| 1 | A complete sequence and comparative analysis of a SARS-associated virus(Isolate BJ01)显示文摘The genome sequence of the Severe Acute Respiratory Syndrome (SARS)-associated virus provides essential information for the identification of pathogen(s), exploration of etiology and evolution, interpretation of transmission and pathogenesis, development of diagnostics, prevention by future vaccination, and treatment by developing new drugs. We report the complete genome sequence and comparative analysis of an isolate (BJ01) of the coronavirus that has been recognized as a pathogen for SARS. The genome is 29725 nt in size and has 11 ORFs (Open Reading Frames). It is composed of a stable region encoding an RNA-dependent RNA polymerase (composed of 2 ORFs) and a variable region representing 4 CDSs (coding sequences) for viral structural genes (the S, E, M, N proteins) and 5 PUPs (putative uncharacterized proteins). Its gene order is identical to that of other known coronaviruses. The sequence alignment with all known RNA viruses places this virus as a member in the family of Coronaviridae. Thirty putative substitutions have been identified by comparative analysis of the 5 SARS- associated virus genome sequences in GenBank. Fifteen of them lead to possible amino acid changes (non-synonymous mutations) in the proteins. Three amino acid changes, with predicted alteration of physical and chemical features, have been detected in the S protein that is postulated to beinvolved in the immunoreactions between the virus and its host. Two amino acid changes have been detected in the Mprotein, which could be related to viral envelope formation. Phylogenetic analysis suggests the possibility of non-human origin of the SARS-associated viruses but provides noevidence that they are man-made. | 2 | Gastric cancer: Epidemiology, risk factors and prevention strategies显示文摘Gastric cancer(GC) is a global health problem, with more than 1 million people newly diagnosed with GC worldwide each year. GC is more prevalent in less developed countries than in more developed countries. About half of all GC cases worldwide occur in East Asia, notably China. Globally, overall incidence rates of GC are declining, which is potentially attributed to a decrease in Helicobacter pylori(H. pylori) infection and the use of refrigeration to preserve foods rather than salt. GC is a multifactorial disease, and its occurrence and development were impacted by environmental and genetic factors. H. pylori infection is the primary risk factor for GC, especially for non-cardia. The prognosis of GC is poor due to stages at the first diagnosis. The 5-year survival rate is less than 10% when patients are diagnosed at an advanced stage, but the rate is as high as 85% if patients are detected at an earlier stage. Endoscopic screening can potentially prevent GC by early diagnosis and early treatment and has been widely adopted in screening programs in East Asian countries, such as Japan and Korea. | 3 | The effects of land consolidation on the ecological connectivity based on ecosystem service value: A case study of Da'an land consolidation project in Jilin province显示文摘Land consolidation has a profound impact on landscape patterns and ecological functions at various scales through engineering and biological measures. In recent years, China invests more than 100 billion RMB yuan on land consolidation each year. To understand how land consolidation affects landscape patterns and ecosystems, we investigated the ecosystem service value and the ecological connectivity in a consolidated area of Da'an city from 2008 to 2014 using a revised ecological connectivity index. The results indicated that land consolidation has certain negative influences on the ecosystem services in this area. The total ecosystem service value will decrease by nearly 30% in the late stage of consolidation. This decrease is caused by the loss of ecosystem service of the wetland and grassland, despite a sensible increase of cultivated land. In addition, land consolidation could change the ecological connectivity as well as the land use structure. Up to 85% of the entire area will be in low connectivity in the late stage of consolidation, representing a 6.23% increase in the total coverage compared to pre-consolidation. Finally, the different connectivity landscape and their key areas can be identified by the revised ecological connectivity index effectively. This study is helpful to trace out the relationships between landscape pattern and ecological process, and provides insights for ecological planning and designing of land consolidation in this area. | 4 | Three-dimensional Finite Element Analysis of the Mechanical Properties of Helical Thread Connection显示文摘Conventional analytical and numerical methods for the mechanical properties of helical threads are relied on many assumptions and approximations and thus hardly yield satisfied results. A parameterized 3D finite element model of bolted joints with real helical thread geometry is established and meshed with refined hexahedral elements. The Von Mises plasticity criterion, kinematic hardening rule of materials and interfacial contacts are employed to make it possible for the suggested model be able to approach real assembly conditions. Then, the mechanical properties of bolted joints with different thread pitches, thread numbers and modular ratios are investigated, including the contact pressure distribution at joint interfaces, the axial load distribution and stress concentration in screw threads during the loading and unloading process. Simulation results indicate that the load distribution in screw threads produced by the suggested model agrees well the results from CHEN's photoelastic tests. In addition, an interesting phenomenon is found that tightening the bolt with a large preload first and then adjusting the clamping force by unloading can make the load distribution more uniform and reduce the maximum residual equivalent stress in thread roots by up to 40%. | 8 | A nonlinear multi-field coupled model for soils显示文摘A nonlinear multi-field coupled model for multi-constituent three-phase soils is derived by using the hybrid mixture theory. The balance equations with three levels (constituents, phases and the whole mixture soil) are set up under the assumption that soil is composed of multi-constituent elastic-plastic solid skeleton (which is different from the linearization method) and viscous liquid and ideal gas. With reasonable constitutive assumptions in such