1Orthogonal analysis of water model study on the optimization of flow control devices in a six-strand tundish显示文摘Improper flow control devices in a multi-strand tundish can cause some problems, for example, liquid steel cannot reach every nozzle at the same time and the liquid steel in nozzles far away from the entry zone has a lower temperature. The water model experiment of a six-strand tundish of Tianjin Iron & Steel Co. Ltd. was performed, a new 'U' type baffle was obtained, and its parameters were defined by perpendicular analysis. 3Psychological impact of the COVID-19 pandemic on healthcare workers:a cross-sectional study in China显示文摘Background Healthcare workers fighting against the coronavirus disease 2019(COVID-19)pandemic are under tremendous pressure,which puts them at an increased risk of developing psychological problems.Aims This study aimed to investigate the prevalence of psychological problems in different healthcare workers(ie,physicians,medical residents,nurses,technicians and public health professionals)during the COVID-19 pandemic in China and explore factors that are associated with the onset of psychological problems in this population during this public health crisis.Methods A cross-sectional,web-based survey was conducted in February 2020 among healthcare workers during the COVID-19 pandemic.Psychological problems were assessed using the Generalized Anxiety Disorder Scale,Patient Health Questionnaire and Insomnia Severity Index.Logistic regression analyses were used to explore the factors that were associated with psychological problems.Results The prevalence of symptoms of anxiety,depression,insomnia and the overall psychological problems in healthcare workers during the COVID-19 pandemic in China was 46.04%,44.37%,28.75%and 56.59%,respectively.The prevalence of the overall psychological problems in physicians,medical residents,nurses,technicians and public health professionals was 60.35%,50.82%,62.02%,57.54%and 62.40%,respectively.Compared with healthcare workers who did not participate in front-line work,front-line healthcare workers had a higher risk of anxiety,insomnia and overall psychological problems.In addition,attention to negative or neutral information about the pandemic,receiving negative feedback from families and friends who joined front-line work,and unwillingness to join front-line work if given a free choice were three major factors for these psychological problems.Conclusions Psychological problems are pervasive among healthcare workers during the COVID-19 pandemic.Receiving negative information and participating in front-line work appear to be.important risk factors for psychological problems.The psychological health of different healthcare workers should be protected during the COVID-19 pandemic with timely interventions and proper information feedback.Jianyu Que Le Shi Jiahui Deng Jiajia Liu Li Zhang Suying Wu Yimiao Gong Weizhen Huang Kai Yuan Wei Yan Yankun Sun Maosheng Ran Yanping Bao Lin Lu 2020General Psychiatry2020,33,3:8

5Groundwater contributions in water-salt balances of the lakes in the Badain Jaran Desert,China显示文摘Groundwater-fed lakes are essential for the ecology in arid and semiarid regions.As a typical arid region,the Badain Jaran Desert (BJD) is famous in the world for the presence of a large number of groundwater-fed saline lakes among the mega dunes.Based on the up to date geological surveys and observations,this study analyzed the groundwater contributions in water-salt balances of the lakes in the desert.We found different types of springs,including the sublacustrine springs that indicate an upward flow of groundwater under the lakebed.A simplified water balance model was developed to analyze the seasonal variations of water level in the Sumu Barun Jaran Lake,which revealed an approximately steady groundwater discharge in the lake and explained why the amplitude of seasonal changes in lake level is less than 0.5 m.In addition,a salt balance model was developed to evaluate the salt accumulations in the groundwater-fed lakes.The relative salt accumulation time is 800–7,000 years in typical saline lakes,which were estimated from the concentration of Cl-,indicating a long history evolution for the lakes in the BJD.Further researches are recommended to provide comprehensive investigations on the interactions between the lakes and groundwater in the BJD.GONG Yanping WANG Xusheng HU B Xiao ZHOU Yangxiao HAO Chunbo WAN Li 2016Journal of Arid Land2016,8,5:7
