| 1 | Connexin 43 hemichannels protect bone loss during estrogen deficiency显示文摘Estrogen deficiency in postmenopausal women is a major cause of bone loss,resulting in osteopenia,osteoporosis,and a high risk for bone fracture.Connexin 43 (Cx43) hemichannels (HCs) in osteocytes play an important role in osteocyte viability,bone formation,and remodeling.We showed here that estrogen deficiency reduced Cx43 expression and HC function.To determine if functional HCs protect osteocytes and bone loss during estrogen deficiency,we adopted an ovariectomy model in wild-type (WT) and two transgenic Cx43 mice:R76W (dominant-negative mutant inhibiting only gap junction channels) and Cx43 Δ130–136 (dominant-negative mutant compromising both gap junction channels and HCs).The bone mineral density (BMD),bone structure,and histomorphometric changes of cortical and trabecular bones after ovariectomy were investigated.Our results showed that the Δ130–136 transgenic cohort had greatly decreased vertebral trabecular bone mass compared to WT and R76W mice,associated with a significant increase in the number of apoptotic osteocyte and empty lacunae.Moreover,osteoclast surfaces in trabecular and cortical bones were increased after ovariectomy in the R76W and WT mice,respectively,but not in Δ130–136 mice.These data demonstrate that impairment of Cx43 HCs in osteocytes accelerates vertebral trabecular bone loss and increase in osteocyte apoptosis,and further suggest that Cx43 HCs in osteocytes protect trabecular bone against catabolic effects due to estrogen deficiency. | Liang Ma Rui Hua Yi Tian Hongyun Cheng Roberto Jose Fajardo Joseph J. | 2 | Characterization and separation of pyrite from Abu Tartur black shale显示文摘This work aimed for pyrite separation from Abu Tartur black shale as a source of sulfur to be an added economic value of Abu Tartur area. The considered samples in the present work were collected from a core drilled in Abu Tartur plateau representing the pyrite-rich black shale of the U. Cretaceous age.Sample characterization was carried out using petrographic microscope, XRD, DTA/DTG, C/S and XRF techniques. Clay minerals, silt-sized quartz, calcite, and hematite were the main minerals associating pyrite(5.34%). Liberation behavior of the sample was about 80% below 5 lm.Sample processing was achieved through one-day soaking followed by classification using 1 inch Mozley hydro-cyclone where about 35.5% by weight went to underflow and 64.5% went to overflow.The underflow product was subjected to an advanced gravity separation process using SB-40 Falcon Concentrator through a CCD statistical design prepared by Design-Expert 6.0 software proposed to optimize the separation process through a study for the effects of frequency(Hz) and water pressure(Psi) on both assay and recovery of the sulfur-rich heavy fraction.A heavy concentrate weighed 10.90% with inorganic sulfur content reached 11.37%(21.24% pyrite) with overall recovery(50.01%) was obtained after two cleaning at the optimum conditions. | Ibrahim S.S. | 3 | Mutations in TP53, ZNF750, and RB1 typify ocular sebaceous carcinoma显示文摘Sebaceous carcinoma (SC) of the eyelid is a rare but aggressive malignancy, accounting for 3%—5% of eyelid malignancies in the United States, and up to 35% in Asian populations (Deprez and Uffer, 2009;Xu et al., 2018;Yu et al., 2018). It is frequently mistaken for benign conditions or less aggressive malignancies such as basal cell carcinoma (Muqit et al., 2013), and the effects of delay in diagnosis can be devastating to patients. Aggressive surgical resection is the primary treatment of these tumors and often invoIves orbital exenteration with significant morbidity to patients, and these tumors frequently recur, with local recurrence rate as high as 18%. | 4 | Exosomes derived from differentiated human ADMSC with the Schwann cell phenotype modulate peripheral nerve-related cellular functions显示文摘Peripheral nerve regeneration remains a significant clinical challenge due to the unsatisfactory functional recovery and public health burden.Exosomes,especially those derived from mesenchymal stem cells(MSCs),are promising as potential cell-free therapeutics and gene therapy vehicles for promoting neural regeneration.In this study,we reported the differentiation of human adipose derived MSCs(hADMSCs)towards the Schwann cell(SC)phenotype(hADMSC-SCs)and then isolated exosomes from hADMSCs with and without differentiation(i.e.,dExo vs uExo).We assessed and compared the effects of uExo and dExo on antioxidative,angiogenic,anti-inflammatory,and axon growth promoting properties by using various peripheral nerve-related cells.Our results demonstrated that hADMSC-SCs secreted more neurotrophic factors and other growth factors,compared to hADMSCs without differentiation.The dExo isolated from hADMSC-SCs protected rat SCs from oxidative stress and enhanced HUVEC migration and angiogenesis.Compared to uExo,dExo also had improved performances in downregulating pro-inflammatory gene expressions and cytokine secretions and promoting axonal growth of sensory neurons differentiated from human induced pluripotent stem cells.Furthermore,microRNA(miRNA)sequencing analysis revealed that exosomes and their parent cells shared some similarities in their miRNA profiles and exosomes displayed a distinct miRNA signature.Many more miRNAs were identified in dExo than in uExo.Several upregulated miRNAs,like miRNA-132-3p and miRNA-199b-5p,were highly related to neuroprotection,anti-inflammation,and angiogenesis.The dExo can effectively modulate various peripheral nerve-related cellular functions and is promising for cell-free biological therapeutics to enhance neural regeneration. | Bo Liu Yunfan Kong Wen Shi Mitchell Kuss Ke Liao Guoku Hu Peng Xiao Jagadesan Sankarasubramanian Chittibabu Guda Xinglong Wang Yuguo Lei Bin Duan | 2022 | Bioactive Materials2022,7,8: | 3 |
