1A Highly Pathogenic Strain of Porcine Deltacoronavirus Caused Watery Diarrhea in Newborn Piglets显示文摘Porcine deltacoronavirus(PDCoV) is a newly identified virus that causes watery diarrhea in newborn piglets and results in significant economic losses to the pig industry. Since first reported in Hong Kong in 2012, PDCoV has been subsequently detected in USA, South Korea, Thailand, and China's Mainland. Here we isolated a strain of PDCoV, named CHN-GD-2016,from the intestinal content of a diseased newborn piglet with severe diarrhea in a pig farm in Guangdong, China. PDCoV CHN-GD-2016 could be identified by immunofluorescence with PDCoV specific rabbit antisera, and typical crown-shaped particles with spiky surface projections of this PDCoV were observed with electron microscopy. Genomic analysis showed that the PDCoV CHN-GD-2016 was closely related to other Chinese PDCoV strains, with the highest sequence similarity with the strain CHN/Tianjin/2016. Importantly, inoculation of newborn piglets with 1×10~5 TCID_(50) of CHN-GD-2016 by oral feeding successfully reproduced clear clinical symptoms, including vomiting, dehydration, and severe diarrhea in piglets. In addition, the virus RNA in rectal swabs from 1 to 7 days post inoculation was detected, macroscopic and microscopic lesions in small intestine were observed, and viral antigen was also detected in the small intestines with immunohistochemical staining. 3A better carbon-water flux simulation in multiple vegetation types by data assimilation显示文摘Background:The accurate estimation of carbon-water flux is critical for understanding the carbon and water cycles of terrestrial ecosystems and further mitigating climate change.Model simulations and observations have been widely used to research water and carbon cycles of terrestrial ecosystems.Given the advantages and limitations of each method,combining simulations and observations through a data assimilation technique has been proven to be highly promising for improving carbon-water flux simulation.However,to the best of our knowledge,few studies have accomplished both parameter optimization and the updating of model state variables through data assimilation for carbon-water flux simulation in multiple vegetation types.And little is known about the variation of the performance of data assimilation for carbon-water flux simulation in different vegetation types.Methods:In this study,we assimilated leaf area index(LAI)time-series observations into a biogeochemical model(Biome-BGC)using different assimilation algorithms(ensemble Kalman filter algorithm(EnKF)and unscented Kalman filter(UKF))in different vegetation types(deciduous broad-leaved forest(DBF),evergreen broad-leaved forest(EBF)and grassland(GL))to simulate carbon-water flux.Results:The validation of the results against the eddy covariance measurements indicated that,overall,compared with the original simulation,assimilating the LAI into the Biome-BGC model improved the carbon-water flux simulations(R^(2)increased by 35%,root mean square error decreased by 10%;the sum of the absolute error decreased by 8%)but more significantly,improved the water flux simulations(R^(2)increased by 31%,root mean square error decreased by 18%;the sum of the absolute error decreased by 16%).Among the different forest types,the data assimilation techniques(both EnKF and UKF)achieved the best performance towards carbon-water flux in EBF(R^(2)increased by 44%,root mean square error decreased by 24%;the sum of the absolute error decreased by 28%),and the performances of EnKF and UKF showed slightly different when simulating carbon fluxes.Conclusion:We suggest that to reduce the uncertainty in global carbon-water flux quantification,forthcoming data assimilation treatment should consider the vegetation types where the data assimilation experiments are carried out,the simulated objectives and the assimilation algorithms.Qiuyu Liu Tinglong Zhang Mingxi Du Huanlin Gao Qingfeng Zhang Rui Sun 2022Forest Ecosystems2022,9,1:1

