1Computational Prediction and Identification of Epstein-Barr Virus Latent Membrane Protein 2A Antigen-Specific CD8^+ T-Cell Epitopes显示文摘Epstein-Barr virus (EBV) associated nasopharyngeal carcinoma (NPC) is a high incidence tumor in Southeast Asia. Among EBV encoded proteins, latent membrane protein 2A (LMP2A) is an important antigen for T cell therapy of EBV. In this study, we predicted six HLA-A2 restricted CTL candidate epitopes of LMP2A by SYFPEITHI, NetMHC and MHCPred methods combined with the polynomial method. Subsequently, biological functions of these peptides were tested by experiments in vitro. In ELISPOT assay, the positive response of the LMP2A specific CTL stimulated by three (LMP2A264-272, LMP2A426-434 and LMP2A356-364) of six peptides respectively showed that the numbers of spots forming cells (SFC) ranged from 55.7 to 80.6 SFC/5 × 104 CD8+ T cells and the responding index (RI) ranged from 5.4 to 7. These three epitope-specific CTLs could effectively kill specific HLA-A2-expressing target cells. As a result, LMP2A264-272 (QLSPLLGAV), LMP2A426-434 (CLGGLLTMV) and LMP2A356-364 (FLYALALLL) were identified as LMP2A-specific CD8+ T-cell epitopes. 3Immunotherapy of Epstein-Barr Virus Associated Malignancies Using Mycobacterial HSP70 and LMP2A356-364 Epitope Fusion Protein显示文摘Epstein-Barr virus infection is strongly associated with a number of malignancies.The EBV latent membrane protein 2A has been implicated as one of the most attractive candidates for immunotherapy of related malignancies.In previous studies,the T cell epitopes of LMP2A have been identified systematically.However,the epitope-based vaccine generally meets inefficient immunogenicity when used in vivo directly,which could be overcome by combination with appropriate adjuvants.Heat shock protein is a natural chaperon,which is able to activate the classical major histocompatibility complex class I antigen-processing pathway(cross-presentation).In this study,a minigene encoding LMP2A356-364(FLYALALLL)was genetically fused to the carboxy-terminal of mycobacterial heat shock protein 70.The epitope fusion protein was expressed and purified,and the cross-presentation of LMP2A_(356-364) by monocyte-derived dendritic cells pulsed with the epitope fusion protein was evaluated.Results showed that the epitope fusion protein-pulsed mDCs were much more efficient than the single peptide-pulsed mDCs on CTL activation.Immunization of HLA-A2.1 transgenic mice with MtHsp70-LMP2A_(356-364) generated peptide specific CTL more effectively than a single peptide plus incomplete Freund's adjuvant(IFA).Growth of LMP2A expressing B16 melanoma tumor cells was suppressed in the vaccinated groups.Our results suggested that MtHsp70-LMP2A_(356-364) fusion protein was more effective than the CD8^(+)T cell epitope alone on anti-tumor immunity.As a result,the MtHsp70-LMP2A_(356-364) fusion protein is considered to be a promising candidate vaccine for EBV related malignancies.Genyan Liu Kun Yao Bing Wang Yun Chen Feng Zhou Yidi Guo Jian Xu Hongzhen Shi 2009Cellular & Molecular Immunology2009,6,6:6
4Potent Dendritic Cell Vaccine Loaded with Latent Membrane Protein 2A (LMP2A)显示文摘Epstein-Barr virus (EBV), a potential oncogenic herpesvirus, has been found to be associated with several malignancies. It's critical to elicit cellular immunity of the body to fight against EBV-associated tumor development. Using dendritic cells (DCs) loaded with latent membrane protein 2A (LMP2A) to elicit T cell response against tumor may be one of the most direct and safest immunotherapy approaches. The present study aimed to develop DCs-based cancer vaccine (DC loaded with LMP2A protein) and study its biological characteristics and immune functions. Purified LMP2A protein was extracted from a cell line L929/LMP2A stably expressing LMP2A. LMP2A could be loaded on DCs with no significant changes of the DC surface markers and cytomorphology. The percentage of DCs loaded with LMP2A was above 80%. LMP2A-loaded DCs markedly enhanced the proliferation of antigen-specific CD8+ T and CD4+ T cells by 3H-TdR incorporation assay. Besides, the specific cytotoxicity of the CTLs against LMP2A target cells was also significantly increased. 5Prevalence and trends of aminoglycoside resistance in Shigella worldwide, 1999-2010显示文摘Shigellosis causes diarrheal disease in humans in both developed and developing countries, and multi-drug resistance in Shigella is an emerging problem. Understanding changing resistance patterns is important in determining appropriate antibiotic treatments. This meta-analysis systematically evaluated aminoglycoside resist-ance in Shigella. A systematic review was constructed based on MEDLINE and EMBASE databases. Randomeffect models or fixed-effect models were used based on P value considering the possibility of heterogeneity between studies for meta-analysis. Data manipulation and statistical analyses were performed using software STATA 11.0. By means of meta-analysis, we found a lower resistance to three kinds of aminoglycosides in the Europe-America areas during the 12 year study period than that of the Asia-Africa areas. Kanamycin resistance was observed to be the most common drug resistance among Shigella isolates with a prevalence of 6.88% (95%CI: 6.36%-7.43%). Comparison of data from Europe-America and Asia-Africa areas revealed that Shigella flexneri resistance was greater than the resistance calculated for Shigella sonnei. Importantly,Shigella sonnei has played a significant role in aminoglycoside-resistance in recent years. Similarly, data showed that resistance to these drugs in children was higher than the corresponding data of adults. In conclusion, aminoglycoside-resistant Shigella is not an unusual phenomenon worldwide. Distribution in Shigella resistance differs sharply based on geographic areas, periods of time and subtypes. 