1Fault plane parameters of Sanhe-Pinggu M8 earthquake in 1679 determined using present-day small earthquakes显示文摘The great Sanhe-Pinggu M8 earthquake occurred in 1679 was the largest surface rupture event recorded in history in the northern part of North China plain. This study determines the fault geometry of this earthquake by inverting seismological data of present-day moderate-small earthquakes in the focal area. We relocated those earthquakes with the double-difference method.Based on the assumption that clustered small earthquakes often occur in the vicinity of fault plane of large earthquake, and referring to the morphology of the long axis of the isoseismal line obtained by the predecessors, we selected a strip-shaped zone from the relocated earthquake catalog in the period from 1980 to 2009 to invert fault plane parameters of this earthquake. The inversion results are as follows: the strike is 38.23°, the dip angle is 82.54°,the slip angle is-156.08°, the fault length is about 80 km,the lower-boundary depth is about 23 km and the buried depth of upper boundary is about 3 km. This shows that the seismogenic fault is a NNE-trending normal dip-slip fault,southeast wall downward and northwest wall uplift, with the right-lateral strike-slip component. 2Kinematics and trajectory planning of a cucumber harvesting robot manipulator显示文摘In order to reduce cucumber harvesting cost and improve economic benefits,a cucumber harvesting robot was developed.The cucumber harvesting robot consists of a vehicle,a 4-DOF articulated manipulator,an end-effector,an upper monitor,a vision system and four DC servo drive systems.The Kinematics of the cucumber harvesting robot manipulator was constructed using D-H coordinate frame model.And the inverse kinematics which provides a foundation for trajectory planning has been solved with inverse transform technique.The cycloidal motion,which has properties of continuity and zero velocity and acceleration at the ports of the bounded interval,was adopted as a feasible approach to plan trajectory in joint space of the cucumber harvesting robot manipulator.Moreover,hardware and software based on CAN-bus communication between the upper monitor and the joint controllers have been designed.Experimental results show that the upper monitor communicates with the four joint controllers efficiently by CAN-bus,and the integrated errors of four joint angles do not exceed four degrees.Probable factors resulting in the errors were analyzed and the corresponding solutions for improving precision are proposed.Zhang Libin Wang Yan Yang Qinghua Bao Guanjun Gao Feng Xun Yi 2009International Journal of Agricultural and Biological Engineering2009,2,1:7
3Pharmacological Activation of RXR-a Promotes Hematoma Absorption via a PPAR-y-dependent Pathway After Intracerebral Hemorrhage显示文摘Endogenously eliminating the hematoma is a favorable strategy in addressing intracerebral hemorrhage(ICH).This study sought to determine the role of retinoid X receptor-ot(RXR-a)in the context of hematoma absorption after ICH.Our results showed that pharmacologically activating RXR-a with bexarotene significantly accelerated hematoma clearance and alleviated neurological dysfunction after ICH.RXR-ot was expressed in microglia/macro-phages,neurons,and astrocytes.Mechanistically,bexarotene promoted the nuclear translocation of RXR-a and PPAR-y,as well as reducing neuroinflammation by modulating microglia/macrophage reprograming from the Ml into the M2 phenotype.Furthermore,all the beneficial effects of RXR-a in ICH were reversed by the PPAR-y inhibitor GW9662.In conclusion,the pharmacological activation of RXR-a confers robust neuroprotection against ICH by accelerating hematoma clearance and repolarizing microglia/macrophages towards the M2 phenotype through PPAR-y-related mechanisms.Our data support the notion that RXR-ot might be a promising therapeutic target for ICH.Chaoran Xu Huaijun Chen Shengjun Zhou Chenjun Sun Xiaolong Xia Yucong Peng Jianfeng Zhuang Xiongjie Fu Hanhai Zeng Hang Zhou Yang Cao Qian Yu Yin Li Libin Hu Guoyang Zhou Feng Yan Gao Chen Jianru Li 2021Neuroscience Bulletin2021,37,10:4
