2Defined tumor antigen-specific T cells potentiate personalized TCR-T cell therapy and prediction of immunotherapy response显示文摘Personalized immunotherapy targeting tumor-specific antigens(TSAs)could generate efficient and safe antitumor immune response without damaging normal tissues.Although neoantigen vaccines have shown therapeutic effect in clinic trials,precise prediction of neoantigens from tumor mutations is still challenging.The host antitumor immune response selects and activates T cells recognizing tumor antigens.Hence,T cells engineered with T-cell receptors(TCRs)from these naturally occurring tumor antigen-specific T(Tas)cells in a patient will target personal TSAs in his/her tumor.To establish such a personalized TCR-T cell therapy,we comprehensively characterized T cells in tumor and its adjacent tissues by single-cell mRNA sequencing(scRNA-seq),TCR sequencing(TCR-seq)and in vitro neoantigen stimulation.Compared to bystander T cells circulating among tissues,Tas cells were characterized by tumor enrichment,tumor-specific clonal expansion and neoantigen specificity.We found that CXCL13 is a unique marker for both CD4^(+)and CD8^(+)Tas cells.Importantly,TCR-T cells expressing TCRs from Tas cells showed significant therapeutic effects on autologous patient-derived xenograft(PDX)tumors.Intratumoral Tas cell levels measured by CXCL13 expression precisely predicted the response to immune checkpoint blockade,indicating a critical role of Tas cells in the antitumor immunity.We further identified CD200 and ENTPD1 as surface markers for CD4^(^(+))and CD8^(^(+))Tas cells respectively,which enabled the isolation of Tas cells from tumor by Fluorescence Activating Cell Sorter(FACS)sorting.Overall,our results suggest that TCR-T cells engineered with Tas TCRs are a promising agent for personalized immunotherapy,and intratumoral Tas cell levels determine the response to immunotherapy.Jingjing He Xinxin Xiong Han Yang Dandan Li Xuefei Liu Shuo Li Shuangye Liao Siyu Chen Xizhi Wen Kuai Yu Lingyi Fu Xingjun Dong Kaiyu Zhu Xiaojun Xia Tiebang Kang Chaochao Bian Xiang Li Haiping Liu Peirong Ding Xiaoshi Zhang Zhenjiang Liu Wende Li Zhixiang Zuo Penghui Zhou 2022Cell Research2022,32,6:10
3Progress of regional oceanography study associated with western boundary current in the South China Sea显示文摘Recent progress of physical oceanography in the South China Sea (SCS) associated with the western boundary current (WBC) and eddies is reviewed in this paper. It includes Argo observations of the WBC, eddy detection in the WBC based on satellite images, cross-continental shelf exchange in the WBC, eddy-current interaction, interannual variability of the WBC, air-sea interaction, the SCS throughflow (SCSTF), among others. The WBC in the SCS is strong, and its structure, variability and dynamic processes on seasonal and interannual time scales are yet to be fully understood. In this paper, we summarize progresses on the variability of the WBC, eddy-current interaction, air-sea interaction, and the SCSTF achieved in the past few years. Firstly, using the drifting buoy observations, we point out that the WBC becomes stronger and narrower after it reaches the central Vietnam coast. The possible mechanisms influencing the ocean circulation in the northern SCS are discussed, and the dynamic mechanisms that induce the countercurrent in the region of northern branch of WBC in winter are also studied quantitatively using momentum balance. The geostropic component of the WBC was diagnosed using the ship observation along 18°N, and we found that the WBC changed significantly on interannual time scale. Secondly, using the ship observations, two anti-cyclonic eddies in the winter of 2003/2004 in the northern SCS, and three anti-cyclonic eddies in the summer of 2007 along 18°N were studied. The results show that the two anti-cyclonic eddies can propagate southwestward along the continental shelf at the speed of first Rossby wave (~0.1 ms-1 ) in winter, and the interaction between the three anti-cyclonic eddies in summer and the WBC in the SCS is preliminarily revealed. Eddies on the continental shelf of northern SCS propagated southeastward with a maximum speed of 0.09 ms-1 , and those to the east of Vietnam coast had the largest kinetic energy, both of which imply strong interaction between eddy activity and WBC in the SCS. Thirdly, strong intraseasonal variability (ISV) of sea surface temperature (SST) near the WBC regions was found, and the ISV signal of SST in winter weakens the ISV signal of latent heat flux by 20%. 