| 1 | Improved Brassica rapa reference genome by single-molecule sequencing and chromosome conformation capture technologies显示文摘Brassica rapa comprises several important cultivated vegetables and oil crops.Current reference genome assemblies of Brassica rapa are quite fragmented and not highly contiguous,thereby limiting extensive genetic and genomic analyses.Here,we report an improved assembly of the B.rapa genome(v3.0)using single-molecule sequencing,optical mapping,and chromosome conformation capture technologies(Hi-C).Relative to the previous reference genomes,our assembly features a contig N50 size of 1.45 Mb,representing a~30-fold improvement.We also identified a new event that occurred in the B.rapa genome~1.2 million years ago,when a long terminal repeat retrotransposon(LTR-RT)expanded.Further analysis refined the relationship of genome blocks and accurately located the centromeres in the B.rapa genome.The B.rapa genome v3.0 will serve as an important community resource for future genetic and genomic studies in B.rapa.This resource will facilitate breeding efforts in B.rapa,as well as comparative genomic analysis with other Brassica species. | Lei Zhang Xu Cai Jian Wu Min Liu Stefan Grob Feng Cheng Jianli Liang Chengcheng Cai Zhiyuan Liu Bo Liu Fan Wang Song Li Fuyan Liu Xuming Li Lin Cheng Wencai Yang Mai-he Li Ueli Grossniklaus Hongkun Zheng Xiaowu Wang | 2018 | Horticulture Research2018，5，1： | 10 |
| 2 | Branched tetraether lipids in Chinese soils:Evaluating the fidelity of MBT/CBT proxies as paleoenvironmental proxies显示文摘The ubiquitous occurrence of branched glycerol dialkyl glycerol tetraethers(br GDGTs) in soils has allowed development of new proxies for reconstruction of past climate and environment. The methylation and cyclization degrees of br GDGTs, expressed as MBT and CBT, respectively, are reported to be mainly controlled by mean annual air temperature(MAAT) and soil p H. However, the br GDGT-derived temperatures and soil p H scatter widely when data from different environmental conditions are considered. In this study, we collected over 300 soil samples from China, which are representative of humid(Xishuangbanna, Guangzhou, and Shanghai), semi-arid(Dongying) and semi-arid/arid(Lanzhou, Tibetan Plateau) regions. Collectively we have the most extensive dataset that broadly characterizes the distribution of br GDGTs according to climate zones in China. The overall data demonstrate that the MBT/CBT derived temperatures better match the measured MAATs in humid and non-alkaline regions than those from regions of low MAP(<400 mm/yr) and above neutral soil p H(>7.0–7.5). Similarly, CBT describes soil p H much better in humid and non-alkaline soils than in semi-arid/arid and alkaline soils; the semi-arid/arid and alkaline soils tend to show a positive correlation between soil p H and CBT, which contradicts that in the humid and non-alkaline soils. While soil p H, MAAT and mean annual precipitation(MAP) are dominating factors controlling the br GDGT distribution across all climate zones, conductivity, total organic carbon and total nitrogen, as well as soil water content can also play an important role locally. Removing br GDGT-II resulted in a revised CBT index that provides more accurate estimation of p H, especially in semi-arid/arid and alkaline soils. | 3 | Comparison of the short-term and long-term outcomes of laparoscopic hysterectomies and of abdominal hysterectomies: a case study of 4,895 patients in the Guangxi Zhuang Autonomous Region, China显示文摘Background: To evaluate the short-term and long-term outcomes after laparoscopic hysterectomy(LH) compared with abdominal hysterectomy(AH) in case of benign gynecological disease. Methods: A multi-center cohort retrospective comparative study of population among 4,895 hysterectomies(3,539 LH vs.1,356 AH) between 2007 and 2013 was involved. Operative time(OT), estimated blood loss(EBL), intra-operative and post-operative complications, passing flatus; days with indwelling catheter, questionnaires covering pelvic floor functions and sexual functions were assessed.Results: The EBL(174.1±157.4 vs. 263.1±183.2 cc, LH and AH groups, respectively), passing flatus(38.7±14.1 vs. 48.1±13.2 hours), days with indwelling catheter(1.5±0.6 vs. 2.2±0.8 days), use of analgesics(6.5% vs. 73.1%), intra-operative complication rate(2.4% vs. 4.1%), post-operative complication rate(2.3% vs. 5.7%), post-operative constipation(12.1% vs. 24.6%), mild and serious stress urinary incontinence(SUI) post-operative(P<0.001; P=0.014), and proportion of Female Sexual Functioning Index(FSFI) total score <26.55 post-operative(P<0.001) of the LH group were significantly less than those of AH group. There wereno significant differences in OT(106.5±34.5 vs. 106.2±40.3 min) between the two groups.Conclusions: LH is a safe and efficient operation for improving patients' long-term quality of life(Qo L), and LH is a cost-effectiveness procedure for treating benign gynecological disease. | 4 | Biogenic synthesis of silver nanoparticles using ginger(Zingiber officinale)extract and their antibacterial properties against aquatic pathogens显示文摘With the development of aquaculture, there is an urgent demand for an alternative antibacterial agent to reduce the drug resistance and environmental pollution caused by