4Domesticated cynomolgus monkey embryonic stem cells allow the generation of neonatal interspecies chimeric pigs显示文摘Blastocyst complementation by pluripotent stem cell(PSC)injection is believed to be the most promising method to generate xenogeneic organs.However,ethical issues prevent the study of human chimeras in the late embryonic stage of development.Primate embryonic stem cells(ESCs),which have similar pluripotency to human ESCs,are a good model for studying interspecies chimerism and organ generation.However,whether primate ESCs can be used in xenogenous grafts remains unclear.In this study,we evaluated the chimeric ability of cynomolgus monkey(Macaca fascicularis)ESCs(cmESCs)in pigs,which are excellent hosts because of their many similarities to humans.We report an optimized culture medium that enhanced the anti-apoptotic ability of cmESCs and improved the development of chimeric embryos,in which domesticated cmESCs(D-ESCs)injected into pig blastocysts differentiated into cells of all three germ layers.In addition,we obtained two neonatal interspecies chimeras,in which we observed tissue-specific D-ESC differentiation.Taken together,the results demonstrate the capability of D-ESCs to integrate and differentiate into functional cells in a porcine model,with a chimeric ratio of 0.001-0.0001 in different neonate tissues.We believe this work will facilitate future developments in xenogeneic organogenesis,bringing us one step closer to producing tissue-specific functional cells and organs in a large animal model through interspecies blastocyst complementation.Rui Fu Dawei Yu Jilong Ren Chongyang Li Jing Wang Guihai Feng Xuepeng Wang Haifeng Wan Tianda Li Libin Wang Ying Zhang Tang Hai Wei Li Qi Zhou 2020Protein & Cell2020,11,2:4
5Overview of research on agricultural robots in China显示文摘As the representation of new concept agricultural machinery,agricultural robots possess great advantages of improving agricultural productivity,enhancing production environment and solving the problem of labor shortage.Therefore,the strategy for application of agricultural robots and precision agriculture to improve the intelligence and information level of agriculture is the inevitable trend for China’s agriculture in the twentieth century.Based on the developmental status of agricultural robots in China,the agricultural robots are categorized and the performances,structures and characteristics of various agricultural robots such as vegetable grafting robots,transplanting robots,spraying robots,mowing robots,harvesting robots,grading and detecting robots are amply introduced.It can be seen that vegetable grafting robots,spraying robots,harvesting robots,grading and detecting robots have already been put into production while others are still at experimental stage.At present,there are several problems such as low popularization,great limitations,high cost and low intelligence,which greatly restrict the development of agricultural robots in China.Thus,open agricultural robot system with good expansibility,generality and flexibility should be developed and adopted to decrease its cost and shorten developing cycle.The mechanical structure of robots should also be designed as simply as possible.Finally,multi-robot system would become another important development direction of agricultural robots in the future.Zhang Libin Yang Qinghua Bao Guanjun Wang Yan Qi Liyong Gao Feng Xu Fang 2008International Journal of Agricultural and Biological Engineering2008,1,1:4
6DC-CIK联合化疗治疗非小细胞肺癌的临床疗效评价(英文)显示文摘Objective: The aim of the study was to evaluate the safety and therapeutic effects of autologous dendritic cells co-cultured with cytokine-induced killer cells (DC-CIK) combined with chemotherapy in advanced non-small cell lung cancer (NSCLC) patients. Methods: Fifty patients with advanced NSCLC (stages III to IV), who had received therapies in our Center (Department of Biotherapy, Affiliated to Cancer Hospital of Shanxi Medical University, Taiyuan, China) from August 2008 to January 2010, were treated by DC-CIK + chemotherapy as the combined treatment group; fifty advanced NSCLC patients treated with chemotherapy at the same time served as controls. The immunologic function, short-term therapeutic effects, the 1-year survival rate, the life