| 1 | Anti-inflammatory mechanisms of the novel cytokine interleukin-38 in allergic asthma显示文摘We elucidated the anti-inflammatory mechanisms of IL-38 in allergic asthma.Human bronchial epithelial cells and eosinophils were cocultured upon stimulation with the viral RLR ligand poly(I:C)/LyoVec or infection-related cytokine TNF-αto induce expression of cytokines/chemokines/adhesion molecules.House dust mite(HDM)-induced allergic asthma and humanized allergic asthma NOD/SCID murine models were established to assess anti-inflammatory mechanisms in vivo.IL-38 significantly inhibited induced proinflammatory IL-6,IL-1β,CCL5,and CXCL10 production,and antiviral interferon-βand intercellular adhesion molecule-1 expression in the coculture system.Mass cytometry and RNA-sequencing analysis revealed that IL-38 could antagonize the activation of the intracellular STAT1,STAT3,p38 MAPK,ERK1/2,and NF-κB pathways,and upregulate the expression of the host defense-related gene POU2AF1 and anti-allergic response gene RGS13.Intraperitoneal injection of IL-38 into HDM-induced allergic asthma mice could ameliorate airway hyperreactivity by decreasing the accumulation of eosinophils in the lungs and inhibiting the expression of the Th2-related cytokines IL-4,IL-5,and IL-13 in the bronchoalveolar lavage fluid(BALF)and lung homogenates.Histological examination indicated lung inflammation was alleviated by reductions in cell infiltration and goblet cell hyperplasia,together with reduced Th2,Th17,and innate lymphoid type 2 cell numbers but increased proportions of regulatory T cells in the lungs,spleen,and lymph nodes.IL-38 administration suppressed airway hyperreactivity and asthma-related IL-4 and IL-5 expression in humanized mice,together with significantly decreased CCR3^(+) eosinophil numbers in the BALF and lungs,and a reduced percentage of human CD4^(+)CRTH2^(+)Th2 cells in the lungs and mediastinal lymph nodes.Together,our results demonstrated the anti-inflammatory mechanisms of IL-38 and provided a basis for the development of a regulatory cytokine-based treatment for allergic asthma. | Xiaoyu Sun Tianheng Hou Edwin Cheung Tiffany Nga-Teng Iu Victor Wai-Hou Tam Ida Miu-Ting Chu Miranda Sin-Man Tsang Paul Kay-Sheung Chan Christopher Wai-Kei Lam Chun-Kwok Wong | 2020 | Cellular & Molecular Immunology2020,17,6: | 22 |
| 2 | Cystic meconium peritonitis with jejunoileal atresia:Is it associated with unfavorable outcome?显示文摘AIM To compare the outcome between patients with jejunoileal atresia(JIA) associated with cystic meconium peritonitis(CMP) and patients with isolated JIA(JIA without CMP).METHODS A retrospective study was conducted for all neonates with JIA operated in our institute from January 2005 to January 2016.Demographics including the gestation age,sex,birth weight,age at operation,the presence of associated syndrome was recorded.Clinical outcome including the type of operation performed,operative time,the need for reoperation and mortality were studied.The demographics and the outcome between the 2 groups were compared.RESULTS During the study period,53 neonates had JIA underwent operation in our institute.Seventeen neonates(32%) were associated with CMP.There was no statistical difference on the demographics in the two groups.Patients with CMP had earlier operation than patients with isolated JIA(mean 1.4 d vs 3 d,P = 0.038).Primaryanastomosis was performed in 16 patients(94%) with CMP and 30 patients(83%) with isolated JIA(P = 0.269).Patients with CMP had longer operation(mean 190 min vs 154 min,P = 0.004).There were no statistical difference the need for reoperation(3 vs 6,P = 0.606) and mortality(2 vs 1,P = 0.269) between the two groups.CONCLUSION Primary intestinal anastomosis can be performed in94% of patients with JIA associated with CMP.Although patients with CMP had longer operative time,the mortality and reoperation rates were low and were comparable to patients with isolated JIA. | Kin Wai Edwin Chan Kim Hung Lee Hei Yi Vicky Wong Siu Yan Bess Tsui Yuen Shan Wong Kit Yi Kristine Pang Jennifer Wai Cheung Mou Yuk Him Tam | 2017 | World Journal of Clinical Pediatrics2017,6,1: | 3 |

| 4 | Use of laparoscopy as the initial surgical approach of impalpable testes:10-year experience显示文摘AIM: To review the experience in the management of impalpable testes using laparoscopy as the initial approach and the need for inguinal exploration.METHODS: From January 2004 to June 2014, 339 patients with undescended testes underwent operation in our institute. Fifty patients(15%) had impalpable testes. All children with impalpable testes underwent initial laparoscopy. A retrospective review was conducted on this group of patients and the outcome was analyzed.RESULTS: Forty children had unilateral impalpable testis. Ten children had bilateral impalpable testes. Thirty-one children(78%) in the unilateral group underwent subsequent inguinal exploration while 4 children(40%) in the bilateral group underwent inguinal exploration(P < 0.05). Orchidopexy was performed in 16 children(40%) in the unilateral group and 9 children(90%) in the bilateral group(P < 0.05). Regarding the 24 children with unilateral impalpable testis and underwent orchidectomy for