4Arabidopsis ANAC092 regulates auxin-mediated root development by binding to the ARF8 and PIN4 promoters显示文摘Auxin is an important plant hormone that is essential for growth and development due to its effects on organogenesis, morphogenesis, tropisms, and apical dominance. The functional diversity of auxin highlights the importance of its biosynthesis, transport, and associated responses. In this study, we show that a NAC transcription factor, ANAC092(also named At NAC2 and ORESARA1),known to positively regulate leaf senescence and contribute to abiotic stress responses, also affects primary root development. Plants overexpressing ANAC092 had altered root meristem lengths and shorter primary roots compared with the wild-type control. Additionally, expression of the pro ANAC092::GUS was strongly induced by indole-3-acetic acid. Quantitative real-time RT-PCR(q RT-PCR) analysis revealed that the YUCCA2, PIN, and ARF expression levels were downregulated in ANAC092-overexpressing plants. Moreover, yeast one-hybrid and chromatin immunoprecipitation assays confirmed that ANAC092 binds to the promoters of AUXIN RESPONSE FACTOR 8(ARF8) and PIN-FORMED 4(PIN4). Furthermore, a dual-luciferase assay indicated that ANAC092 decreases ARF8 and PIN4 promoter activities. We also applied a CRISPR/Cas9 system to mutate ANAC092. The roots of three of the analyzed mutants were longer than normal. 5ICG@ZIF-8:One-step encapsulation of indocyanine green in ZIF-8 and use as a therapeutic nanoplatform显示文摘How to fabricate zeolitic imidazole framework-8(ZIF-8) based therapeutic nanoplatform will be of significance in biomedicine considering its good biocompatibility. Herein, we report a one-step encapsulation of indocyanine green(ICG) in ZIF-8 nanoparticles(NPs). The as-prepared ICG@ZIF-8 NPs possess an absorption band in the near infrared region and have the good photothermal conversion efficiency. The in vivo and in vitro studies show that, after loading chemotherapy agent hydrophobic doxorubicin(DOX), ICG@ZIF-8-DOX NPs exhibit the chem-and photothermal synergistic therapy for tumor. 6Synthesis of a reduction-sensitive Bletilla striata polysaccharide amphiphilic copolymer显示文摘Recently, much special attention has been focused on the development of amphiphilic copolymers.Herein, a reduction-sensitive stearic acid grafted-Bletilla striata polysaccharide copolymer(BSP-ss-SA)was synthesized and characterized. BSP-ss-SA copolymer could spontaneously form micelles with an average particle size of(106 ? 4.36) nm. Particle sizes were dramatically larger under 10 mmol/L dithiothreitol(DTT) than that in absence of 10 mmol/L DTT. Docetaxel loading content and encapsulation efficiency of micelles were 5.94 wt% and 72.75 wt%, respectively. The docetaxel release rate in vitro in micelles could be remarkably accelerated in pH 7.4 phosphate buffer solution(PBS) containing 10 mmol/L DTT compared with that of micelles(0 mmol/L DTT). Furthermore, MTT results demonstrated the anticancer activities in vitro of docetaxel-loaded BSP-ss-SA micelles on HepG2 were superior to that of docetaxel injection. 7LRP6 downregulation promotes cardiomyocyte proliferation and heart regeneration显示文摘The adult mammalian heart is thought to be a terminally differentiated organ given the postmitotic nature of cardiomyocytes.Consequently,the potential for cardiac repair through cardiomyocyte proliferation is extremely limited.Low-density lipoprotein receptor-related protein 6(LRP6)is a Wnt co-receptor that is required for embryonic heart development.In this study we investigated the role of LRP6 in heart repair through regulation of cardiomyocyte proliferation.Lrp6 deficiency increased cardiomyocyte cell cycle activity in neonatal,juvenile and adult mice.Cardiomyocyte-specific deletion of Lrp6 in the mouse heart induced a robust regenerative response