6Molecular cytogenetic analyses of two new wheat-rye 6RL translocation lines with resistance to wheat powdery mildew显示文摘Rye(Secale cereale)is a valuable gene donor for wheat improvement,especially for its resistance to diseases.Developing rye-derived resistance sources is important for wheat breeding.In the present study,two wheat-rye derivatives,designated JS016 and JS110,were produced by crossing common wheat cultivar Yangmai 23 with Pakistani rye accession W2A.Using sequential genomic in situ hybridization(GISH)and multicolor fluorescence in situ hybridization(mc-FISH),JS016 and JS110 were identified as a T6BS.6RL translocation line and a T6BS.6BL6RL translocation line,respectively.Ten newly 6RL chromosome arm-specific markers were developed and used to confirm the 6RL translocation.The wheat 55K single-nucleotide polymorphism(SNP)array further verified the molecular cytogenetic identification results above and clarified their breakpoints at 430.9 and 523.0 Mb of chromosome 6B in JS016 and JS110,respectively.Resistance spectrum and allelism test demonstrated that JS016 and JS110 possessed novel powdery mildew resistance gene(s)that was derived from the 6RL translocation but differed from Pm20.Moreover,JS016 and JS110 had better agronomic traits than the previously reported 6RL translocation line carrying Pm20.To efficiently transfer and detect the 6RL translocation from JS016 and JS110,one 6RL-specific Kompetitive allele specific PCR(KASP)marker was developed and validated in high throughput marker-assisted selection(MAS).Shanying Zhu Haonan Du Fuyu Su Jin Wang Qingfeng Meng Tianlei Liu Rui Guo Zhaozhao Chen Huanhuan Li Wenxuan Liu Pengtao Ma Huagang He 2023The Crop Journal2023,11,2:1

9Dissection of cellular and molecular mechanisms of aristolochic acid-induced hepatotoxicity via single-cell transcriptomics显示文摘Background:Aristolochic acids(AAs),a class of carcinogenic and mutagenic natural products from Aristolochia and Asarum plants,are well-known to be responsible for inducing nephrotoxicity and urothelial carcinoma.Recently,accumulating evidence suggests that exposure to AAs could also induce hepatotoxicity and even hepatocellular carcinoma,though the mechanisms are poorly defined.Methods:Here,we aimed to dissect the underlying cellular and molecular mechanisms of aristolochic acid I(AAI)-induced hepatotoxicity by using advanced single-cell RNA sequencing(scRNA-seq)and proteomics techniques.We established the first single-cell atlas of mouse livers in response to AAI.Results:In hepatocytes,our results indicated that AAI activated NF-κB and STAT3 signaling pathways,which may contribute to the inflammatory response and apoptosis.In liver sinusoidal endothelial cells(LSECs),AAI activated multiple oxidative stress and inflammatory associated signaling pathways and induced apoptosis.Importantly,AAI induced infiltration of cytotoxic T cells and activation of proinflammatory macrophage and neutrophil cells in the liver to produce inflammatory cytokines to aggravate inflammation.Conclusions:Collectively,our study provides novel knowledge of AAs-induced molecular characteristics of hepatotoxicity at a singlecell level and suggests future treatment options for AAs associated hepatotoxicity.Piao Luo Jiayun Chen Qian Zhang Fei Xia Chen Wang Yunmeng Bai Huan Tang Dandan Liu Liwei Gu Qingfeng Du Wei Xiao Chuanbin Yang Jigang Wang 2022Precision Clinical Medicine2022,5,4:0
10A novel protein complex that regulates active DNA demethylation in Arabidopsis显示文摘Active DNA demethylation is critical for altering DNA methylation patterns and regulating gene expression.The 5-methylcytosine DNA glycosylase/lyase ROS1 initiates a base-excision repair pathway for active DNA demethylation and is required for the prevention of DNA hypermethylation at 1000 s of genomic regions in Arabidopsis.How ROS1 is regulated and targeted to specific genomic regions is not well understood.Here,we report the discovery of an Arabidopsis protein complex that contains ROS1,regulates ROS1 gene expression,and likely targets the ROS1 protein to specific genomic regions.ROS1 physically interacts with a WD40 domain protein(RWD40),which in turn interacts with a methyl-DNA binding protein(RMB1)as well as with a zinc finger and homeobox domain protein(RHD1).RMB1 binds to DNA that is methylated in any sequence context,and this binding is necessary for its function in vivo.Loss-of-function mutations in RWD40,RMB1,or RHD1 cause DNA hypermethylation at several tested genomic regions independently of the known ROS1 regulator IDM1.Because the hypermethylated genomic regions include the DNA methylation monitoring sequence in the ROS1 promoter,plants mutated in RWD40,RMB1,or RHD1 show increased ROS1 expression.Importantly,ROS1 binding to the ROS1 promoter requires RWD40,RMB1,and RHD1,suggesting that this complex dictates ROS1 targeting to this locus.Our results demonstrate that ROS1 forms a protein complex with RWD40,RMB1,and RHD1,and that this novel complex regulates active DNA demethylation at several endogenous loci in Arabidopsis.Pan Liu Wen-Feng Nie Xiansong Xiong Yuhua Wang Yuwei Jiang Pei Huang Xueqiang Lin Guochen Qin Huan Huang Qingfeng Niu Jiamu Du Zhaobo Lang Rosa Lozano-Duran Jian-Kang Zhu 2021Journal of Integrative Plant Biology2021,63,4:0