testicular nubbin(n = 19) or atrophic testes(n = 2) or has vanishing testes(n = 3); contralateral testicular hypertrophy was noticed in 10(41%). No intra-operative complication was encountered. Two children after staged Fowler-Stephensprocedure and 1 child after inguinal orchidopexy had atrophic testes.CONCLUSION: The use of laparoscopy in children with impalpable testes is a safe procedure and can guide the need for subsequent inguinal exploration. Children with unilateral impalpable testis were associated with an increased need for inguinal exploration after laparoscopy. | 6 | Production of Hypoxia-induced Corneal Edema in Aged Eyes显示文摘Purpose:Corneal thickness assessment is a common clinical procedure applied in corneal and contact lens care. This study aims to investigate the effect of age on hypoxia-induced corneal swelling.Methods:Eighteen male subjects were equally divided into the younger [(23.7 ± 0.8) years old] and older [(74.4 ± 2.5) years old] groups. Each subject wore a thick soft contact lens (uniform thickness of 0.3 mm) on the left cornea. With the contact lens in place, the baseline central corneal thickness was measured using a specially designed photo-pachometer. The lens was then patched behind the closed eyelids, producing an extremely hypoxic stress to the cornea. The change in central corneal thickness was monitored every 20 minutes with momentary disruptions to the hypoxic stress over the next 2 hours. The increase in thickness was taken as an index of corneal edema. The rate of change in corneal thickness, as derived from a non-linear mathematical model, was compared between groups.Results:The corneal thickness of both age groups increased significantly with time (P<0.000 1). The mean corneal swelling constant for the older subjects was 16.5 × 10-3 (S.E.M. = 2.65 × 10-3) and the value for the younger subjects was 46.5 × 10-3 (S.E.M. = 3.25 × 10-3). The difference was statistically significant (P < 0.000 1).Conclusion:Aging cornea has a slower hypoxia-induced edema response compared with the younger group. | 7 | 围术期收缩压变异性是否可预测心脏手术后死亡率:ECLIPSE研究的探索性分析显示文摘背景目前,有关围术期血压稳定性-9手术预后关系的研究很少,本研究旨在验证心脏手术患者中收缩压(systolic blood pressure,SBP)变异性与术后30天死亡率具有相关性这一假设。方法本研究从ECLIPSE试验中随机选取1512例围术期高血压患者,分析其SBP变异性。通过超出既定SBP可变范围的偏差×持续时间(曲线下面积)衡量SBP变异性。手术中既定的SBP可变范围为65—135mmHg,手术前和手术后为75—145mmHg,按10mmHg递增提高SBP低限,将既定的SBP可变汜44-围逐步缩窄到手术中105-135mmHg及手术前手术后115—145mmHg。应用多元逻辑回归分析评价收缩压变异性与从ECLIPSE试验结果中得到手术后30天死亡率的关系。结果超过既定范围的SBP变异性(手术中75—135mmHg及手术前手术后85。145mmHg)与手术后30天的死亡率显著相关。每增加60mmHg·min^-1·h^-1的收缩压偏差,手术后30天死亡风险的比值比为1.16(95%可信区间1.04—1.30)。当曲线下的面积从0增加到300mmHg·min^-1·h^-1对于低危组患者,30天死亡率的预测概率从0.2%增加到0.5%,而高危患者从42.4%增加到60.7%。结论心脏手术患者围术期血压变异性与30天死亡率相关,并与超出既定的SBP可变范围(手术中75—135mlnng及手术前手术后85—145mmHg)的偏差程度成正比。与低危组患者相比,高危组患者死亡率的预测值更高。 | Solomon Aronson Cornelius M. | 8 | Exploring the utility of organo-polyoxometalate hybrids to inhibit SOX transcription factors显示文摘Background:SOX transcription factors constitute an attractive target class for intervention with small molecules as they play a prominent role in the field of regenerative biomedicine and cancer biology.However,rationally engineering specific inhibitors that interfere with transcription factor DNA interfaces continues to be a monumental challenge in the field of transcription factor chemical biology.Polyoxometalates(POMs)are inorganic compounds that were previously shown to target the high-mobility group(HMG)of SOX proteins at nanomolar concentrations.In continuation of this work,we carried out an assessment of the selectivity of a panel of newly synthesized organo-polyoxometalate hybrids in targeting different transcription factor families to enable the usage of polyoxometalates as specific SOX transcription factor drugs.Results:The residual DNA-binding activities of 15 different transcription factors were measured after treatment with a panel of diverse polyoxometalates.Polyoxometalates belonging to the Dawson structural class were found to be more potent inhibitors than the Keggin class.Further,organically modified Dawson polyoxometalates were found to be the most potent in inhibiting transcription factor DNA binding activity.The size of the polyoxometalates and its derivitization were found to be the key determinants of their potency.Conclusion:Polyoxometalates are highly potent,nanomolar range inhibitors of the DNA binding activity of the Sox-HMG family.However,binding assays involving a limited subset of structurally diverse polyoxometalates revealed a low selectivity profile against different transcription factor families.Further progress in achieving selectivity and deciphering structure-activity relationship of POMs require the identification of POM binding sites on transcription factors using elaborate approaches like X-ray crystallography and multidimensional NMR.In summary,our report reaffirms that transcription factors are challenging molecular architectures and that future polyoxometalate chemistry must consider further modification strategies,to address the substantial challenges involved in achieving target selectivity. | Kamesh Narasimhan Kevin Micoine Emmanuel Lacôte Serge Thorimbert Edwin Cheung Bernold Hasenknopf Ralf Jauch | 2014 | Cell Regeneration2014,3,1: | 0 |
