| 1 | Altered expression of mitochondrial related genes in the native Tibetan placents by mitochondrial cDNA array analysis显示文摘Objective: To explore the mechanism of native Tibetan fetuses adaptation to hypoxia, we tried to find the different expression genes about mitochondrial function in the native Tibetan placents. Methods: In this study, the placents of native Tibetan and the high-altitude Han (ha-Han) were collected. After the total RNA extraction, the finally synthesized cDNAs were hybridized to mitochondrial array to find the altered expression genes between them. Then, the cytochrome c oxidase 17 (Cox17), dynactin 2 (DCTN2, also known as p50), and vascular endothelial growth factor receptor (VEGFR, also known as KDR) were chosen from the altered expression genes to further verify the array results using the SYBR Green real-time PCR. Because the altered expression genes (such as Cybb and Cox17) in the array results related to the activities of COXI and COXIV, the placental mitochondria activities of COXI and COXIV were measured to find their changes in the hypoxia. Results: By a standard of ≥1.5 or ≤0.67, there were 24 different expressed genes between the native Tibetan and the ha-Han placents, including 3 up-regulated genes and 21 down-regulated genes. These genes were related to energy metabolism, signal transduction, cell proliferation, electron transport, cell adhesion, nucleotide-excision repair. The array results of Cox17, DCTN2 and KDR were further verified by the real-time RT-PCR. Through the mitochondria respiration measurements, the activity of COXI in the native Tibetan placents were higher than that of ha-Han, there was no difference in COXIV activity between them. | 3 | Wall shear stress in intracranial aneurysms and adjacent arteries显示文摘Hemodynamic parameters play an important role in aneurysm formation and growth. However, it is difficult to directly observe a rapidly growing de novo aneurysm in a patient. To investigate possible associations between hemodynamic parameters and the formation and growth of intracranial aneurysms, the present study constructed a computational model of a case with an internal carotid artery aneurysm and an anterior communicating artery aneurysm, based on the CT angiography findings of a patient. To simulate the formation of the anterior communicating artery aneurysm and the growth of the internal carotid artery aneurysm, we then constructed a model that virtually removed the anterior communicating artery aneurysm, and a further two models that also progressively decreased the size of the internal carotid artery aneurysm. Computational simulations of the fluid dynamics of the four models were performed under pulsatile flow conditions, and wall shear stress was compared among the different models. In the three aneurysm growth models, increasing size of the aneurysm was associated with an increased area of low wall shear stress, a significant decrease in wall shear stress at the dome of the aneurysm, and a significant change in the wall shear stress of the parent artery. The wall shear stress of the anterior communicating artery remained low, and was significantly lower than the wall shear stress at the bifurcation of the internal carotid artery or the bifurcation of the middle cerebral artery. After formation of the anterior communicating artery aneurysm, the wall shear stress at the dome of the internal carotid artery aneurysm increased significantly, and the wall shear stress in the upstream arteries also changed significantly. | 4 | Type I interferon:From innate response to treatment for COVID-19显示文摘Effective prophylactic and therapeutic interventions are urgentlyneeded to address the coronavirus disease 2019(COVID-19)pandemic.Various antiviral drugs have recently been tested.Type I interferon(IFN)is a regulatory protein involved in the innate immune response,with broad-spectrum antiviral activities and the ability to directly block viral replication and support the immune response to eliminate virus infection.Insufficient virus-induced type I IFN production is characteristic of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection,because SARS-CoV-2 suppresses the IFN response by interacting with essential IFN signaling pathways.Exogenous type I IFN is recommended for treating COVID-19.Unexpectedly however,angiotensin converting enzyme-2(ACE2)receptor,which acts as a SARS-CoV-2 receptor,was shown to be stimulated by IFN,raising doubts about the suitability of IFN use.However,further studies have excluded concerns regarding IFN administration.Type I IFNs,including IFN-α1b,have been used clinically as antiviral drugs for many years and have shown strong antiviral activity against SARS-CoV-2in vitro.Preliminary clinical studies of type I IFNs,especially when delivered via aerosol inhalation,have demonstrated efficacy for the treatment and prevention of COVID-19.Randomized controlled trials of IFN for COVID-19 treatment are ongoing. | Fuyu Lin Kunling Shen | 2020 | Pediatric Investigation2020,4,4: | 5 |

