1Connexin 43 hemichannels protect bone loss during estrogen deficiency显示文摘Estrogen deficiency in postmenopausal women is a major cause of bone loss,resulting in osteopenia,osteoporosis,and a high risk for bone fracture.Connexin 43 (Cx43) hemichannels (HCs) in osteocytes play an important role in osteocyte viability,bone formation,and remodeling.We showed here that estrogen deficiency reduced Cx43 expression and HC function.To determine if functional HCs protect osteocytes and bone loss during estrogen deficiency,we adopted an ovariectomy model in wild-type (WT) and two transgenic Cx43 mice:R76W (dominant-negative mutant inhibiting only gap junction channels) and Cx43 Δ130–136 (dominant-negative mutant compromising both gap junction channels and HCs).The bone mineral density (BMD),bone structure,and histomorphometric changes of cortical and trabecular bones after ovariectomy were investigated.Our results showed that the Δ130–136 transgenic cohort had greatly decreased vertebral trabecular bone mass compared to WT and R76W mice,associated with a significant increase in the number of apoptotic osteocyte and empty lacunae.Moreover,osteoclast surfaces in trabecular and cortical bones were increased after ovariectomy in the R76W and WT mice,respectively,but not in Δ130–136 mice.These data demonstrate that impairment of Cx43 HCs in osteocytes accelerates vertebral trabecular bone loss and increase in osteocyte apoptosis,and further suggest that Cx43 HCs in osteocytes protect trabecular bone against catabolic effects due to estrogen deficiency.Liang Ma Rui Hua Yi Tian Hongyun Cheng Roberto Jose Fajardo Joseph J. 2Mechanotransduction via the coordinated actions of integrins,PI3K signaling and Connexin hemichannels显示文摘Mechanical loading opens connexin 43(Cx43)hemichannels(HCs),leading to the release of bone anabolic molecules,such as prostaglandins,from mechanosensitive osteocytes,which is essential for bone formation and remodeling.However,the mechanotransduction mechanism that activates HCs remains elusive.Here,we report a unique pathway by which mechanical signals are effectively transferred between integrin molecules located in different regions of the cell,resulting in HC activation.Both integrinα5 andαV were activated upon mechanical stimulation via either fluid dropping or flow shear stress(FSS).Inhibition of integrinαV activation or ablation of integrinα5 prevented HC opening on the cell body when dendrites were mechanically stimulated,suggesting mechanical transmission from the dendritic integrinαV toα5 in the cell body during HC activation.In addition,HC function was compromised in vivo,as determined by utilizing an antibody blockingαV activation andα5-deficient osteocyte-specific knockout mice.Furthermore,inhibition of integrinαV activation,but not that ofα5,attenuated activation of the phosphoinositide 3-kinase(PI3K)-protein kinase B(AKT)signaling pathway upon mechanical loading,and the inhibition of PI3K/AKT activation blocked integrinα5 activation and HC opening.Moreover,HC opening was blocked only by an anti-integrinαV antibody at low but not high FSS levels,suggesting that dendriticαV is a more sensitive mechanosensor thanα5 for activating HCs.Together,these results reveal a new molecular mechanism of mechanotransduction involving the coordinated actions of integrins and PI3K/AKT in osteocytic dendritic processes and cell bodies that leads to HC opening and the release of key bone anabolic factors.Manuel A.Riquelme Sumin Gu Rui Hua Jean X.Jiang 2021Bone Research2021,9,1:7
3Peptidome analysis of human intrauterine adhesion tissues and the identification of antifibrotic peptide显示文摘Intrauterine adhesion(IUA)is a common clinical endometrial disease,which can severely damage the fertility and quality of life in women.This study aims to find the differentially expressed endogenous peptides and their possible roles in IUA.Liquid chromatography-mass spectrometry was used to identify the peptidomic profiling of IUA tissues,and the differentially expressed peptides were screened out.Using real-time quantitative PCR,Western blotting,and immunocytochemistry staining,the function of six endogenous peptides was verified in vitro.It was found that peptide 6(T6)(peptide sequence:TFGGAPGFPLGSPLSSVFPR)could inhibit the expression of TGF-β1-induced cell fibrosis in human endometrial stromal cell line and primary human endometrial stromal cell at a concentration of 50μmol/L.This study provides new targets for further clarifying the formation and prevention of IUA.Xiangdong Hua Yan Zhang Juan Xu Lu Xu Yaqian Shi Dazhen Yang Xiaoyan Gu Sumin Wang Xuemei Jia Feng Xu Jie Chen Xiaoyan Ying 2022The Journal of Biomedical Research2022,36,4:3