after myocardial infarction(MI),led to reduced MI area and improvement in left ventricular systolic function.In vivo genetic lineage tracing revealed that the newly formed cardiomyocytes in Lrp6-deficient mouse hearts after MI were mainly derived from resident cardiomyocytes.Furthermore,we found that the pro-proliferative effect of Lrp6 deficiency was mediated by the ING5/P21 signaling pathway.Gene therapy using the adeno-associated virus(AAV)9 miRNAi-Lrp6 construct promoted the repair of heart injury in mice.Lrp6 deficiency also induced the proliferation of human induced pluripotent stem cell-derived cardiomyocytes(iPSC-CMs).Our study identifies LRP6 as a critical regulator of cardiomyocyte proliferation,which may lead to the development of a novel molecular strategy to promote myocardial regeneration and repair.Yahan Wu Liping Zhou Hongyu Liu Ran Duan Huixing Zhou Fulei Zhang Xiaoyu He Dongbo Lu Ke Xiong Maolin Xiong Jinzhu Zhuang Yi Liu Li Li Dandan Liang Yi-Han Chen 2021Cell Research2021,31,4:5

9A comparative study of hydrothermal aging effect on cerium and lanthanum doped Cu/SSZ-13 catalysts for NH_(3)-SCR显示文摘Ce-or La-doped Cu/SSZ-13 catalysts were prepared by a hydrothermal method and Cu,Ce or La ions were incorporated through stepwise ion exchange,The catalyst activity was measured for the ammonia selective catalytic reduction reaction.The structure and composition of catalyst were characterized by using X-ray diffraction,scanning electron microscopy,inductively coupled plasma mass spectrometry solid-state NMR,NH_(3)-TPD techniques,and the active components were examined by XPS and XANES.The results indicate that the Ce and La doping can both completely preserve the SCR activity of Cu/SSZ-13 above 300℃,but there is also a decrease of activity below 200℃.On the other hand,Ce doping is beneficial to the formation of framework aluminum,stabilizes molecular sieve framework and Cu active sites of Cu/SSZ-13,thereby improves the catalyst hydrothermal stability.But La doping will decrease the amount of framework aluminum and Cu active sites of Cu/SSZ-13 after hydrothermally aging,even destroy zeolite CHA structure.This is quite harmful to the catalyst hydrothermal stability.Di Deng Shujun Deng Dandan He Zhiheng Wang Zhipeng Chen Yi Ji Guoping Yan Guangjin Hou Licheng Liu Hong He 2021Journal of Rare Earths2021,39,8:5
10Effect of direct current pulse stimulating acupoints of JiaJi (T10-L3) and Ciliao (BL 32) with Han's Acupoint Nerve Stimulator on labour pain in women: a randomized controlled clinical study显示文摘OBJECTIVE: To assess the clinical effect and safety of direct current(DC) pulse produced by Han's Acupoint Nerve Stimulator in reduction(HANS) of labor pain.METHODS: Totally 120 participants were enrolled in this clinical trial, and were randomly divided into4 groups including: HANS group, patient controlled intravenous analgesia(PCIA) group, patient-controlled epidural analgesia(PCEA) group and control group. The HANS group was treated by stimulating the acupoints of Jia Ji(T10-L3) and Ciliao(BL 32)with DC pulse of 100 Hz and 15-30 m A produced by a portable battery-powered Han's Acupoint Nerve Stimulator for 30 min. The PCIA group was intravenously infused Ondansetron(8 mg) for 5 min,then tramadol injection(1.5 mg/kg) was slowly dripped by using Baxter APⅡ electronic pump with50 m L tramadol(0.70%) + ondansetron(8 mg),background infusion 2 m L/h, PCA dose of 2 m L,lockout interval of 10 min. In PCEA group, women received intrathecal injection ropivacaine(3 mg) in L2-3, and epidural catheter was connected to Baxter APⅡ electronic pump, with 100 m L Ropivacaine(0.1%) and Sufentanil(50 ug), background infusion5 m L, Patient controlled analgesia(PCA) dose of 5m L, lockout interval of 10 min. The control group was not received analgesia. The visual analogue scale(VAS), stage and manner of labor, Apgar score of newborn, neonatal weights, oxytocin dosage,postpartum hemorrhage and side effects were monitored in all groups.RESULTS: The vital signs were all stable in the four analgesic groups. After analgesia, there was statistical difference in VAS score between HANS group and control group, between PCEA group and the control