12Mechanistic engineering of celastrol liposomes induces ferroptosis and apoptosis by directly targeting VDAC2 in hepatocellular carcinoma显示文摘Hepatocellular carcinoma(HCC)is one of most common and deadliest malignancies.Celastrol(Cel),a natural product derived from the Tripterygium wilfordii plant,has been extensively researched for its potential effectiveness in fighting cancer.However,its clinical application has been hindered by the unclear mechanism of action.Here,we used chemical proteomics to identify the direct targets of Cel and enhanced its targetability and antitumor capacity by developing a Cel-based liposomes in HCC.We demonstrated that Cel selectively targets the voltage-dependent anion channel 2(VDAC2).Cel directly binds to the cysteine residues of VDAC2,and induces cytochrome C release via dysregulating VDAC2-mediated mitochondrial permeability transition pore(mPTP)function.We further found that Cel induces ROS-mediated ferroptosis and apoptosis in HCC cells.Moreover,coencapsulation of Cel into alkyl glucoside-modified liposomes(AGCL)improved its antitumor efficacy and minimized its side effects.AGCL has been shown to effectively suppress the proliferation of tumor cells.In a xenograft nude mice experiment,AGCL significantly inhibited tumor growth and promoted apoptosis.Our findings reveal that Cel directly targets VDAC2 to induce mitochondria-dependent cell death,while the Cel liposomes enhance its targetability and reduces side effects.Overall,Cel shows promise as a therapeutic agent for HCC.Piao Luo Qian Zhang Shuo Shen Yehai An Lixia Yuan Yin-Kwan Wong Sizhe Huang Shaohui Huang Jingnan Huang Guangqing Cheng Jiahang Tian Yu Chena Xiaoyong Zhang Weiguang Li Songqi He Jigang Wang Qingfeng Du 2023Asian Journal of Pharmaceutical Sciences2023,18,6:0
13A high-safety, flame-retardant cellulose-based separator with encapsulation structure for lithium-ion battery显示文摘The safety issues of lithium-ion batteries have received attention because flammable organic electrolytes are used.Also,the commercial polyolefin separator will undergo severe thermal shrinkage when the internal temperature of the battery increases to 130-160°C,which increases the risk.Therefore,the development of a high thermal stability and high-safety separator is an effective strategy to improve battery safety.Herein,we design a green,cellulose-based separator(Cel@DBDPE)with a unique encapsulation structure for lithium-ion batteries,in which functional flame retardants(DBDPE)are wrapped in microscrolls formed by the self-rolling of 2D cellulose nanosheets upon freeze-drying.This structure can firmly anchor DBDPE particles in the separator to prevent them from undergoing exfoliation and does not affect the properties of the separator,such as the thickness and the pore structure.Compared with commercial polypropylene,Cel@DBDPE has excellent thermal stability and flame retardancy.The former makes it less prone to thermal shrinkage and the latter can effectively prevent the combustion of the electrolyte,showing an efficient self-extinguishing ability.Moreover,the Cel@DBDPE is only 15μm in size and has competitive properties comparable to polypropylene.Thus,there is no sacrifice in the electrochemical performance of battery when the Cel@DBDPE is used as separator.This study provides a new structural design for the construction of a high-safety separator.Jinzhou Fu Hanwei Wang Zhichen Du Yao Liu Qingfeng Sun Huiqiao Li 2023SmartMat2023,4,5:0
14The effect of Lorentz-like force on collective flows of K+in Au+Au collisions at 1.5 Ge V/nucleon显示文摘Producing kaon mesons in heavy-ion collisions at beam energies below their threshold energy is an important way to investigate the properties of dense nuclear matter. In this study, based on the newly updated version of the ultrarelativistic quantum molecular dynamics model, we introduce the kaon-nucleon(KN) potential, including both the scalar and vector(also dubbed Lorentz-like)aspects. We revisit the influence of the KN potential on the collective flow of K+mesons produced in Au+Au collisions at Elab= 1.5 Ge V/nucleon and find that the contribution of the newly included Lorentz-like force is very important, particulary for describing the directed flow of K+.