6Physiological functions and clinical implications of fibrinogen-like 2:A review显示文摘Fibrinogen-like 2(FGL2) encompasses a transmembrane(m FGL2) and a soluble(s FGL2) form with differential tertiary structure and biological activities. Typically, m FGL2 functions as prothrombinase that is capable of initiating coagulation in tissue without activation of the blood clotting cascade, whereas s FGL2 largely acts as an immunosuppressor that can repress proliferation of alloreactive T lymphocytes and maturation of bone marrow dendritic cells. Protein sequences of FGL2 exhibit evolutionary conservation across wide variety of species, especially at the carboxyl terminus that contains fibrinogen related domain(FRED). The FRED of FGL2 confers specificity and complexity in the action of FGL2, including receptor recognition, calcium affiliation, and substrate binding. Constitutive expression of FGL2 during embryogenesis and in mature tissues suggests FGL2 might be physiologically important. However, excessive induction of FGL2 under certain medical conditions(e.g. pathogen invasion) could trigger complement activation, inflammatory response,cellular apoptosis, and immune dysfunctions. On the other hand, complete absence of FGL2 is also detrimental as lack of FGL2 can cause autoimmune glomerulonephritis and acute cellular rejection of xenografts. All these roles involve m FGL2, s FGL2, or their combination. Although it is not clear how m FGL2 is cleaved off its host cells and secreted into the blood, circulating s FGL2 has been found correlated with disease severity and viral loading among patients with human hepatitis B virus or hepatitis C virus infection. Further studies are warranted to understand how FGL2 expression is regulated under physiological and pathological conditions. Even more interesting is to determine whether m FGL2 can fulfill an immunoregulatory role through its FRED at carboxyl end of the molecule and, and vice versa, whether s FGL2 is procoagulant upon binding to a target cell. 9Preparation of nanostructured mesoporous Ni Co_2O_4 and its electrocatalytic activities for water oxidation显示文摘Nanostructured mesoporous Ni Co2O4,Co3O4 and Ni O spinel oxides were prepared through the hydroxide coprecipitation method using EG(ethylene glycol) as solvent without any template.The Ni Co2O4 powders were analyzed by TGA,XRD,N2adsorption-desorption,SEM and EDS.And the electrocatalytic properties of Ni Co2O4/Ti,Co3O4/Ti and Ni O/Ti electrodes for oxygen evolution in 0.1 mol/L KOH(p H = 13)were tested by cyclic voltammetry,Tafel curves and electrolysis.The result shows that Ni Co2O4/Ti electrode has higher electrocatalytic activity in oxygen evolution reaction than Co3O4/Ti and NiO/Ti electrodes.Ni Co2O4/Ti electrode prepared at 350°C has the highest apparent current density and the highest true current density.The Tafel slope of Ni Co2O4/Ti electrode prepared at 350°C is 68.49 m V/dec.Genyan Hao Wei Wang Guofeng Gao Qiang Zhao Jinping Li 2015Journal of Energy Chemistry2015,24,3:1

11Strengthen metal-oxygen covalency of CoFe-layered double hydroxide for efficient mild oxygen evolution显示文摘Oxygen evolution reaction(OER)is crucial for hydrogen production as well as other energy storage technologies.CoFe-layered double hydroxide(CoFe-OH)has been widely considered as one of the most efficient electrocatalysts for OER in basic aqueous solution.However,it still suffers from low activity in neutral electrolyte.This paper describes partially oxidized CoFe-OH(PO-CoFe-OH)with enhanced covalency of M-O bonds and displays enhanced OER performance under mild condition.Mechanism studies reveal the suitably enhanced M-O covalency in PO-CoFe-OH shifts the OER mechanism to lattice oxygen oxidation mechanism and also promotes the rate-limiting deprotonation,providing superior OER performance.It just requires the overpotentials of 186 and 365 mV to drive the current density densities of 1 and 10 mA·cm^(-2) in 0.1 M KHCO_(3) aqueous solution(pH=8.3),respectively.It provides a new process for rational design of efficient catalysts for water oxidation in mild conditions.Dazhong Zhong Tan Li Dong Wang Lina Li Jiancheng Wang Genyan Hao Guang Liu Qiang Zhao Jinping Li 2022Nano Research2022,15,1:1

13Ag nanoparticles anchored on MIL-100/nickel foam nanosheets as an electrocatalyst for efficient oxygen evolution reaction performance显示文摘Metal-organic frameworks(MOFs)exhibit excellent application potential in the field of electrocatalysis.In this study,we first prepare MIL-100 nanosheets on nickel foam(MIL-100/NF)and then successfully anchor Ag nanoparticles(NPs)on the nanosheets(Ag@MIL-100/NF)for oxygen evolution reaction(OER)catalysis.This strategy dramatically improves the conductivity of MIL-100 and the Ag NPs are uniformly dispersed on the nanosheets.The Ag@MIL-100/NF catalyst has excellent electrocatalytic performance and long-term corrosion resistance,with a low overpotential of 207 mV and a long-term stability of at least 100 h at a current density of 50 mA·cm^(-2).The experimental results demonstrate that this high OER catalytic performance is due to the improved charge transfer after loading Ag NPs,the combination of nanosheets and highly dispersed Ag NPs that expose more active sites and the adjusted chemical valence states of Fe and Ni in MIL-100.This work provides a surface decoration approach for the preparation of excellent catalysts directly used in the OER.Tao Zhao Dazhong Zhong Genyan Hao Guang Liu Jinping Li Qiang Zhao 2022Chinese Journal of Chemical Engineering2022,35,1:0