group, between PCIA group and control group. The analgesic effect in the PCEA group was significantly better than that of other two groups.The second stage of labor in the PCEA group was longer than the other three groups, showing significant difference between them. The Apgar score of newborn 1min after birth in the PCIA group was slightly lower than that of the other two groups,showing significant difference between them. The neonatal weights between four groups were not significantly different. The rate of cesarean sectionin the control group was significantly higher than that of the labor analgesia group, there was statistically difference in four groups. The number of PCIA group that used oxytocin was lower than that of other three groups. There was no significant difference in postpartum hemorrhage between four groups. The side effects of the PCEA group were itching, uroschesis and neonatal asphyxia and PCIA group were nausea and vomiting and neonatal asphyxia. 11Design of depth-control planting unit with single-side gauge wheel for no-till maize precision planter显示文摘No-till planters are very popular for maize seeding in fields covered with residue in annual wheat-maize double cropping system in North China Plain.However,there is no suitable depth control mechanism for existing no-till maize planters,and as a result,it is hard to obtain consistent planting depth,uniform emergence,and good passing ability at the same time.For the above reasons,a proper planting unit with a new type of depth-control mechanism was developed in this study.The mechanism consists of a single-side gauge wheel,a parallel four-bar linkage,a pair of double-disc opener,a V-shape press wheel and a depth regulator,which can adjust the planting depth from 30 mm to 90 mm to satisfy the agronomic requirement under different field conditions.Based on analysis and calculation,the width of gauge wheel was set to 50 mm while the length of parallel four-bar linkage was set to 350 mm.Field experiment was conducted and the results indicated that the newly designed planting unit with single-side gauge wheel performed well with regard to planting depth uniformity and anti-blocking ability.The planting depth uniformity and seed spacing quality were 95.45%and 91.90%,respectively,when the average height of stubble was 22.5 cm and residue amount was 0.64 kg/m^(2)in the field.It can satisfy the requirement of no-till maize planting on the cropland with residue and stubble in North China Plain.Zhang Rui Cui Tao Han Dandan Zhang Dongxing Li Kehong Yin Xiaowei Wang Yunxia He Xiantao Yang Li 2016International Journal of Agricultural and Biological Engineering2016,9,6:4
12Expression Pattern of Peptide and Amino Acid Genes in Digestive Tract of Transporter Juvenile Turbot(Scophthalmus maximus L.)显示文摘Turbot(Scophthalmus maximus L.),a carnivorous fish species with high dietary protein requirement,was chosen to examine the expression pattern of peptide and amino acid transporter genes along its digestive tract which was divided into six segments including stomach,pyloric caeca,rectum,and three equal parts of the remainder of the intestine.The results showed that the expression of two peptide and eleven amino acid transporters genes exhibited distinct patterns.Peptide transporter 1(Pep T1) was rich in proximal intestine while peptide transporter 2(PepT2) was abundant in distal intestine.A number of neutral and cationic amino acid transporters expressed richly in whole intestine including B^0-type amino acid transporter 1(B^0AT1),L-type amino acid transporter 2(LAT2),T-type amino acid transporter 1(TAT1),proton-coupled amino acid transporter 1(PAT1),y^+L-type amino acid transporter 1(y^+LAT1),and cationic amino acid transporter 2(CAT2) while ASC amino acid transporter 2(ASCT2),sodium-coupled neutral amino acid transporter 2(SNAT2),and y^+L-type amino acid transporter 2(y^+LAT2) abundantly expressed in stomach.In addition,system b^(0,+) transporters(rBAT and b^(0,+)AT) existed richly in distal intestine.These findings comprehensively characterized the distribution of solute carrier family proteins,which revealed the relative importance of peptide and amino acid absorption through luminal membrane.Our findings are helpful to understand the mechanism of the utilization of dietary protein in fish with a short digestive tract.XU Dandan HE Gen MAI Kangsen ZHOU Huihui XU Wei SONG Fei 2016Journal of Ocean University of China2016,15,2:3