quality, the chemotherapy side effects were compared between the two groups, the safety and therapeutic effects of DC-CIK cells therapy were observed too. Results: There was no obvious change of subsets of T cells in peripheral blood before and after therapy in DC-CIK + chemotherapy group, and IFN-γ was improved after therapy in this group (P < 0.05); in chemotherapy alone group, the ratios of CD3+CD4+, CD3+CD8+, CD3-CD56+ cells and the secretion of IL-2, TNF-α decreased significantly after therapy (P < 0.05); the ratios of CD3+CD8+, CD3+CD56+ were improved after cell culture (P < 0.05). The disease control rate (DCR) of DC-CIK + chemotherapy group was higher than that in the chemotherapy alone group (78.0% vs 56.0%, P < 0.05); the 1-year survival rates of DC-CIK + chemotherapy group and chemotherapy alone group were 50% and 44% respectively, had no significant difference. Compared with chemotherapy alone group, the occurrence of chemotherapy side effects (including bone marrow suppression, nausea and vomiting, peripheral nerve toxicity) was less in the DC-CIK + chemotherapy group (P < 0.05). The physical and appetite were better in DC-CIK + chemotherapy group after therapy. 8Occurrence and Epidemic Regularity of Wheat Stripe Rust in Nanchong City of Sichuan Province显示文摘In order to realize monitoring and early warning and comprehensive management of wheat stripe rust and to reduce its occurrence in Nanchong City,the occurrence and epidemic regularity of wheat stripe rust in Nanchong was studied by system monitoring and general survey,resistance identification,physiological race monitoring and meteorological data analysis. The initial occurrence location and spreading pathway of Puccinia striiformis f. sp. tritici( Pst) were first verified;there were two infection peaks of wheat stripe rust in Nanchong and one to three epidemic peaks in fields,in which the occurrence area of the first epidemic peak played a pivotal role in disease prevalence in that year; the cumulative occurrence area in late January was positively correlated with annual occurrence area,with the correlation coefficient of 0. 769; the prediction model for infected field rate,diseased plant rate and annual occurrence area was established. The internal reason for heavy occurrence and prevalence of wheat stripe rust in Nanchong was the decline or loss of wheat resistance against stripe rust,as well as the appearance of physiological races of Pst,which later became dominant races. Large fluctuation of temperature in warm winter and spring and more fog and dew days were external reasons responsible for prevalence of stripe rust. 9A joint call for actions to advance taxonomy in China显示文摘Taxonomy plays an important role in understanding the origin, evolution, and ecological functionality of biodiversity. There are large number of unknown species yet to be described by taxonomists, which together with their ecosystem services cannot be effectively protected prior to description. Despite this, taxonomy has been increasingly underrated insufficient funds and permanent positions to retain young talents. Further, the impact factordriven evaluation systems in China exacerbate this downward trend, so alternative evaluation metrics are urgently necessary. When the current generation of outstanding taxonomists retires,there will be too few remaining taxonomists left to train the next generation. 10Identification of Global DNA Methylation Signatures in Glioblastoma-Derived Cancer Stem Cells显示文摘Glioblastoma(GBM)is the most common and most aggressive primary brain tumor in adults.The existence of a small population of stem-like tumor cells that efficiently propagate tumors and resist cytotoxic therapy is one proposed mechanism leading to the resilient behavior of tumor cells and poor prognosis.In this study,we performed an in-depth analysis of the DNA methylation landscape in GBMderived cancer stem cells(GSCs).Parallel comparisons of primary tumors and GSC lines derived from these tumors with normal controls(a neural stem cell(NSC)line and normal brain tissue)identified groups of hyper- and hypomethylated genes that display a trend of either increasing or decreasing methylation levels in the order of