8Molecular structure parameters and thermal stabilities of benzocarbazoles显示文摘The distance between 4-H and adjacent H on the substitutional benzene ring in benzo[c]carbazole was calculated in terms of molecular structure parameters. The result showed that the distance is shorter than the sum of van der Vaals' radii of two hydrogen atoms. This means that there is a steric hinderance between them. This effect will make the bond angle stretch and the bond length extend, or cause the molecule to distort, thus making benzo[c]carbazoles become unstable in thermodynamics. On the contrary there is no such effect for benzo[a]carbazoles. This conclusion has been confirmed by the result of calculation of the distance between the same two atoms with software GaussView. So the skeleton of benzo[c]carbazoles in sediments may be breached or isomerized with increasing maturity. 9Novel Zr-doped β-Li_(3)PS_(4) solid electrolyte for all-solid-state lithium batteries with a combined experimental and computational approach显示文摘All-solid-state lithium batteries(ASSLBs)are promising for safety and high-energy-density large-scale energy storage.In this contribution,we propose a Li_(3–4x)Zr_(x)PS_(4)(LZPS)by Zr-dopedβ-Li_(3)PS_(4)(LPS)as a novel solid electrolyte(SE)for ASSLBs based on experimental and simulation methods.The structure,electronic property,mechanical property,and ionic transport properties of LZPS(x=0,0.03,0.06,and 0.1)are investigated with first-principles calculations.Meanwhile,LZPS is prepared by solid states reaction method.By combining experimental analysis and first-principles calculations,it is confirmed that a small amount of Zr4+can be successfully doped into the framework ofβ-LPS composites without significantly compromising structural integrity.When the Zr^(4+)concentration is x=0.03,the doped material Li_(2.88)Zr_(0.03)PS_(4)exhibits the highest ionic conductivity(5.1×10^(−4)S·cm^(−1))at 30℃,and the Li-ion migration energy barrier is the lowest.The Li_(2.88)Zr_(0.03)PS_(4)SE has obtained the best mechanical properties,the good ductility,and shear deformation resistance,which can better maintain the structural stability of the battery.In addition,the Li/Li symmetrical cell is assembled,which shows excellent electrochemical stability of electrolyte against lithium.The constructed all-solid-state batteries(LiCoO_(2)-Li_(6)PS_(5)Cl|Li_(2.88)Zr_(0.03)PS_(4)|Li-In)delivers an initial discharge capacity of 130.4 mAh·g^(−1)at 0.2 C and a capacity retention of 85.1%after 100 cycles at room temperature.This study provides a promising electrolyte for the application of ASSLBs with high ionic conductivity and excellent stability against lithium.Junbo Zhang Guoxi Zhu Han Li Jiangwei Ju Jianwei Gu Renzhuang Xu Sumin Jin Jianqiu Zhou Bingbing Chen 2023Nano Research2023,16,2:0
10Osteocytes regulate bone anabolic response to mechanical loading in male mice via activation of integrinα5显示文摘Physical mechanical stimulation can maintain and even increase bone mass.Here,we report an important role of osteocytic integrinα5 in regulating the anabolic response of bone to mechanical loading using an Itga5 conditional gene knockout(cKO)mouse model.Integrinα5 gene deletion increased apoptotic osteocytes and reduced cortical anabolic responses to tibial compression including decreased endosteal osteoblasts and bone formation,and increased endosteal osteoclasts and bone resorption,contributing to the decreased bone area fraction and biomechanical properties,leading to an enlarged bone marrow area in cKO mice.Similar disruption of anabolic responses to mechanical loading was also detected in cKO trabecular bone.Moreover,integrinα5 deficiency impeded load-induced Cx43 hemichannel opening,and production and release of PGE2,an anabolic factor,resulting in attenuated effects of the loading on catabolic sclerostin(SOST)reduction and anabolicβ-catenin increase.Together,this study shows an indispensable role of integrinα5 in osteocytes in the anabolic action of mechanical loading on skeletal tissue through activation of hemichannels and PGE2-evoked gene expression.Integrinα5 could act as a potential new therapeutic target for bone loss,especially in the elderly population with impeded mechanical sensitivity.Dezhi Zhao Rui Hua Manuel A.Riquelme Hongyun Cheng Teja Guda Huiyun Xu Sumin Gu Jean X.Jiang 2022Bone Research2022,10,4:0