| 6 | Assemblies of covalent organic framework microcrystals:multiple-dimensional manipulation for enhanced applications显示文摘Covalent organic frameworks (COFs) are well known as the next generation of shape-persistent zeolite analogues, which have brought new impetus to the development of porous organic materials as well as two-dimensional polymers. Since the advent of COFs in 2005, many striking findings have definitely proven their great potentials expanding applications across energy,environment and healthcare fields. With thorough exploration over a decade, research interest has been drawn on the scientific challenges on chemistry, while making full play of COF values has remained far from satisfactory yet. Thus opening an avenue to modulating COF assemblies on the multi-scale is no longer just an option, but a necessity for matching the application requirements with enhanced performances. In this mini-review, we summarize the recent progress on design of nanoscale COFs with varying forms. | 7 | Effects on Agronomic Traits,Yield and Benefit of One-year Triple Cropping 'Maize-soybean' Strip Intercropping System Under Reduced Fertilizer Application显示文摘[Objectives] This study was conducted to investigate the effects on agronomic traits,yield and benefit of one-year triple cropping ' maize-soybean' strip intercropping system under reduced fertilizer application. [Methods]A single factor randomized block design was used,and wide and narrow strip intercropping was adopted. Three different fertilization treatments,namely fertilizers for conventional field fertilization( A1),80% A1( A2) and 60% A1( A3) were applied to the field production. The fertilizing sites were about 25 cm away from maize and repeated three times. [Results]Compared with A1,the change laws of the agronomic traits and yield in the A2 and A3 intercropping systems were described as below. Agronomic traits: The plant height became shorter gradually for the spring soybean,but increased gradually for the summer soybean,the bottom pod height gradually reduced,and the node number,pods per plant and seeds per plant increased;and the plant height,ear length,rows per ear,seeds per row and 1 000-seed weight of the spring maize decreased. Yield: The yield of maize as the main crop decreased but not significant,by 3. 20% and 3. 99%,respectively in A2 and A3,both smaller than 5. 00%;the yield of the spring soybean significantly increased by 9. 70% and 11. 84%,respectively;the yield of the summer soybean increased by 5. 18% and 8. 98%,respectively;and the total yield increased by 0. 20% and 0. 92%,respectively. Benefit: The total output value increased by 2. 97 % and 4. 91 %,respectively,and the total benefit increased by 6. 39% and 11. 22%,respectively. | 8 | Developing controllable hypermutable Clostridium cells through manipulating its methyl-directed mismatch repair system显示文摘Development of controllable hypermutable cells can greatly benefit understanding and harnessing microbial evolution.However,there have not been any similar sys-tems developed for Clostridium,an important bacterial genus.Here we report a novel two-step strategy for de-veloping controllable hypermutable cells of Clostridium acetobutylicum,an important and representative indus-trial strain.Firstly,the mutS/L operon essential for methyl-directed mismatch repair(MMR)activity was inactivated from the genome of C.acetobutylicum to generate hy-permutable cells with over 250-fold increased mutation rates.Secondly,a proofreading control system carrying an inducibly expressed mutS/L operon was constructed.The hypermutable cells and the proofreading control system were integrated to form a controllable hypermut-able system SMBMutC,of which the mutation rates can be regulated by the concentration of anhydrotetracycline(aTc).Duplication of the miniPthl-tetR module of the proof-reading control system further signifi cantly expanded the regulatory space of the mutation rates,demonstrating hypermutable Clostridium cells with controllable mutation rates are generated.The developed C.acetobutylicum strain SMBMutC2 showed higher survival capacities than the control strain facing butanol-stress,indicating greatly increased evolvability and adaptability of the controllable hypermutable cells under environmental challenges. | Guodong Luan Zhen Cai Fuyu Gong Hongjun Dong Zhao Lin Yanping Zhang Yin Li | 2013 | Protein & Cell2013,4,11: | 4 |

