| 1 | Use of testicular sperm in couples with SCSA-defined high sperm DNA fragmentation and failed intracytoplasmic sperm injection using ejaculated sperm显示文摘Sperm DNA fragmentation(SDF)has been linked with male infertility,and previous studies suggest that SDF can have negative influence on pregnancy outcomes with assisted reproduction.We performed a retrospective review of consecutive couples with a high SDF level that had intracytoplasmic sperm injection(ICSI)using testicular sperm(T-ICSI).We compared the T-ICSI outcomes to that of two control groups:87 couples with failed first ICSI cycle and who had a second ICSI cycle using ejaculated sperm(Ej-ICSI),and 48 consecutive couples with high sperm chromatin structure assay(SCSA)-defined SDF(>15%)that underwent an ICSI cycle using ejaculated sperm after one or more failed ICSI cycles(Ej-ICSI-high SDF).The mean number of oocytes that were retrieved and the total number of embryos were not different among the three groups.The mean number of transferred embryos in the T-ICSI group was higher than the Ej-ICSI group but not significantly different than the Ej-ICSI-high SDF group(1.4,1.2,and 1.3,respectively,P<0.05).Clinical pregnancy rate in the T-ICSI group was not significantly different than the Ej-ICSI and Ej-ICSI-high SDF groups(48.6%,48.2%,and 38.7%,respectively,P>0.05).No significant difference was found in live birth rate when comparing T-ICSI to Ej-ICSI and Ej-ICSI-high SDF groups.The results suggest that pregnancy outcomes and live birth rates with T-ICSI are not significantly superior to Ej-ICSI in patients with an elevated SCSA-defined sperm DNA fragmentation and prior ICSI failure(s). | Mohannad Alharbi Fadl Hamouche Simon Phillips Jacques Isaac Kadoch Armand Zini | 2020 | Asian Journal of Andrology2020,22,4: | 4 |
| 2 | AFM-compatible microfluidic platform for affinity- based capture and nanomechanical characterization of circulating tumor cells显示文摘Circulating tumor cells(CTCs)carried by the patient’s bloodstream are known to lead to the metastatic spread of cancer.It is becoming increasingly clear that an understanding of the nanomechanical characteristics of CTCs,such as elasticity and adhesiveness,represents advancements in tracking and monitoring cancer progression and metastasis.In the present work,we describe a combined microfluidic–atomic force microscopy(AFM)platform that uses antibody–antigen capture to routinely isolate and nanomechanically characterize CTCs present in blood samples from prostate cancer patients.We introduce the reversible assembly of a microfluidic device and apply refined and robust chemistry to covalently bond antibodies onto its glass substrate with high density and the desired orientation.As a result,we show that the device can efficiently capture CTCs from patients with localized and metastatic prostate cancer through anti-EpCAM,anti-PSA,and anti-PSMA antibodies,and it is suitable for AFM measurements of captured intact CTCs.When nanomechanically characterized,CTCs originating from metastatic cancer demonstrate decreased elasticity and increased deformability compared to those originating from localized cancer.While the average adhesion of CTCs to the AFM tip surface remained the same in both the groups,there were fewer multiple adhesion events in metastatic CTCs than there were in their counterparts.The developed platform is simple,robust,and reliable and can be useful in the diagnosis and prognosis of prostate cancer as well as other forms of cancer. | Muhammedin Deliorman Farhad K.Janahi Pavithra Sukumar Ayoub Glia Roaa Alnemari Samar Fadl Weiqiang Chen Mohammad A.Qasaimeh | 2020 | Microsystems & Nanoengineering2020,6,1: | 2 |
| 3 | Determination of optimum tapping date for gum arabic production in South Kordofan State, Sudan显示文摘Acacia senegal, the gum arabic producing tree, is the most important tree species for the livelihood of the people in South Kordofan State, Sudan. The objective of this study was to determine the optimum tapping date for gum arabic production in the study area. A randomized complete block design experiment with three replications was conducted at (Meari) area for two continuous growing seasons 2008/2009 - 2009/2010. The treatments comprised six tapping dates (1 Oct, 15 Oct, 1 Nov, 15 Nov, 1 Dec, and 15 Dec). Results showed highly significant differences (p < 0.001) in gum arabic yield (g\tree) in all pickings and in the total gum yield between the tapping dates. The results also showed that tapping of trees on 15 October and 1 November gave a higher yield compared to the other dates. The highest gum yield of 1086.6 and 661.2 g/tree was recorded on 15 October and 15 November, while the lowest gum yield of 297.9 g/tree was recorded when the trees were tapped on 1 October. The two highest-yield dates of tapping (15 Oct and 1 Nov) are recommended as the best time for tapping for gum arabic production in South Kordofan State.
