2Ciliary parathyroid hormone signaling activates transforming growth factor-βto maintain intervertebral disc homeostasis during aging显示文摘Degenerative disc disease(DDD) is associated with intervertebral disc degeneration of spinal instability. Here, we report that the cilia of nucleus pulposus(NP) cells mediate mechanotransduction to maintain anabolic activity in the discs. We found that mechanical stress promotes transport of parathyroid hormone 1 receptor(PTH1 R) to the cilia and enhances parathyroid hormone(PTH) signaling in NP cells. PTH induces transcription of integrin α_vβ_6 to activate the transforming growth factor(TGF)-β-connective tissue growth factor(CCN2)-matrix proteins signaling cascade. Intermittent injection of PTH(iPTH) effectively attenuates disc degeneration of aged mice by direct signaling through NP cells, specifically improving intervertebral disc height and volume by increasing levels of TGF-β activity, CCN2, and aggrecan. PTH1 R is expressed in both mouse and human NP cells. Importantly,knockout PTH1 R or cilia in the NP cells results in significant disc degeneration and blunts the effect of PTH on attenuation of aged discs. 3Fractional order nonsingular terminal sliding mode control for flexible spacecraft attitude tracking显示文摘This paper investigates a fractional terminal sliding mode control for flexible spacecraft attitude tracking in the presence of inertia uncertainties and external disturbances. The controller is based on the fractional calculus and nonsingular terminal sliding mode control technique,and it guarantees the convergence of attitude tracking error in finite time rather than in the asymptotic sense. With respect to the controller,a fractional order sliding surface is given,the corresponding control scheme is proposed based on Lyapunov stability theory to guarantee the sliding condition,and the finite time stability of the whole close loop system is also proven. 4Metal release/accumulation during the decomposition of Potamogeton crispus in a shallow macrophytic lake显示文摘Changes in metal concentrations in the litter of Potamogeton crispus were monitored during a consecutive 40-day in situ decomposition experiment using the litterbag method.The accumulation index was calculated and used to indicate the changes in the metals in litter.The results showed that the concentrations of Al,Cd,Cr,Fe,Mn,and Pb in litter increased significantly during the decomposition,while Cu and Zn concentrations decreased dramatically.Significant positive correlations were found between the concentrations of Al,Cr,Fe,and Mn and between Cu and Zn.Moreover,Cu and Zn both negatively correlated with Al and Fe.The remaining dry mass was negatively correlated with Al and Fe concentrations but positively correlated with Cu and Zn concentrations.Generally the accumulation index values of metals other than Al were less than one,indicating that the litter of P.crispus acted as a source of metals to the surrounding water body.Al was the only metal that showed continuous net accumulation in litter.The net accumulation of Fe and Mn in litter during the last 10 days of the experiment may indicate the precipitation of Feand Mn-oxides.It was estimated that 160 g/m^2(dry weight)P.crispus was decomposed in40 days.This was equivalent to releasing the following amounts of metals:0.01 mg Cd,0.03 mg Cr,0.71 mg Cu,0.55 mg Mn,0.02 mg Pb and 13.8 mg Zn into surrounding water,and accumulating 149 mg Al and 11 mg Fe,in a 1 m^2 area.Huanguang Deng Ju Zhang Shiyue Chen Liwei Yang Dongqi Wang Shiyong Yu 2016Journal of Environmental Sciences2016,28,4:10