1418beta-glycyrrhetinic acid induces ROS-mediated apoptosis to ameliorate hepatic fibrosis by targeting PRDX1/2 in activated HSCs显示文摘Hepatic stellate cells(HSCs)are essential drivers of fibrogenesis.Inducing activated-HSC apoptosis is a promising strategy for treating hepatic fibrosis.18beta-glycyrrhetinic acid(18b-GA)is a natural compound that exists widely in herbal medicines,such as Glycyrrhiza uralensis Fisch,which is used for treating multiple liver diseases,especially in Asia.In the present study,we demonstrated that 18b-GA decreased hepatic fibrosis by inducing the apoptosis in activated HSCs.18b-GA inhibited the expression of a-smooth muscle actin and collagen type Ⅰ alpha-1.Using a chemoproteomic approach derived from activity-based protein profiling,together with cellular thermal shift assay and surface plasmon resonance,we found that 18b-GA covalently targeted peroxiredoxin 1(PRDX1)and peroxiredoxin 2(PRDX2)proteins via binding to active cysteine residues and thereby inhibited their enzymatic activities.18b-GA induced the elevation of reactive oxygen species(ROS),resulting in the apoptosis of activated HSCs.PRDX1 knockdown also led to ROS-mediated apoptosis in activated HSCs.Collectively,our findings revealed the target proteins and molecular mechanisms of 18b-GA in ameliorating hepatic fibrosis,highlighting the future development of 18b-GA as a novel therapeutic drug for hepatic fibrosis.Qian Zhang Piao Luo Liuhai Zheng Jiayun Chen Junzhe Zhang Huan Tang Dandan Liu Xueling He Qiaoli Shi Liwei Gu Jiahao Li Qiuyan Guo Chuanbin Yang Yin Kwan Wong Fei Xia Jigang Wang 2022Journal of Pharmaceutical Analysis2022,12,4:3
15Decreased Soil Nitrification Rate with Addition of Biochar to Acid Soils显示文摘This study was conducted to investigate the effects of mixed biochar on the nitrification rate in acidic soils. A15 N tracer experiment with(15 NH4)2 SO4 was conducted to determine the nitrification rates of 4 acidic agricultural soils with p H 4.03-6.02 in Yunnan Province, Southern China. The accumulation of15 N-NO3-and nitrification rates decreased with the addition of biochar at the end of incubation, suggesting that biochar could be a nitrification inhibitor in acidic fertilized soil. Nitrification rates in soil with p H 4.03 were evidently lower than those in soil with p H 4.81-6.02 with or without biochar. Decreased nitrification rates were detected in the acidic soils with biochar. 16Effects of Angelica sinensis extracts on lipid oxidation in fish feeds and growth performance of juvenile Jian carp(Cyprinus carpio var. Jian)显示文摘The study was to explore the effect of the extracts of Angelica sinensis(EAs) on lipid oxidation in fish feeds compared with ethoxyquin(EQ) and the effect of dietary EAs on growth performance of carp(Cyprinus carpio var. Jian). Firstly, fish feeds were respectively added with EQ, and ethyl ether extract ethyl acetate extract(EAE), acetone extract, ethanol extract(EE) and aqueous extract(AQE) of Angelica sinensis, except for the control. The results showed that EAs and EQ inhibited lipid oxidation in fish feed(P < 0.05). Of all of the examined EAs, EAE showed the strongest protective effects against lipid oxidation(P < 0.05). Moreover, EAE at high concentrations showed a stronger effect on lipid oxidation compared with EQ(P < 0.05). Then, 7 experimental diets respectively supplemented with 0.0, 1.0, 2.0, 3.0, 4.0, 5.0 and 6.0 g/kg of EAE were fed to the respective treatment groups for 30 d. Four replicates were performed for each treatment group; 20 carp(mean weight: 12.10 ± 0.13 g) were in each replicate. The result indicated that dietary EAE improved the growth performance in carp(P < 0.05). The appropriate con centration of EAE for carp growth was estimated to be 3.643 g/kg diet. 17Characterization of viromes within mosquito species in China显示文摘Mosquitoes are vectors of multiple disease-causing agents that cause worldwide outbreaks of serious infectious diseases in both humans and animals. These arboviral agents include Dengue virus (DENV), Zika virus (ZIKV), chikungunya virus, yellow fever virus, Japanese encephalitis virus (JEV),and Rift Valley fever virus. 