| 1 | A phase I study of different doses and frequencies of pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) in patients with standard-dose chemotherapy-induced neutropenia显示文摘Objective: The recommended dose of prophylactic pegylated recombinant human granulocyte-colony stimulating factor(PEG rhG-CSF) is 100 μg/kg once per cycle for patients receiving intense-dose chemotherapy.However, few data are available on the proper dose for patients receiving less-intense chemotherapy. The aim of this phase I study is to explore the proper dose and administration schedule of PEG rhG-CSF for patients receiving standard-dose chemotherapy.Methods:Eligible patients received 3-cycle chemotherapy every 3 weeks.No PEG rhG-CSF was given in the first cycle.Patients experienced grade 3 or 4 neutropenia would then enter the cycle 2 and 3.In cycle 2,patients received a single subcutaneous injection of prophylactic PEG rhG-CSF on d 3,and received half-dose subcutaneous injection in cycle 3 on d 3 and d 5,respectively.Escalating doses(30,60,100 and 200μg/kg)of PEG rhG-CSF were investigated.Results:A total of 26 patients were enrolled and received chemotherapy,in which 24 and 18 patients entered cycle 2 and cycle 3 treatment,respectively.In cycle 2,the incidence of grade 3 or 4 neutropenia for patients receiving single-dose PEG rhG-CSF of 30,60,100 and 200 μg/kg was 66.67%,33.33%,22.22% and 0,respectively,with a median duration less than 1(0–2)d.No grade 3 or higher neutropenia was noted in cycle 3 in all dose cohorts.Conclusions:The pharmacokinetic and pharmacodynamic profiles of PEG rhG-CSF used in cancer patients were similar to those reported,as well as the safety.Double half dose administration model showed better efficacy result than a single dose model in terms of grade 3 neutropenia and above.The single dose of 60 μg/kg,100 μg/kg and double half dose of 30 μg/kg were recommended to the phase Ⅱ study,hoping to find a preferable method for neutropenia treatment. | Yan Qin Xiaohong Han Lin Wang Ping Du Jiarui Yao Di Wu Yuanyuan Song Shuxiang Zhang Le Tang Yuankai Shi | 2017 | Chinese Journal of Cancer Research2017,29,5: | 11 |
| 2 | Candidate genes involved in the biosynthesis of lignan in Schisandra chinensis fruit based on transcriptome and metabolomes analysis显示文摘Schisandra chinensis Turcz.(Baill.) is a plant species with fruits that have been well known in Far Eastern medicine for a long time. It has traditionally been used as a stimulating and fortifying agent in cases of physical exhaustion and to inhibit fatigue.The major bioactive compounds found in S. chinensis are lignans with a dibenzocyclooctadiene skeleton, but little is known about their biosynthesis in plants. S. chinensis is the ideal medicinal plant for studying the biosynthesis of lignans, especially the dibenzocyclooctadiene skeleton. Genomic information for this important herbal plant is unavailable. To better understand the lignan biosynthesis pathway, we generated transcriptome sequences from the fruit during ripening and performed de novo sequence assembly, yielding136 843 unique transcripts with N50 of 1778 bp. Putative functions could be assigned to 41 824 transcripts(51.57%) based on BLAST searches against annotation databases including GO(Gene ontology) and KEGG(Kyoto Encyclopedia of Genes and Genomes). Furthermore, 22 candidate cytochrome P450 genes and 15 candidate dirigent proteins genes that were most likely involved in the lignan biosynthesis pathway were discovered based on transcriptome sequencing of S. chinensis. The genomic data obtained from S. chinensis, especially the identification of putative genes involved in the lignan biosynthesis pathway, will facilitate our understanding of lignan biosynthesis at the molecular level. The lignan metabolite profiles were analyzed by metabolomes, the accumulation patterns of 30 metabolites involved in the lignan pathway were studied. Co-expression network of lignan contents and transcriptional changes showed355 strong correlations(correlation coefficient, R^2 > 0.9) between 21 compounds and 153 transcripts. | 3 | Separation and Manipulation of Rare-earth Oxide Particles by Dielectrophoresis显示文摘A challenge in chemical engineering is the separation and purification of rare-earth elements and their compounds. We report the design and manufacture of a dielectrophoresis(DEP) microchip of microelectrode arrays. This microchip device is constructed in order to use DEP to capture micro-particles of rare-earth oxides in petro-leum. Dielectrophoretic behavior of micro-particles of rare-earth oxides in oil media is explored. The dielectropho-retic effects of particles under different conditions are investigated. It is showed that the prepared microchip is suit-able for use in the investigation of dielectrophoretic responses of the rare-earth oxides in oil media. The factors such as frequency,particle size and valence of rare-earth metal are discussed. When the frequency is fixed,the transla-tion voltage decreases as particle size increases. Lower frequencies are more effective for manipulation of inorganic particles in oil media. Particles of the same rare-earth oxide with different size,as well as particles of different rare-earth oxides,are captured in different regions of the field by regulating DEP conditions. | 14 | Prototype of single-event effect localization system with CMOS pixel sensor显示文摘The single-event effect(SEE) is a serious threat to electronics in radiation environments. The most important issue in radiation-hardening studies is the localization of the sensitive region in electronics to the SEE. To solve this problem, a prototype based on a complementary metal oxide semiconductor(CMOS) pixel sensor, i.e., TopmetalM, was designed for SEE localization. A beam test was performed on the prototype at the radiation terminal of the Heavy Ion Research Facility in Lanzhou(HIRFL). The results indicated that the inherent deflection angle of the prototype to the beam was 1.7°, and the angular resolution was 0.6°.
