2Gene editing in plants:progress and challenges显示文摘The clustered regularly interspaced short palindromic repeat(CRISPR)-associated protein 9(Cas9)genome editing system is a powerful tool for targeted gene modifications in a wide range of species,including plants.Over the last few years,this system has revolutionized the way scientists perform genetic studies and crop breeding,due to its simplicity,flexibility,consistency and high efficiency.Considerable progress has been made in optimizing CRISPR/Cas9 systems in plants,particularly for targeted gene mutagenesis.However,there are still a number of important challenges ahead,including methods for the efficient delivery of CRISPR and other editing tools to most plants,and more effective strategies for sequence knock-ins and replacements.We provide our viewpoint on the goals,potential concerns and future challenges for the development and application of plant genome editing tools.Yanfei Mao Jose Ramon Botella Yaoguang Liu Jian-Kang Zhu 2019National Science Review2019,6,3:17

4MAPPING OF THE RICE (ORYZA SATIVA L.) THERMO-SENSITIVE GENIC MALE STERILE GENE TMS5 WITH EST AND SSR MARKERS显示文摘With the cDNA suppression subtraction hybridization method, a spikelet-specific cDNA library was constructed that expressed at meiosis stage in rice. A total of 121 cDNA fragments were selected from the library and used as EST (expressed se-quence tags) markers to detect the polymorphism between Annong N, a normal fertile Indica rice line and Annong S-1, its spontaneous mutant with thermo-sensitive genic male sterility, using the RFLP (restriction fragment length polymorphism) technique. HN57, one of the EST probes, could detect poly-morphism between them. The results of segregation analysis with the F2 population developed from An-nong S-1 and Annong N indicate that HN57 co-seg- regates with the thermo-sensitive genic male-sterility controlled by tms5, the recessive gene in Annong S-1. This marker is located on the 31.2-cM region of the chromosome 2 of RGP (rice genome research pro-gram) genetic map. To further determine the location of tms5, 80 SSR (simple sequence repeat) markers around this region were developed, and 12 of them were polymorphic. 5Low-dose real-time X-ray imaging with nontoxic double perovskite scintillators显示文摘X-rays are widely used in probing inside information nondestructively,enabling broad applications in the medical radiography and electronic industries.X-ray imaging based on emerging lead halide perovskite scintillators has received extensive attention recently.However,the strong self-absorption,relatively low light yield and lead toxicity of these perovskites restrict their practical applications.Here,we report a series of nontoxic double-perovskite scintillators of Cs_(2)Ag_(0.6)Na_(0.4)In_(1-y)Bi_(y)Cl_(6).By controlling the content of the heavy atom Bi^(3+),the X-ray absorption coefficient,radiative emission efficiency,light yield and light decay were manipulated to maximise the scintillator performance.A light yield of up to 39,000±7000 photons/MeV for Cs_(2)Ag_(0.6)Na_(0.4)In_(0.85)Bi_(0.15)Cl_(6) was obtained,which is much higher than that for the previously reported lead halide perovskite colloidal CsPbBr_(3)(21,000 photons/MeV).The large Stokes shift between the radioluminescence(RL)and absorption spectra benefiting from self-trapped excitons(STEs)led to a negligible selfabsorption effect.Given the high light output and fast light decay of this scintillator,static X-ray imaging was attained under an extremely low dose of ∼1μGy_(air),and dynamic X-ray imaging of finger bending without a ghosting effect was demonstrated under a low-dose rate of 47.2μGy_(air) s^(−1).After thermal treatment at 85℃ for 50 h followed by X-ray irradiation for 50 h in ambient air,the scintillator performance in terms of the RL intensity and X-ray image quality remained almost unchanged.Our results shed light on exploring highly competitive scintillators beyond the scope of lead halide perovskites,not only for avoiding toxicity but also for better performance.Wenjuan Zhu Wenbo Ma Yirong Su Zeng Chen Xinya Chen Yaoguang Ma Lizhong Bai Wenge Xiao Tianyu Liu Haiming Zhu Xiaofeng Liu Huafeng Liu Xu Liu Yang(Michael)Yang 2020Light(Science & Applications)2020,9,1:9