5Epigenetic regulation in B-cell maturation and its dysregulation in autoimmunity显示文摘B cells have a critical role in the initiation and acceleration of autoimmune diseases, especially those mediated byautoantibodies. In the peripheral lymphoid system, mature B cells are activated by self or/and foreign antigens andsignals from helper T cells for differentiating into either memory B cells or antibody-producing plasma cells.Accumulating evidence has shown that epigenetic regulations modulate somatic hypermutation and class switchDNA recombination during B-cell activation and differentiation. Any abnormalities in these complex regulatoryprocesses may contribute to aberrant antibody production, resulting in autoimmune pathogenesis such as systemiclupus erythematosus. Newly generated knowledge from advanced modern technologies such as next-generationsequencing, single-cell sequencing and DNA methylation sequencing has enabled us to better understand B-cellbiology and its role in autoimmune development. 6Photo-induced transformations of Hg(Ⅱ) species in the presence of Nitzschia hantzschiana,ferric ion,and humic acid显示文摘Effects of algae Nitzschia hantzschiana, Fe(Ⅲ) ions, humic acid, and pH on the photochemical reduction of Hg(Ⅱ) using the irradiation of metal halide lamps (λ 365 nm, 250 W) were investigated. The photoreduction rate of Hg(Ⅱ) was found to increase with increasing concentrations of algae, Fe(Ⅲ) ions, and humic acid. Alteration of pH value affected the photoreduction of Hg(Ⅱ) in aqueous solution with or without algae. The photoreduction rate of Hg(II) decreased with increasing initial Hg(Ⅱ) concentration in aqueous solution in the presence of algae. The photochemical kinetics of initial Hg(Ⅱ) and algae concentrations on the photoreduction of Hg(Ⅱ) were studied at pH 7.0. 7IL-17 sustains the plasma cell response via p38-mediated Bcl-xL RNA stability in lupus pathogenesis显示文摘Recent studies have demonstrated a central role for plasma cells in the development of autoimmune diseases,such as systemic lupus erythematosus(SLE).Currently,both the phenotypic features and functional regulation of autoreactive plasma cells during SLE pathogenesis remain largely unclear.In this study,we first found that a major subset of IL-17 receptor-expressing plasma cells potently produced anti-dsDNA IgG upon IL-17A(IL-17)stimulation in SLE patients and lupus mice.Using a humanized lupus mouse model,we showed that the transfer of Th17 cell-depleted PBMCs from lupus patients resulted in a significantly reduced plasma cell response and attenuated renal damage in recipient mice compared to the transfer of total SLE PBMCs.Moreover,long-term BrdU incorporation in lupus mice detected highly enriched long-lived BrdU+subsets among IL-17 receptor-expressing plasma cells.Lupus mice deficient in IL-17 or IL-17 receptor C(IL-17RC)exhibited a diminished plasma cell response and reduced autoantibody production with attenuated renal damage,while the adoptive transfer of Th17 cells triggered the plasma cell response and renal damage in IL-17-deficient lupus mice.In reconstituted chimeric mice,IL-17RC deficiency resulted in severely impaired plasma cell generation but showed no obvious effect on germinal center B cells.Further mechanistic studies revealed that IL-17 significantly promoted plasma cell survival via p38-mediated Bcl-xL transcript stabilization.Together,our findings identified a novel function of IL-17 in enhancing plasma cell survival for autoantibody production in lupus pathogenesis,which may provide new therapeutic strategies for the treatment of SLE.Kongyang Ma Wenhan Du Fan Xiao Man Han Enyu Huang Na Peng Yuan Tang Chong Deng Lixiong Liu Yulan Chen Jingjing Li Shiwen Yuan Qin Huang Xiaoping Hong Dajun Hu Xiaoyan Cai Quan Jiang Dongzhou Liu Liwei Lu 2021Cellular & Molecular Immunology2021,18,7:6
8A critical role of IL-17 in modulating the B-cell response during H5N1 influenza virus infection显示文摘Interleukin-17(IL-17),a member of the IL-17 cytokine family,plays a crucial role in mediating the immune response against extracellular bacteria and fungi in the lung.Although there is increasing evidence that IL-17 is involved in protective immunity against H1 and H3 influenza virus infections,little is known about the role of IL-17 in the highly pathogenic H5N1 influenza virus infection.In this study,we show that H5N1-infected IL-17 knockout(KO)mice exhibit markedly increased weight loss,more pronounced lung immunopathology and significantly reduced survival rates as compared with infected wild-type controls.Moreover,the frequency of B cells in the lung were substantially decreased in IL-17 KO mice after virus infection,which correlated with reduced CXCR5 expression in B cells and decreased CXCL13 production in the lung tissue of IL-17 KO mice.Consistent with this observation,B cells from IL-17 KO mice exhibited a significant reduction in chemokine-mediated migration in culture.Taken together,these findings demonstrate a critical role for IL-17 in mediating the recruitment of B cells to the site of pulmonary influenza virus infection in mice.Xiaohui Wang Chris CS Chan Min Yang Jun Deng Vincent KM Poon Virtual HC Leung King-Hung Ko Jie Zhou Kwok Yung Yuen Bo-Jian Zheng Liwei Lu 2011Cellular & Molecular Immunology2011,8,6:2
