing He Boyu Li Fusheng Wang Changliang Yang Shiming Yang 2022World Journal of Otorhinolaryngology-Head and Neck Surgery2022,8,4:2
9Distributed Low-Complexity Channel Assignment for Opportunistic Routing显示文摘Opportunistic Routing (OR) involves multiple forwarding candidates to relay packets by taking advantage of the broadcast nature and multi-user diversity of a wireless medium. Compared with Traditional Routing (TR), OR is more suitable in the case of an unreliable wireless link and can evidently improve the end-to-end throughput of Wireless Mesh Networks (WMNs). In this paper, we focus on OR in Multi-Radio Multi-Channel WMNs (MRMC-WMNs). This problem has not been well examined and is considerably more challenging than the OR in single-radio wireless networks considered in the existing literature. First, we validate the advantage of OR in MRMC-WMNs. Second, we propose Low-complexity Channel Assignment for Opportunistic Routing (LcCAOR), which assigns channels to flows according to the interference state of every node. Third, we implement the LcCOAR in a fully distributed manner. 12All-optical two-channel polarization-multiplexing format conversion from QPSK to BPSK signals in a silicon waveguide显示文摘All-optical two-channel format conversion is proposed and experimentally demonstrated from a 40 Gbit/s polarization multiplexing(Pol-MUX) non-return-to-zero quadrature phase-shift keying(QPSK) signal to Pol-MUX binary phase-shift keying(BPSK) signals by using phase-doubled four-wave mixing effects with two polarization-angled pumps in a silicon waveguide. The eye diagrams and constellation diagrams of the original QPSK sequences and the converted BPSK sequences of each channel are clearly observed on the two polarization states. Moreover,the bit error rates(BERs) of the two converted idlers are measured. 18LogUAD: Log Unsupervised Anomaly Detection Based on Word2Vec显示文摘System logs record detailed information about system operation and areimportant for analyzing the system's operational status and performance. Rapidand accurate detection of system anomalies is of great significance to ensure system stability. However, large-scale distributed systems are becoming more andmore complex, and the number of system logs gradually increases, which bringschallenges to analyze system logs. Some recent studies show that logs can beunstable due to the evolution of log statements and noise introduced by log collection and parsing. Moreover, deep learning-based detection methods take a longtime to train models. Therefore, to reduce the computational cost and avoid loginstability we propose a new Word2Vec-based log unsupervised anomaly detection method (LogUAD). LogUAD does not require a log parsing step and takesoriginal log messages as input to avoid the noise. LogUAD uses Word2Vec togenerate word vectors and generates weighted log sequence feature vectors withTF-IDF to handle the evolution of log statements. At last, a computationally effi-cient unsupervised clustering is exploited to detect the anomaly. We conductedextensive experiments on the public dataset from Blue Gene/L (BGL).
