2Validation of Zebrafish (Danio rerio) Reference Genes for Quantitative Real-time RT-PCR Normalization显示文摘The normalization of quantitative real time RT-PCR (qRT-PCR) is important to obtain accurate gene expression data. The most common method for qRT-PCR normalization is to use reference, or house- keeping genes. However, there is emerging evidence that even reference genes can be regulated under different conditions, qRT-PCR has only recently been used in terms of zebrafish gene expression studies and there is no validated set of reference genes. This study characterizes the expression of nine possible reference genes during zebrafish embryonic development and in a zebrafish tissue panel. All nine reference genes exhibited variable expression. The β-actin, EF1α and Rp113α genes comprise a validated reference gene panel for zebrafish developmental time course studies, and the EF1α, Rp113α and 18S rRNA genes are more suitable as a reference gene panel for zebrafish tissue analysis. 7Long-distance ABA transport can mediate distal tissue responses by affecting local ABA concentrations显示文摘Environmental stresses that perturb plant Hwater relations influence abscisic acid(ABA) concentrations, but it is unclear whether long-distance ABA transport contributes to changes in local ABA levels. To determine the physiological relevance of ABA transport, we made reciprocal-and self-grafts of ABA-deficient flacca mutant and wild-type(WT) tomato plants, in which low phosphorus(P) conditions decreased ABA concentrations while salinity increased ABA concentrations. Whereas foliar ABA concentrations in the WT scions were rootstock independent under control conditions, salinity resulted in long-distance transport of ABA: flacca scions had approximately twice as much ABA when grafted on WT rootstocks compared to flacca rootstocks. Root ABA concentrations were scion dependent: both WT and flacca rootstocks had less ABA with the flacca mutant scion than with the WT scion under control conditions. In WT scions, whereas rootstock genotype had limited effects on stomatal conductance under control conditions, a flacca rootstock decreased leaf area of stressed plants, presumably due to attenuated root-to-shoot ABA transport. In flacca scions, a WT rootstock decreased stomatal conductance but increased leaf area of stressed plants, likely due to enhanced root-to-shoot ABA transport. 9Vibration induced flow in hoppers:DEM 2D polygon model显示文摘A two-dimensional discrete element model (DEM) simulation of cohesive polygonal particles has been developed to assess the benefit of point source vibration to induce flow in wedge-shaped hoppers. The particle-particle interaction model used is based on a multi-contact principle. The first part of the study investigated particle discharge under gravity without vibration to determine the critical orifice size (Bc) to just sustain flow as a function of particle shape. It is shown that polygonal-shaped particles need a larger orifice than circular particles. It is also shown that Bc decreases as the number of particle vertices increases. Addition of circular particles promotes flow of polygons in a linear manner. The second part of the study showed that vibration could enhance flow, effectively reducing Bc. The model demonstrated the importance of vibrator location (height), consistent with previous continuum model results, and vibration amplitude in enhancing flow.Feras Y. Fraige Paul A. 13Detection of low energy solar neutrinos by a two-phase cryogenic e-bubble detector显示文摘A new two-phase cryogenic neutrino detector using electron bubble (e-bubble) specifically in liquid helium is proposed and being developed for real time, high rate measurements of low-energy p-p reac- tion neutrinos from the sun. The e-bubble detector is a time projection chamber-like (TPC) tracking detector. The task of such a neutrino detector is to detect the ionization of the elastically scattered target electrons by incident neutrinos, and then to characterize their energy and direction and to dis- tinguish them from radioactive backgrounds. The ionization signals are expected to be small and hence undergo avalanche amplification in the saturated vapor above the liquid phase by gas electron multi- pliers (GEMs) at high gain. Higher granularity and intrinsically suppressed ion feedback give a good spatial resolution and are the major advantages of this technology. It should be possible to construct such a detector to track charged particles down to 100―200 keV in a massive liquid helium target with fractional millimeter spatial resolution in three-dimensional space, using the GEM-based TPC with a high-resolution CCD camera, for both the electronic and light readout.JU YongLin GU Yan J.