| 10 | Synthetic biology for CO2 fixation显示文摘Recycling of carbon dioxide(CO_2) into fuels and chemicals is a potential approach to reduce CO_2 emission and fossil-fuel consumption. Autotrophic microbes can utilize energy from light, hydrogen, or sulfur to assimilate atmospheric CO_2 into organic compounds at ambient temperature and pressure. This provides a feasible way for biological production of fuels and chemicals from CO_2 under normal conditions. Recently great progress has been made in this research area, and dozens of CO_2-derived fuels and chemicals have been reported to be synthesized by autotrophic microbes. This is accompanied by investigations into natural CO_2-fixation pathways and the rapid development of new technologies in synthetic biology. This review first summarizes the six natural CO_2-fixation pathways reported to date, followed by an overview of recent progress in the design and engineering of CO_2-fixation pathways as well as energy supply patterns using the concept and tools of synthetic biology. | 11 | Bloodletting at Jing-well points decreases interstitial fluid flow in the thalamus of rats显示文摘OBJECTIVE: To investigate the changes in the neuronal microenvironment of the middle cerebral artery(MCA) territory induced by Jing-well points bloodletting acupuncture(WPBA) and to explore the neuroprotective mechanism of WPBA in stroke.METHODS: Adult male Sprague Dawley(n = 32)rats were randomly divided into four groups of eight animals each: WPBA-thalamus group(WT),WPBA-caudate nucleus group(WC), sham-control thalamus group(ST) and sham-control caudate nucleus group(SC). Animals in the WT and WC groups received 2 μL of the extracellular tracer gadolinium-diethylene triamine pentaacetic acid(Gd-DTPA)injected into the thalamus or caudate nucleus, respectively, and 12 Jing-well points in the distal ends of the rats' digits were used for WPBA. Although 2 μL of Gd-DTPA was injected into the thalamus or caudate nucleus, respectively, for animals in the two sham groups(ST and SC), no acupuncture or bloodletting was performed. Brain extracellular space and interstitial fluid flow parameters were measured using Gd-DTPA-enhanced magnetic resonance imaging.RESULTS: The brain interstitial fluid flow speed was decreased in the thalamus after WPBA, with a significantly lower Gd-DTPA clearance rate and longer half-life of Gd-DTPA in the thalamus of treated rats than those in sham-control rats [WPBA-treated rats' clearance rate,(7.47 ± 3.15) × 10^(-5)/s(P = 0.009);half-life,(1.52 ± 0.13) h, P = 0.000]. By contrast, no significant changes in brain extracellular space and interstitial fluid flow parameters were detected in the caudate nucleus after WPBA(P = 0.649). | 12 | Keshan disease and mitochondrial cardiomyopathy显示文摘Keshan disease (KD) is a potentially fatal form of cardiomyopathy (disease of the heart muscle) endemic in certain areas of China. From 1984 to 1986, a national comprehensive scientific investigation on KD in Chuxiong region of Yunnan Province in the southwest China was conducted. The investigation team was composed of epidemiologists, clinic doctors, pathologists, biochemists, biophysicists and specialists in ecological environment. Results of pathological, biochemical and biophysical as well as clinical studies showed: an obvious increase of enlarged and swollen mito- chondria with distended crista membranes in myocardium from patients with KD; significant reduc- tions in the activity of oxidative phosphorylation (succinate dehydrogenase, cytochrome oxidase, succinate oxidase, H+-ATPase) of affected mitochondria; decrease in CoQ, cardiolipin, Se and GSHPx activity, while obvious increase in the Ca2+ content. So, it was suggested that mitochondria are the predominant target of the pathogenic factors of KD. Before Chuxiong KD survey only a few cases of mitochondrial cardiomyopathy were studied. During the multidisciplinary scientific investiga- tion on KD in Chuxiong a large amount of samples from KD cases and the positive controls were examined. On the basis of the results obtained it was suggested that KD might be classified as a “Mitochondrial Cardiomyopathy” endemic in China. This is one of the achievements in the three years’ survey in Chuxiong and is valuable not only to the deeper understanding of pathogenic mechanism of KD but also to the study of mitochondrial cardiomyopathy in general. Keshan disease is not a genetic disease, but is closely related to the malnutrition (especially mi- croelement Se deficiency). KD occurs along a low Se belt, and Se supplementation has been effective in prevention of such disease. The incidence of KD has sharply decreased along with the steady raise of living standard and realization of preventive measures. At present, patients of KD are very sparse. In recent years the research on the non-KD mitochondrial cardiomyopathy has progressed rapidly.