controls,primary GBMs,and their counterpart GSC lines,respectively.Interestingly,concurrent promoter hypermethylation and gene body hypomethylation were observed in a subset of genes including MGMT,AJAP1 and PTPRN2.These unique DNA methylation signatures were also found in primary GBM-derived xenograft tumors indicating that they are not tissue culture-related epigenetic changes.Integration of GSC-specific epigenetic signatures with gene expression analysis further identified candidate tumor suppressor genes that are frequently down-regulated in GBMs such as SPINT2,NEFM and PENK.Forced re-expression of SPINT2 reduced glioma cell proliferative capacity,anchorage independent growth,cell motility,and tumor sphere formation in vitro.The results from this study demonstrate that GSCs possess unique epigenetic signatures that may play important roles in the pathogenesis of GBM.Eun-Joon Lee Prakash Rath Jimei Liu Dungsung Ryu Lirong Pei Satish K.Noonepalle Austin Y.Shull Qi Feng N.Scott Litofsky Douglas C.Miller Douglas C.Anthony Mark D.Kirk John Laterra Libin Deng Hong-Bo Xin Xinguo Wang Jeong-Hyeon Choi Huidong Shi 2015Journal of Genetics and Genomics2015,42,7:1
11PKD3 promotes metastasis and growth of oral squamous cell carcinoma through positive feedback regulation with PD-L1 and activation of ERK-STAT1/3-EMT signalling显示文摘Oral squamous cell carcinoma(OSCC)has a high incidence of metastasis.Tumour immunotherapy targeting PD-L1 or PD-1 has been revolutionary;however,only a few patients with OSCC respond to this treatment.Therefore,it is essential to gain insights into the molecular mechanisms underlying the growth and metastasis of OSCC.In this study,we analysed the expression levels of protein kinase D3(PKD3)and PD-L1 and their correlation with the expression of mesenchymal and epithelial markers.We found that the expression of PKD3 and PD-L1 in OSCC cells and tissues was significantly increased,which correlated positively with that of mesenchymal markers but negatively with that of epithelial markers.Silencing PKD3 significantly inhibited the growth,metastasis and invasion of OSCC cells,while its overexpression promoted these processes.Our further analyses revealed that there was positive feedback regulation between PKD3 and PD-L1,which could drive EMT of OSCC cells via the ERK/STAT1/3 pathway,thereby promoting tumour growth and metastasis.Furthermore,silencing PKD3 significantly inhibited the expression of PD-L1,and lymph node metastasis of OSCC was investigated with a mouse footpad xenograft model.Thus,our findings provide a theoretical basis for targeting PKD3 as an alternative method to block EMT for regulating PD-L1 expression and inhibiting OSCC growth and metastasis.Bomiao Cui Jiao Chen Min Luo Yiying Liu Hongli Chen Die Lü Liwei Wang Yingzhu Kang Yun Feng Libin Huang Ping Zhang 2021International Journal of Oral Science2021,13,1:1
12Effect of moxibustion on autophagy and the inflammatory response of synovial cells in rheumatoid arthritis model rats显示文摘OBJECTIVE:To investigate the effect of moxibustion on synovitis and the autophagy of synoviocytes in rheumatoid arthritis(RA).METHODS:Forty Sprague-Dawley rats were randomly divided into a normal group,model group,moxibustion group,cigarette moxibustion group,and medicine group,with eight rats included in each group.The RA model was established by subcutaneous injection of complete Freund's adjuvant into the left posterior toe.Rats in the model group were not interfered with.In the moxibustion group,rats were treated by moxibustion,where a 1-cm diameter moxa stick was applied at the left Zusanli(ST 36)point.The distance of the moxa stick to the skin was 2 cm and moxibustion was completed for 20 min daily for 15 d total.In the cigarette moxibustion group,the moxa stick was replaced by a common cigarette.In the medicine group,rats were treated with a tripterygium glycoside suspension(8 mg/kg)once a day for 15 d total.In each group,the left hind limb toe volume was measured with a toe volume meter;the synovial cells were observed by hematoxylin and eosin staining;the interleukin(IL)-4,IL-6,IL-10,IL-1β,IL-23,IL-17,and tumor necrosis factor(TNF)-αlevels in serum were measured by enzyme-linked immunosorbent