restrictive conditions as the principles of determinism, equipresence, material frame-indifference and the compatible principle in continuum mechanics, a theoretical framework of constitutive relations modeling three-phase soil in both non-equilibrium and equilibrium states is established, thus the closed field equations are formed. In the theoretical framework, the concept of effective generalized thermodynamic forces is introduced, and the nonlinear coupling constitutive relations between generalized dissipation forces and generalized flows within the system at nonequilibrium state are also presented. On such a basis, four special coupling relations, i.e., solid thermal elastic-plastic constitutive relation, liquid visco-elastic-plastic constitutive relation, the generalized Fourier’s law, and the generalized Darcy’s law are put forward. The generalized or nonlinear results mentioned above can degenerate into the linear coupling results given by Bennethum and Singh. Based on a specific dissipation function, the concrete form of generalized Darcy’s law is deduced, which may degenerate into the traditional form of Darcy’s law by neglecting the influence of skeleton deformation and temperature. | 12 | Drug nanoclusters formed in confined nano-cages of CD-MOF: dramatic enhancement of solubility and bioavailability of azilsartan显示文摘Tremendous efforts have been devoted to the enhancement of drug solubility using nanotechnologies, but few of them are capable to produce drug particles with sizes less than a few nanometers. This challenge has been addressed here by using biocompatible versatile γ-cyclodextrin(γ-CD) metal-organic framework(CD-MOF) large molecular cages in which azilsartan(AZL) was successfully confined producing clusters in the nanometer range. This strategy allowed to improve the bioavailability of AZL in Sprague–Dawley rats by 9.7-fold after loading into CD-MOF. The apparent solubility of AZL/CD-MOF was enhanced by 340-fold when compared to the pure drug. Based on molecular modeling, a dual molecular mechanism of nanoclusterization and complexation of AZL inside the CD-MOF cages was proposed, which was confirmed by small angle X-ray scattering(SAXS) and synchrotron radiation-Fourier transform infrared spectroscopy(SR-FTIR) techniques. In a typical cage-like unit of CD-MOF, three molecules of AZL were included by the γ-CD pairs, whilst other three AZL molecules formed a nanocluster inside the 1.7 nm sized cavity surrounded by six γ-CDs. | 16 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. | 18 | Apoptotic mechanism of premature ovarian failure and rescue effect of Traditional Chinese Medicine: a review显示文摘Social and environmental factors render premature ovarian failure(POF) as a major cause of decline or loss of female fertility. The natural pregnancy rate of patients with POF is only 5%-10%. Follicular atresia is the main factor in the pathogenesis of POF.Due to the unique ovarian physiological environment and follicular developmental processes, the apoptosis of ovarian granulosa cells and oocytes together cause follicular atresia, which involves the apoptosis-related internal and external pathways.Furthermore, during POF, apoptosis and oxidative stress forms a 'vicious circle', which involves a variety of changes between the molecules. The existing pharmaceutical preparations such as gonadal hormones are the basic methods for the treatment of POF, and the curative effect was affirmative;however, it was ineffective after withdrawn, while the long-term application led to adverse reactions. Traditional Chinese Medicine(TCM) has a history of treating gynecological diseases and infertility and has gained increasing attention. Studies have shown that compounds isolated from herbal medicine exerted a positive effect on follicular atresia caused by cell apoptosis that also improved the POF. | 19 | The High Energy X-ray telescope (HE) onboard the Insight-HXMT astronomy satellite显示文摘The Insight-Hard X-ray Modulation Telescope(Insight-HXMT) is a broadband X-ray and γ-ray(1-3000 ke V) astronomy satellite. One of its three main telescopes is the High Energy X-ray telescope(HE). The main detector plane of HE comprises 18 Na I(Tl)/Cs I(Na) phoswich detectors, where Na I(Tl) is used as the primary detector to measure ~ 20-250 ke V photons incident from the field of view(FOV) defined by collimators, and Cs I(Na) is used as the active shielding detector to Na I(Tl) by pulse shape discrimination. Additionally, Cs I(Na) is used as an omnidirectional γ-ray monitor. The HE collimators have a diverse FOV,i.e. 1.1°×5.7°(15 units), 5.7°×5.7°(2 units), and blocked(1 unit). Therefore, the combined FOV of HE is approximately5.7°×5.7°. Each HE detector has a diameter of 190 mm resulting in a total geometrical area of approximately 5100 cm2, and the energy resolution is ~15% at 60 ke V. For each recorded X-ray event by HE, the timing accuracy is less than 10 μs and the deadtime is less than 10 μs. HE is used for observing spectra and temporal variability of X-ray sources in the 20-250 ke V band either by pointing observations for known sources or scanning observations to unveil new sources. Additionally, HE is used for monitoring the γ-ray burst in 0.2-3 Me V band. | 20 | Leakage performance of floating ring seal in cold/hot state for aero-engine显示文摘Rotating experimental investigations were carried out to study the oil sealing capability of two different floating ring seals in cold/hot state for aero-engine. High-speed Floating Ring Seal(HFRS) is a seal with the inner diameter of 83.72 mm and maximum speed of 38000 r/min, and Low-speed Floating Ring Seal(LFRS) is another seal with the inner diameter of 40.01 mm and maximum speed of 18000 r/min. In hot state, sealing air with the temperature of 371 K and oil with the temperature of 343 K was employed to model the working conditions of an aero-engine. Comparisons between floating ring seal and labyrinth seal were done to inspect the leakage performance.More attention was paid to the critical pressure ratio where the oil leakage began. Results show that the critical pressure ratio in cold state is obviously larger than that in hot state for both seals. An underlying sealing mechanism for floating ring seal is clarified by the fluid film, which closely associates with the dimensionless parameter of clearance over rotating diameter(2 c/Dr). Another fantastic phenomenon is that the leakage coefficient in hot state, not the leakage magnitude, is unexpectedly larger than that in cold state.