6Developing controllable hypermutable Clostridium cells through manipulating its methyl-directed mismatch repair system显示文摘Development of controllable hypermutable cells can greatly benefit understanding and harnessing microbial evolution.However,there have not been any similar sys-tems developed for Clostridium,an important bacterial genus.Here we report a novel two-step strategy for de-veloping controllable hypermutable cells of Clostridium acetobutylicum,an important and representative indus-trial strain.Firstly,the mutS/L operon essential for methyl-directed mismatch repair(MMR)activity was inactivated from the genome of C.acetobutylicum to generate hy-permutable cells with over 250-fold increased mutation rates.Secondly,a proofreading control system carrying an inducibly expressed mutS/L operon was constructed.The hypermutable cells and the proofreading control system were integrated to form a controllable hypermut-able system SMBMutC,of which the mutation rates can be regulated by the concentration of anhydrotetracycline(aTc).Duplication of the miniPthl-tetR module of the proof-reading control system further signifi cantly expanded the regulatory space of the mutation rates,demonstrating hypermutable Clostridium cells with controllable mutation rates are generated.The developed C.acetobutylicum strain SMBMutC2 showed higher survival capacities than the control strain facing butanol-stress,indicating greatly increased evolvability and adaptability of the controllable hypermutable cells under environmental challenges.Guodong Luan Zhen Cai Fuyu Gong Hongjun Dong Zhao Lin Yanping Zhang Yin Li 2013Protein & Cell2013,4,11:4
7The disbalance of LRP1 and SIRPα by psychological stress dampens the clearance of tumor cells by macrophages显示文摘The relationship between chronic psychological stress and tumorigenesis has been well defined in epidemiological studies;however, the underlying mechanism remains underexplored. In this study, we discovered that impaired macrophage phagocytosis contributed to the psychological stressevoked tumor susceptibility, and the stress hormone glucocorticoid(GC) was identified as a principal detrimental factor. Mechanistically, GC disturbed the balance of the 'eat me' signal receptor(low-density lipoprotein receptor-related protein-1, LRP1) and the 'don’t eat me' signal receptor(signal regulatory protein alpha, SIRPa). Further analysis revealed that GC led to a direct, glucocorticoid receptor(GR)-dependent trans-repression of LRP1 expression, and the repressed LRP1, in turn, resulted in the elevatedgene level of SIRPa by down-regulating mi RNA-4695-3 p. These data collectively demonstrate that stress induces the imbalance of the LRP1/SIRPa axis and entails the disturbance of tumor cell clearance by macrophages. 12A single-center retrospective study on epidemiological and Traditional Chinese Medicine syndrome characteristics of 21010 patients with reflux/heartburn symptoms显示文摘OBJECTIVE:To provide a basis for the clinical identification of true and false reflux,integrated traditional Chinese and Western medicine,and psychosomatic treatment,we conducted a retrospective study of the etiology and epidemiological and Traditional Chinese Medicine(TCM) syndrome characteristics of patients with reflux/heartburn symptoms.METHODS:The 210 10 patients with reflux/heartburn treated at Tianjin Nankai Hospital from January 1,2016,to December 31,2019,were divided into four groups according to their pathogenesis.Sex,age,course of disease,incidence rate,gastroscopy,24-h p Himpedance,esophageal manometry,Hamilton Anxiety Scale(HAMA)/Hamilton Depression Scale(HAMD) score,8-week proton pump inhibitor(PPI) treatment effect,and TCM syndrome characteristics were statistically analyzed.RESULTS:A total of 21010 patients(8864 men and 12146 women),with reflux/heartburn symptoms were screened,including 6284(29.9%) patients with reflux esophagitis(RE),10427(49.6%) patients with nonerosive reflux esophagitis(NERD),2430(11.6%) patients with reflux hypersensitivity(RH),and 1870(8.9%) patients with functional heartburn(FH).The incidence of the disease was higher in women than in men(P <0.0001).The ranking of the incidence of anxiety and depression in these four groups was FH > RH > NERD > RE(P < 0.0001).There were more women than men in the groups with anxiety and more men than women in the groups with depression(P < 0.0001),and there was no significant difference in the distribution of anxiety and depression between men and women(P = 0.5689).There were significant differences in TCM syndrome characteristics between NERD,RE,and functional esophageal diseases(P < 0.01).The highest proportion of functional esophageal disease TCM symptoms was Qi stagnation and phlegm obstruction syndrome(36.16%),and there was no significant difference between RH and FH.The effective rates