assay;the erythrocyte sedimentation rate(ESR)were detected by Westergren sedimentation rate testing;the C-reactive protein(CRP)and rheumatoid factor(RF)levels in serum were detected by rate nephelometry;the expression levels of ULK1,autophagy-associated protein(Atg)3,Atg5,and Atg12 messenger RNA(m RNA)in synovium were detected by real time-quantitative polymerase chain reaction(RT-q PCR);and the protein expression levels of phosphatidylinositol-3-kinase(PI3K),protein kinase B(Akt),mammalian target of rapamycin(mTOR),LC3-Ⅱ,beclin-1,phosphorylated-PI3K(p-PI3K),p-Akt,p-mTOR in synovium were detected by Western blotting.RESULTS:Among the RA model rats,joint swelling,an inflammatory reaction,and the proliferation of synovial tissue were obvious and the signal of the PI3K/Akt/mTOR pathway was active,while autophagy was inhibited.Moxibustion at Zusanli(ST36)or intragastric administration of Tripterygium wilfordii glycosides could alleviate the inflammatory reaction of RA rats;relieve the swelling of the toes;downregulate the levels of ESR,CRF,RF;lower the levels of IL-6,IL-1β,TNF-α,and IL-17;and increase the IL-4 and IL-10.At the same time,the m RNA expression levels of ULK1,Atg3,Atg5,and Atg12 and those of LC3-Ⅱand beclin-1 were increased,while the PI3K,Akt,mTOR,p-PI3K,p-Akt,p-mTOR were decreased.Cigarette moxibustion did not significantly reduce the swelling of the toe joint in RA rats,and was not as good as that of moxibustion or Tripterygium wilfordii polyglycosides in the effects of inflammation relief and the influences of the levels of ESR,CRF,RF.While cigarette moxibustion has a weak effect to affect the expression of corresponding molecules in autophages and the expression level of the autophagy biomaker in synovial tissue.Moxibustion and tripterygium glycosides can significantly reduce the joint swelling,relieve synovitis and synovial hyperplasia,and inhibit the PI3K/Akt/mTOR signaling pathway to increase autophagy in a manner superior to cigarette moxibustion.CONCLUSION:Moxibustion can limit the proliferation of synoviocytes in RA rats by inhibiting the PI3K/Akt/mTOR signaling pathway,promoting autophagy,effectively reducing synovitis,and alleviating joint swelling.HAO Feng WANG Qiang LIU Lei WU Libin CAI Ronglin SANG Jiajia HU Jun WANG Jie YU Qing HE Lu SHEN Yingchao MIAO Yiming HU Ling WU Zijian 2022Journal of Traditional Chinese Medicine2022,42,1:1
13The ties of brotherhood between japonica and indica rice for regional adaptation显示文摘Selection of beneficial genomic variants was crucial for regional adaptation of crops during domestication,but the underlying genomic basis remains largely unexplored.Here we report a genome-wide selective-sweep analysis of 655 japonica and 1,205 indica accessions selected from 2,673 landraces through principal component analysis to identify 5,636 non-synonymous single nucleotide polymorphisms(SNPs)fixed in at least one subspecies.We classified these SNPs into three groups,jiS(japonica-and indica-selected),jS(japonica-selected only),and iS(indica-selected only),and documented evidence for selection acting on these groups,their relation to yield-related traits,such as heading date,and their practical value in cropping area prediction.We also demonstrated the role of a jiS-SNP-containing gene in temperature adaptability.Our study informs genes underpinning adaptation that may shape Green Super Rice and proposes a time-saving,cost-reducing selection strategy of genomic breeding,sweep-SNP-guided selection,for developing regionally-adapted heterosis.Man Wang Jiehu Chen Feng Zhou Jianming Yuan Libin Chen Rongling Wu Yaoguang Liu Qunyu Zhang 2022Science China(Life Sciences)2022,65,7:1