9Finite-time coordination control for formation flying spacecraft显示文摘This paper investigates a distributed coordination control scheme using an adaptive terminal sliding mode for formation flying spacecraft with coupled attitude and translational dynamics. In order to overcome the singularity of the traditional fast terminal sliding manifold, a novel fast terminal sliding manifold is given. And then, based on the adaptive control method, a continuous robust coordinated controller is designed to compensate external disturbances and to alleviate the chattering phenomenon. 12Thermal equation of state of a natural kyanite up to 8.55 GPa and 1273 K显示文摘The thermal equation of state of a natural kyanite has been investigated with a DIA-type,cubic-anvil apparatus(SAM85)combined with an energy-dispersive synchrotron X-ray radiation technique up to 8.55 GPa and 1273 K.No phase transition was observed in the studied pressure-temperature(P-T)range.The Le Bail full profile refinement technique was used to derive the unit-cell parameters.By fixing the bulk modulus K 0 as 196 GPa and its pressure derivative K0 as 4,our P-V(volume)-T data were fitted to the high temperature BircheMurnaghan equation of state.The obtained parameters for the kyanite are:V_(0)=294.05(9)Å^(3),a=2.53(11)×10^(-5)K^(-1) and(K/T)P=-0.021(8)GPa∙K^(-1).These parameters have been combined with other experimentally-measured thermodynamic data for the relevant phases to calculate the P-T locus of the reaction kyanite¼stishoviteþcorundum.With this thermodynamically constrained phase boundary,previous high-pressure phase equi-librium experimental studies with the multi-anvil press have been evaluated.Qiang He Xi Liu Baosheng Li Liwei Deng Wei Liu Liping Wang 2016Matter and Radiation at Extremes2016,1,5:1

15Equation of state of CAS phase to pressure of the uppermost lower mantle at ambient temperature显示文摘The CAS phase is a major constituent phase for the continental crust and basaltic compositions at the P-T conditions of the Earth’s mantle, and potentially plays an important role in the geodynamic processes related to slab subduction. Its equation of state has been investigated here at ambient temperature up to about 25 GPa by using a diamond-anvil cell and synchrotron X ray radiation. Its P-V data, fitted to the third-order Birch-Murnaghan equation, yield an isothermal bulk modulus (KT) of 185 (9) GPa and first pressure derivative ( KT′ ) of 7.2 (12). If KT′ is fixed at 4, the derived KT is 212 (4) GPa. 16Aryl hydrocarbon receptor activation drives polymorphonuclear myeloid-derived suppressor cell response and efficiently attenuates experimental Sjögren’s syndrome显示文摘Myeloid-derived suppressor cells(MDSCs)comprise heterogeneous myeloid cell populations with immunosuppressive capacity that contribute to immune regulation and tolerance induction.We previously reported impaired MDSC function in patients with primary Sjögren’s syndrome(pSS)and mice with experimental SS(ESS).However,the molecular mechanisms underlying MDSC dysfunction remain largely unclear.In this study,we first found that aryl hydrocarbon receptor(AhR)was highly expressed by human and murine polymorphonuclear MDSCs(PMN-MDSCs).Indole-3-propionic acid(IPA),a natural AhR ligand produced from dietary tryptophan,significantly promoted PMN-MDSC differentiation and suppressive function on CD4^(+)T cells.In contrast,feeding a tryptophan-free diet resulted in a decreased PMN-MDSC response,a phenotype that could be reversed by IPA supplementation.The functional importance of PMN-MDSCs was demonstrated in ESS mice by using a cell-depletion approach.Notably,AhR expression was reduced in PMN-MDSCs during ESS development,while AhR antagonism resulted in exacerbated ESS pathology