10Dynamic changes of peptidome and release of polysaccharide in sea cucumber(Apostichopus japonicus) hydrolysates depending on enzymatic hydrolysis approaches显示文摘Enzymatic hydrolysis has been widely used to produce bioactive hydrolysates from sea cucumber body wall.Here,inspired by the clarification of Apostichopus japonicus genome,we investigated the enzymatic hydrolysis of sea cucumber body wall by using the omics strategy.Shared proteins,including major yolk proteins,collagens,extracellular matrix glycoproteins and muscle proteins,were released from the body wall by different hydrolysis condition.A portfolio of 216 shared peptides were detected in the peptidome by papain with different hydrolysis time,while 32 shared peptides were detected in the peptidome by differing proteases.Unshared peptides and the relative abundance distribution profiles of shared peptides changed depending on hydrolysis approaches,indicating dynamic changes of peptidome during hydrolysis.Moreover,release of sulfated fucan and fucosylated chondroitin sulfate changed with the hydrolysis condition.The monitoring of dynamic enzymatic hydrolysis process at a molecular scale would contribute to production and quality control of sea cucumber hydrolysates.Yanchao Wang Yan Song Yaoguang Chang Yanyan Liu Guangning Chen Changhu Xue 2022Food Science and Human Wellness2022,11,5:2

15Highly oriented MAPbI3 crystals for efficient hole-conductor-free printable mesoscopic perovskite solar cells显示文摘Highly crystalline perovskite films with large grains and few grain boundaries are conducive for efficient and stable perovskite solar cells.Current methods for preparing perovskite films are mostly based on a fast crystallization process,with rapid nucleation and insufficient growth.In this study,MAPbI3 perovskite with inhibited nucleation and promoted growth in the TiO_(2)/ZrO_(2)/carbon triple mesoscopic scaffold was crystallized by modulating the precursor and the crystallization process.N-methylformamide showed high solubility for both methylammonium iodide and Pbl2 and hampered the formation of large colloids in the MAPbI3 precursor solution.Furthermore,methylammonium chloride was added to reduce large colloids,which are a possible source of nucleation sites.During the crystallization of MAPbI3,the solvent was removed at a slow controlled speed,to avoid rapid nucleation and provide sufficient time for crystal growth.As a result,highly oriented MAPbI3 crystals with suppressed non-radiative recombination and promoted charge transport were obtained in the triple mesoscopic layer with disordered pores.The corresponding hole-conductor-free,printable mesoscopic perovskite solar cells exhibited a highest power conversion efficiency of 18.82%.The device also exhibited promising long-term operational sta-bility of 1000 h under continuous illumination at maximum power point at 55±5°C and damp-heat stability of 1340 h aging at 85°C as well as 85%relative humidity.Shuang Liu Deyi Zhang Yusong Sheng Weihua Zhang Zhaotong Qin Minchao Qin Sheng Li Yifan Wang Chenxu Gao Qifei Wang Yue Ming Chao Liu Kai Yang Qingyi Huang Jianhang Qi Qiaojiao Gao Kai Chen Yue Hu Yaoguang Rong Xinhui Lu Anyi Mei Hongwei Han 2022Fundamental Research2022,2,2:1
16The ties of brotherhood between japonica and indica rice for regional adaptation显示文摘Selection of beneficial genomic variants was crucial for regional adaptation of crops during domestication,but the underlying genomic basis remains largely unexplored.Here we report a genome-wide selective-sweep analysis of 655 japonica and 1,205 indica accessions selected from 2,673 landraces through principal component analysis to identify 5,636 non-synonymous single nucleotide polymorphisms(SNPs)fixed in at least one subspecies.We classified these SNPs into three groups,jiS(japonica-and indica-selected),jS(japonica-selected only),and iS(indica-selected only),and documented evidence for selection acting on these groups,their relation to yield-related traits,such as heading date,and their practical value in cropping area prediction.We also demonstrated the role of a jiS-SNP-containing gene in temperature adaptability.Our study informs genes underpinning adaptation that may shape Green Super Rice and proposes a time-saving,cost-reducing selection strategy of genomic breeding,sweep-SNP-guided selection,for developing regionally-adapted heterosis.Man Wang Jiehu Chen Feng Zhou Jianming Yuan Libin Chen Rongling Wu Yaoguang Liu Qunyu Zhang 2022Science China(Life Sciences)2022,65,7:1