18Genome-wide prediction for hybrids between parents with distinguished difference on exotic introgressions in Brassica napus显示文摘Extensive exotic introgression could significantly enlarge the genetic distance of hybrid parental populations to promote strong heterosis.The goal of this study was to investigate whether genome-wide prediction can support pre-breeding in populations with exotic introgressions.We evaluated seed yield,seed yield related traits and seed quality traits of 363 hybrids of Brassica napus (AACC) derived from two parental populations divergent on massive exotic introgression of related species in three environments.The hybrids presented strong heterosis on seed yield,which was much higher than other investigated traits.Five genomic best linear unbiased prediction models considering the exotic introgression and different marker effects (additive,dominance,and epistatic effects) were constructed to test the prediction ability for different traits of the hybrids.The analysis showed that the trait complexity,exotic introgression,genetic relationship between the training set and testing set,training set size,and environments affected the prediction ability.The models with best prediction ability for different traits varied.However,relatively high prediction ability (e.g.,0.728 for seed yield) was also observed when the simplest models were used,excluding the effects of the special exotic introgression and epistasis effect by5-fold cross validation,which would simplify the prediction for the trait with complex architecture for hybrids with exotic introgression.The results provide novel insights and strategies for genome-wide prediction of hybrids between genetically distinct parent groups with exotic introgressions.Dandan Hu Yusheng Zhao Jinxiong Shen Xiangxiang He Yikai Zhang Yong Jiang Rod Snowdon Jinling Meng Jochen C.Reif Jun Zou 2021The Crop Journal2021,9,5:2

20MicroRNA-activated hydrogel scaffold generated by 3D printing accelerates bone regeneration显示文摘Bone defects remain a major threat to human health and bone tissue regeneration has become a prominent clinical demand worldwide.The combination of microRNA(miRNA)therapy with 3D printed scaffolds has always posed a challenge.It can mimic physiological bone healing processes,in which a biodegradable scaffold is gradually replaced by neo-tissue,and the sustained release of miRNA plays a vital role in creating an optimal osteogenic microenvironment,thus achieving promising bone repair outcomes.However,the balance between two key factors-scaffold degradation behavior and miRNA release profile-on osteogenesis and bone formation is still poorly understood.Herein,we construct a series of miRNA-activated hydrogel scaffolds(MAHSs)generated by 3D printing with different crosslinking degree to screened the interplay between scaffold degradation and miRNA release in the osteoinduction activity both in vitro and in vivo.Although MAHSs with a lower crosslinking degree(MAHS-0 and MAHS-0.25)released a higher amount of miR-29b in a sustained release profile,they degraded too fast to provide prolonged support for cell and tissue ingrowth.On the contrary,although the slow degradation of MAHSs with a higher crosslinking degree(MAHS-1 and MAHS-2.5)led to insufficient release of miR-29b,their adaptable degradation rate endowed them with more efficient osteoinductive behavior over the long term.MAHS-1 gave the most well-matched degradation rate and miR-29b release characteristics and was identified as the preferred MAHSs for accelerated bone regeneration.This study suggests that the bio-adaptable balance between scaffold degradation behavior and bioactive factors release profile plays a critical role in bone regeneration.These findings will provide a valuable reference about designing miRNAs as well as other bioactive molecules activated scaffold for tissue regeneration.Ting Pan Wenjing Song Hongbao Xin Haiyue Yu He Wang Dandan Ma Xiaodong Cao Yingjun Wang 2022Bioactive Materials2022,7,4:2
