| 1 | Orthotopic transplantation model of human gastrointestinal cancer and detection of micrometastases显示文摘AIM To establish a relevant animal model ofhuman gastrointestinal cancer, which can beused for repetitive investigations, so as toimprove our understanding and management ofcarcinogenesis and cancer metastasis.METHODS Intact tissues of human colorectaland pancreatic cancers were transplanted innude mice. The biological characteristics of theoriginal and the corresponding transplantedtumors were investigated by HE staining, PASstaining and immunostaining. The metastases inthe livers and lungs of nude mice wereinvestigated by immunostaining withbiotinylated mab KL-1 and by RT-PCR using CK20specific primers.RESULTS There were totally 9 of 16 surgicalspecimens growing in nude mice subcutaneouslyand/.or orthotopically (4 of 6 colorectal and 5 of10 pancreatic cancer). Tumor cell content of thespecimens and freezing of tissue specimens areimportant factors influencing the growth oftransplanted tumor. In the group of fresh tumortissues with greater than 50% tumor cellcontent, the success rate of the transplantationwas 100% (3 cases of pancreatic cancer and 3cases of colorectal cancer). The orthotopicallytransplanted tumors resemble the original tumormorphologically and biologically, including TAAexpression such as CEA byimmunohistochemistry, and CEA level in theserum of mice. Ki-67 labeling index and theexpression of TAA especially K-ras, 17-lA andRA-96, are associated with the potential of tumorgrowth in nude mice. Micrometastases in thelungs and livers of tumor bearing mice can bedetected by immunostaining with biotinylatedmab KL-1 and CK20-specific RT-PCR.CONCLUSION An orthotopic transplantationmodel for human colon and pancreatic cancer innude mice has been set up. We have alsoestablished sensitive detection methods withCK-immunohistochemistry and CK20-RT-PCR tostudy xenotransplanted human cancer and itsmetastatic cancer cells in the liver and lung ofnude mice. | 2 | Mycoplasma infections and different human carcinomas显示文摘AIM To explore relationships between human carcinomas and mycoplasma infection.METHODS Monoclonal antibody PD4, which specifically recognizes a distinct protein from mycoplasma hyorhinis, was used to detect mycoplasma infection in different paraffinembedded carcinoma tissues with immunohistochemistry. PCR was applied to amplify the mycoplasma DNA from the positive samples for confirming immunohistochemistry.RESULTS Fifty of 90 cases (56%) of gastric carcinoma were positive for mycoplasma hyorhinis. In other gastric diseases, the mycoplasma infection ratio was 28% (18/49) in chronic superficial gastritis, 30% (14/ 46) in gastric ulcer and 37% (18/ 49) in intestinal metaplasia. The difference is significant with gastric cancer (X2=12.06, P<0.05). In colon carcinoma, the mycoplasma infection ratio was 55.1% (32/58), but it was 20.9% (10/49) in adenomarous polyp (X2=13.46, P<0.005).Gastric and colon cancers with high differentiation had a higher mycoplasma infection ratio than those with low differentiation (P< 0.05). Mycoplasma infection in esophageal cancer, lung cancer, breast cancer and glioma was 50.9% (27/53), 52.6% (31/ 59), 39.7%(25/63) and 41% (38/91), respectively. The mycoplasma DNA was successfully amplified with the DNA extracted from the cancer tissues that were positive for mycoplasma infection (detected with antibody PD4).CONCLUSION There was high correlation between mycoplasma infection and different cancers, which suggests the possibility of an association between the two. | 5 | Building damage in Dujiangyan during Wenchuan Earthquake显示文摘A field damage survey of 1,005 buildings damaged by the Wenchuan Earthquake in Dujiangyan City was carried out and the resulting data was analyzed using the statistical method. It is shown that buildings that were seismically designed achieved the desired seismic fortification target; they sustained less damage than the non-seismically designed buildings. Among the seismically designed buildings investigated, RC frame buildings performed the best in terms of seismic resistance. Masonry buildings with a ground story of RC frame structure were the second best, and masonry buildings performed the worst. Considering building height, multistory buildings sustained more severe damage than high-rise buildings and 2- and 3-story buildings. The effects of the mass, damping, stiffness, natural period, and impulse duration (or oscillation period) on the translation, rotation, sliding force, overturning moment, and corresponding dynamic amplifying factors are studied. It is concluded that the ampli-ying factors only depend on the ratio of the system natural period to impulse duration (or oscillation period) under a certain damping ratio. In this study, the generalized forms of Pierson-Moskowitz spectrum and JONSWAP spectrum are used to describe the local wave spectrum by selecting suitable spectral form parameters. It is shown that, at a specific site, the similarity of wave spectral form exists. Thus it is possible to use a representative spectral form for a given nearshore region to describe the wave spectrum at this nearshore. The functional form of the eddy viscosity is evaluated based on computational results from a two-equation turbulence model in which the eddy viscosity varies with time and space. The present eddy viscosity model simplifies much of the mathematical complexity in many existing models. Predictions from the present model have been compared with a wide range of experimental data. | 20 | Action of Cnoidal Waves on Vertical Walls显示文摘Based on the 2nd order cnoidal wave theory, the characters of shallow water standing waves and their action on vertical walls are studied in this paper. The theoretical expressions of the wave surface elevation in front of and the wave pressure on the vertical wall are obtained. In order to verify the theoretical results, model tests were made in the State Key Laboratory of Coastal and Offshore Engineering at DUT. For the wave surface elevation in front of the wall and the wave forces on the wall at the moment when the wave surface at the wall surface goes down to the bottom of the wave trough, the calculated results coincide quite well with the experimental results.