the abuse of antibiotics. Recently, silver nanoparticles(Ag NPs) have been viewed as a novel type of antimicrobial agents due to their unique advantages. In this study,Ag NPs were biosynthesized with the ginger rhizomes extract. The biosynthesized Ag NPs were characterised by UV–visible spectroscopy, transmission electron microscopy, X-ray diffraction and fourier transform infrared spectroscopy. Furthermore, the antimicrobial activities of the Ag NPs were fully analyzed against six typical aquatic pathogens. The results indicated that the components in ginger extract could function as the chemical reductant to synthesize Ag NPs. Moreover, compared with the Ag NPs synthesized by chemical methods, the biosynthesized Ag NPs were smaller, and had higher stability and antibacterial activity. | 6 | Effects of the physical and chemical properties of petroleum coke on its slurryability显示文摘In this study, the effects of particle size distribution (PSD), chemical composition and pore structure of petroleum coke on the slurryability of petroleum coke water slurry (PCWS) were investigated. Four petroleum cokes were studied, and they showed completely different slurryability. The solid concentration at fixed viscosity (i.e. apparent viscosity of 1000 mPa) (SCFV) of four PCWSs is different from each other, with the highest value of 70.9%, and lowest of 62.1%. The apparent viscosity of the four PCWSs all increased with an increase of the solid concentration. The results showed that the PSD, inherent moisture content, specific surface area and pore volume of petroleum coke were key factors to affect the slurryability. | 9 | Mobile ARGONAUTE 1d binds 22-nt miRNAs to generate phasiRNAs important for low-temperature male fertility in rice显示文摘Phased small interfering RNAs(phasiRNAs) are abundantly expressed in anthers and linked to environment-related male fertility in grasses, yet how they function under different environmental conditions remains unclear. Here, we identified a rice(Oryza sativa) low temperature-induced Argonaute(AGO) protein, OsAGO1d, that is responsible for generating phasiRNAs and preserving male fertility at low temperature. Loss of OsAGO1d function causes low-temperature male sterility associated with delayed programmed cell death of tapetal cells during anther development. OsAGO1d binds miR2118 and miR2275 family members and triggers phasiRNA biogenesis;it also binds 21-nt phasiRNAs with a 5′ terminal U. In total, phasiRNAs from 972loci are OsAGO1d-dependent. OsAGO1d protein moves from anther wall cells into meiocytes, where it loads miR2275 to produce 24-nt phasiRNAs. In this paper, permutation and sub- stitution methods are incorporated to present a stronger image encryption algorithm. Spatial chaotic maps are used to realize the position permutation, and to confuse the relationship between the ci- pher-image and the plain-image. A failed denoising may introduce many artifacts, and lead to the failure of final processing results. Seislet transform is a wavelet-like transform that analyzes seismic data following variable slopes of seismic events. The local slope is the key of seismic data. An earlier work used traditional normal moveout(NMO) equation to construct velocity-dependent(VD) seislet transform, which only adapt to hyperbolic condition. In this work, we use shifted hyperbola NMO equation to obtain more accurate slopes in nonhyperbolic situation. Self-adaptive threshold method was used to remove random noise while preserving useful signal. | 20 | Design of a high-sampling-rate electronic module for array-detector positron annihilation lifetime measurements显示文摘Introduction In this study,a high-time-resolution electronic module with a high channel density and low power consumption was designed for the measurement of the multi-detector array positron annihilation lifetimes.This electronic module consisted of 32 input channels,and each channel provided a high sampling rate up to 5.12 GSPS based on a Domino Ring Sampler 4(DRS4)chip.Compared to the high-speed flash analog-digital converter(FADC),DRS4 chip has a higher channel density with an affordable lower price and power consumption.Methods The developed electronic module was also capable of real-time data analysis for directly extracting the time information of input signals at the data acquisition site,thereby significantly decreasing the data rate.The digital constant fraction discriminator(DCFD)algorithm was implemented in the field programmable gate array(FPGA)for performing the time pick-up.Results The coincidence time resolution of the electronic module was measured,and the test results revealed a value of 26 ps.A prototypical 16-pixel detector module of the multi-detector system was evaluated using this electronic module,and the coincidence time resolution of the prototypical module was 411.84 ps.Conclusions The electronic module was confirmed to satisfy the severe requirements of the multi-array-detector positron annihilation lifetime measurement system.It was also suitable for other high-time-resolution,high-channel-density,costeffective,and low-power-consumption applications. | Jiale Cai Daowu Li Yingjie Wang Zhiming Zhang Peilin Wang Haohui Tang Baoyi Wang Xingzhong Cao Fuyan Liu Long Wei | 2019 | Radiation Detection Technology and Methods2019，3，3： | 0 |