15MeCP2 deficiency promotes cell reprogramming by stimulating IGF1/AKT/mTOR signaling and activating ribosomal protein-mediated ceil cycle gene translation显示文摘The generation of induced pturipotent stem cells (iPSCs)offers a great opportunity in research and regenerative medicine.The current poor efficiency and incomplete mechanistic understanding of the reprogramming process hamper the clinical application of iPSCs. MeCP2 connects histone modification and DNA methyiation,which are key changes of somatic cell reprogramming.However,the rote of MeCP2 in celt reprogramming has not been examined.In this study,we found that MeCP2 deficiency enhanced reprogramming efficiency and stimulated cell proliferation through regulating cell cycle protein expression in the earLy stage of reprogramming.MeCP2 deficiency enhanced the expression of ribosomal protein genes,thereby enhancing reprogramming efficiency through promoting the translation of ceLl cycle genes.In the end,MeCP2 deficiency stimulated IGF1/AKT/mTOR signaling and activated ribosomal protein gene expression.Taken together,our data indicate that MeCP2 deficiency promoted cell reprogramming through stimulating IGF1/AKT/mTOR signaling and activating ribosomal protein-mediated cell cycle gene translation in the early stage of reprogramming.Wei Zhang Guihai Feng Libin Wang Fei Teng Liu Wang Wei Li Ying Zhang Qi Zhou 2018Journal of Molecular Cell Biology2018,10,6:0
1640 Hz light flickering promotes sleep through cortical adenosine signaling显示文摘Flickering light stimulation has emerged as a promising non-invasive neuromodulation strategy to alleviate neuropsychiatric disorders.However,the lack of a neurochemical underpinning has hampered its therapeutic development.Here,we demonstrate that light flickering triggered an immediate and sustained increase(up to 3 h after flickering)in extracellular adenosine levels in the primary visual cortex(V1)and other brain regions,as a function of light frequency and intensity,with maximal effects observed at 40 Hz frequency and 4000 lux.We uncovered cortical(glutamatergic and GABAergic)neurons,rather than astrocytes,as the cellular source,the intracellular adenosine generation from AMPK-associated energy metabolism pathways(but not SAM-transmethylation or salvage purine pathways),and adenosine efflux mediated by equilibrative nucleoside transporter-2(ENT2)as the molecular pathway responsible for extracellular adenosine generation.Importantly,40 Hz(but not 20 and 80 Hz)light flickering for 30 min enhanced non-rapid eye movement(non-REM)and REM sleep for 2–3 h in mice.This somnogenic effect was abolished by ablation of V1(but not superior colliculus)neurons and by genetic deletion of the gene encoding ENT2(but not ENT1),but recaptured by chemogenetic inhibition of V1 neurons and by focal infusion of adenosine into V1 in a dose-dependent manner.Lastly,40 Hz light flickering for 30 min also promoted sleep in children with insomnia by decreasing sleep onset latency,increasing total sleep time,and reducing waking after sleep onset.Collectively,our findings establish the ENT2-mediated adenosine signaling in V1 as the neurochemical basis for 40 Hz flickering-induced sleep and unravel a novel and non-invasive treatment for insomnia,a condition that affects 20%of the world population.Xuzhao Zhou Yan He Tao Xu Zhaofa Wu Wei Guo Xi Xu Yuntao Liu Yi Zhang Huiping Shang Libin Huang Zhimo Yao Zewen Li Lingya Su Zhihui Li Tao Feng Shaomin Zhang Olivia Monteiro Rodrigo A.Cunha Zhi-Li Huang Kang Zhang Yulong Li Xiahong Cai Jia Qu Jiang-Fan Chen 2024Cell Research2024,34,3:0

20Differential gene expression in the body wall of the sea cucumber(Apostichopus japonicus)under strong lighting and dark conditions显示文摘Sea cucumber, Apostichopus japonicus is very sensitive to light changes. It is important to study the influence of light on the molecular response of A. japonicus. In this study, RNA-seq provided a general overview of the gene expression profiles of the body walls of A. japonicus exposed to strong light('light'), normal light('control') and fully dark('dark') environment. In the comparisons of 'control' vs. 'dark', 'control' vs. 'light' and 'dark' vs.'light', 1 161, 113 and 1 705 differentially expressed genes(DEGs) were identified following the criteria of|log_2 ratio|≥1 and FDR≤0.001, respectively. Gene ontology analysis showed that 'cellular process' and 'binding'enriched the most DEGs in the category of 'biological process' and 'molecular function', while 'cell' and 'cell part' enriched the most DEGs in the category of 'cellular component'. And the DEGs were mapped to 214, 41 and229 pathways in the Kyoto Encyclopedia of Genes and Genomes database, and 51, 2 and 57 pathways were significantly enriched, respectively.