and dysregulated T effector cells,which could be phenocopied by a tryptophan-free diet.Interferon regulatory factor 4(IRF4),a repressive transcription factor,was upregulated in PMN-MDSCs during ESS progression.Chromatin immunoprecipitation analysis revealed that IRF4 could bind to the promoter region of AhR,while IRF4 deficiency markedly enhanced AhR-mediated PMN-MDSC responses.Furthermore,dietary supplementation with IPA markedly ameliorated salivary glandular pathology in ESS mice with restored MDSC immunosuppressive function.Together,our results identify a novel function of AhR in modulating the PMN-MDSC response and demonstrate the therapeutic potential of targeting AhR for the treatment of pSS.Yanxia Wei Na Peng Chong Deng Futao Zhao Jie Tian Yuan Tang Sulan Yu Yacun Chen Yu Xue Fan Xiao Yingbo Zhou Xiaomei Li Hejian Zou Ke Rui Xiang Lin Liwei Lu 2022Cellular & Molecular Immunology2022,19,12:1

19A Novel Unsupervised MRI Synthetic CT Image Generation Framework with Registration Network显示文摘In recent years,radiotherapy based only on Magnetic Resonance(MR)images has become a hot spot for radiotherapy planning research in the current medical field.However,functional computed tomography(CT)is still needed for dose calculation in the clinic.Recent deep-learning approaches to synthesized CT images from MR images have raised much research interest,making radiotherapy based only on MR images possible.In this paper,we proposed a novel unsupervised image synthesis framework with registration networks.This paper aims to enforce the constraints between the reconstructed image and the input image by registering the reconstructed image with the input image and registering the cycle-consistent image with the input image.Furthermore,this paper added ConvNeXt blocks to the network and used large kernel convolutional layers to improve the network’s ability to extract features.This research used the collected head and neck data of 180 patients with nasopharyngeal carcinoma to experiment and evaluate the training model with four evaluation metrics.At the same time,this research made a quantitative comparison of several commonly used model frameworks.We evaluate the model performance in four evaluation metrics which achieve Mean Absolute Error(MAE),Root Mean Square Error(RMSE),Peak Signal-to-Noise Ratio(PSNR),and Structural Similarity(SSIM)are 18.55±1.44,86.91±4.31,33.45±0.74 and 0.960±0.005,respectively.Compared with other methods,MAE decreased by 2.17,RMSE decreased by 7.82,PSNR increased by 0.76,and SSIM increased by 0.011.The results show that the model proposed in this paper outperforms other methods in the quality of image synthesis.The work in this paper is of guiding significance to the study of MR-only radiotherapy planning.Liwei Deng Henan Sun Jing Wang Sijuan Huang Xin Yang 2023Computers, Materials & Continua2023,77,11:0
20Ultrafast flash memory with large self-rectifying ratio based on atomically thin MoS_(2)-channel transistor显示文摘Flash memory with high operation speed and stable retention performance is in great demand to meet the requirements of big data.In addition,the realisation of ultrafast flash memory with novel functions offers a means of combining heterogeneous components into a homogeneous device without considering impedance matching.This report proposes a 20 ns programme flash memory with 10^(8) self-rectifying ratios based on a 0.65 nm-thick MoS_(2)-channel transistor.A high-quality van der Waals heterojunction with a sharp interface is formed between the Cr/Au metal floating layer and h-BN tunnelling layer.In addition,the large rectification ratio and low ideality factor(n=1.13)facilitate the application of the MoS_(2)-channel flash memory as a bit-line select transistor.Finally,owing to the ultralow MoS_(2)/h-BN heterojunction capacitance(50 fF),the memory device exhibits superior performance as a high-frequency(up to 1 MHz)sine signal rectifier.These results pave the way toward the potential utilisation of multifunctional memory devices in ultrafast two-dimensional NAND-flash applications.Liwei Liu Yibo Sun Xiaohe Huang Chunsen Liu Zhaowu Tang Senfeng Zeng David Wei Zhang Shaozhi Deng Peng Zhou 2022Materials Futures2022,1,2:0
