2Small extracellular vesicles in combination with sleep-related circRNA3503:A targeted therapeutic agent with injectable thermosensitive hydrogel to prevent osteoarthritis显示文摘Osteoarthritis(OA),characterized by chondrocyte apoptosis and disturbance of the balance between catabolism and anabolism of the extracellular matrix(ECM),is the most common age-related degenerative joint disease worldwide.As sleep has been found to be beneficial for cartilage repair,and circular RNAs(circRNAs)have been demonstrated to be involved in the pathogenesis of OA,we performed RNA sequencing(RNA-seq),and found circRNA3503 was significantly increased after melatonin(MT)-induced cell sleep.Upregulation of circRNA3503 expression completely rescued the effects of interleukin-1β(IL-1β),which was used to simulate OA,on apoptosis,ECM degradation-and synthesis-related genes.Mechanistically,circRNA3503 acted as a sponge of hsa-miR-181c-3p and hsa-let-7b-3p.Moreover,as we previously showed that small extracellular vesicles(sEVs)derived from synovium mesenchymal stem cells(SMSCs)can not only successfully deliver nucleic acids to chondrocytes,but also effectively promote chondrocyte proliferation and migration,we assessed the feasibility of sEVs in combination with sleep-related circRNA3503 as an OA therapy.We successfully produced and isolated circRNA3503-loaded sEVs(circRNA3503-OE-sEVs)from SMSCs.Then,poly(D,L-lactide)-b-poly(ethylene glycol)-b-poly(D,L-lactide)(PDLLA-PEG-PDLLA,PLEL)triblock copolymer gels were used as carriers of sEVs.Through in vivo and in vitro experiments,PLEL@circRNA3503-OE-sEVs were shown to be a highly-effective therapeutic strategy to prevent OA progression.Through multiple pathways,circRNA3503-OE-sEVs alleviated inflammation-induced apoptosis and the imbalance between ECM synthesis and ECM degradation by acting as a sponge of hsa-miR-181c-3p and hsa-let-7b-3p.In addition,circRNA3503-OE-sEVs promoted chondrocyte renewal to alleviate the progressive loss of chondrocytes.Our results highlight the potential of PLEL@circRNA3503-OE-sEVs for preventing OA progression.Shi-Cong Tao Ji-Yan Huang Yuan Gao Zi-Xiang Li Zhan-Ying Wei Helen Dawes Shang-Chun Guo 2021Bioactive Materials2021,6,12:6

4Sedation-associated hiccups in adults undergoing gastrointestinal endoscopy and colonoscopy显示文摘AIM:To investigate whether the incidence of hiccups in patients undergoing esophagogastroduodenoscopy (EGD) or same-day bidirectional endoscopy (EGD and colonoscopy;BDE) with sedation is different from those without sedation in terms of quantity,duration and typical onset time.METHODS:Consecutive patients scheduled for elective EGD or same-day BDE at the gastrointestinal endoscopy unit or the health examination center were allocated to two groups:EGD without sedation (Group A) and BDE with sedation (Group B).The use of sedation was based on the patients' request.Anesthesiologists participated in this study by administrating sedative drugs as usual.A single experienced gastroenterologist performed both the EGD and the colonoscopic examinations for all the patients.The incidence,duration and onset time of hiccups were measured in both groups.In addition,the association between clinical variables and hiccups were analyzed.RESULTS:A total of 435 patients were enrolled in the study.The incidences of hiccups in the patients with and without sedation were significantly different (20.5% and 5.1%,respectively).The use of sedation for patients undergoing endoscopy was still significantly associated with an increased risk of hiccups (adjusted odds ratio:8.79,P < 0.001) after adjustment.The incidence of hiccups in males under sedation was high (67.4%).The sedated patients who received 2 mg midazolam developed hiccups more frequently compared to those receiving 1 mg midazolam (P = 0.0028).The patients with the diagnosis of gastroesophageal reflux disease (GERD) were prone to develop hiccups (P = 0.018).CONCLUSION:Male patients undergoing EGD or BDE with sedation are significantly more likely to suffer from hiccups compared to those without sedation.Midazolam was significantly associated with an increased risk of hiccups.Furthermore,patients with GERD are prone to develop hiccups.Chien Cheng Liu Cheng Yuan Lu Chih Fang Changchien Ping Hsin Liu Daw Shyong Perng 2012World Journal of Gastroenterology2012,18,27:4
5结核分枝杆菌H37Ra FadD3基因克隆与表达研究显示文摘为探索FadD3在结核分枝杆菌胆固醇分解代谢中的作用,从结核分枝杆菌H37Ra的全基因组中克隆了FadD3基因,并在大肠杆菌BL21中表达.根据NCBI公布的结核分枝杆菌全基因组序列设计一对引物,PCR扩增FadD3基因.PCR扩增产物与克隆载体pGEM3Zf(+)进行拼接,得到重组基因FadD3-pGEM3Zf(+),再转化到大肠杆菌DH5ɑ得到克隆.PCR检测阳性克隆,再与表达载体pYUB28b拼接,得到重组质粒FadD3-pYUB28b.阳性重组质粒亚克隆到大肠杆菌BL21宿主菌进行自体诱导表达.经过表型筛选及鉴定分析,已成功构建了重组表达质粒FadD3-pYUB28b.SDS-PAGE和Western blotting证实,重组基因FadD3-pYUB28b在大肠杆菌BL21中有表达产物.实验结果表明,以pYUB28b为表达载体,FadD3重组基因在大肠杆菌表达体系于温度分别为18,28℃有包涵体形式的表达蛋白,在37℃无表达产物.袁方 陈元晓 Stephanie Dawes Edward N.Baker 2014云南大学学报(自然科学版)2014,36,4:4