of PPI treatment at 8 weeks in patients in the RE,NERD,RH,and FH groups were 89%,72%,54%,and 0%,respectively.RE was classified into grades A,B,C,and D according to the Los Angeles grading system.The ranking of the incidence of these four grades was A > B > C > D(P < 0.0001).The effective rates of PPI treatment at 8 weeks were 91%,81%,69%,and 63% in patients with grade A,B,C,and D RE,respectively(P < 0.0001).The highest proportion of TCM syndrome types of NERD and RE was the stagnated heat syndrome in the liver and stomach syndrome,38.99% and 33.90%,respectively.CONCLUSION:Reflux/heartburn symptoms are relatively common in middle-aged women,and NERD is the most common etiology,followed by RE,RH,and FH.The most common TCM syndrome characteristics in NERD and RE were stagnated heat syndrome in the liver and stomach syndrome,and Qi stagnation and phlegm obstruction syndrome in functional esophageal diseases.Most patients with reflux/heartburn symptoms also experienced anxiety and depression.TANG Yanping LI Peicai LIU Xi LIU Lei GONG Yanxia WEI Xiaodong LIU Lina YANG Li 2023Journal of Traditional Chinese Medicine2023,43,3:2

15CRISPR/Cas9-mediated mutagenesis of CiBG1 decreased seed size and promoted seed germination in watermelon显示文摘Abscisic acid(ABA)is a critical regulator of seed development and germination.β-glucosidases(BGs)have been suggested to be contributors to increased ABA content because they catalyze the hydrolysis of ABA-glucose ester to release free ABA.However,whether BGs are involved in seed development is unclear.In this study,a candidate gene,CiBG1,in watermelon was selected for targeted mutagenesis via the CRISPR/Cas9 system.Seed size and weight were significantly reduced in the Clbg1-mutant watermelon lines,which was mainly attributed to decreased cell number resulting from decreased ABA levels.A transcriptome analysis showed that the expression of 1015 and 1429 unique genes was changed 10 and 18 days after pollination(DAP),respectively.Cytoskeleton-and cell cycle-related genes were enriched in the differentially expressed genes of wild type and Clbg1-mutant lines during seed development.Moreover,the expression of genes in the major signaling pathways of seed size control was also changed.In addition,seed germination was promoted in the Cibg1-mutant lines due to decreased ABA content.These results indicate that ClBG1 may be critical for watermelon seed size regulation and germination mainly through the modulation of ABA content and thereby the transcriptional regulation of cytoskeleton-,cell cycle-and signaling-related genes.Our results lay a foundation for dissecting the molecular mechanisms of controlling watermelon seed size,a key agricultural trait of significant economic importance.Yanping Wang Jinfang Wang Shaogui Guo Shouwei Tian Jie Zhang Yi Ren Maoying Li Guoyi Gong Haiying Zhang Yong Xu 2021Horticulture Research2021,8,1:1

18Oxidized galectin-1 in SLE fails to bind the inhibitory receptor VSTM1 and increases reactive oxygen species levels in neutrophils显示文摘Inhibitory immune receptors set thresholds for immune cell activation,and their deficiency predisposes a person to autoimmune responses.However,the agonists of inhibitory immune receptors remain largely unknown,representing untapped sources of treatments for autoimmune diseases.Here,we show that V-set and transmembrane domain-containing 1(VSTM1)is an inhibitory receptor and that its binding by the competent ligand soluble galectin-1(Gal1)is essential for maintaining neutrophil viability mediated by downregulated reactive oxygen species production.However,in patients with systemic lupus erythematosus(SLE),circulating Gal1 is oxidized and cannot be recognized by VSTM1,leading to increased intracellular reactive oxygen species levels and reduced neutrophil viability.Dysregulated neutrophil function or death contributes significantly to the pathogenesis of SLE by providing danger molecules and autoantigens that drive the production of inflammatory cytokines and the activation of autoreactive lymphocytes.Interestingly,serum levels of glutathione,an antioxidant able to convert oxidized Gal1 to its reduced form,were negatively correlated with SLE disease activity.Taken together,our findings reveal failed inhibitory Gal1/VSTM1 pathway activation in patients with SLE and provide important insights for the development of effective targeted therapies.Xu Jiang Xinyue Xiao Hao Li Yiyi Gong Min Wang Huaxia Yang Lidan Zhao Ying Jiang Yanping Wei Chongchong Zhao Jin Li Yuling Chen Shan Feng Haiteng Deng Shiliang Ma Yue Xu Yudong Liu George C.Tsokos Minghong Jiang Xuan Zhan 2023Cellular & Molecular Immunology2023,20,11:1
